Description

この実施例においては、HIV-1トランス活性化タンパク質(TAT)のタンパク質形質導入ドメインおよびMYCを含むPTD-MYC融合ポリペプチドの、インビボでメラノーマ細胞に対する免疫応答をモジュレートする能力を調査した。具体的には、メラノーマ保持マウスに由来し、TAT-MYCによって処理されたリンパ系細胞の、メラノーマ腫瘍を保有しているマウスを処置する能力を研究した。これらの研究の目的は、メラノーマ保持マウスに由来し、TAT-MYCリンパ球を生成するため、TAT-MYCによって処理された免疫細胞が、メラノーマ保持マウスへ移植されると、メラノーマ腫瘍のための有効な処置になるか否かを判定することであった。
C57BL/6Jは、最も広く使用されている近交系であり、そのゲノムが配列決定された最初の系統である。この系統は、多くの腫瘍に対して難治性であるが、大部分の変異の最大発現を許容するバックグラウンドである。C57BL/6Jマウスは、聴原発作に対して抵抗性であり、比較的低い骨密度を有し、年齢に関連した難聴を発症する。それらは、食事によって誘導される肥満、2型糖尿病、およびアテローム性動脈硬化症にもかかりやすい。この系統に由来するマクロファージは、炭疽菌致死毒素の効果に対して抵抗性である。
体重がおよそ25gであり、メラノーマ腫瘍を保有している15匹のC57BL/6マウス(Jackson Laboratory Stock#000664)を生成し、5匹の3コホート、未処置対照としての1匹の1コホート、腫瘍保持マウスに由来し、対照TAT融合タンパク質によって処理されたリンパ系細胞によって処置される1コホート、およびTAT-MYCリンパ球によって処置される1コホートに分割した。
植え込みのためのB16-F10メラノーマ細胞(ATCC CRL 6475、マウス皮膚メラノーマ)を、D10培地(DMEM、10%FBS、Pen/Strep(10,000単位/ml)(Gibco Cat#15140);L-グルタミン(200mM)(Gibco Cat#25030);MEM非必須アミノ酸(Gibco Cat#11140))において培養した。
