技术领域
本发明涉及一种含有植物萃取物的组合物,尤其涉及一种用以提升转谷氨酰胺酶1基因、角蛋白基因、水通道蛋白3基因、聚角蛋白微丝基因、葡萄糖脑苷酯酶基因、及玻尿酸合成酶基因表达的含有植物萃取物的组合物的用途。
背景技术
表皮层为皮肤的最外层,由外往内依序为角质层、颗粒层、有棘层及基底层,表皮层主要由基底层中未分化的圆柱型角质细胞持续向上进行分化形成,此过程称为角质化。角质细胞内含水量高,随着细胞向上代谢分化,角质细胞形状会逐渐变成扁平状,且细胞核及细胞器开始退化萎缩,并在角质层形成不具细胞核与细胞器的死细胞。表皮层的主要功能为使皮肤保水,并形成皮肤屏障以抵御各种外来伤害,其中表皮层最外层由弱酸性的皮脂膜以及如砖墙结构的角质层所构成,此屏障能锁住皮肤的水分和油脂、抵抗皮肤表面病菌入侵,及对抗外界异物及紫外光等伤害,对人体有非常重要的保护作用。
表皮层中的角质层,其角质细胞虽然为死细胞,但其主要成分为角蛋白(keratin),角蛋白能吸收水分使皮肤保持湿润,角质细胞也会分泌如玻尿酸等物质作为细胞间质,以维持表皮层皮肤屏障的结构完整,以防止皮肤水分散失及形成完整防护。当皮肤接触过冷或过热的环境以及照射紫外光等刺激,会导致角质细胞无法维持正常的代谢循环,并使得皮肤表皮层屏障受损,而 让皮肤变得粗糙、干燥脱屑、脆弱易受刺激、敏感泛红且会使皮肤保水能力下降,因此角质层的健康对于皮肤保湿以及抵御外来伤害着实非常重要。
综合上面所述,为了改善因角质细胞受损导致皮肤变得脆弱、易敏,以及皮肤保水能力下降的问题,开发一种能有效维持角质细胞排列、维持角质层结构完整以及使角质细胞分泌更多保湿因子,以提升皮肤屏障以及保湿功能的组合物,着实有其必要性。
然而,目前用于该用途的成分或药物有限,若为某种植物萃取物,其必须施予相当高的剂量,在使用成本上以及效果上有其局限,因此,若能施予多种萃取物的少量组分,即可产生单一成分的高剂量效果时,对于前述功效的运用,不但可大幅提升,也同时降低了使用成本。
发明内容
缘此,本发明的一目的在提供一种植物萃取物用于制备提升转谷氨酰胺酶1(Transglutaminase 1,TGM1)基因、角蛋白(Keratin,KRT)基因、水通道蛋白3(Aquaporin 3,AQP3)基因、聚角蛋白微丝(Filaggrin,FLG)基因、葡萄糖脑苷酯酶(Glucocerebrosidase,GBA)基因、及玻尿酸合成酶(Hyaluronan synthase,HAS)基因表达的组合物的用途,其中该植物萃取物包含选自由普洱茶萃取物与蓝莓萃取物、普洱茶萃取物与菠菜萃取物、四季春茶萃取物与蓝莓萃取物、及四季春茶萃取物与菠菜萃取物所组成的组中至少任一组合。
在本发明的一实施例中,其中该角蛋白基因包含角蛋白1(Keratin1,KRT1)基因、角蛋白10(Keratin10,KRT10)基因及角蛋白14(Keratin14,KRT14)基因;该玻尿酸合成酶基因包含玻尿酸合成酶2(Hyaluronan synthase 2,HAS2)基因及玻尿酸合成酶3(Hyaluronan synthase 3,HAS3)基因。
在本发明的一实施例中,该植物萃取物包含至少0.125mg/mL的普洱茶萃取物及至少0.125mg/mL的菠菜萃取物、至少0.125mg/mL的四季春茶萃取物及至少0.125mg/mL的菠菜萃取物、至少0.25mg/mL的普洱茶萃取物及至少0.25mg/mL的蓝莓萃取物、或至少0.25mg/mL的普洱茶萃取物及至少0.25mg/mL的四季春茶萃取物。
在本发明的一实施例中,该组合物具有促进皮肤保湿的功效。
在本发明的一实施例中,该组合物进一步包含医药上可接受的载体,且该组合物为粉末、颗粒、液体、胶状或膏状的剂型,又该组合物被制备成医药品或食品产品的形式。

以下将配合附图进一步说明本发明的实施方式,下述所列举的实施例用以阐明本发明的发明特点及应用,而非以限定本发明的范围,对于本领域技术人员来说,在不脱离本发明的精神和范围内,还可以做出若干改进和润饰,因此本发明的保护范围应当以本申请权利要求书的内容为准。
实施例1植物萃取物的制备
实施例1-1本发明四季春茶萃取物的制备方法
四季春茶(Four Seasons Spring tea)萃取物的制备方法如下:首先,将四季春茶茶叶(leaves of Four Seasons Spring tea plant)洗净并进行干燥,再用粉碎机将四季春茶茶叶进行粗碎,接着以水为溶剂对四季春茶茶叶的粗碎物进行萃取,该溶剂与四季春茶茶叶粗碎物以液固比5~20:1~5均匀混合,萃取温度为50℃~100℃,优选为75℃~95℃,萃取时间约为0.5~3小时,得到四季春茶的粗萃取物,将该粗萃取物冷却至室温后,经由400目的滤网过滤以移除残余固体物,再进一步于45℃~70℃进行减压浓缩而获得浓缩产物,即为本发明的四季春茶萃取物。
实施例1-2本发明菠菜萃取物的制备方法
菠菜萃取物通过对菠菜(Spinacia oleracea)进行萃取而获得,本实施例中使用的菠菜萃取物可购自弘祥农产加工厂。
实施例1-3本发明蓝莓萃取物的制备方法
蓝莓萃取物通过对北美蓝莓(Vaccinium Cyanococcus)果实进行萃取而获得,本实施例中使用的蓝莓萃取物可购自元弘草本科研有限公司。
实施例1-4本发明普洱茶萃取物的制备方法
普洱茶(Puerh tea)萃取物通过对普洱茶茶叶(post-fermented leaves of Camellia sinensis)进行萃取而获得,本实施例中使用的普洱茶萃取物可购自南京泽朗生物科技有限公司。
实施例2本发明植物萃取组合物提升TGM1、KRT1、KRT10、KRT14、APQ3及FLG基因表达的效果
本发明以人类初代皮肤角质细胞(human primary epidermal keratinocytes,HPEK)进行TGM1、KRT1、KRT10、KRT14、APQ3及FLG的基因分析。该人类初代皮肤角质细胞购自CELLnTEC公司(瑞士)编号HPEK-50,将该细胞在无血清的角质细胞培养液(keratinocyte-SFM)(Gibco公司,编号为#10724-011,美国)中培养。
首先,将1.5×10 5个人类初代皮肤角质细胞种植于含2mL的上述培养液的6孔培养板中,将人类初代角质细胞分成十组各别加入:(1)0.125mg/mL的普洱茶萃取物、(2)0.125mg/mL的菠菜萃取物、(3)0.125mg/mL的四季春茶萃取物、(4)0.125mg/mL的以1:1比例混合的普洱茶萃取物与菠菜萃取物的组合物、(5)0.125mg/mL的以1:1比例混合的四季春茶萃取物与菠菜萃取物的组合物、(6)0.25mg/mL的普洱茶萃取物、(7)0.25mg/mL的蓝莓萃取物、(8)0.25mg/mL的四季春茶萃取物、(9)0.25mg/mL的以1:1比例混合的普洱茶萃取物与蓝莓萃取物的组合物、(10)0.25mg/mL的以1:1比例混合的四季春茶萃取物与蓝莓萃取物的组合物,并以没有加入任何萃取物的组别为空白对照组。加入植物萃取物后置于37℃培养箱,使角质细胞形成受植物萃取物刺激后的角质细胞,于第6小时与24小时更换培养液,培养液为角质细胞培养液及植物萃取液。接着,将角质细胞加入裂解液(lysis buffer)收集成管后,使用RNA萃取试剂试剂盒(购自Geneaid公司,台湾,Lot No.FC24015-G)分别收集两组角质细胞内的RNA,接着利用

FAST qPCR Kits(购自Sigma公司,美国,编号38220000000)将六组反转录后产物分别以表1的组合引物进行定量实时反转录聚合酶链式反应(quantitative  real-time reverse transcription polymerase chain reaction)试验,定量TGM1基因、KRT1基因、KRT10基因、KRT14基因、APQ3基因、FLG基因、GBA基因、HAS2基因及HAS3基因的mRNA表达量,条件为95℃反应1秒,60℃反应20秒,总共40个循环,其中定量数值取自阈值循环数(Ct),而目标基因的mRNA相对量推导自方程式2 -△Ct,其中△Ct=Ct 目标基因-Ct ACTB(β-肌动蛋白,beta-actin),接着以SCORE法来定量该十组中个别基因表达量的分数,再将各基因的表达分数求和作为该组的保湿分数,其中SCORE法是利用ACTB基因及基准基因(空白对照组中相应的个别基因)的循环阈值来计算。

本发明的各植物萃取组合物的保湿分数如表2及图1所示,保湿分数越高代表保湿能力越高。先前研究指出转谷氨酰胺酶(Transglutaminase1,TGM)会使角质化细胞的细胞膜与结构蛋白间形成强力键结,并能增加表皮层的强度与稳定性;角蛋白(Keratin,KRT)会形成角蛋白微丝,而聚角蛋白微丝(Filaggrin,FLG)会帮助角蛋白微丝组装成坚固的网络,为皮肤提供强度和弹性;水通道蛋白(Aquaporin,AQP)则会增加角质细胞内水的通透性,以提高角质细胞的含水量;玻尿酸合成酶(Hyaluronan synthase,HAS)会促进角质细胞分泌玻尿酸的能力,并使皮肤角质层结构完整且提升皮肤屏障功能,能使皮肤保水力提升;而葡萄糖脑苷酯酶(Glucocerebrosidase,GBA)则与维持角质结构完整有关。单独以0.125mg/mL普洱茶萃取物、四季春茶萃取物、及菠菜萃取物处理角质细胞所获得的保湿分数分别为2.16、4.2、及2.04;然而以0.125mg/mL的1:1比例混合的普洱茶萃取物与菠菜萃取物的组合物、及四季春茶萃取物与菠菜萃取物的组合物则分别为7.2及10.2;单独以0.25mg/mL普洱茶萃取物、四季春茶萃取物、及蓝莓萃取物处理角质细胞所获得的保湿分数分别为3.5、3.8、及4.7;然而以0.25mg/mL的1:1比例混合的普洱茶萃取物与蓝莓萃取物的组合物、及四季春茶萃取物与蓝莓萃取物的组合物则分别为7.4及6.6,由以上结果可知普洱茶萃取物与蓝莓萃取物的组合物、普洱茶萃取物与菠菜萃取物的组合物、四季春茶萃取物与蓝莓萃取物的组合物、及四季春茶萃取物与菠菜萃取物的组合物对于提升皮肤保湿基因表达的效果相当显著,皆较单独普洱茶萃取物、四季春茶萃取物、菠菜萃取物及蓝莓萃取物更为有效地提升TGM1、KRT1、KRT10、KRT14、APQ3及FLG基因的表达,并能更有效的维持皮肤角质细胞排列,使皮肤角质层结构完整,也能有效地提升皮肤屏障功能以及使皮肤形成更多保湿因子。

综上所述,本发明的普洱茶萃取物与蓝莓萃取物的组合物、普洱茶萃取物与菠菜萃取物的组合物、四季春茶萃取物与蓝莓萃取物的组合物、或四季春茶萃取物与菠菜萃取物的组合物能够产生超乎预期的相乘效果,使皮肤保湿基因的表达较单纯普洱茶萃取物或四季春茶萃取物显著增加,并能更有效地维持皮肤角质细胞排列,使皮肤角质层结构完整,并提升皮肤屏障功能以及使皮肤形成更多保湿因子。
因此,本发明的用于提升转谷氨酰胺酶1基因、角蛋白基因、水通道蛋白3基因、聚角蛋白微丝基因、葡萄糖脑苷酯酶基因、及玻尿酸合成酶基因表达的含有植物萃取物的组合物的用途,也可进一步添加于食品、保健食品、膳食补充品或饮品中,且也可将本发明的植物萃取物的组合物用于制备医药组合物,该医药组合物也可进一步加入所属技术领域所熟知的载体或其他辅剂,该医药组合物的剂型,可为但不限于溶液、胶囊、或锭剂。

  1. 植物萃取物在制备提升转谷氨酰胺酶1(Transglutaminase 1,TGM1)基因、角蛋白(Keratin,KRT)基因、水通道蛋白3(Aquaporin 3,AQP3)基因、聚角蛋白微丝(Filaggrin,FLG)基因、葡萄糖脑苷酯酶(Glucocerebrosidase,GBA)基因、及玻尿酸合成酶(Hyaluronan synthase,HAS)基因表达的组合物中的用途,其中所述植物萃取物包含选自由普洱茶萃取物与蓝莓萃取物、普洱茶萃取物与菠菜萃取物、四季春茶萃取物与蓝莓萃取物、及四季春茶萃取物与菠菜萃取物所组成的组中的至少任一组合。
  2. 如权利要求1所述的用途,其中所述角蛋白(KRT)基因包含角蛋白1(Keratin1,KRT1)基因、角蛋白10(Keratin10,KRT10)基因及角蛋白14(Keratin14,KRT14)基因。
