Claims (1)
- 【特許請求の範囲】 1. 式 Pr(−X−S−S−W)m 「式中: Prは蛋白質性キャリアーであり、 Xは、蛋白質性キャリアーと反応することができるあらゆる反応性の基の生成 物を含んでなるリンカーであり、 Wは、天然に存在するメチルトリスルフィド基の除去により形成されるカリキ アミシン基であり;そして mは0.5から15の数である] を有する、より高い薬剤配合量/収率及び減少された凝集を伴う、モノマー性カ リキアミシン誘導体/キャリアー複合体を調製するための方法であって、 (1) 約4.0から8.5の範囲内のpHをもつ、非求核性の、蛋白と相容性 の緩衝溶液中のカリキアミシン誘導体及び蛋白質性キャリアーをインキュベーシ ョンさせること、 ただし、前記溶液は更に、 (a) プロピレングリコール、エタノール、DMSO、及びそれらの組み合わ せ物からなる群から選ばれる1種類の補助溶媒、並びに (b) 少なくとも1種類のC6−C18カルボン酸を含んでなる添加剤を含んで なり、ここで、 前記インキュベーションは、約25℃から約37℃の範囲の温度で、約15分 間から約24時間の範囲の期間で実施される;並びに (2) 段階(1)で生成された複合体を精製してモノマー性複合体を 生成すること、 の段階を含んでなる方法。 2. Xが式 Z−Sp [式中: Spは直鎖もしくは分枝鎖状の2価もしくは3価(C1−C18)基、2価もし くは3価アリールもしくはヘテロアリール基、2価もしくは3価(C3−C18) シクロアルキルもしくはヘテロシクロアルキル基、2価もしくは3価アリール− もしくはヘテロアリール−アリール(C1−C18)基、2価もしくは3価シクロ アルキル−もしくはヘテロシクロアルキル−アルキル(C1-C18)基、又は2価 もしくは3価(C2−C18)不飽和アルキル基であり、そこで、 ヘテロアリールは好ましくはフリル、チエニル、N−メチルピロリル、ピリジ ニル、N−メチルイミダゾリル、オキサゾリル、ピリミジニル、キノリル、イソ キノリル、N−メチルカルバゾイル、アミノクマリニル、又はフェナジニルであ り、そしてそこで、 Spが3価の基である場合は、Spは更に、低級(C1−C5)ジアルキルアミ ノ、低級(C1−C5)アルコキシ、ヒドロキシ、又は低級(C1−C5)アルキル チオ基で置換されてもよく;そして Zは−NHC(=O)−、−CH=NNHC(=O)−、−CH2NHNHC (=O)−、−CH=NNHC(=O)NH−、−CH2NHNHC(=O)N H−、−CH=NNHC(=S)NH−、−CH2NHNHC(=S)NH−、 −CH=N−、−CH2NH−、−OC(=O)−、−SS−、 である] を有する、請求の範囲第1項の方法。 3. Xが式 (CO−Alk1−Sp1−Ar−Sp2−Alk2−C(Z1)=Q−Sp) [式中、 Alk1及びAlk2は独立して、1個の結合又は分枝もしくは非分枝(C1− C10)アルキレン鎖であり; Sp1は、1個の結合、−S−、−O−、−CONH−、−NHCO−、−N R’−、−N(CH2CH2)2N−、又は−X−Ar’−Y−(CH2)n−Zで あり、 (ここで、 X、Y、及びZは独立して、1個の結合、−NR’−、−S−、又は−O−で ある、ただし、 n=0の場合は、Y及びZの少なくとも1個は、1個の結合でなければならず 、そして Ar’は、場合によっては(C1−C5)アルキル、(C1−C4)アルコキシ、 (C1−C4)チオアルコキシ、ハロゲン、ニトロ、−COOR’、 −CONHR’、−O(CH2)nCOOR’、−S(CH2)nCOOR’、−O (CH2)nCONHR’、又は−S(CH2)nCONHR’のうちの1、2、又 は3個の基で置換されている、1,2−、1,3−、又は1,4−フェニレンで あり、 ただし、Alk1が1個の結合である場合は、Sp1は1個の結合であり; nは0から5の整数であり; R’は、場合によっては、−OH、(C1−C4)アルコキシ、(C1−C4)チ オアルコキシ、ハロゲン、ニトロ、(C1−C3)ジアルキルアミノ、又は(C1 −C3)トリアルキルアンモニウム−A-(ここで、A-は、塩を完成させている 、製薬学的に許容できる陰イオンである)のうちの1又は2個の基により置換さ れている、分枝もしくは非分枝(C1−C5)鎖である); Arは、場合によっては、(C1−C6)アルキル、(C1−C5)アルコキシ、 (C1−C4)チオアルコキシ、ハロゲン、ニトロ、−COOR’、−CONHR ’、−O(CH2)nCOOR’、−S(CH2)nCOOR’、−O(CH2)nC ONHR’、又は−S(CH2)nCONHR’(ここで、n及びR’は、前記に 定義されたものである)のうちの1、2、又は3個の基で置換されている、1, 2−、1,3−、又は1,4−フェニレン、あるいは1,2−、1,3−、1, 4−、1,5−、1,6−、1,7−、1,8−、2.3−、2,6−、又は2 ,7−ナフチリデン、あるいは 場合によっては、(C1−C6)アルキル、(C1−C5)アルコキシ、(C1−C4 )チオアルコキシ、ハロゲン、ニトロ、−COOR’、−CONHR’、−O( CH2)nCOOR’、−S(CH2)nCOOR’、又は−S(CH2)nCONH R’(ここで、n及びR’は前記に定義のものである)のうちの1、2、3、又 は4種の基で置換されている、各ナフチリデン又はフェノチアジアンであり、た だし、 Arがナフチリデンである場合は、Z1は水素でなくそして、 Arがフェノチアジンである場合は、Sp1は窒素にのみ結合している1個の 結合である; Sp2は1個の結合、−S−又は−O−であり、ただし、 Alk2が1個の結合である場合は、Sp2は1個の結合である; Z1は、H、(C1−C5)アルキル、あるいは、場合によっては、(C1−C5 )アルキル、(C1−C5)アルコキシ、(C1−C5)チオアルコキシ、ハロゲン 、ニトロ、−COOR’、−CONHR’、−O(CH2)nCOOR’、−S( CH2)nC0OR’、−O(CH2)nCONHR’、又は−S(CH2)nCON HR’(ここで、n及びR’は前記のようなものである)のうちの1、2、又は 3個の基で置換されているフェニルであり; Spは、直鎖もしくは分枝鎖の2価もしくは3価(C1−C18)基、2価もし くは3価アリールもしくはヘテロアリール基、2価もしくは3価(C3−C18) シクロアルキルもしくはヘテロシクロアルキル基、2価もしくは3価のアリール −もしくはヘテロアリール−アリール(C1−C18)基、2価もしくは3価のシ クロアルキル−もしくはヘテロシクロアルキル−アルキル(C1−C18)基、又 は2価もしくは3価(C2−C18)不飽 和アルキル基であり、ここで、 ヘテロアリールは好ましくは、フリル、チエニル、N−メチルピロリル、ピリ ジニル、N−メチルイミダゾリル、オキサゾリル、ピリミジニル、キノリル、イ ソキノリル、N−メチルカルバゾイル、アミノクマリニル、又はフェナジニルで あり、そしてそこで、 Spが3価の基である場合は、Spは更に、低級(C1−C5)ジアルキルアミ ノ、低級(C1−C5)アルコキシ、ヒドロキシ、又は低級(C1−C5)アルキル チオ基で置換されてもよい;そして、 Qは=NHNCO−、=NHNCS−、=NHNCONH−、=NHNCSN H−、又は=NHO−である] を有する、請求の範囲第1項の方法。 4. Sp1が1個の結合、−S−、−O−、−CONH−、−NHCO−、又 は−NR’−(ここで、n及びR’は請求の範囲第3項に定義されたものである )であり、ただし、Alk1が1個の結合である場合は、Sp1が1個の結合であ る; Arが、場合によっては、(C1−C6)アルキル、(C1−C5)アルコキシ、 (C1−C4)チオアルコキシ、ハロゲン、ニトロ、−COOR’、−CONHR ’、−O(CH2)nCOOR’、−S(CH2)nCOOR’、−O(CH2)nC ONHR’、又は−S(CH2)nCONHR’(ここで、n及びR’は請求の範 囲第3項に定義されているものである)のうちの1、2、又は3個の基で置換さ れている、1,2−、1,3−、又は1,4−フェニレンであるか、あるいは Arが、それぞれ場合によっては、(C1−C6)アルキル、(C1−C5)アル コキシ、(C1−C4)チオアルコキシ、ハロゲン、ニトロ、−CO OR’、−CONHR’、−O(CH2)nCOOR’、−S(CH2)nCOOR ’、−O(CH2)nCONHR’、又は−S(CH2)nCONHR’のうちの1 、2、3、又は4個の基で置換されている、1,2−、1,3−、1,4−、1 ,5−、1,6−、1,7−、1,8−、2,3−、2,6−、又は2,7−ナ フチリデンであり; Alk2が分枝もしくは非分枝(C1−C10)アルキレン鎖であり; Z1が(C1−C5)アルキル、あるいは、場合によっては、(C1−C5)アル キル、(C1−C4)アルコキシ、(C1−C4)チオアルコキシ、ハロゲン、ニト ロ、−COOR’、−CONHR’、−O(CH2)nCOOR’、−S(CH2 )nCOOR’、−O(CH2)nCONHR’、又は−S(CH2)nCONHR ’のうちの1、2、又は3個の基で置換されているフェニルであり;そして Alk2及びSp2がともに1個の結合である、 請求の範囲第3項の方法。 5. Sp1が−O−であり、Alk1がC3アルキレンであり、Arが1,4− フェニレンであり、そしてZ1がC1アルキルである、請求の範囲第4項の方法。 6. Qが=NHNCO−であり、そしてSpが−CH2C(CH3)2−である 、請求の範囲第3項の方法。 7. Zが−NHC(=O)−であり、そしてSpが−CH2CH2C(CH3)2 −である、請求の範囲第2項の方法。 8.
