Description
Die vorliegende Erfindung betrifft das im Anspruch 1 angegebene Verfahren zum
Schneiden doppelsträngiger DNA. Die Ansprüche 2 und 3 betreffen
Ausgestaltungen dieses Verfahrens.
Mit dem Aufkommen der DNA-Rekombinations-Technologie ist
die kontrollierte bakterielle Produktion einer enormen
Vielfalt nützlicher Polypeptide möglich geworden. Man hat
bereits Bakterien in der Hand, die durch diese Technologie
verändert wurden, um die Produktion derartiger Polypeptid-
Produkte zu ermöglichen, wie z. B. Somatostatin (K. Itakura,
et al., Science 198, 1056 (1977)), die jeweils einzelnen
A- und B-Ketten des menschlichen Insulins (D. V. Goeddel,
et al., Proc Nat'l Acad Sci, USA 76, 106 (1979)) und menschliches
Wachstumshormon (D. V. Goeddel et al., Nature 281,
544 (1979)). Kürzlich wurde die DNA-Rekombinations-Technik
dazu verwendet, um die bakterielle Herstellung von Thymosin α1
zu ermöglichen, einer immunverstärkenden Substanz, die in
der Thymusdrüse gebildet wird (US-Patentanmeldung von
Roberto Crea und Ronald Wetzel vom 28. Februar 1980, übertragen
auf die Anmelderin der vorliegenden Anmeldung).
Die Möglichkeiten dieser Technologie sind derartig durchschlagend,
daß so gut wie jedes nützliche Polypeptid
bakteriell hergestellt werden kann und die kontrollierte
Herstellung von Hormonen, Enzymen, Antikörpern und Impfstoffen
gegen eine große Vielfalt von Krankheiten in
Reichweite gebracht wird. Die zitierten Veröffentlichungen,
die im Detail die obengenannten repräsentativen Beispiele
beschreiben, sowie weitere, im folgenden genannte Veröffentlichungen,
welche den Hintergrund der Erfindung bilden,
sind durch Verweis in die Offenbarung mit einbezogen.
Das Arbeitspferd der DNA-Rekombinations-Technik ist
das Plasmid, ein nichtchromosomaler Ring doppelsträngiger
DNA, der in Bakterien gefunden wird und oft in vielfacher
Kopie pro Bakterienzelle vorliegt. Die DNA des Plasmids
enthält eine Information, die benötigt wird, um das Plasmid
in Tochterzellen zu reproduzieren (d. h. ein "Replikon") und
normalerweise ein oder mehr Selektions-Charakteristika, wie
z. B. Resistenz gegen Antibiotika, die es erlauben, diejenigen
Clone der Wirtszelle, die das interessierende Plasmid
enthalten, zu erkennen und bevorzugt in selektivem Medium
wachsen zu lassen. Der Nutzen der bakteriellen Plasmide
liegt in der Tatsache, daß sie durch die eine oder andere
Restriktionsendonuklease oder "Restriktionsenzym" spezifisch
geschnitten werden können, von denen jedes eine andere
Stelle auf der Plasmid-DNA erkennt. Danach können heterologe
Gene oder Genfragmente in das Plasmid eingefügt werden, wobei
entweder die Enden der Schnittstellen oder unmittelbar
an die Schnittstellen angefügte Enden miteinander verbunden
werden. Im folgenden wird der Terminus "heterolog" für ein
Gen verwendet, das normalerweise nicht in E. coli gefunden
wird oder eine Polypeptidsequenz, die normalerweise nicht
durch E. coli hergestellt wird. Der Terminus "homolog"
wird für ein Gen oder Polypeptid verwendet, das in Wildtyp
E. coli hergestellt wird. Die DNA-Rekombination wird außerhalb
der Bakterie durchgeführt. Das resultierende "rekombinante"
Plasmid kann durch ein Verfahren, das als Transformation
bekannt ist, in Bakterien eingeführt werden,
und man erhält große Mengen des rekombinanten Plasmids,
das das heterologe Gen enthält, indem man die Transformanten
wachsen läßt. Wenn das Gen im Hinblick auf Bereiche
des Plasmids, die die Transkription und Translation der
codierten DNA-Information regeln, richtig eingefügt ist,
kann der resultierende Expressionsvektor dazu verwendet
werden, tatsächlich die Polypeptidsequenz zu produzieren,
für die das inserierte Gen codiert, ein Prozeß, der Expression
genannt wird.
Die Expression wird in einer Gegend initiiert, die als
Promoter bekannt ist, der von der RNA-Polymerase erkannt
wird und an den diese bindet. In einigen Fällen, wie z. B.
beim trp-Operon, das im nachfolgenden noch ausführlich
diskutiert werden wird, werden Promoterregionen von "Operator"-
Regionen überlappt, wodurch ein kombinierter Promoter-
Operator gebildet wird. Operatoren sind DNA-Sequenzen, die
von sogenannten Repressorproteinen erkannt werden, die dazu
dienen, die Häufig der Initiation der Transkription
an einem speziellen Promoter zu regulieren. Die Polymerase
arbeitet sich an der DNA entlang, wobei sie die in dem codierenden
Strang enthaltene Information vom 5′- zum 3′-Ende
in Messenger-RNA (mRNA) transkribiert, die ihrerseits wieder
in ein Polypeptid translatiert wird, das die Aminosäuresequenz
aufweist, für die die DNA codiert. Jede Aminosäure
wird durch ein einziges Nukleotidtriplet oder "Codon" codiert,
das innerhalb eines "Strukturgens" liegt, wie man es
für die vorliegenden Zwecke nennen mag, d. h. in dem Teil,
der für die Aminosäuresequenz des exprimierten Produkts
codiert. Nach dem Binden an den Promoter transkribiert die
RNA-Polymerase vorerst Nukleotide, die für eine Ribosomen-
Bindestelle codieren, sodann ein Initiations- oder "Start"-
Signal (normalerweise ATG, das in der resultierenden mRNA
zu AUG wird) und schließlich die Nukleotidcodons innerhalb
des Strukturgens selbst. Sogenannte Stop-Codons werden am
Ende des Strukturgens transkribiert, wonach die Polymerase
eine zusätzliche Sequenz von mRNA bilden kann, die, aufgrund
der Anwesenheit des Stopsignals, von den Ribosomen
nicht mehr translatiert wird. Ribosomen binden an die auf
der mRNA zur Verfügung gestellten Bindestelle, bei Bakterien
normalerweise wenn die mRNA gebildet wird, und stellen
selbst das codierte Polypeptid her, wobei sie am Translations-
Startsignal beginnen und an dem oben erwähnten
Stopsignal enden. Das gewünschte Produkt wird hergestellt,
wenn die Sequenzen, die für die Ribosomen-Bindestelle codieren,
im Hinblick auf das AUG-Initiatorcodon richtig
liegen und wenn alle übrigen Codons dem Initiatorcodon
in Phase folgen. Das resultierende Produkt kann erhalten
werden, indem man die Wirtszelle lysiert und das Produkt
durch geeignete Reinigungsschritte von anderen bakteriellen
Proteinen trennt.
Die durch die Anwendung der DNA-Rekombinations-Technik
exprimierten Polypeptide können vollkommen heterolog sein,
wie im Falle der direkten Expression des menschlichen
Wachstumshormons, können aber auch andererseits ein heterologes
Polypeptid enthalten, das zumindest mit einem Teil
der Aminosäuresequenz eines homologen Peptids verbunden ist,
wie im Fall der Herstellung von Zwischenprodukten für Somatostatin
und die Bestandteile des menschlichen Insulins.
Im letzteren Fall enthält beispielsweise das angefügte homologe
Polypeptid einen Teil der Aminosäuresequenz für β-Galactosidase.
In diesen Fällen ist das angestrebte bioaktive
Produkt durch das angefügte homologe Polypeptid so lange
bioinaktiv, bis dieses durch extrazelluläre Maßnahmen abgeschnitten
wird. Die ebengenannten "Fusionsproteine" können
so hergestellt werden, daß ein hochspezifisches Schneiden
des Vorläuferproteins von dem angestrebten Produkt
ermöglicht ist, beispielsweise durch die Einwirkung von
Bromcyan auf Methionin oder aber durch enzymatisches
Schneiden; siehe auch GB-PS Nr. 20 07 676 A.
Wenn die DNA-Rekombinations-Technologie ihr Versprechen
voll halten soll, müssen Systeme erdacht werden, die die
Expression der Geninsertionen optimieren, so daß das angestrebte
Polypeptid-Produkt in großen Mengen erhalten werden
kann. Die β-Lactamase- und Lactose-Promoter-Operator-
Systeme, die in der Vergangenheit üblicherweise verwendet
wurden, haben, obwohl sie gut zu verwenden sind, vom
Standpunkt der Menge her die Kapazität der Technologie
nicht voll ausgenutzt. Es besteht ein Bedürfnis für
einen bakteriellen Expressionsvektor, der fähig ist, das
gewünschte Polypeptid-Produkt in größerer Menge kontrolliert
zu exprimieren.
Tryptophan ist eine Aminosäure, die von Bakterien als Bestandteil
eines homologen Polypeptids verwendet wird und
in einem Biosyntheseweg hergestellt wird, der folgende
Schritte enthält: Chorisminsäure → Anthranilsäure →
Phosphoribosylanthranilsäure → CDRP (Enol-1-(o-Carboxyphenylamino)-
1-desoxy-D-Ribulose-5-Phosphat) → Indol-
3-Glycerinphosphat und letztlich Tryptophan selbst. Die
enzymatischen Reaktionen dieses Syntheseweges werden durch
Produkte des Tryptophan- oder "trp"-Operons katalysiert,
einem polycistronischen DNA-Segment, das unter der Kontrolle
des trp-Promoter-Operator-Systems transkribiert wird.
Die resultierende polycistronische mRNA codiert die sogenannte
trp-Führer- (im folgenden "Leader"- genannte)
Sequenz und dann, der Reihe nach, die Polypeptide, genannt
trpE, trpD, trpC, trpB und trpA. Diese Polypeptide katalysieren
und kontrollieren auf verschiedene Weise einzelne
Schritte des Syntheseweges ausgehend von der Chorisminsäure
bis zum Tryptophan.
Im Wildtyp E. coli ist das Tryptophan-Operon unter der
Kontrolle mindestens dreier, deutlich zu unterscheidender
Arten. Im ersten Fall, einer Promoter-Operator-Repression,
handelt das Tryptophan als Corepressor und bindet an seinen
Aporepressor, um einen aktiven Repressorkomplex zu bilden,
der seinerseits an den Operator bindet, wodurch das Tryptophan
seinen eigenen Syntheseweg vollkommen verschließt.
Zweitens bindet Tryptophan in einem Prozeß von Rückkopplungshemmung
an einen Komplex der trpE- und trpD-Polypeptide
und verhindert dadurch deren Beteiligung an dem Syntheseweg.
Letztlich wird eine Kontrolle ausgeübt durch
einen Prozeß, der als "Attenuierung" bekannt ist. Dieser
Vorgang läuft unter der Kontrolle einer "Attenuator-Region"
des Gens ab, einer Region, die innerhalb der trp-Leader-
Sequenz liegt. Die Attenuierung ist ein Vorgang, der
phänotypisch als eine Art Verdünnung des Tryptophans in
der Zelle in Erscheinung tritt (siehe dazu allgemein
G. F. Miozzari et al., J. Bacteriology 133, 1457 (1978);
The Operon 263-302, Cold Spring Harbor Laboratory (1978),
Miller und Reznikoff, eds.; F. Lee et al., Proc Nat'l
Acad Sci, USA 74, 4365 (1977) und K. Bertrand et al.,
J. Mol. Biol. 103, 319 (1976)). Das Ausmaß der Attenuierung
scheint durch die intrazelluläre Konzentration des Tryptophans
geregelt zu werden, und in Wildtyp E. coli beendet
der Attenuator die Expression in ungefähr neun von zehn
Fällen, möglicherweise durch die Bildung einer Sekundärstruktur,
eines "Terminationsrings" in der mRNA, welcher
die RNA-Polymerase dazu veranlaßt, sich verfrüht von der
DNA zu lösen.
Andere Autoren haben das trp-Operon dazu verwendet, um
bis zu einem gewissen Grad heterologe Polypeptide zu exprimieren.
Mit diesen Arbeiten hat man sich versuchsweise
mit Problemen der Repression und Attenuierung befaßt,
wobei Indolacrylsäure zugegeben wird, ein Induktor und
Analogon, das mit Tryptophan um das trp-Repressor-Molekül
konkurriert, wobei durch kompetitive Hemmung auf Derepression
abgezielt wurde. Gleichzeitig verringert der Induktor durch
Hemmung der enzymatischen Umwandlung von Indol zu Tryptophan
die Attenuierung und bewirkt dadurch, daß der Zelle Tryptophan
entzogen wird. Als Ergebnis davon lesen mehr Polymerasen
erfolgreich über den Attenuator hinweg. Vom Standpunkt der
vollständig durchgehenden Translation und der angestrebten
großen Mengen erscheint dieser Versuch jedoch problematisch,
da die Tryptophan-enthaltenden Proteinsequenzen
während der Synthese infolge des Mangels von nutzbarem
Tryptophan vorzeitig beendet werden. Tatsächlich ist bei
diesem Versuch eine wirkungsvolle Unterstützung der Attenuierung
vollständig von einer strengen Tryptophan-Aushungerung
abhängig.
In der Stammanmeldung der vorliegenden Anmeldung (P 31
11 405.9) werden neue, von Plasmiden abgeleitete
Expressionsvektoren für das Herstellen von heterologen
Polypeptid-Produkten in Bakterien bereitgestellt. Diese
Vektoren haben eine Sequenz von doppelsträngiger DNA,
die in Phase von einem ersten 5′- zu einem zweiten
3′-Ende des codierenden Strangs folgende
Bestandteile enthält: einen trp-Promoter-Operator,
Nukleotide, die für die trp-Leader-Ribosomen-Bindestelle
codieren und Nukleotide, die für die Translationsinitiatoren
zur Expression eines Strukturgens codieren, welches für
die Aminosäuresequenz eines heterologen Polypeptids codiert.
Diese beschriebene DNA-Sequenz enthält weder eine trp-
Attenuator-Region noch Nukleotide, die für die trpE-Ribosomen-
Bindestelle codieren. Statt dessen wird die trp-Leader-
Ribosomen-Bindestelle wirkungsvoll dazu verwendet, die
Expression von Information zu bewirken, die durch inserierte
Gene codiert wird.
Mit einem, einen trp-Promoter-Operator enthaltenden
Plasmid, das keinen Attenuator aufweist,
werden Zellen transformiert und in Gegenwart von zugegebenem
Tryptophan kultiviert. Mit der Verwendung von Tryptophan-
reichem Medium steht ausreichend Tryptophan zur Verfügung,
um im wesentlichen vollständig den trp-Promoter-
Operator durch trp/Repressorinteraktionen zu reprimieren,
so daß das Zellwachstum, ungehemmt durch vorzeitige Expression
großer Mengen von heterologen Polypeptiden,
fortschreiten kann, wobei dieses Polypeptid durch eine
Insertion codiert wird, die ansonsten unter der Kontrolle
des trp-Promoter-Operator-Systems steht. Sobald einerseits
die Kultur der Rekombinanten zu einer Dichte angewachsen
ist, die für eine industrielle Herstellung des Polypeptids
geeignet ist und andererseits das von außen zugeführte
Tryptophan entfernt wurde, läßt man die Zellen lediglich
mit dem Tryptophan wachsen, das sie selbst produzieren.
Das Ergebnis ist eine schwache Tryptophanlimitierung, wodurch
folglich der Syntheseweg dereprimiert wird und eine
überaus wirksame Expression der heterologen Insertion auftritt,
unbehindert durch Attenuierung, da die Attenuator-
Region aus dem System entfernt wurde. Auf diese Weise wird
den Zellen niemals ernstlich Tryptophan entzogen, und alle
Proteine, ob sie Tryptophan enthalten oder nicht, können
in einem ausreichenden Umfang produziert werden.
Die vorliegende Erfindung stellt nun ein Verfahren zur Verfügung, um
doppelsträngige DNA an jeder beliebigen, gewünschten Stelle
zu schneiden, auch wenn eine Erkennungsstelle für ein
Restriktionsenzym fehlt, eine Technik, die unter anderem
für das Herstellen eines trp-Operons brauchbar ist, welches
Attenuator-Deletionen aufweist, die sich von denjenigen
unterscheiden, die man früher durch Selektion von
Mutanten erhalten hat.
Die folgende Beschreibung sowie Zeichnungen erläutern
die Erfindung. Es zeigen dabei die
Fig. 1a, 1b und 2 ein Schema für die Herstellung
eines Plasmides unter Verwendung des erfindungsgemäßen
Verfahrens, wobei das Plasmid fähig ist, heterologe Gene
(wie in dem dargestellten Fall für Somatostatin),
fusioniert mit einem Teil des trpD-Polypeptides zu
exprimieren, wobei die Fusion später abgespalten werden
kann;
Fig. 3 das Ergebnis einer Polyacrylamid-Gel-Trennung
von Zellprotein, das homolog(trpE)-heterologe
Fusionsproteine zum Herstellen von Somatostatin
bzw. Thymosinα1, menschlichem Proinsulin und
der A- und B-Ketten des menschlichen Insulins
enthält.
In den Figuren werden aus Gründen der Klarheit der Darstellung
in den meisten Fällen nur der codierende Strang
des doppelsträngigen Plasmids und lineare DNA-Stränge
gezeichnet. Gene, die für die Resistenz gegen ein Antibiotikum
codieren, werden als apR (Ampicillin-Resistenz)
und tcR (Tetracyclin-Resistenz) bezeichnet. Die Bezeichnung
tcS bedeutet ein Gen für Tetracyclin-Resistenz, das nicht
unter der Kontrolle eines Promoter-Operator-Systems steht,
so daß Plasmide, die dieses Gen enthalten, dennoch Tetracyclin-
sensitiv sind. Die Bezeichnung apS bezeichnet eine
Ampicillin-Sensitivität, die durch die Deletion eines Teils
des Gens entstanden ist, das für Ampicillin-Sensitivität
codiert. Promotoren und Operatoren des Plasmids werden mit
p und o bezeichnet. Die Buchstaben A, T, G bzw. C bezeichnen
die Nukleotide, die die Basen Adenin, Thymin, Guanin
und Cytosin enthalten. Weitere Legenden der Figuren werden
aus dem Text ersichtlich.
Die verwendeten Restriktionsendonukleasen sind im
folgenden bezeichnet und mit ihrer zugehörigen
Erkennungssequenz sowie dem Schneidemuster (angezeigt
durch Pfeile) beschrieben.
Wo die Schnittpunkte auf den entsprechenden Strängen voneinander
entfernt sind, werden die geschnittenen Enden
"sticky", d. h. überstehend, und insofern in der Lage sein,
sich wieder neu zu verbinden, bzw. diese "sticky-ends"
können mit einer anderen, komplementären "sticky-end"-DNA
durch Watson-Crick-Basenpaarung (A mit T und G mit C)
korrekt miteinander verzapft werden. Einige Restriktionsenzyme,
beispielsweise die oben beschriebenen HpaI und
PvuII, hinterlassen nach dem Schnitt stumpfe Enden. Die
oben gezeigten Nukleotidsequenzen sind gemäß einer diesbezüglichen
Übereinkunft in der folgenden Weise dargestellt:
Der obere Strang ist der Protein-codierende Strang,
und man bewegt sich fortschreitend von links nach rechts
vom 5′- zum 3′-Ende dieses Strangs, d. h. in Richtung der
Transkription von einem proximalen zu einem distalen Punkt.
Schließlich bezeichnet, im Hinblick auf die Übereinkunft,
das Symbol "Δ" eine Deletion. Daher bezeichnet eine Benennung
eines Plasmids, wie beispielsweise "ΔEcoRI-XbaI", dasjenige
Plamids, aus welchem die Nukleotidsequenz zwischen
den EcoRI- und XbaI-Erkennungsstellen der entsprechenden
Restriktionsenzyme durch Verdauung durch diese Enzyme entfernt
wurde. Der Einfachheit halber wurden verschiedene
Deletionen durch Zahlen bezeichnet. Die Bezeichnung "Δ1"
bedeutet daher, beginnend vom ersten Basenpaar (Bp) der
EcoRI-Erkennungsstelle, die auf das Gen für Tetracyclin-
Resistenz in dem Elternplasmid pBR322 folgt, eine Deletion
vom Bp 1-30 (d. h. ΔEcoRI-HindIII) und daraus folgend ein
Ausschalten des Tetracyclin-Promoter-Operator-Systems.
Als "Δ2" wird eine Deletion der Bp 1-375 verstanden (d. h.
ΔEcoRI-BamHI) und infolgedessen ein Entfernen sowohl des
Tetracyclin-Promoter-Operators und der Strukturgene, die
für Tetracyclin-Resistenz codieren. Die Bezeichnung "Δ3"
bedeutet eine Deletion der Bp 3611-4359 (d. h. ΔPstI-EcoRI)
und die Eliminierung der Ampicillin-Resistenz. "Δ4" wird
verwendet, um das Entfernen der Bp ∼900-∼1500 vom trp-
Operon-Fragment 5 (Fig. 1) zu bezeichnen, in dem die Strukturgene
für das trpD-Polypeptid eliminiert sind.
Die trp-Leader-Sequenz ist aus den Basenpaaren (Bp)
1-162 aufgebaut, angefangen vom Startpunkt für die trp-
mRNA. Ein auf 14 Aminosäuren geschätztes trp-Leader-Polypeptid
wird durch die Bp 27-71 codiert, die den ATG-
Nukleotiden folgen, die für das Translations-Startsignal
codieren. Die trp-Attenuator-Region enthält aufeinanderfolgend
GC-reiche und AT-reiche Sequenzen, die zwischen
den Bp 114 und 156 liegen. Die Attenuierung wird offensichtlich
durch mRNA-Nukleotide bewirkt, die durch die
Bp ∼134-141 der Leader-Sequenz codiert werden. Um heterologe
Polypeptide unter dem Einfluß der trp-Leader-Ribosomen-
Bindestelle zu exprimieren und gleichzeitig die Attenuierung
zu verhindern, müssen die folgenden Kriterien beachtet werden:
- 1. Die Basenpaare 134-141 oder noch mehr über das Bp 141 hinaus müssen deletiert werden;
- 2. das ATG-Codon des inserierten Gens muß, wie bekannt, in der richtigen Lage zu einer Ribosomen-Bindestelle ausgerichtet sein (siehe z. B. J. A. Steitz "Genetic signals and nucleotide sequences in messenger RNA" in Biological Regulation and Control (ed. R. Goldberger) Plenum Press, N. Y. (1978));
- 3. wo ein homolog-heterologes Fusionsprotein produziert werden soll, sollte das Translations-Startsignal einer homologen Polypeptidsequenz verfügbar bleiben, und die Codons für den homologen Teil des Fusionsproteins müssen in Phase ohne dazwischenliegende Translations-Stopsignale inseriert werden.
Wenn beispielsweise alle Basenpaare innerhalb der Leader-
Sequenz distal vom Bp 70 deletiert werden, wird die
Attenuator-Region entfernt, die ATG-Sequenz, die für das
Translations-Startsignal codiert, verbleibt und der dazwischenliegende
Translations-Stop, codiert durch TCA (Bp
69-71), wird entfernt durch Eliminieren von A und die
darauffolgenden Nukleotide. Eine derartige Deletion hätte
als Ergebnis die Expression eines Fusionsproteins, das mit
dem Leader-Polypeptid beginnt und mit demjenigen Polypeptid
endet, das durch irgendeine heterologe Insertion codiert
wird. Von diesen Polypeptiden wird eine distale Region
eines an die Leader-Sequenz anschließenden trp-Operon-
Polypeptids eingeschlossen, die durch das Ausmaß der Deletion
in der 3′-Richtung bestimmt wird. Eine in das Gen E
hineinreichende Deletion würde daher zur Expression eines
homologen Vorläufers führen, der die L-Sequenz und die
distale Region des Gens E (jenseits des Endpunkts der
Deletion) enthält, verbunden mit der Sequenz, die durch
irgendeine folgende Insertion codiert wird, usw.
Zwei Plasmide, aus denen die Attenuator-
Region entfernt wurde, sind die Plasmide pGM1 und pGM3
(G. F. Miozzari et al., J. Bacteriology 133, 1457 (1978)).
Diese Plasmide tragen die Deletionen trp ΔLE 1413 und
trp ΔLE 1417 und exprimieren (unter der Kontrolle des trp-
Promoter-Operators) ein Polypeptid, das ungefähr die ersten
sechs Aminosäuren des trp-Leaders und distale Regionen des
E-Polypeptids enthält. In dem Fall
des Plasmids pGM1 wird lediglich etwa das letzte Drittel
des E-Polypeptids exprimiert, wohingegen pGM3 nahezu die
gesamte distale Hälfte des E-Polypeptid-Codons exprimiert.
Der Stamm E. coli K-12 W3110 tna 2 - trp -Δ102, der pGM1 enthält,
ist bei der American Type Culture Collection (ATCC)
unter der Nummer 31622 hinterlegt. Das Plasmid pGM1 kann
auf herkömmliche Weise aus dem genannten Stamm zur Verwendung
in den unten beschriebenen Verfahren entfernt werden.
Erfindungsgemäß werden nun Deletionen durch das
spezifische Schneiden von doppelsträngiger DNA an jeder
gewünschten Stelle eingeführt. Diese Schneidetechnik
wird im folgenden durch ein Beispiel erläutert.
Erfindungsgemäß wird doppelsträngige DNA in einem den
beabsichtigten Schneidepunkt umgebenden Bereich durch
Reaktion mit λ-Exonuklease in einzelsträngige DNA umgewandelt.
Daraufhin wird ein synthetischer oder anderer
Einzelstrang-DNA-Primer an das vorher gebildete Einzelstrangstück
durch Watson-Crick-Basenpaarung hybridisiert.
Dabei muß sichergestellt sein, daß das 5′-Ende der Primer-
Sequenz exakt gegenüber demjenigen Nukleotid auf dem
ersten Strang endet, das genau vor dem beabsichtigten
Schneidepunkt liegt. Als nächstes wird der Primer durch
Reaktion mit DNA-Polymerase in 3′-Richtung verlängert,
wodurch derjenige Teil der ursprünglich doppelsträngigen
DNA, der vor dem beabsichtigten Schnittpunkt liegt, wieder
hergestellt wird, der im ersten Schritt verlorengegangen
war. Gleichzeitig oder danach wird derjenige Teil
des ersten Stranges, der jenseits des beabsichtigten
Schnittpunkts liegt, wegverdaut. Das nachfolgende Schema
veranschaulicht diesen Vorgang noch einmal, wobei "v"
den beabsichtigten Schnittpunkt kennzeichnet:
In einer besonders bevorzugten Ausführungsform werden
die Schritte (d) und (e) gleichzeitig ausgeführt, wobei
eine Polymerase verwendet wird, die gleichzeitig die vorstehenden
Einzelstrangenden in der 3′ → 5′-Richtung verdaut
und den Primer (in Anwesenheit von dATP, dGTP, dTTP
und dCTP) in der 5′ → 3′-Richtung verlängert. Besonders
geeignet für diese Zwecke ist die Klenow-Polymerase I,
d. h. dasjenige Fragment, das durch proteolytisches
Spalten der DNA-Polymerase I erhalten wird und die
5′ → 3′-Polymerisierungsaktivität sowie die 3′ → 5′-
Exonukleaseaktivität des Elternenzyms aufweist, dagegen
seine 5′ → 3′-Exonukleaseaktivität verloren hat (A. Kornberg,
DNA Synthesis, 98, W. H. Freeman und Co., SFO (1974)).
Bei Verwendung des erfindungsgemäßen Verfahrens können
Attenuator-Deletionen in einem ein trp-Operon enthaltenden
Plasmid auf jede nur gewünschte Weise eingeführt werden.
Das Plasmid wurde vorher in die lineare Form überführt,
beispielsweise durch einen Schnitt an einer
Restriktionserkennungsstelle stromabwärts von dem beabsichtigten
Schnittpunkt, an dem das Molekül stumpfendig
sein soll (siehe Bereich "v" im oben gezeigten Schema).
Nach dem Deletieren der Attenuator-Region wird das Plasmid
wieder in die Ringform gebracht. Dies kann beispielsweise
durch Verbinden der stumpfen Enden oder auch durch andere
Methoden geschehen, die dem Fachmann bekannt sind.
Die DNA-Rekombinations-Experimente, die in dem folgenden
Beispiel beschrieben werden, wurden bei
Genentech Inc. in Übereinstimmung mit den Richtlinien des
National Institutes of Health (NIH) bezüglich der Forschung
mit rekombinanter DNA durchgeführt.
Hinsichtlich der verwendeten Bakterienstämme und
Plasmide sowie den Kultivierungsbedingungen
einschließlich Repression und Expression sowie
für alle weiteren Details der Durchführung der
Experimente wird auf die korrespondierende
Stammanmeldung P 31 11 405.9 verwiesen.
Herstellen eines Plasmids, das das Tryptophan-Operon
enthält, fähig zur Expression eines spezifisch spaltbaren
Fusionsproteins, das aus sechs Aminosäuren des
trp-Leader-Peptids, dem letzten Drittel des trpE-Polypeptids
(genannt LE′) sowie einem heterologen Strukturgen-
Produkt besteht.
Die Strategie für das Herstellen eines Plasmids, das
ein LE′-Fusionsprotein exprimiert, beinhaltet folgende
Schritte:
- a) Bereitstellen eines Genfragments, das aus Codons für die distale Region des LE′-Polypeptids besteht und überstehende Enden einer BglII- Erkennungsstelle am 5′-Ende und einer EcoRI- Erkennungsstelle am 3′-Ende des codierenden Strangs aufweist;
- b) Eliminieren der Codons von der distalen Region des LE′-Genfragments und derjenigen für die trpD- Gene aus dem Plasmid Som7Δ2 und Inserieren des in Stufe 1 gebildeten Fragments, wodurch die LE′- Codonsequenz unmittelbar stromaufwärts von der für das heterologe Gen für Somatostatin codierenden Codonsequenz wiederhergestellt wird.
Fig. 1a zeigt die HindIII-Verdauung des Plasmids
pSom7Δ2 (siehe Stammanmeldung P 31 11 405.9), gefolgt von einer λ-Exonuklease-Verdauung
(einer 5′ → 3′-Exonuklease) unter Bedingungen, die
so gewählt werden, daß jenseits der BglII-Restriktionserkennungsstelle
innerhalb der LE′ codierenden Region
verdaut wird. 20 µg des HindIII-verdauten pSom7Δ2 werden
in Puffer gelöst (20 mM Glycinpuffer pH 9,6,
1 mM MgCl₂, 1 mM β-Mercaptoäthanol). Das erhaltene
Gemisch wird mit 5 Einheiten λ-Exonuklease 60 Minuten
lang bei Raumtemperatur behandelt. Die so erhaltene
Reaktionsmischung wird dann Phenol-extrahiert,
Chloroform-extrahiert und mit Äthanol gefällt.
Um letztlich einen EcoRI-Rest am distalen Ende des
LE′-Genfragments herzustellen, wird mittels der bewährten
Phosphotriester-Methode (R. Crea et al., Proc
Nat'l Acad Sci USA 75, 5765 (1978)) ein Primer
³²pCCTGTGCATGAT synthetisiert und an ein Einzelstrangende
des LE′-Genfragments hybridisiert, das durch
λ-Exonuklease-Verdauung erhalten wurde. Die Hybridisierung
wird im folgenden beschrieben.
20 µg des λ-Exonuklease-behandelten HindIII-Verdauungsprodukts
des Plasmids pSom7Δ2 wird in 20 µl H₂O gelöst
und mit 6 µl einer Lösung vereint, die etwa 80 Picomole
der 5′-phosphorylierten Oligonukleotide, wie oben beschrieben,
enthält. Das synthetische Fragment wird an
das 3′-Ende der LE′ codierenden Sequenz hybridisiert,
und der verbleibende Einzelstrangteil des LE′-Fragments
wird durch das Klenow-Polymerase-I-Verfahren, wie oben
beschrieben, aufgefüllt, wobei dATP, dTTP, dGTP und
dCTP verwendet werden.
Die Reaktionsmischung wird auf 50°C erhitzt und langsam
auf 10°C abgekühlt, wonach 4 µl Klenow-Enzym zugegeben
werden. Nach 15minütiger Inkubation bei Raumtemperatur,
gefolgt von 30minütiger Inkubation bei
37°C, wird die Reaktion durch Zugabe von 5 µl 0,25
molares EDTA gestoppt. Die Reaktionsmischung wird
Phenol-extrahiert. Chloroform-extrahiert und mit
Äthanol gefällt. Anschließend wird die DNA mit dem
Restriktionsenzym BglII geschnitten. Die Fragmente
werden durch PAGE getrennt. Ein aus dem Gel erhaltenes
Autoradiogramm zeigt ein ³²P-markiertes Fragment der
erwarteten Länge von annähernd 470 Bp, das durch Elektroeluierung
rückgewonnen wird. Wie bereits hervorgehoben,
hat das Fragment LE′(d) ein BglII- und ein stumpfes
Ende, das mit dem Anfang des Primers übereinstimmt.
Das Plasmid pThα1, das im Abschnitt I. (C) der Stammanmeldung
(P 31 11 405.9) beschrieben wurde, enthält ein Strukturgen für
Thymosinα1, das an seinem 5′-Ende des codierenden
Strangs in eine EcoRI-Erkennungsstelle und an seinem
3′-Ende in eine BamHI-Erkennungsstelle geclont ist.
Wie in Fig. 1b gezeigt, enthält das Gen für Thymosin
eine zusätzliche BglII-Erkennungsstelle.
Das Plasmid pThα1 enthält weiterhin ein Gen für
Ampicillin-Resistenz. Um ein Plasmid herzustellen,
das in der Lage ist, das, wie oben geschildert, hergestellte
LE′(d)-Fragment aufzunehmen, wird das Plasmid
pThα1 mit EcoRI verdaut, gefolgt von einer Klenow-
Polymerase-I-Reaktion mit dTTP und dATP, um die EcoRI-
Reste stumpfendig zu machen. Durch BglII-Verdauung
des so erhaltenen Produkts wird ein lineares DNA-
Fragment 33 (Fig. 1b) hergestellt, das die Gene für
Ampicillin-Resistenz und, jeweils an den entgegengesetzten
Enden, einen überstehenden BglII-Rest und ein
stumpfes Ende enthält. Das so erhaltene Produkt kann
durch Reaktion mit dem LE′(d)-Fragment, das ein überstehendes
BglII-Ende und ein stumpfes Ende enthält,
in der Gegenwart von T₄-Ligase rezirkularisiert werden,
um das Plasmid pTrp24 (Fig. 1b) zu bilden. Während
dieses Vorgangs wird eine EcoRI-Erkennungsstelle
an der Position wiederhergestellt, wo die Verknüpfung
der stumpfen Enden erfolgte.
Im weiteren Verlauf wird, wie in Fig. 2 gezeigt,
nacheinander das Plasmid pTrp24 mit BglII und EcoRI
verdaut, danach PAGE behandelt und elektroeluiert,
wodurch ein Fragment entsteht, das die Codons für das
LE′(d)-Polypeptid mit einem überstehenden BglII-Ende
und einem überstehenden EcoRI-Ende, angrenzend an
seinen 3′-Terminus des codierenden Strangs aufweist.
Das LE′(d)-Fragment 38 (Fig. 2) kann in die BglII-
Erkennungsstelle des Plasmids pSom7Δ2 geclont werden,
um ein LE′-Polypeptid/Somatostatin-Fusionsprotein zu
bilden, das unter der Kontrolle des Tryptophan-Promoter-
Operators exprimiert wird, wie in Fig. 2 gezeigt.
Ein solches Verfahren erfordert erstens eine partielle
EcoRI-Verdauung des Plasmids pSom7Δ2, um die EcoRI-
Erkennungsstelle distal vom Tryptophan-Promoter-
Operator zu schneiden, wie in Fig. 2 gezeigt, und
zweitens eine korrekte Wahl der Primer-Sequenz
(Fig. 9), um den korrekten Codon-Leseraster zu erhalten
und eine EcoRI-Schneidestelle wiederherzustellen.
Dazu werden 16 µg des Plasmids pSom7Δ2 in 200 µl
Puffer, bestehend aus 20 mM Tris, pH 7,5, 5 mM MgCl₂,
0,02% NP40 Detergens, 100 mM NaCl verdünnt und mit
0,5 Einheiten EcoRI behandelt. Nach 15minütiger
Behandlungszeit bei 37°C wird die Reaktionsmischung
Phenol-extrahiert, Chloroform-extrahiert und mit
Äthanol gefällt und anschließend mit BlgII verdaut.
Das größere, so erhaltene Fragment 36 (Fig. 2) wird
durch die PAGE-Behandlung isoliert, gefolgt von Elektroeluierung.
Dieses Fragment enthält die Codons LE′(p)
für das proximale Ende des LE′-Polypeptids, d. h.,
diejenigen Codons, die stromaufwärts von der BglII-
Erkennungsstelle liegen. Das Fragment 36 (Fig. 2)
wird als nächstes an das Fragment 38 (Fig. 2) in Anwesenheit
von T₄-Ligase verknüpft, um das Plasmid
pSom7Δ2Δ4 zu bilden, das, nach Transformation in den
Stamm E. coli 294, wirksam
unter die Kontrolle des Tryptophan-Promoter-Operators
ein Fusionsprotein produziert, das aus dem vollständig rekonstituierten
LE′-Polypeptid und Somatostatin besteht.
Das Fusionsprotein, von dem das Somatostatin dank der
Anwesenheit eines Methionins am 5′-Ende der Somatostatinsequenz
spezifisch gespalten werden kann, wird durch
SDS-Polyacrylamid-Gel-Elektrophorese, wie bereits beschrieben,
segregiert. Das Fusionsprotein-Produkt
stellt die am deutlichsten sichtbare Bande in der
Spalte 6 der Fig. 3 dar.
Claims (3)
1. Verfahren zum Schneiden doppelsträngiger DNA, dadurch
gekennzeichnet, daß zum Schneiden an
jeder beliebigen Stelle
- a) die doppelsträngige DNA in einem die zu schneidende Stelle umgebenden Bereich in einzelsträngige DNA umgewandelt wird;
- b) an die in Schritt a) gebildete Einzelstrangregion ein komplementäres Primerstück von einzelsträngiger DNA hybridisiert wird, wobei das 5′-Ende des Primers gegenüber demjenigen Nukleotid liegt, das an die beabsichtigte Schnittstelle angrenzt;
- c) derjenige Teil des zweiten, in Schritt a) eliminierten Strangs wiederhergestellt wird, der in 3′- Richtung des Primers liegt, und zwar durch eine Reaktion mit DNA-Polymerase in Anwesenheit von Adenin-, Thymin-, Guanin- und Cytosin-enthaltenden Desoxynukleotidtriphosphaten; und
- d) das verbleibende DNA-Einzelstrangstück, das über die beabsichtigte Schnittstelle vorsteht, verdaut wird.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet,
daß die Schritte c) und d) gleichzeitig
durchgeführt werden, durch Reaktion mit einer
DNA-Polymerase, die in 5′ → 3′-Richtung polymerisiert,
die Exonukleasewirkung in 3′ → 5′-Richtung
aufweist, jedoch keine Nukleasewirkung in 5′ → 3′-
Richtung zeigt.
3. Verfahren nach Anspruch 2, dadurch gekennzeichnet,
daß die Polymerase die Klenow-
Polymerase I ist.
