米国特許法(35 U.S.C.)第119(e)条に準拠し、本出願は、2001年6月21日に出願された米国特許仮出願第60/341,723号の出願日に対する優先権を主張するものであり;その開示は本明細書に参照として組入れられている。

発明の分野
本発明の分野は、蛍光性タンパク質である。
標識は、関心対象のタンパク質、細胞、または生物に印をつけるための道具であり、多くの生化学、分子生物学、および医学的診断適用において顕著な役割を果たしている。放射性標識、色素標識、蛍光標識、化学発光標識等を含む多様な異なる標識が開発されている。しかしながら、新規の標識の開発への関心は依然としてある。特に関心があるのは、色素タンパク質および/または蛍光性タンパク質標識を含む、新規のタンパク質標識の開発である。
急速に成熟する蛍光タンパク質をコードしている核酸組成物に加え、それらの非凝集型(およびそれらの変異体)ならびにこれによりコードされたタンパク質が提供される。関心対象のタンパク質は、蛍光を発するタンパク質であり、ここでこの特徴は、このタンパク質の2残基以上の相互作用から生じる。更に本発明のタンパク質は、ある態様において、花虫類(Anthozoan)のような非生物発光刺胞動物(Cnidarian)、もしくは花虫類非ウミエラ(Pennatulacean)(シーペン(sea pen))種のいずれかから得られた野生型タンパク質中に認められるかまたはそれらの変異体であるという点でさらに特徴付けられる。ある態様において、この本発明のタンパク質は、「DsRed」として市販されている野生型イソギンチャクモドキ(Discosoma)種「赤色」蛍光タンパク質の変異体である。先の特異的タンパク質に実質的に類似した、またはその変異体であるタンパク質も関心がある。同じくこれらの核酸の断片およびそれらによりコードされたペプチドに加え、本発明のタンパク質に対する抗体ならびにトランスジェニック細胞および生物も提供される。本発明のタンパク質および核酸の組成物は、様々な異なる適用における使用が認められる。最後に、例えば本発明の核酸組成物を含む、そのような適用において使用するためのキットが提供される。

核酸組成物
先にまとめられたように、本発明は、急速に成熟する色素/蛍光タンパク質およびそれらの変異体、更にはこれらのタンパク質の断片および相同体をコードしている核酸組成物を提供する。急速に成熟する色素/蛍光タンパク質は、発色するおよび/または蛍光を発するタンパク質を意味し、例えばこれは励起波長の光の照射時に、低、中または高蛍光を発することができる。更に、このタンパク質は、急速に成熟するので、その最終的色素/蛍光特性に、約72時間未満で、時には48時間未満で、および時には24時間未満で到達する。ある態様において、このタンパク質は、20時間未満、例えば18時間、16時間、14時間、12時間、10時間、8時間などの期間内に、成熟することができる。

本発明により、急速に成熟する色素および/または蛍光タンパク質ならびにそれらの変異体に加え、それらに関連したポリペプチド組成物も提供される。本発明のタンパク質は色素タンパク質であるので、これらは、発色したタンパク質であり、これは蛍光、低蛍光または非蛍光であってもよい。本明細書に使用される色素タンパク質および蛍光タンパク質という用語は、レニラルシフェラーゼのような、ルシフェラーゼを含まず、および例えば白色光もしくは特定波長光(または励起波長のような波長の狭いバンド)の光が照射された場合に、着色または発色および/または発蛍光されるいずれかのタンパク質を意味する。本明細書において使用される用語ポリペプチド組成物は、完全長タンパク質に加え、それらの部分または断片の両方を意味する。同じくこの用語は、天然に存在するタンパク質の変種も含み、ここでこのような変種は、以下により詳細に説明されるように、天然に存在するタンパク質に相同または実質的に類似しており、ならびに天然に存在するタンパク質の変異体である。本発明のポリペプチドは、それらの天然に存在する環境以外に存在する。
本発明の蛍光タンパク質に特異的に結合する抗体も提供される。適当な抗体は、本発明のタンパク質の全てまたは一部を含むペプチドによる宿主動物の免疫処置により得られる。適当な宿主動物は、マウス、ラット、ヒツジ、ヤギ、ハムスター、ウサギなどを含む。タンパク質免疫原の起源は、一般に刺胞動物種、特に生物発光しない刺胞動物種、例えば花虫類またはウミエラでない花虫類であろう。宿主動物は、一般に例えばマウスなどのその免疫原以外の様々な種であろう。
本発明の核酸は、トランスジェニック非ヒト植物もしくは動物、または細胞株における部位特異的遺伝子改変を生じるために使用され得る。本発明のトランスジェニック細胞は、導入遺伝子として存在する本発明による核酸を1個または複数含んでいる(この定義には、導入遺伝子を含むよう形質転換された親細胞およびそれらの子孫が含まれる)。多くの態様において、トランスジェニック細胞は、本発明に係る核酸を通常は保有または含有していない細胞である。トランスジェニック細胞が本発明の核酸を天然に含有している態様においては、その核酸は、細胞内の天然の位置ではない位置に存在すると考えられ、即ち細胞のゲノム物質内に非天然の位置で組み込まれているであろう。内因性遺伝子座を改変したトランスジェニック動物は、相同的組み換えを介して作製され得る。または、核酸構築物は、ゲノム内へ無作為に組み込まれる。安定的な組み込みのためのベクターには、プラスミド、レトロウイルスおよびその他の動物ウイルス、YAC等が含まれる。
本発明の色素タンパク質およびその蛍光性変異体は、多様な異なる適用において有益であるが、適用は、タンパク質がが色素タンパク質であるかまたは蛍光性タンパク質であるかに依って必然的に異なる。これらの各タンパク質型の代表的な用途を以下に記載するが、以下に記載された用途は、単なる代表的なものであり、本発明のタンパク質の使用を下記のものに制限するものでは決してない。
本発明の色素タンパク質は、多様な異なる適用において有益である。一つの関心対象の適用は、問題としている特定の組成物に色または色素を付与することができる着色料としての本発明のタンパク質の使用である。ある種の態様において特に関心対象であるのは、無毒の色素タンパク質である。本発明の色素タンパク質は、問題とする多様な異なる組成物に取り込まれ得る(問題とする組成物の代表には、食物組成物、医薬品、化粧品、生物、例えば動物および植物等が含まれる)。着色料または色素として使用される場合、所望の色または色素を付与するために十分な量の色素タンパク質が、問題とする組成物に取り込まれる。色素タンパク質は、任意の便利なプロトコルを使用して、問題とする組成物に取り込まれてもよく、利用される特定のプロトコルは、少なくとも部分的には、着色すべき問題とする組成物の性質に必然的に依存すると考えられる。利用され得るプロトコルには、混和、拡散、摩擦、噴霧、注入、および入れ墨等が含まれるが、これらに制限はされない。

赤色蛍光タンパク質DsRedは、緑色蛍光タンパク質(GFP)との二色実験にとって理想的なスペクトル特性を有する。しかし野生型DsRedは、いくつかの欠点を有し、これには発色団の成熟が遅いことおよび溶解度が低いことが含まれる。成熟が遅いことを克服するために、本発明者らは、ランダムおよび位置指定突然変異誘発を使用し、野生型タンパク質よりも10〜15倍早く成熟するDsRed変種を作成した。42位のアスパラギンのグルタミンへの置換により、DsRedの成熟は大きく促進するが、緑色発光レベルも増加する。追加のアミノ酸置換により、この緑色発光は抑制されるが、他方で成熟は更に促進する。DsRedの溶解度を高めるために、このタンパク質のN末端近傍の実効電荷を低下させた。得られたDsRed変種は、酵母のような急激に増殖する生物においても、明蛍光をもたらした。
A. 突然変異誘発およびスクリーニング
突然変異誘発に関して、pDsRed1-N1ベクター(Clontech社、パロアルト、CA)中に存在する野生型または変異型DsRed遺伝子は、NheIおよびHpaIで切出し、およびエラープローンPCR(Cadwell, R.C.およびJoyce, G.F.、In PCR Primer.「A laboratory manual.」 (Dieffenbach, C.W.およびDveksler, G.S.編)583-589頁(Cold Spring Harbor Laboratory Press社、コールドスプリングハーバー、NY;1995年))における鋳型として使用した。増幅された生成物は、BamHIおよびBsaBIで切断し、ゲル精製し、およびN末端ヘキサヒスチジンタグをコードしているpQE31発現ベクター(Qiagen社、バレンシア、CA)のBamHI部位とEcl136II部位との間にライゲーションした。成熟したDsRed遺伝子のライブラリーは、大腸菌株DH10Bへ形質転換した。突然変異誘発の1〜3回目のために、50,000〜100,000個のコロニーを、本文中に説明されたスライドプロジェクターアッセイ法を用い、明蛍光についてスクリーニングした。4回目に関して、4,000個の明蛍光コロニーを、96ウェルプレートのウェルに採取し、その後飽和まで増殖させ、B-PER II試薬(Pierce社、ロックフォード、IL)で溶解し、2,500gで5分間遠心した。上清を、96ウェルプレートの第二セットに移し、ペレットおよび上清からの蛍光シグナルを、スライドプロジェクターアッセイ法を用い、肉眼により比較した。可溶性蛍光の不溶性蛍光に対する比に上昇がみられるクローンを更に分析した。5回目に関して、10,000個の変異体各クローンの10プールを、形質転換プレートから回収し、ベクトンディッキンソン(Becton Dickinson)FACStarPlusフローサイトメーターを用いる細胞選別に供した。蛍光シグナルは、緑色(FL-1)および赤色(FL-2)チャネルにおいて同時に測定し、およびこれらが強力な赤色蛍光を発するが緑色蛍光の低下を示した場合に、細胞を収集した。
ヘキサヒスチジンタグを付けたタンパク質を精製するために、pQE31ベクター中の蛍光タンパク質遺伝子を、pREP4リプレッサープラスミド(Qiagen社)を保持する大腸菌細胞に形質転換した。培養液250mlを、OD600が0.5となるまで増殖し、その後1mMイソプロピル-β-D-チオガラクトシド(IPTG)により6〜8時間37℃で誘導した。これらの細胞を、10mlのB-PER IIで溶解し、27,000gで20分間遠心した。NaClを300mMまで添加し、2,500gで10分間遠心することにより、界面活性剤を上清から除去した。Ni2+-NTA-アガロースビーズ(Qiagen社)1mlを添加後、このチューブを上下反転させて(end-over-end)1時間混合した。ビーズを、300mM NaCl、20mMイミダゾール-HCl(pH7.4)、0.5% Triton X-100の10mlで3回洗浄し、その後Triton X-100を含まない同じ緩衝液で3回洗浄した。この蛍光タンパク質は、300mMイミダゾール-HCl(pH7.4)2.5mlで溶出し、50mM Na+-HEPES(pH7.5)、100mM NaCl、1mM EDTAに透析した。
DsRed変種をコードしている遺伝子を、pQE81発現ベクター(Qiagen社)にクローニングし、および大腸菌へ形質転換した。37℃で曝気しながら増殖している細菌培養物は、1mM IPTGで30分間誘導し、各DsRed変種について発現のパルスを作成した。その後170μg/mlクロラムフェニコール、30μg/mlカナマイシン、および50μg/mlテトラサイクリンの混合物でタンパク質合成を阻害することにより、追跡を開始した。指定された時点で、これらの培養物のアリコートを除去し、15%グリセロールに調節し、-80℃で凍結した。これらのアリコートを後に、急速解凍し、およびベクトンディッキンソンFACScanフローサイトメーターを用いて評価し、細胞1個当りの赤色蛍光(チャネルFL-2)の平均強度を決定した。各アリコートの一部は、トリクロロ酢酸で沈殿させ、その後SDS-PAGEおよび抗ヘキサヒスチジンモノクローナル抗体(Qiagen社)による免疫ブロッティングを施し、培養物中のDsRedポリペプチドの総量を測定した。酵母の蛍光顕微鏡観察。ADH1プロモーターの制御下、pCox4-DsRed1融合遺伝子を保持するpRS315(Sikorski、Genetics、122:19-27(1989))由来のCENプラスミドを使用した。このプラスミドの誘導体は、DsRed1コード配列を、DsRed.T3またはDsRed.T4コード配列で置換えることにより作成した。これらのプラスミドは、染色体SEC7-EGFPx3遺伝子20を保持するサッカロマイセス・セレビシエ株BGY101に導入した。得られた酵母株からの細胞を、最小グルコース培地において増殖し、固定し、および投射による蛍光像を、Rossaneseらの論文(J. Cell Biol.、145:69-81(1999))に説明されたように得た。
最近蛍光タンパク質のファミリーが説明されている。これらの新たに発見されたタンパク質の最も有用なものは、DsRedであり、これはサンゴのイソギンチャクモドキ(Discosoma)に由来している。DsRedは、最大発光が583nmであるオレンジ色〜赤色の蛍光を有する。生物物理試験およびX線結晶解析試験は、DsRedが安定した四量体を形成すること、および各単量体は構造的にGFPに非常に類似していることを明らかにした。GFPに対し赤色にシフトしたDsRedの蛍光は、より広範に共役するπシステムをもつ発色団から生じた。DsRed蛍光は、最適に558nmで励起するが、しかし標準の488nmレーザーで励起することもでき、DsRedをレーザーベースの共焦点顕微鏡およびフローサイトメーターで使用することができる。DsRedは、高い量子収量を有し、光学的に安定している。これらの特徴により、DsRedは、蛍光造影、特にGFPおよびその変種が関与する多色実験において理想的な候補となる。DsRedのコドンを最適化した型は、現在DsRed1の名称で入手可能である。

b.輝度は、吸光係数および量子収量の積により決定した。相対輝度は、DsRed1の輝度を1.00と定義することにより計算した。
c.成熟の半減期は、図2の実験プロトコールを用い、グラフにより推定した。列記した値は、2回の個別の実験の平均である;各DsRed変種について、2回の実験で得られた数値は互いに15%以内であった。
本項は、最適化されたDsRed変種を得るために使用された、多工程突然変異誘発戦略を説明する。概要は、補遺表2に提供される。
a.開始コドンに対し位置-4のプロリンコドンは、pDsRed1-N1ベクターにおける複数のクローニング部位によりもたらされる。
b.R2A置換は、DsRed1中の開始コドンの後に存在する余分なバリンコドンも除去する。
c.T217Aは、DsRed.T1およびDsRed.T4に存在するが、DsRed.T3には存在しない。
d.DsRed.T1は、C117Sを含むが、A145Pは含まないのに対し、DsRed.T3およびDsRed.T4はA145Pを含むが、C117Sは含まない。
Claims (11)
- 野生型DsRedの色素変異体または蛍光変異体をコードしているポリヌクレオチドであって、前記野生型DsRedの色素変異体または蛍光変異体が配列番号:2に対して、N42Q、H41T、V44A、T21S、R2A、K5E、N6D、T217AおよびC117Sのアミノ酸置換、またはN42Q、H41T、V44A、T21S、R2A、K5E、N6DおよびA145Pのアミノ酸置換を含み、
前記変異体が野生型DsRedに比べて急速に成熟する、ポリヌクレオチド。 - 変異体が変異N42Q、H41T、V44A、T21S、R2A、K5E、N6D、T217AおよびC117Sを含む、請求項1に記載のポリヌクレオチド。
- 変異体が変異N42Q、H41T、V44A、T21S、R2A、K5E、N6DおよびA145Pを含む、請求項1に記載のポリヌクレオチド。
- 作動可能なようにプロモーターに連結した、請求項1〜3のいずれかに記載のポリヌクレオチドを含む、ポリヌクレオチド。
- 請求項1〜4のいずれかに記載のポリヌクレオチドを含む細胞。
- 請求項5に記載の細胞を培養する工程を含む、野生型DsRedの色素変異体または蛍光変異体の製造方法であって、ここで請求項1〜4のいずれかに記載のポリヌクレオチドが色素変異体または蛍光変異体の製造のために発現される方法。
- 請求項1〜4のいずれかに記載のポリヌクレオチドを含む、非ヒトトランスジェニック生物。
- 請求項1〜4のいずれかに記載のポリヌクレオチドによってコードされるポリペプチド。
- 請求項1〜4のいずれかに記載のポリヌクレオチドを含むキット。
- 色素タンパク質または蛍光タンパク質を使用する用途における、請求項8に記載のポリペプチドの使用。
- 色素ポリペプチドまたは蛍光ポリペプチドをコードする核酸を使用する用途における、請求項1〜4のいずれかに記載のポリヌクレオチドの使用。
