Procedimiento para producir lípidos que contienen ácidos grasos polienoicos a partir de microorganismos eucariotas capaces de producir por lo menos aproximadamente el 20% de su biomasa en forma de lípidos, que comprende añadir a un medio de fermentación que comprende dichos microorganismos una fuente de carbono no alcohólica y una fuente de nutrientes limitadora en una proporción suficiente para incrementar la densidad de biomasa de dicho medio de fermentación hasta por lo menos aproximadamente 100 g/l.
Claims (47)
1. Procedimiento para producir lípidos que
contienen ácidos grasos polienoicos a partir de microorganismos
eucariotas capaces de producir por lo menos aproximadamente el 20%
de su biomasa en forma de lípidos, que comprende añadir a un medio
de fermentación que comprende dichos microorganismos una fuente de
carbono no alcohólica y una fuente de nutrientes limitadora en una
proporción suficiente para incrementar la densidad de biomasa de
dicho medio de fermentación hasta por lo menos aproximadamente 100
g/l.
2. Procedimiento para cultivar microorganismos
eucariotas capaces de producir por lo menos aproximadamente el 20%
de su biomasa en forma de lípidos, comprendiendo dicho
procedimiento añadir a un medio de fermentación que comprende dichos
microorganismos una fuente de carbono y una fuente de nutrientes
limitadora en una relación suficiente para incrementar la densidad
de biomasa de dicho medio de fermentación hasta por lo menos
aproximadamente 100 g/l, en el que dicho procedimiento produce por
lo menos una proporción de aproximadamente 0,5 g/l/hora de lípidos,
y en el que por lo menos aproximadamente el 15% de los lípidos
totales producidos por dichos microorganismos son lípidos
poliinsaturados.
3. Procedimiento para cultivar microorganismos
eucariotas que comprende añadir a un medio de fermentación que
comprende dichos microorganismos una fuente de carbono y una
fuente de nutrientes limitadora en una proporción suficiente para
incrementar la densidad de biomasa de dicho medio de fermentación
hasta por lo menos aproximadamente 100 g/l, en el que se
seleccionan dichos microorganismos de entre el grupo que comprende
algas, hongos (incluyendo la levadura), protistas, bacterias y
mezclas de los mismos, y en el que dichos microorganismos son
capaces de producir por lo menos aproximadamente el 20% de su
biomasa en forma de lípidos que comprenden unos ácidos grasos
polienoicos.
4. Procedimiento para producir lípidos
microbianos eucariotas, que comprende:
- (a)
- cultivar microorganismos eucariotas en un medio de fermentación para incrementar la densidad de biomasa de dicho medio de fermentación hasta por lo menos aproximadamente 100 g/l;
- (b)
- proporcionar condiciones de fermentación suficientes para permitir que dichos microorganismos produzcan dichos lípidos; y
- (c)
- recuperar dichos lípidos, en el que más de aproximadamente el 15% de dichos lípidos son lípidos poliinsaturados.
5. Procedimiento para producir lípidos
microbianos eucariotas, que comprende:
- (a)
- añadir una fuente de carbono no alcohólica y una fuente de nutrientes limitadora a un medio de fermentación que comprende dichos microorganismos;
- (b)
- proporcionar condiciones suficientes para que dichos microorganismos produzcan dichos lípidos microbianos; y
- (c)
- recuperar dichos lípidos microbianos,
en el que por lo menos aproximadamente el 15% de
dichos lípidos microbianos son lípidos poliinsaturados.
6. Procedimiento para producir lípidos
microbianos eucariotas, que comprende:
- (a)
- añadir una fuente de carbono y una fuente de nutrientes limitadora a un medio de fermentación que comprende microorganismos eucariotas y proporcionar condiciones suficientes para mantener un nivel de oxígeno disuelto de por lo menos aproximadamente el 4% en dicho medio de fermentación para producir una densidad de biomasa de por lo menos aproximadamente 100 g/l;
- (b)
- proporcionar condiciones suficientes para mantener un nivel de oxígeno disuelto de aproximadamente el 1% o menos en dicho medio de fermentación y proporcionar condiciones suficientes para permitir que dichos microorganismos produzcan dichos lípidos; y
- (c)
- recuperar dichos lípidos microbianos,
en el que por lo menos aproximadamente el 15% de
dichos lípidos microbianos son lípidos poliinsaturados.
7. Procedimiento para enriquecer el contenido en
ácidos grasos polienoicos de un microorganismo, que comprende
fermentar dicho microorganismo en un medio de cultivo que presenta
un nivel de oxígeno disuelto menor del 10%.
8. Procedimiento según la reivindicación 1, en el
que dicho procedimiento comprende una fase de aumento de la
densidad de biomasa y una fase de producción, en el que dicha fase
de aumento de la densidad de biomasa comprende añadir dicha fuente
de carbono y dicha fuente de nutrientes limitadora y dicha fase de
producción comprende añadir dicha fuente de carbono sin adición o
con adición reducida de dicha fuente de nutrientes limitadora, para
inducir condiciones de nutrientes limitadoras que induzcan la
producción de lípidos.
9. Procedimiento según cualquiera de las
reivindicaciones 4, 5 ó 8, en el que la cantidad de oxígeno
disuelto presente en dicho medio de fermentación durante dicha fase
de producción o dicha etapa de producción de lípidos es menor que
la cantidad de oxígeno disuelto presente en dicho medio de
fermentación durante dicha fase de aumento de la densidad de
biomasa o dicha etapa de cultivo de microorganismos.
10. Procedimiento según cualquiera de las
reivindicaciones 4, 5 ó 8, en el que la cantidad de oxígeno
disuelto en dicho medio de fermentación durante dicha etapa de
aumento de la densidad de biomasa o dicha etapa de cultivo de
microorganismos es por lo menos aproximadamente el 4%.
11. Procedimiento según cualquiera de las
reivindicaciones 4, 5 ó 8, en el que la cantidad de oxígeno
disuelto en dicho medio de fermentación durante dicha fase de
producción o dicha etapa de producción de lípidos es menor de
aproximadamente el 1%.
12. Procedimiento según cualquiera de las
reivindicaciones 1 ó 5, en el que dicha fuente de carbono no
alcohólica comprende un carbohidrato.
13. Procedimiento según cualquiera de las
reivindicaciones 1, 2, 3, 5 ó 36, en el que dicha fuente de
nutrientes limitadora se selecciona de entre el grupo que comprende
fuentes de nitrógeno, fuentes de carbono, fuentes de fosfato,
fuentes de vitaminas (tales como fuentes de vitamina B12, fuentes
de pantotenato, fuentes de tiamina), fuentes de oligometales (tales
como fuentes de zinc, fuentes de cobre, fuentes de cobalto, fuentes
de níquel, fuentes de hierro, fuentes de manganeso, fuentes de
molibdeno), y fuentes de metales mayores (tales como fuentes de
magnesio, fuentes de calcio, fuentes de sodio, fuentes de potasio),
fuentes de sílice y mezclas de las mismas.
14. Procedimiento según cualquiera de las
reivindicaciones 1, 2, 3, 5 ó 36, en el que dicha fuente de
nutrientes limitadora es una fuente de oligometales o una fuente de
metales mayores seleccionada de entre el grupo que consiste en
sales de sulfato y de cloruro de tales metales (tales como
MgSO_{4}\cdot7H_{2}O; MnCl_{2}\cdot4H_{2}O;
ZnSO_{4}\cdot7H_{2}O; CoCl_{2}\cdot6H_{2}O;
Na_{2}MoO_{4}\cdot2H_{2}O; CuSO_{4}\cdot5H_{2}O;
NiSO_{4}\cdot6H_{2}O; FeSO_{4}\cdot7H_{2}O; CaCl_{2};
K_{2}SO_{4}; KCl; y Na_{2}SO_{4}) y mezclas de los
mismos.
15. Procedimiento según cualquiera de las
reivindicaciones 1, 2, 3, 5 ó 36, en el que dicha fuente de
nutrientes limitadora comprende una fuente de nitrógeno.
16. Procedimiento según cualquiera de las
reivindicaciones 1, 2, 3, 5 ó 36, en el que dicha fuente de
nutrientes limitadora comprende una sal amónica inorgánica.
17. Procedimiento según cualquiera de las
reivindicaciones 1, 2, 3, 5 ó 36, en el que dicha fuente de
nutrientes limitadora comprende hidróxido de amonio.
18. Procedimiento según cualquiera de las
reivindicaciones 1, 2, 3, 5 ó 36, en el que se controla el pH del
medio de fermentación por medio de dicha fuente de nutrientes
limitadora.
19. Procedimiento según cualquiera de las
reivindicaciones 1 a 7, en el que dicho medio de fermentación o
dicho medio de cultivo presenta una temperatura de por lo menos
aproximadamente 20ºC.
20. Procedimiento según cualquiera de las
reivindicaciones 1 a 7, en el que dicho procedimiento produce
lípidos en una proporción media de por lo menos aproximadamente
0,5 g/l/hora.
21. Procedimiento según cualquiera de las
reivindicaciones 1 a 7, en el que dicho procedimiento produce
lípidos en una proporción media de por lo menos aproximadamente
0,5 g/l/hora y en el que por lo menos aproximadamente el 15% de
dichos lípidos son lípidos poliinsaturados.
22. Procedimiento según cualquiera de las
reivindicaciones 1 a 7, en el que dicho procedimiento produce
lípidos en una proporción media de por lo menos aproximadamente
0,5 g/l/hora, y en el que la cantidad total de ácidos grasos
omega-3 y omega-6 es por lo menos
aproximadamente el 20% de dichos lípidos.
23. Procedimiento según cualquiera de las
reivindicaciones 1 a 7, en el que dicho procedimiento produce
lípidos en una proporción media de por lo menos aproximadamente
0,5 g/l/hora y en el que por lo menos aproximadamente el 25% de
dichos lípidos es el ácido docosahexaenoico.
24. Procedimiento según cualquiera de las
reivindicaciones 1 a 7, en el que se seleccionan dichos
microorganismos de entre el grupo que comprende algas, hongos
(incluyendo la levadura), protistas, bacterias y mezclas de los
mismos, en el que dichos microorganismos son capaces de producir
ácidos grasos polienoicos u otros lípidos que anteriormente se
creía que requerían oxígeno para su síntesis.
25. Procedimiento según cualquiera de las
reivindicaciones 1 a 7, en el que se seleccionan dichos
microorganismos de entre el grupo que comprende algas, hongos
(incluyendo la levadura), protistas, bacterias y mezclas de los
mismos, en el que dichos microorganismos son capaces de producir
ácidos grasos polienoicos u otros lípidos que anteriormente se
creía que requerían oxígeno para su síntesis y en el que dichos
microorganismos se cultivan en un procedimiento de lote
alimentado.
26. Procedimiento según cualquiera de las
reivindicaciones 1 a 7, en el que dichos microorganismos se
seleccionan de entre el grupo que comprende algas, hongos
(incluyendo la levadura), protistas, bacterias y mezclas de los
mismos, en el que dichos microorganismos son capaces de producir
ácidos grasos polienoicos u otros lípidos que anteriormente se
creía que requerían oxígeno para su síntesis y en el que se
cultivan dichos microorganismos en un procedimiento de lote
alimentado y que comprende además mantener un nivel de oxígeno
menor que aproximadamente el 3% de saturación en dicho medio de
fermentación durante la fase de producción de dicho procedimiento
de fermentación.
27. Procedimiento según cualquiera de las
reivindicaciones 1 a 7, en el que dichos microorganismos son
microalgas y microorganismos a modo de algas.
28. Procedimiento según cualquiera de las
reivindicaciones 1 a 7, en el que dichos microorganismos son
Stramenopiles.
29. Procedimiento según cualquiera de las
reivindicaciones 1 a 7, en el que dichos microorganismos son del
orden de Traustoquitriales.
30. Procedimiento según cualquiera de las
reivindicaciones 1 a 7, en el que dichos microorganismos se
seleccionan de entre el grupo que comprende Thraustochytrium,
Schizochytrium, y mezclas de los mismos.
31. Procedimiento según la reivindicación 2, en
el que dicho procedimiento además comprende:
- (a)
- una fase de aumento de la densidad de biomasa, en la que el nivel de oxígeno disuelto en el medio de fermentación es por lo menos de aproximadamente el 4%; y
- (b)
- una fase de producción elevada de lípidos, en la que el nivel de oxígeno disuelto en el medio de fermentación es menor de aproximadamente el 1%.
32. Procedimiento según cualquiera de las
reivindicaciones 2, 3, 6 ó 36, en el que dicha fuente de carbono
es una fuente de carbono no alcohólica.
33. Procedimiento según cualquiera de las
reivindicaciones 1 a 7, en el que dicho procedimiento produce de
promedio por lo menos aproximadamente 0,2 g/l/hora de ácido
docosahexaenoico.
34. Procedimiento según la reivindicación 4, en
el que dicha etapa de recuperación de lípidos comprende:
- (d)
- eliminar el agua de dicho medio de fermentación para proporcionar microorganismos secos; y
- (e)
- aislar dichos lípidos de dichos microorganismos secos.
35. Procedimiento según la reivindicación 4, en
el que dicha etapa de recuperación de lípidos comprende:
- (d)
- tratar el caldo de fermentación para permeabilizar, lisar o romper las células microbianas; y
- (e)
- recuperar los lípidos del caldo de fermentación a través de separación por gravedad, con o sin la ayuda de un agente para coadyuvar en romper la emulsión lípido/agua.
36. Procedimiento según la reivindicación 4, en
el que dicha etapa de cultivo de microorganismos comprende añadir
una fuente de carbono y una fuente de nutrientes limitadora.
37. Procedimiento según cualquiera de las
reivindicaciones 5 ó 6, en el que dicha etapa de recuperación de
lípidos comprende:
- (d)
- eliminar el agua de dicho medio de fermentación para proporcionar microorganismos secos; y
- (e)
- aislar dichos lípidos de dichos microorganismos secos.
38. Procedimiento según cualquiera de las
reivindicaciones 5 ó 6, en el que dicha etapa de recuperación de
lípidos comprende:
- (d)
- evaporar el agua de dicho medio de fermentación sin centrifugación previa para proporcionar microorganismos secos; y
- (e)
- aislar dichos lípidos de dichos microorganismos secos.
39. Procedimiento según cualquiera de las
reivindicaciones 5 ó 6, en el que dicha etapa de recuperación de
lípidos comprende:
- (d)
- tratar el caldo de fermentación para permeabilizar, lisar o romper las células microbianas; y
- (e)
- recuperar los lípidos del caldo de fermentación a través de separación por gravedad, con o sin la ayuda de un agente para coadyuvar en romper la emulsión lípido/agua.
40. Procedimiento según cualquiera de las
reivindicaciones 7 ó 44, en el que dicho nivel de oxígeno disuelto
es menor del 5%.
41. Procedimiento según cualquiera de las
reivindicaciones 7 ó 44, en el que dicho nivel de oxígeno disuelto
es menor del 1%.
42. Procedimiento según la reivindicación 7, en
el que durante una fase de cultivo inicial, el nivel de oxígeno
disuelto es mayor de aproximadamente el 10% y durante una fase de
producción posterior el nivel de oxígeno disuelto es menor del
10%.
43. Procedimiento según la reivindicación 7, en
el que dicho microorganismo contiene genes de policeturo
sintetasa.
44. Procedimiento heterótrofo para producir
productos y microorganismos que comprende:
- (a)
- cultivar dichos microorganismos en un medio de cultivo en el que dichos microorganismos contienen genes de policeturo sintetasa; y
- (b)
- mantener el nivel de oxígeno disuelto a un nivel inferior a aproximadamente el 10%.
45. Procedimiento según la reivindicación 44, en
el que dichos genes de policeturo sintetasa se encuentran
naturalmente en dichos microorganismos.
46. Procedimiento según la reivindicación 44, en
el que dichos genes de policeturo sintetasa se introducen
genéticamente en dichos microorganismos.
47. Procedimiento según la reivindicación 44, en
el que durante una fase de cultivo inicial, el nivel de oxígeno
disuelto es mayor de aproximadamente el 10% y durante una fase de
producción posterior el nivel de oxígeno disuelto es menor de
aproximadamente el 10%.
