Abstract
1. Полипептид, содержащий Fc вариант Fc области человеческого IgG дикого типа, указанный Fc вариант содержит аминокислотную замену в положении Pro329 и по меньшей мере еще одну аминокислотную замену, где остатки пронумерованы в соответствии с системой EU index по Кабат, и где указанный полипептид проявляет сниженную аффинность к FcγRIIIA и/или FcγRIIA и/или FcγRI человека по сравнению с полипептидом, содержащим Fc область дикого типа IgG, и где ADCC, индуцированная указанным полипептидом, снижена по меньшей мере на 20% от ADCC, индуцированной полипептидом, содержащим Fc область человеческого IgG дикого типа.2. Полипептид согласно п.1, где Pro329 Fc области дикого типа человеческого происхождения замещен на глицин или аргинин или аминокислотный остаток достаточно большого размера, разрушающий «пролиновый сэндвич» на границе между Fc/Fcγ рецептором.3. Полипептид согласно п.1, где указанная по меньшей мере еще одна аминокислотная замена представляет собой S228P, Е233Р, L234A, L235A, L235E, N297A, N297D или P331S.4. Полипептид согласно п.1, где указанная по меньшей мере еще одна аминокислотная замена представляет собой L234A и L235A Fc области IgG1 человека или S228P и L235E Fc области IgG4 человека.5. Полипептид согласно п.1, где аффинность по меньшей мере к еще одному рецептору группы, состоящей из рецепторов FcγI, FcγIIA и C1q человека, снижена по сравнению с полипептидом, содержащим Fc область человеческого IgG дикого типа.6. Полипептид согласно п.1, где полипептид содержит Fc области IgG1 или IgG4 человека.7. Полипептид согласно п.1, где полипептид представляет собой антитело или белок, гибридизованный с Fc.8. Claims (19)
1. Полипептид, содержащий Fc вариант Fc области человеческого IgG дикого типа, указанный Fc вариант содержит аминокислотную замену в положении Pro329 и по меньшей мере еще одну аминокислотную замену, где остатки пронумерованы в соответствии с системой EU index по Кабат, и где указанный полипептид проявляет сниженную аффинность к FcγRIIIA и/или FcγRIIA и/или FcγRI человека по сравнению с полипептидом, содержащим Fc область дикого типа IgG, и где ADCC, индуцированная указанным полипептидом, снижена по меньшей мере на 20% от ADCC, индуцированной полипептидом, содержащим Fc область человеческого IgG дикого типа.
2. Полипептид согласно п.1, где Pro329 Fc области дикого типа человеческого происхождения замещен на глицин или аргинин или аминокислотный остаток достаточно большого размера, разрушающий «пролиновый сэндвич» на границе между Fc/Fcγ рецептором.
3. Полипептид согласно п.1, где указанная по меньшей мере еще одна аминокислотная замена представляет собой S228P, Е233Р, L234A, L235A, L235E, N297A, N297D или P331S.
4. Полипептид согласно п.1, где указанная по меньшей мере еще одна аминокислотная замена представляет собой L234A и L235A Fc области IgG1 человека или S228P и L235E Fc области IgG4 человека.
5. Полипептид согласно п.1, где аффинность по меньшей мере к еще одному рецептору группы, состоящей из рецепторов FcγI, FcγIIA и C1q человека, снижена по сравнению с полипептидом, содержащим Fc область человеческого IgG дикого типа.
6. Полипептид согласно п.1, где полипептид содержит Fc области IgG1 или IgG4 человека.
7. Полипептид согласно п.1, где полипептид представляет собой антитело или белок, гибридизованный с Fc.
8. Полипептид согласно п.1, где агрегация тромбоцитов, индуцированная полипептидом, снижена по сравнению с агрегацией трмбоцитов, индуцированной полипептидом, содержащим Fc область человеческого IgG дикого типа.
9. Полипептид согласно п.1, где CDC, индуцированная полипептидом, значительно снижена по сравнению с CDC, индуцированной полипептидом, содержащим Fc область человеческого IgG дикого типа.
10. Полипептид согласно п.1 для применения в качестве лекарства.
11. Полипептид согласно п.1, где полипептид представляет собой антитело к CD9, характеризующееся тем, что полипептид, содержащий Fc область дикого типа, содержит вариабельный участок тяжелой цепи по SEQ ID NO:9 и вариабельный участок легкой цепи по SEQ ID NO:8.
12. Полипептид согласно любому из пп.1-11 для применения в лечении заболевания, при котором желательно, чтобы эффекторная функция полипептида была значительно снижена по сравнению с эффекторной функцией, индуцируемой полипептидом, содержащим Fc область человеческого IgG дикого типа.
13. Применение полипептида согласно любому из пп.1-12 в изготовлении лекарства для лечения заболевания, при котором желательно, чтобы эффекторная функция полипептида, содержащего Fc вариант Fc области человеческого IgG дикого типа, была значительно снижена по сравнению с эффекторной функцией, индуцируемой полипептидом, содержащим Fc область человеческого IgG дикого типа.
14. Способ лечения индивида, имеющего заболевание, при котором желательно, чтобы эффекторная функция полипептида, содержащего Fc вариант Fc области человеческого IgG дикого типа, была значительно снижена по сравнению с эффекторной функцией, индуцируемой полипептидом, содержащим Fc область человеческого полипептида дикого типа, включающий введение индивиду эффективного количества полипептида согласно любому из пп.1-12.
15. Применение полипептида, содержащего Fc вариант Fc области человеческого IgG дикого типа, где указанный полипептид имеет замену Pro329 Fc области человеческого IgG на глицин, где нумерация остатков соответствует системе EU index по Кабат, где указанный полипептид проявляет пониженную аффинность к FcγRIIIA и FcγRIIA человека, приводящую к понижению ADCC по меньшей мере на 20% от ADCC, индуцируемой полипептидом, содержащим Fc область человеческого IgG дикого типа, и/или к понижению ADCP.
16. Применение полипептида согласно п.15, где Fc вариант содержит по меньшей мере еще две аминокислотные замены L234A и L235A Fc области IgG1 человека или S228P и L235E Fc области IgG4 человека.
17. Применение согласно пп.15-16, где агрегация тромбоцитов, индуцируемая полипептидом, снижена по сравнению с агрегацией тромбоцитов, индуцируемой полипептидом, содержащим Fc область дикого типа человеческого происхождения, где полипептид представляет собой антитело, активирующее тромбоциты.
18. Способ лечения индивида, имеющего заболевание, полипептидом, причем указанный полипептид имеет замену Pro329 Fc области IgG человека на глицин, где остатки пронумерованы в соответствии с системой EU index по Кабат, где указанный полипептид характеризуется значительно пониженным связыванием с FcγRIIIA и FcγRIIA по сравнению с полипептидом, содержащим Fc область человеческого IgG дикого типа, включающий введение индивиду эффективного количества указанного полипептида.
19. Способ по п.18, где указанный полипептид содержит по меньшей мере еще две аминокислотные замены L234A и L235A Fc области IgG1 человека или S228P и L235E Fc области IgG4 человека.
