Abstract
Изобретение относится к биотехнологии и может быть использовано для получения моноклональных антител и биологически активных веществ. Сущность изобретения: клетки-продуценты помещают в питательную среду и в ходе культивирования в среду дополнительно вводят комбинированную добавку, состоящую из глюкозы и одной или нескольких аминокислот, в частности глутамина. В качестве дополнительного компонента используют холин в концентрации 0,005-0,015 г/л среды. Добавочное питание клеток начинают во второй половине экспоненциальной фазы роста, продолжают в стационарной фазе, и когда рост клеток становится менее эффективным, отделяют целевой продукт. Общее время введения дополнительного питания составляет 30-100 ч. Выход моноклональных антител при использовании предложенного способа по сравнению с известным увеличивается в 3-7 раз. 4 з.п. ф-лы, 2 табл.

Изобретение относится к биотехнологии, в частности к получению моноклональных антител и биологически активных веществ.
Известен способ периодического культивирования клеток, продуцирующих белок или антитела в питательной среде, содержащей избыток аминокислот [1]. Однако при использовании данного способа отмечается недостаточно высокий выход целевого продукта.
Целью изобретения является увеличение выхода моноклональных антител и биологически активных веществ при культивировании клеток-продуцентов.
Способ осуществляют следующим образом.
Клетки-продуценты помещают в питательную среду и в ходе культивирования в среду вводят комбинированную добавку, состоящую из глюкозы и одной или нескольких аминокислот, в частности глутамина. В качестве дополнительного компонента используют холин. Добавочное питание клеток начинают во второй половине экспоненциальной фазы роста, продолжают в стационарной фазе, и когда рост клеток становится менее эффективным, отделяют целевой продукт.
П р и м е р 1. Клетки гибридомы NВI выращивают в воздушном подъемном ферментере емкостью 5 л в среде Игла, модифицированной Дульбекко (ДМЕМ) с добавлением 3% (по объему) сыворотки плода коровы (FCS). Исходная плотность популяции клеток приблизительно составляет 0,2 x 106 кл/мл. Во второй половине экспоненциальной фазы роста при достижении плотности клеток 1 x 106 в 1 мл в среду начинают последовательно вводить дополнительное питание. Состав питания и способ его введения в культуру приведены в табл. 1.
Результаты исследований показывают, что после введения добавки клетки продолжают расти до максимальной плотности популяции жизнеспособных клеток 2,2 x 106 кл/мл3. Затем после фазы роста наблюдается длительная стационарная фаза, в ходе которой происходят лишь относительно небольшие изменения плотности популяции жизнеспособных клеток и далее начинается спад роста клеточной культуры. Анализ культуральной среды показывает, что в ходе всего культивирования большая часть потребляемых питательных веществ находится в избытке. Выход целевого продукта осуществляют иммуносорбентным методом и в реакции агглютинации эритроцитов. Результаты исследований показывают, что антитела накапливаются в культуральной среде в ходе всего процесса культивирования клеток, однако удельная скорость синтеза антител увеличивается от значения 0,3 мкг на 106 клеток в 1 ч в ходе экспоненциальной фазы роста до 0,5-1,5 мкг на 106 клеток в 1ч на более поздних стадиях культивирования. Конечная концентрация антител составляет около 116 мг/л, что в 3-4 раза выше по сравнению с культурой, не получающей дополнительного питания. В табл. 2 приведены дополнительные результаты исследований, полученные при культивировании гибридных клеток известным и предложенным методами.
Данные, приведенные в табл. 2, показывают, что в случае линий клеток грызунов введение дополнительного питания обеспечивает 3-7-кратное повышение выхода антител.
При культивировании линии человеческих клеток периодическое питание приводит к улучшению выхода антител в 1,7 раз по сравнению с известным.
П р и м е р 2. Клетки гибридомы NBI выращивают в воздушном подъемном ферментере емкостью 5 л в среде ДМЕМ с добавлением FCS. Исходная плотность клеточной популяции составляет примерно 0,2x106 жизнеспособных клеток в 1 мл. Во второй половине экспоненциальной фазы роста при достижении плотности клеток 1 x 106 в 1 мл в среду однократно добавляют глутамин до конечной концентрации 4 мМ. Результаты исследований показывают, что культура более длительное время (по сравнению с использованием известного способа культивирования) сохраняет жизнеспособность, что приводит к увеличению общего титра антител.
П р и м е р 3. Клетки линии рРRI 1/10, продуцирующие активатор плазмогена тканевого типа (tРА), выращивают в ферментере с питательной средой, в который снизу вверх подают струю воздуха. Ферментер имеет рабочую емкость 5 л и снабжен устройствами для автоматического контроля упругости паров растворенного кислорода, рН и температуры. Значение рН контролируется путем регулируемого введения двуокиси углерода в газовую смесь или путем подачи в культуру щелочи. Температура регулируется путем подачи потока воды с контролируемой температурой в рубашку реактора. Клетки рНRI 1/10 выращивают в среде ДМЕМ с добавлением альбумина, инсулина, трансферрина, этаноламина, холина витаминов, микроэлементов и защитного полимера. В ходе культивирования во второй половине экспоненциальной фазы роста в культуру вводят дополнительное питание следующим образом:
однократно добавляют глутамин до конечной концентрации 2 мМ;
однократно добавляют "нерастворимые" аминокислоты триптофан, тирозин и цистеин в количестве 0,01-0,07 г/л среды;
с помощью насоса в виде концентрированного раствора в фосфатно-солевом буфере подают комбинированную смесь питательных веществ, состоящую из глюкозы (4-5 г/л), холина (0,005-0,015 г/л) и "растворимых" аминокислот (0,01-0,15 г/л). Подачу дополнительного питания осуществляют в течение 50 ч. Максимальная плотность общей популяции клеток составляет 4 x106 кл в 1 мл. Синтез tРА происходит в ходе всего процесса культивирования, максимальная концентрация продукта составляет 42 мкг/мл.

Таким образом, предложенный способ может быть эффективно использован для культивирования клеток-продуцентов моноклональных антител и биологически активных веществ с хорошим выходом целевого продукта.
Claims (5)
1. СПОСОБ ПЕРИОДИЧЕСКОГО КУЛЬТИВИРОВАНИЯ КЛЕТОК-ПРОДУЦЕНТОВ МОНОКЛОНАЛЬНЫХ АНТИТЕЛ И БИОЛОГИЧЕСКИ АКТИВНЫХ ВЕЩЕСТВ, предусматривающий внесение клеток в питательную среду и их инкубирование в данной среде с последующим отделением целевого продукта, отличающийся тем, что во второй половине экспоненциальной фазы роста среду в течение общего периода времени 30 - 100 ч дополняют комбинацией глюкозы и одной или нескольких аминокислот, культивирование продолжают до фазы спада роста клеток и отделяют целевой продукт.
2. Способ по п.1, отличающийся тем, что общее количество глюкозы составляет 1 - 10 г/л среды.
3. Способ по п.1, отличающийся тем, что питательная среда дополнительно содержит холин в общем количестве 0,005 - 0,015 г/л среды.
4. Способ по п.1, отличающийся тем, что аминокислоты добавляют однократно, а общее количество каждой аминокислоты составляет 0,01 - 0,15 г/л среды.
5. Способ по п.1, отличающийся тем, что в качестве аминокислоты используют глутамин, который добавляют в количестве 0,5 - 3 г/л среды.
