【発明の詳細な説明】
本発明は、ヒトサイトメガロウィルス主要前初期遺伝子からのDNA配列を含有
する発現ベクター、この々フタ−を含有する宿主細胞、上記配列を含有するベク
ターを用いて所望のポリペプチド′t−製造する方法、および上記DNA配列の
使用に関する。
発明の背景
組換えDNA技術の分野における研究者の主たる目的は、特定のポリペプチドの
可能な限り高レベルの製造を達成することである。これは、天然にはあまり豊富
でないポリペゾチドt−製造するために組換えDNA技術一般的にDNA技術の
応用は、所望のポリペプチドをコーrする遺伝子のクローニング、クローン化さ
れた遺伝子の適当な発現ベクター中への配置、このベクターによる宿主細胞糸の
トランスフェクション、およびトランスフェクションを受けた細胞系の培養によ
るポリペプチドの製造が包含される。ある特定のベクターもしくは細胞系または
それらの組合せが有用なレベルの産生を導くかどうかを予測することはほとんど
不可能である。
一般に、トランスフェクションを受けた細胞系によって産生されるポリペプチド
の量に有意に影響する因子は、1)遺伝子コピー数、2)遺伝子の転写およびm
RNAの翻訳の切出、3) mRNAの安定性、ならびに4)タンパク質の分泌
切土である。
組換えポリペプチドの発現レベルの増大を目指す大部分の研究は、転写開始&!
構の改良に集中されてきた。
その結果、切工的な翻訳に影響する因子の理解および触明ははるかに遅れてbて
、ある特定のDNA配列が効!的な翻訳を達成するのに有用であるかを予測する
ことは不可能である。
翻訳を研究する試みは、大部分、コンセンサス翻訳開始シグナルの周囲のDNA
配列が翻訳開始にどのような影qiIIt−もつかを決定するために、これを変
化させるものである〔コデク(KOZaJ M、 ) :セル、第41巻、第2
86〜292頁、1986年〕。
所望の異種遺伝子の発現が関与する研究では通常、異種遺伝子の暗号配列と少な
くとも一部の5′非翻訳配列の両者を、翻訳開始が遺伝子の天然配列に由来する

は産生ずる生成物の量に有害な効果を与える。

上流領域からの配列が使用されていた。しかしながら、これらは、もっばら、プ
ロモーターおよび/またはエンノ〜ンサーとしての配列の使用に関するものであ
った。スペーテ(5paete )およびモカルスキ−(ジャーナル俸オプ令ヴ
アイロロジー、巣56巻、第1号、第135〜143頁、1985年)は、異極
遺伝子の発現のためのプロモーターとして、プロモーター、エンハンサ−および
一部の5′−非翻訳領域を包含するhCMV −MIE遺伝子のPsti〜Ps
t1フラグメントを使用している。翻訳を達成するために、異種遺伝子の天然の
5′−非翻訳領域が使用された。
公告されたヨーロッパ特許出願第260148号には、異種タンパク質の連続的
製造方法が記載されている。構築された発現ベクターは、安定化配列として、h
CMV −M工E遺伝子の5′−非翻訳領域の一部を含有する。
安定化配列は所望の異種タンパク質をコードする遺伝子の5′−非翻訳領域中に
置かれる。すなわち、この場合も、遺伝子の天然5′−非翻訳領域が翻訳に必須
であ第一の特徴として本発明はヒトサイトメガロウィルスの主要前初期遺伝子の
プロモーター、エン−・ンサー、および最初のイントロンを含めて実質的に完全
な5′−非翻訳領域からなるDNA配列を含有するベクター全提供する。
本発明の第一の特徴の好ましい態様においては、ベクターは異種遺伝子の挿入の
ための制限部位を含有する。
本発明は、ヒトサイトメガロウィルスの主要前初期遺伝子(hcMv −M工E
)のプロモーター、エンハンサ−および完全5′−非翻訳領域からなるDIJA
配列?異種遺伝子の上流に含有するベクターが異種遺伝子生成物の高レベルの発
現を生じることの発見に基づくものである。とくに、hCMV−MIEから誘導
されたDNA ’i異種遺伝子の暗号配列に直接連結すると高レベルのmRNA
翻訳が達成することの発見は全く予期し得ないものであった。このmRNAの効
率的な翻訳は一貫して達成され、発現される特定の異種遺伝子に依存するもので
はなりようである。
本発明は、第°二の特徴として、ヒトサイトメガロウィルスの主要前初期遺伝子
のプロモーター、エンハンサ−1および最初のイントロンを含めて実質的に完全
な5′−非翻訳領域からなるDNA配列を異種遺伝子の上流に含有するベクター
を提供する。
本発明の第二の特徴によるhCMV −M工E誘導DNAは、たとえば異種遺伝
子の5′−非翻訳領域のような介在配列によって異種遺伝子の暗号配列から分離
されていてもよ−。hCMV −MIE誘導DNAは異種遺伝子の暗号配列に直
接連結するのが有利である。
本発明の第二の特徴の好ましい態様においては、本発明は、hCMV−M工E遺
伝子のプロモーター、エンハンサ−1および最初のイントロンを含めて実質的に
完全な5′−非翻訳領域に、異種遺伝子のDNA暗号配列が直接連絡してなるD
NA配列を含有するベクターを提供する。
hCMV−MIE誘導配列は、天然のhCMV −MIE m訳開始シグナルと
同一の配列を包含することが好ましい。しかしながら、所望のポリペプチドの暗
号配列への連結を容易にするため、すなわち便利な制限酵素認識部位を導入する
ことにより、天然の翻訳開始シグナルを修飾することが必要であるかまたは有利
である。たとえば、翻訳開始部位はNc酊認識部位を与えるように修飾するのが
有利である。
異種遺伝子は、真核生物ポリペプチド、たとえば、酵素たとえばキモシンもしく
は冑リパーゼ;酵素インヒビターたとえばメタロプロテナーゼの組織インヒビタ
ー (TIMP ) ; ホにモンたとえば成長ホルモン;リンホカインたとえ
ばインターフェロン;プラスミノーデンアクテイベーターたとえば組織プラスミ
ノーデンアクテイベーター(tPA )もしくはプロウロキナーゼのような哺乳
動物ポリペプチド;または抗体−酵素もしくは抗体−トキシンキメ2のような二
元的活性を有するキメラ免疫グロブリンを包含する天然、修飾もしくはキメラ免
疫グロブリンもしくはその7ラグメントをコードする遺伝子とすることができる
。
本発明の第三の特徴によれば、本発明は、本発明の第一または第二の特徴による
ベクターでトランスフェクションされた宿主細胞を提供する。
宿主細胞は任意の真核生物細胞たとえば植物または昆虫細胞であってもよいが、
哺乳動物細胞、たとえばCHO細胞もしくは骨髄起源の細胞たとえば骨髄腫細胞
またはハイプリドーマ細胞であることが好ましい。
本発明は、その第四の特徴として、本発明の第三の特徴によるトランスフェクシ
ョンを受けた細胞の培養によって異種ポリペプチドを製造する方法を提供する。
本発明は、その第五の特徴として、hCMV−MIE遺伝子のプロモーター、エ
ンノ・ンサー、および最初のイントロンを含めた実質的に完全な5′−非翻訳領
域からなるDNA配列の、異種遺伝子の発現のための使用を提供する。
本発明の第五の特徴の好ましい態様においては、hCMV −M工E誘4 DN
A配列は、異種遺伝子のDNA暗号配列に直接連結される。
本発明の範囲には、プラスミドpcMGs XpHT、 1およびpEE6hc
MVも包含される。

hCMVのPst−1mフラグメント(ボシャルトら;セル、第41巻、第52
1〜530頁、1985年;スペーテおよびモカルスキ一二ジャーナル・オプ・
ヴアイロロジー、第56巻、第1号、第165〜146頁、1985年)+!n
工E遺伝子のプロモーター−エンハンサ−と、5′非翻訳リーダーの大部分を最
初のイントロンを含めて含有する。5′非翻訳配列の残りの部分は、約20塩基
対の小さな付加配列を結合させることで再生できる。
多くの真核生物遺伝子は、翻訳開始部位とl復するNco1制限部位(5’−C
CATGG −3′)を有し、この配列が好ましい翻訳開始シグナル5’ AC
CATGPu3’の部分を形成することが多い。h、cMV −M工E遺伝子は
タンパク質暗号配列の最初にNcoi部位をもたないが、単一の塩基対変化で、
「コデク」コンセンサス開シグナルへのより一層の類似と、NCo1認識部位の
導入の両者が、この配列に生じる。したがって、配列
01対の相補性オリゴヌクレオチドを合成した。これは、hCMvのPst−1
mフラグメントに融合させると、翻訳開始コrンに対して位置−10GがCに1
個変化したMIE遺伝子の完全な5′非翻訳配列が再生される。
この合成りNAフラグメントをhCMV Pst−1mプロモーター−エンハン
サ−リーダーフラグメントとグルタミンシンテターゼ(OS)暗号配列の間に、
Pst−inフラグメントと合成オリヒマ−のNco1消化pBV2.GSとの
リゾ−ジョンによって導入し、新しいプラスミr1pcMGsを発生さ°せた(
:PSV2.GSの―造は公告国際特許出願第W0 8704462号に記載
されている)。
pcMGsは第3図に示す。すなわち、pcMGsは次の配列:hCMV−M工
Eプロモーター一エンノ−ンサーの上流に上記合成オリゴマー(ここでは単に便
利なPsti −Ncoi rアダプター」として働く)、hCMV −M工E
プロモーター、および翻訳開始部位でG8暗号配列に直接融合した、最初のイン
トロンを含むM工E遺伝子の完全5′非翻訳領域から構成されるノ・イブリッド
転写単位を含有する。
pcMGs f !Jン酸カルシウム仲介トランスフェクションによってCHO
−に工細胞に導入し、シラスミドt−G5−インヒビター、メチオニンスルホキ
シミン(MSX )に対する抵抗性の付与能力について試験した。psv2.e
sとの比較結果を第1表に示す。
pcMGsがpsv2.osと類似のs度で20 M MSX K対する抵抗性
を付与できることは、このベクターで活性G8酵素が実際に発現されることを示
している。

pTTIMP1に用因られたTIMP cDNAとSV40ポリアゾニレ−ジョ
ンシグナル(ドチャテイら:ネイチャー、第318巻、第66〜69頁、198
5年)をpEE 乙の唯一のH1ndエエエおよびBan H工部位に挿入して
pKK6TIMP t−創製した。pZK 6は、咄乳動物細胞内での覆設を抑
制する配列が除去された細菌ベクターである。これはpCT 54の亜工〜Bc
l工部分(エムターシュら:プロシーデイングズ・オプ・デ・ナショナル・アカ
デミ−・サブ・サイエンシズ・オプ・fφユナイテツP 、スティソ・オブ・ア
メリカ、第80巻、第3671〜3675頁、1983年)をpsF 64(メ
ルトンら:ヌクレインク・アシツズ・リサーチ、から消化によって予め除去し、
フレノウ醪素で充填し、再リゾ−ジョンした。と工〜Bam Hエフラグメント
は237 bp SV 40前期ポリアゾニレージョンシグナル(SV40 2
770〜2533)である。Bam I工〜BglエフラグメントはpBR32
8(375〜2422)から誘導され、拓工とだ1工部位の間(651〜142
5)を欠失させ、ついでと工部位1csallリンカ−を付加したものである。
Bgl N部位からXmn N部位までの配列はpsP 64のβ−2クタマー
ゼ遺伝子に由来するhcMVMIE フロそ一ターーエンハンサーオヨヒ5′非
翻訳配列を含有する2129塩基対のNco1フラグメントを部分Ncoi消化
によってp(MGSから単離し、pBR6゜TIMP中のTIMPの翻訳開始シ
グナルと重複するNcoi部位に挿入して、プラスミドpET−1(第2図に示
す〕を発生させた。 。
GS遺伝子は、Pvui消化psVLGs 、 iに合成りam Hニリンカー
を付加させたのち、psVLGs 、 1の5.5 K Pvu1〜BamHエ
フラグメント(第1図)をpHT、iのBamH工部位に導入してpHT、iG
s ′f:発生させることにより、永久細胞系の選択を可能にするためにpHT
、 iに導入された。
このシラスミドではhCMV −TIMPおよびGS転写単位は同じ配向で転写
する。
pHT 、 1GSはリン酸カルシウム仲介トランスフェクションによってCH
O−に工細胞中に導入され、20μM MSXに対して抵抗性のクローンをトラ
ンスフェクション2〜3週後に単離した。T工MP分泌速度は、培養上澄液を、
捕獲抗体としてヒツジ抗T工MPポリクローナル抗体、認識抗体としてマウスT
工MPモノクローナル抗体、モノクローナル抗体の結合はヒツジ抗マウスエgG
ペルオキシダーゼ抱合体を用いて明らかにする特異的2サイ) EL工SAで試
験することによって測定された。アッセイの検量のための標準としては、a製し
た天然T工Ωを用いた。全曲線が2〜2 D D n97m1の範囲内で直線で
あった。CHO細胞調節培養メジウム中では非特異的反応は検出されなかった。
ひとつの細胞系os、19 kついで再びクローニングした。サブクローンGS
19−12はTIMPを3X10”分子/細胞/日というきわめて高いレベルで
分泌”する。
この細胞系から抽出される総デノムDNAを、標準方法を用いてサデンプロット
解析によりT工MPプローブとハイブリダイゼーションさせて、各細胞ごとにT
工MP転写単位の1個の無傷のコピーを含有することが明らかにされた(ならび
に2個の再配置ゾ2スミrバンド)。
この細胞系を高レベルのMSXで選択し、最初の選択で500μMのMSX i
c抵抗性を示した赴胞のプールを単離し、再クローニングした。クローンG S
−i 9.6(500)14は3X10’分子/絶胞/日のTIMP i分泌
する。このpH胞系におけるベクターコを一数は約20〜30コピー/細胞であ
る。一層遺伝子を増幅させるための選択の次回の反復では、T工MP分泌の増大
を生じなかった。
すなわち、pHT 、 iからのhCMV −T工MP転写転位はCHD−に工
細胞中、細胞あたりほぼ単一のコピーできわめて切土的に発現されることが可能
で、遺伝子増幅1回で現方法を用いて最大の分泌速度が導かれるようでhCMV
−M工Eプロモーター一エンハーサ一一リーダーが他のタンパク°質配列の切藁
的な発現を企図しても使用できるかどうかを試験するために、2種の異なるが類
縁のプラスミノーゲンアクティベータ暗号配列(FA−1およびFA−2と呼ば
れる)t−1このタンパク質暗号配列が直接hCMV配列に融合したベクタニ中
、CHO−XI lf&I胞に導入した。
いずれの場合も、翻訳配列の最初にNcoi部位がなく、シたがって、合成オリ
がヌクレオチドを使用して翻訳領域内の適当な制限部位からその標準暗号配列を
再生した。pHT、iに使用したような修飾hCMV翻訳開始シグナルの配列も
合成オリゴヌクレオチド内に構築し、これをつbでPst−i制限部位で終結さ
せた。hCMVのPst−1mmフラグメントラにこの部位に挿入し、完全ナソ
ロモーターーエンハンサーーリーダー配列を創製した。
hCMV−プラスミノ−ダンアクティベーター転写単位は、CHO−に工細胞中
に、上述のように唯一のBamHJ部位にGS遺伝子を挿入したのちに挿入し、
プラスミノーゲンアクティベーターを分泌するMSX抵抗性糺胞全車離した。
各場合における最善の初期トランスフェクション細胞系の分泌速度を第2表に示
す。これから、kLCMv−プロモーター−エンハンサー−IJ−ターμ、こレ
ラ(7)2種のプラスミノーゲンアクティベータータンパク質の切上的な発現を
企図して同様に使用できることが明らかである。

pEE 6 bcMvは、hCMVのPat−1m 7ラグメント、Hlnd
エエエ消化pEE 6および次の配列:を有する相補性オリゴヌクレオチrをリ
ゾ−ジョンさせて作成した。
免疫グロブリン軽鎖をコーrするcDNA f 、pEK6゜hCMVのEco
RI部位に、hCMV −M工E 7°0モーター−エンハンサ−+ リーダー
がこのcDNAの発現を支配できるようにまた複製の5V4Q起点を含有する選
択可能なマーカー遺伝子が各プラスミドのBamH工部位に挿入されるように、
挿入した。
このシラスミドを標準DEAE−デキストラントランスフェクション法によって
CO3−1?ル腎に細胞にトランスフェクションし、トランスフェクション72
時間後に、短時間、発現をモニタリングした。@釦は少なくとも1000 ng
/mlの速度でメジウム中に分泌され、全hCMV−MIE 5’−非翻訳配列
を翻訳開始ATGまで(翻訳開始ATGは含まない)ついでマウス免疫グロブリ
ン軽鎖遺伝子の天然5′−非細訳領域の15塩基を含有する転写単位から、実際
に軽鎖が発現することを示している。
