Claims (50)
- 微生物の多価不飽和脂肪酸含量を豊富にする方法であって、飽和の1%未満の溶存酸素レベルを有する発酵培地で、前記微生物を発酵させることを含む方法。
- 産物と微生物を生産する従属栄養方法であって、飽和の1%未満の溶存酸素レベルを有する発酵培地で、ポリケチドシンターゼ遺伝子を有する前記微生物を培養することを含む方法。
- 微生物により生産される脂質の合計の少なくとも15%を多価不飽和脂肪酸として生産することができる、スラウストキトリアレス(Thraustochytriales)目である微生物から、多価不飽和脂肪酸を含む脂質を生産する方法であって、非アルコール炭素源と制限栄養素源とを含む培地で前記微生物の発酵を行う段階を含む方法であり、
発酵培地の溶存酸素含量が飽和の少なくとも4%である場合に、非アルコール炭素源と制限栄養素源とが発酵培地のバイオマス密度を増大させるために培地に添加され、かつ
発酵培地の溶存酸素含量が飽和の3%未満である場合に、培地の発酵が多価不飽和脂肪酸を含む脂質を生産する、
多価不飽和脂肪酸を含む脂質を生産する方法。 - そのバイオマスの少なくとも20%を脂質として生産することができる、スラウストキトリアレス(Thraustochytriales)目である微生物から、多価不飽和脂肪酸を含む脂質を生産する方法であって、炭素源を含む培地で前記微生物の発酵を行う段階を含む方法であり、
発酵培地の溶存酸素含量が飽和の少なくとも4%である場合に、炭素源が発酵培地のバイオマス密度を増大させるために供給バッチ方法の発酵培地に添加され、かつ
発酵培地の溶存酸素含量が飽和の3%未満である場合に、培地の発酵が多価不飽和脂肪酸を含む脂質を生産する、
多価不飽和脂肪酸を含む脂質を生産する方法。 - そのバイオマスの少なくとも20%を脂質として生産することができる、スラウストキトリアレス(Thraustochytriales)目である微生物から、多価不飽和脂肪酸を含む脂質を生産する方法であって、非アルコール炭素源と制限栄養素源とを含む培地で前記微生物の発酵を行う段階を含む方法であり、

前記方法が少なくとも0.5g/L/hrの平均速度で脂質を生産し、かつ
前記微生物により生産された脂質の合計の少なくとも15%が多価不飽和脂肪酸である、
微生物を生育する方法。 - スラウストキトリアレス(Thraustochytriales)目である微生物を生育する方法であって、炭素源と制限栄養素源とを含む培地で前記微生物の発酵を行う段階を含む方法であり、
発酵培地の溶存酸素含量が飽和の少なくとも4%である場合に、炭素源と制限栄養素源とが発酵培地のバイオマス密度を増大させるために培地に添加され、
発酵培地の溶存酸素含量が飽和の3%未満である場合に、培地の発酵が多価不飽和脂肪酸を含む脂質を生産し、かつ
前記微生物がそのバイオマスの少なくとも20%をいくつかの多価不飽和脂肪酸を含む脂質として生産することができる、
微生物を生育する方法。 - 発酵培地のバイオマス密度を増大させるために、発酵培地でスラウストキトリアレス(Thraustochytriales)目である微生物を生育するステップと、
発酵培地の溶存酸素含量が飽和の3%未満である場合に、前記微生物に脂質を生産させるステップと、
少なくとも15%が多価不飽和脂肪酸である脂質を回収するステップとを含む、微生物脂質を生産する方法。 - 炭素源と制限栄養素源とを含む培地で微生物の発酵を行うステップと、
発酵培地の溶存酸素レベルが飽和の少なくとも4%である場合に、炭素源と制限栄養素源とを発酵培地に添加するステップと、
発酵培地の溶存酸素レベルが飽和の3%以下である場合に、脂質を生産するステップと、
少なくとも15%が多価不飽和脂肪酸である微生物脂質を回収するステップとを含む、微生物脂質を生産する方法。 - 供給バッチ方法である、請求項1〜3、5、または7〜10のいずれか一項に記載の方法。
- バッチ方法または連続方法である、請求項1〜3、5、または7〜10のいずれか一項に記載の方法。
- 発酵培地の溶存酸素含量が飽和の3%未満である場合に、脂質生産を引き起こす栄養素制限状態を誘導するために、制限栄養素源を全く加えないかあるいはほどんど加えない状態で炭素源を添加する、請求項1〜10のいずれか一項に記載の方法。
- 培地の発酵が多価不飽和脂肪酸を含む脂質を生産する場合、発酵培地の溶存酸素含量は飽和の1%未満である、請求項1〜10のいずれか一項に記載の方法。
- 発酵培地中の溶存酸素レベルが最初の24時間は8%、24時間目から40時間目までは4%、40時間目からプロセスの終りまでは0.5%以下に維持され少なくとも90時間行われる、請求項1〜10のいずれか一項に記載の方法。
- 発酵培地中の溶存酸素レベルがプロセスの40時間目以降からプロセスの終りまで0.5%以下に維持される、請求項1〜10のいずれか一項に記載の方法。
- 非アルコール炭素源が炭水化物を含む、請求項3または5に記載の方法。
- 制限栄養素源が、窒素源、炭素源、リン酸塩源、ビタミン源、微量金属源、主要金属源、珪酸源、およびそれらの混合物から成る群より選択される栄養素源を含む、請求項3、5、または7〜10のいずれか一項記載の方法。

- 少なくとも0.5g/L/hrの平均速度で脂質を生産し、ω−3およびω−6脂肪酸の合計量が前記脂質の少なくとも20%である、請求項1〜10のいずれか一項に記載の方法。
- 少なくとも0.5g/L/hrの平均速度で脂質を生産し、前記脂質の少なくとも25%がドコサヘキサエン酸である、請求項1〜10のいずれか一項に記載の方法。
- 微生物が、好気的条件下で生産することができる多価不飽和脂肪酸または他の脂質を生産することができる、請求項1〜10のいずれか一項に記載の方法。
- 微生物が、好気的条件下で生産される多価不飽和脂肪酸または他の脂質を生産する、請求項31に記載の方法。
- 微生物が好気的条件下で生産することができる多価不飽和脂肪酸または他の脂質を生産することができる方法であって、前記微生物が供給バッチ方法において生育する、請求項1〜10のいずれか一項に記載の方法。

- 微生物が好気的条件下で生産することができる多価不飽和脂肪酸または他の脂質を生産することができる方法であって、前記微生物が供給バッチ方法において生育する、請求項1〜10のいずれか一項に記載の方法。
- 微生物がスラウストキトリウム(Thraustochytrium)属、シゾキトリウム(Schizochytrium)属およびそれらの混合物から成る群より選択される、請求項1〜10のいずれか一項に記載の方法。
- 発酵培地中の溶存酸素が調節される、請求項1〜10のいずれか一項に記載の方法。

(b)脂質/水エマルションの破壊を援助する作用物質の支援があるかまたは支援がない状態で比重選別法により発酵培地から脂質を回収するステップであって、微生物脂質の少なくとも15%が多価不飽和脂肪酸であるステップとをさらに含む、請求項1〜10のいずれか一項に記載の方法。 - 微生物脂質の少なくとも15%がドコサヘキサエン酸である、請求項42に記載の方法。
