Claims (29)
1. Мультиспецифическое антитело, содержащее:
а) две модифицированные тяжелые цепи, при этом каждая тяжелая цепь содержит в направлении от С-конца к N-концу константные домены тяжелой цепи 3, 2 и 1 (СН3, СН2 и СН1) и вариабельный домен легкой цепи (VL), где вариабельный домен легкой цепи (VL) представляет собой вариабельный домен одинаковой легкой цепи;
б) одну модифицированную легкую цепь, при этом модифицированная легкая цепь содержит в направлении от С-конца к N-концу константный домен легкой цепи (CL) изотипа каппа и вариабельный домен тяжелой цепи (VH1), происходящей из антитела, специфически связывающегося с первым антигеном; и
в) одну модифицированную легкую цепь, при этом модифицированная легкая цепь содержит в направлении от С-конца к N-концу константный домен легкой цепи изотипа лямбда и вариабельный домен тяжелой цепи (VH2), происходящей из антитела, специфически связывающегося со вторым антигеном;
при этом вариабельные домены VH1 и VL образуют первый антигенсвязывающий сайт, который специфически связывается с первым антигеном, и вариабельные домены VH2 и VL образуют второй антигенсвязывающий сайт, который специфически связывается со вторым антигеном.
2. Мультиспецифическое антитело по п. 1, где антитело является двухвалентным биспецифическим антителом.
3. Мультиспецифическое антитело по п. 1, где антитело относится к классу IgG.
4. Мультиспецифическое антитело по п. 3, где антитело относится к подклассу IgG1 или IgG4.
5. Нуклеиновая кислота, кодирующая мультиспецифическое антитело по любому из пп. 1-4.
6. Экспрессирующий вектор, содержащий нуклеиновую кислоту по п. 5, способный экспрессировать указанную нуклеиновую кислоту в прокариотической или эукариотической клетке-хозяине.
7. Прокариотическая или эукариотическая клетка-хозяин, содержащая экспрессиирующий вектор по п. 6.
8. Способ получения мультиспецифического антитела по любому из пп. 1-4, включающий следующие стадии:
а) трансформацию клетки-хозяина экспрессирующими векторами, содержащими молекулы нуклеиновой кислоты, кодирующей мультиспецифическое антитело по любому из пп. 1-4;
б) культивирование клетки-хозяина в условиях, обеспечивающих синтез указанной молекулы мультиспецифического антитела; и
в) выделение указанной молекулы мультиспецифического антитела из указанной культуры.
9. Фармацевтическая композиция, содержащая мультиспецифическое антитело по любому из пп. 1-4 и по меньшей мере один фармацевтически приемлемый эксципиент.
10. Мультиспецифическое антитело по любому из пп. 1-4 для применения в качестве лекарственного средства.
11. Применение мультиспецифического антитела по любому из пп. 1-4 для изготовления лекарственного средства.
12. Способ лечения пациента, нуждающегося в терапии, характеризующийся введением пациенту терапевтически эффективного количества мультиспецифического антитела по любому из пп. 1-4.
13. Способ получения мультиспецифического антитела на основе первого антитела, специфически связывающегося с первым антигеном, и второго антитела, специфически связывающегося со вторым антигеном, где первое антитело и второе антитело содержат одинаковую легкую цепь, включающий следующие стадии:
а) модификацию легкой цепи, происходящей из указанного первого антитела, для получения легкой цепи, содержащей в направлении от С-конца к N-концу константный домен легкой цепи изотипа каппа и вариабельный домен тяжелой цепи (VH1), путем замены вариабельного домена легкой цепи (VL) вариабельным доменом тяжелой цепи (VH1), происходящей из указанного первого антитела и, возможно, замену исходного константного домена легкой цепи константным доменом легкой цепи изотипа каппа;
б) модификацию легкой цепи, происходящей из указанного второго антитела, для получения легкой цепи, содержащей в направлении от С-конца к N-концу константный домен легкой цепи изотипа лямбда и вариабельный домен тяжелой цепи (VH2), путем замены вариабельного домена легкой цепи (VL) вариабельным доменом тяжелой цепи (VH2), происходящей из указанного второго антитела и, возможно, замену исходного константного домена легкой цепи константным доменом легкой цепи изотипа лямбда;
в) модификацию тяжелой цепи, происходящей из указанного первого антитела, для получения тяжелой цепи, содержащей в направлении от С-конца к N-концу константные домены тяжелой цепи 3, 2 и 1 (СН3, СН2 и СН1) и вариабельный домен легкой цепи (VL), путем замены вариабельного домена тяжелой цепи (VH1) вариабельным доменом легкой цепи (VL), происходящей из указанного первого антитела и/или
модификацию тяжелой цепи, происходящей из указанного второго антитела, для получения тяжелой цепи, содержащей в направлении от С-конца к N-концу константные домены тяжелой цепи 3, 2 и 1 (СН3, СН2 и СН1) и вариабельный домен легкой цепи (VL), путем замены вариабельного домена тяжелой цепи (VH2) вариабельным доменом легкой цепи (VL), происходящей из указанного второго антитела.
14. Способ по п. 13, дополнительно включающий следующие стадии:
а) трансформацию клетки-хозяина экспрессирующими векторами, содержащими молекулы нуклеиновой кислоты, кодирующей полученное мультиспецифическое антитело,
б) культивирование клетки-хозяина в условиях, обеспечивающих синтез указанной молекулы мультиспецифического антитела; и
в) выделение указанной молекулы мультиспецифического антитела из указанной культуры.
15. Мультиспецифическое антитело, полученное способом по п. 13 или 14.
