Abstract
1. Биспецифическое антитело, содержащее по меньшей мере два Fab-фрагмента, в которомпервый Fab-фрагмент содержит по меньшей мере один антигенсвязывающий центр, специфический в отношении первого антигена; и второй Fab-фрагмент содержит по меньшей мере один антигенсвязывающий центр, специфический в отношении второго антигена, где либо вариабельные области, либо константные области тяжелой и легкой цепи второго Fab заменены друг на друга; и два Fab-фрагмента соединены друг с другом двумя пептидными линкерами, и где биспецифическое антитело лишено Fc-домена.2. Биспецифическое антитело по п. 1, дополнительно содержащее третий Fab-фрагмент.3. Биспецифическое антитело по п. 2, в котором третий Fab-фрагмент содержит по меньшей мере один антигенсвязывающий центр, специфический в отношении первого или второго антигена.4. Биспецифическое антитело по п. 3, в котором третий Fab-фрагмент содержит по меньшей мере один антигенсвязывающий центр, специфический в отношении первого антигена.5. Биспецифическое антитело по одному из п.п. 2-4, в котором третий Fab-фрагмент соединен с первым Fab-фрагментом с помощью одного или двух пептидных линкеров.6. Биспецифическое антитело по одному из п.п. 2-4, в котором третий Fab-фрагмент соединен со вторым Fab-фрагментом с помощью одного или двух пептидных линкеров.7. Биспецифическое антитело по одному из п.п. 1-4, в котором первый Fab-фрагмент содержит по меньшей мере один антигенсвязывающий центр, специфический в отношении опухолевого антигена; а второй Fab-фрагмент содержит по меньшей мере один антигенсвязывающий центр, специфический в отношении активирующего Т-клетку антигена.8. Биспецифическое антитело по п. 7, где активирующий Т клетку антиген представляет собой CD3-антиген
Claims (17)
1. Биспецифическое антитело, содержащее по меньшей мере два Fab-фрагмента, в котором
первый Fab-фрагмент содержит по меньшей мере один антигенсвязывающий центр, специфический в отношении первого антигена; и второй Fab-фрагмент содержит по меньшей мере один антигенсвязывающий центр, специфический в отношении второго антигена, где либо вариабельные области, либо константные области тяжелой и легкой цепи второго Fab заменены друг на друга; и два Fab-фрагмента соединены друг с другом двумя пептидными линкерами, и где биспецифическое антитело лишено Fc-домена.
2. Биспецифическое антитело по п. 1, дополнительно содержащее третий Fab-фрагмент.
3. Биспецифическое антитело по п. 2, в котором третий Fab-фрагмент содержит по меньшей мере один антигенсвязывающий центр, специфический в отношении первого или второго антигена.
4. Биспецифическое антитело по п. 3, в котором третий Fab-фрагмент содержит по меньшей мере один антигенсвязывающий центр, специфический в отношении первого антигена.
5. Биспецифическое антитело по одному из п.п. 2-4, в котором третий Fab-фрагмент соединен с первым Fab-фрагментом с помощью одного или двух пептидных линкеров.
6. Биспецифическое антитело по одному из п.п. 2-4, в котором третий Fab-фрагмент соединен со вторым Fab-фрагментом с помощью одного или двух пептидных линкеров.
7. Биспецифическое антитело по одному из п.п. 1-4, в котором первый Fab-фрагмент содержит по меньшей мере один антигенсвязывающий центр, специфический в отношении опухолевого антигена; а второй Fab-фрагмент содержит по меньшей мере один антигенсвязывающий центр, специфический в отношении активирующего Т-клетку антигена.
8. Биспецифическое антитело по п. 7, где активирующий Т клетку антиген представляет собой CD3-антиген корецептора Т-клеточного рецептора (CD3)
9. Биспецифическое антитело по п. 8, где опухолевый антиген выбран из группы, включающей ассоциированный с меланомой хондроитинсульфат-протеогликан (MCSP), рецептор эпидермального фактора роста (EGFR), карциноэмбриональный антиген (СЕА), фибробласт-активирующий белок (FAP) и CD33.
10. Биспецифическое антитело по п. 9, где опухолевый антиген представляет собой MCSP.
11. Фармацевтическая композиция, содержащая биспецифическое антитело по п.п. 1-10.
12. Биспецифическое антитело по п.п. 1-4, 8, 9 и 10, предназначенное для лечения рака.
13. Биспецифическое антитело по п.п. 1-4, 8, 9 и 10, предназначенное для применения в качестве лекарственного средства.
14. Применение биспецифического антитела по п.п. 1-10 для приготовления лекарственного средства.
15. Применение по п. 14, в котором лекарственное средство предназначено для лечения рака.
16. Прокариотическая или эукариотическая клетка-хозяин, содержащая векторы, которые содержат молекулы нуклеиновых кислот, кодирующие легкие цепи и тяжелые цепи биспецифического антитела по одному из п.п. 1-10.
17. Способ получения антитела, предусматривающий культивирование клетки-хозяина по п. 16 таким образом, чтобы получать антитело.
