Claims (16)
- Y g/Lの細胞乾燥物質(CDM)(ここで、Yは、100〜300g/Lの範囲で存在する)で、好気性条件下で発酵槽において栄養要求性海洋微生物を連続して培養するための方法であって、培養液1L当たり(Y×h)g(hは、1.1〜3.0の範囲である)の量で徐々に添加され、そして20〜100時間の滞留時間をもって、炭素源を含んで成る培養培地において、前記栄養要求性海洋微生物を培養することを含んで成り、こので前記炭素源がバイオマスの生成のために制限的であり、当該生成されるバイオマスの少なくとも40%が、クロロホルム:メタノールの混合物により抽出できる成分から構成される、前記の方法。
- 前記培養培地が、培養液1L当たり(Y×h×f)(ここで、fは、0.002〜0.2の範囲である)の量で徐々に添加される、窒素源を含んで成る請求項1記載の方法。
- 前記培養培地が、達成されるバイオマス濃度のために制限的でない量で、徐々に添加される、バイオマス形成のために必要とされる塩、鉱物及び任意には、ビタミンを含んで成る請求項1記載の方法。
- hが、1.1〜2.5の範囲、特に1.2〜2.0の範囲である請求項1記載の方法。
- fが、0.004〜0.1の範囲、特に0.01〜0.04の範囲である請求項2〜4のいずれか1項記載の方法。
- 前記栄養要求性海洋微生物が、藻類である請求項1〜5のいずれか1項記載の方法。
- 前記栄養要求性海洋微生物が、トラウストキトリドsp.(Thraustochytrids sp.)である請求項1〜6のいずれか1項記載の方法。
- 前記トラウストキトリドsp.が、シゾキトリウム(Schizochytrium)又はトラウストキトリウム(Thraustochytrium)から成る群から選択される請求項1〜7のいずれか1項記載の方法。
- 前記培養温度が、20〜35℃の範囲である請求項1〜8のいずれか1項記載の方法。
- 前記培養培地のpHが、3.0〜9.0の範囲である請求項1〜9のいずれか1項記載の方法。
- 前記クロロホルム及びメタノールが、2:1(v/v)の比で混合される請求項1記載の方法。
- 少なくとも0.2gのDHA/l/hのポリエン酸生産性が達成される請求項1〜11のいずれか1項記載の方法。
- 前記連続培養工程における培養液の滞留時間が、一定に維持され、そして20〜100hの範囲である請求項1〜12のいずれか1項記載の方法。
- 前記連続培養工程における培養液の滞留時間が、20〜100の範囲内で変化する請求項1〜13のいずれか1項記載の方法。
- 前記連続的培養が、次の3培養段階:
a)初期バッチ工程、続いて
b)フェドバッチ工程、続いて
c)連続工程、
を含んで成る請求項1記載の方法。 - 前記溶解される酸素のレベルが、段階c)の開始から10%以下の飽和で維持される請求項15記載の方法。
