Claims (38)
1. Набор, содержащий
(A) молекулу биспецифического антитела, которая содержит
(i) первый связывающий домен, связывающий антиген на CD8+ Т-клетках, не существующий в естественных условиях в или на CD8+ Т-клетках; и
(ii) второй связывающий домен, связывающий опухолеспецифический антиген, существующий в естественных условиях на поверхности опухолевых клеток; и
(B) материал, необходимый для трансдукции CD8+ Т-клеток, полученных у субъекта, подлежащего лечению, антигеном, не существующим в естественных условиях в или на указанных CD8+ Т-клетках.
2. Набор по п. 1, в котором материал, необходимый для трансдукции CD8+ Т-клеток, представляет собой нуклеиновую кислоту, кодирующую антиген, не существующий в естественных условиях в или на указанных CD8+ Т-клетках.
3. Набор по п. 1 или 2, где указанный антиген, существующий в естественных условиях на поверхности опухолевых клеток, выбран из группы, состоящей из ЕрСАМ, HER-1, HER-2, HER-3, CD20, CD22, CD33, CD52, СА-12-5, HLA-DR, MUC-1 (муцина), АЗ3-антигена, PSMA (простат-специфического мембранного антигена), рецептора трансферрина, тенасцина и CA-IX.
6. Набор по п. 1 или 2, где указанный первый домен и/или второй связывающий домен является человеческим и/или гуманизированным.
7. Набор по п. 1 или 2, где трансдуцированные CD8+ Т-клетки дополнительно содержат Т-клеточный рецептор, существующий в естественных условиях на указанных Т-клетках, и/или Т-клеточный рецептор, который был введен в указанные Т-клетки при помощи генной инженерии.
8. Набор по п. 1 или 2, где молекула биспецифического антитела кодируется последовательностью нуклеиновой кислоты.
9. Фармацевтическая композиция, содержащая молекулу биспецифического антитела, которая содержит
(i) первый связывающий домен, связывающий антиген на CD8+ Т-клетках, не существующий в естественных условиях в или на CD8+ Т-клетках; и
(ii) второй связывающий домен, связывающий опухолеспецифический антиген, существующий в естественных условиях на поверхности опухолевых клеток,
которую следует вводить в комбинации с трансдуцированными CD8+ Т-клетками, содержащими антиген, не существующий в естественных условиях в или на CD8+ Т-клетках, причем указанную молекулу биспецифического антитела следует вводить перед, одновременно или после введения трансдуцированных CD8+ Т-клеток, и причем указанные CD8+ Т-клетки получены у субъекта, подлежащего лечению.
10. Фармацевтическая композиция по п. 9, где указанная молекула биспецифического антитела выбрана из группы, состоящей из полноразмерного антитела, F(ab)-, Fab′-SH-, Fv-, Fab′-, F(ab′)2- фрагмента, химерного антитела, антитела с пересаженными CDR, полностью человеческого антитела, конструкции бивалентного антитела, гибридного антитела, синтетического антитела, бивалентного антитела, тривалентного антитела, тетравалентного антитела, бивалентного одноцепочечного антитела, тривалентного одноцепочечного антитела и мультивалентного одноцепочечного антитела.
11. Фармацевтическая композиция по п. 9, где указанный антиген, не существующий в естественных условиях в или на CD8+ Т-клетках, выбран из группы, состоящей из белка Cripto (cryptic family protein), представителей семейства (не T-клеточных) молекул CD (кластера дифференцировки), EGFR и TSH-R.
12. Фармацевтическая композиция по п. 9, где указанный антиген, существующий в естественных условиях на поверхности опухолевых клеток, выбран из группы, состоящей из ЕрСАМ, HER-1, HER-2, HER-3, CD20, CD22, CD33, CD52, CA-12-5, HLA-DR, MUC-1 (муцина), АЗ3-антигена, PSMA (простат-специфического мембранного антигена), рецептора трансферрина, тенасцина и CA-IX.
13. Фармацевтическая композиция по п. 9, где указанный первый домен и/или второй связывающий домен является человеческим и/или гуманизированным.
14. Фармацевтическая композиция по п. 9, где трансдуцированные CD8+ Т-клетки дополнительно содержат Т-клеточный рецептор, существующий в естественных условиях на указанных Т-клетках, и/или Т-клеточный рецептор, который был введен в указанные Т-клетки при помощи генной инженерии.
15. Фармацевтическая композиция по п. 9, где молекула биспецифического антитела кодируется последовательностью нуклеиновой кислоты.
16. Способ лечения онкологического заболевания, включающий введение нуждающемуся в этом субъекту молекулы биспецифического антитела, которая содержит
(i) первый связывающий домен, связывающий антиген на CD8+ Т-клетках, не существующий в естественных условиях в или на CD8+ Т-клетках; и
(ii) второй связывающий домен, связывающий опухолеспецифический антиген, присутствующий в естественных условиях на поверхности опухолевых клеток,
в котором указанную молекулу биспецифического антитела следует вводить перед, одновременно или после введения трансдуцированных CD8+ Т-клеток, полученных у указанного субъекта и содержащих антиген, который не присутствует в естественных условиях в или на CD8+ Т-клетках.
17. Способ по п. 16, в котором указанное онкологическое заболевание выбрано из злокачественного новообразования эпителиального, эндотелиального или мезотелиального происхождения и злокачественного новообразования кроветворной ткани.
18. Способ по п. 16, где указанная молекула биспецифического антитела выбрана из группы, состоящей из полноразмерного антитела, F(ab)-, Fab′-SH-, Fv-, Fab′-, F(ab′)2- фрагмента, химерного антитела, антитела с пересаженными CDR, полностью человеческого антитела, конструкции бивалентного антитела, гибридного антитела, синтетического антитела, бивалентного антитела, тривалентного антитела, тетравалентного антитела, бивалентного одноцепочечного антитела, тривалентного одноцепочечного антитела и мультивалентного одноцепочечного антитела.
19. Способ по п. 16, где указанный антиген, не существующий в естественных условиях в или на CD8+ Т-клетках, выбран из группы, состоящей из белка Cripto (cryptic family protein), представителей семейства (не Т-клеточных) молекул CD (кластера дифференцировки), EGFR и TSH-R.
20. Способ по п. 16, где указанный антиген, существующий в естественных условиях на поверхности опухолевых клеток, выбран из группы, состоящей из ЕрСАМ, HER-1, HER-2, HER-3, CD20, CD22, CD33, CD52, СА-12-5, HLA-DR, MUC-1 (муцина), А33-антигена, PSMA (простат-специфического мембранного антигена), рецептора трансферрина, тенасцина и CA-IX.
21. Способ по п. 16, где указанный первый домен и/или второй связывающий домен является человеческим и/или гуманизированным.
22. Способ по п. 16, где трансдуцированные CD8+ Т-клетки дополнительно содержат Т-клеточный рецептор, существующий в естественных условиях на указанных Т-клетках, и/или Т-клеточный рецептор, который был введен в указанные Т-клетки при помощи генной инженерии.
23. Способ по п. 16, где молекула биспецифического антитела кодируется последовательностью нуклеиновой кислоты.
24. Вектор, содержащий последовательность нуклеиновой кислоты по п. 23.
25. Вектор по п. 24, где указанный вектор также содержит регуляторную последовательность, функционально связанную с указанной последовательностью нуклеиновой кислоты по п. 23.
26. Вектор по п. 24 или 25, где указанный вектор представляет собой экспрессирующий вектор.
27. Клетка-хозяин, трансформированная вектором, охарактеризованным в любом из пп. 24-26.
28. Способ получения молекулы биспецифического антитела, описанной в любом из пп. 1-6, указанный способ включает
(a) культивирование клетки-хозяина, описанной в п. 27, в условиях, обеспечивающих экспрессию молекулы биспецифического антитела, описанной в любом из пп. 1-6; и
(b) выделение полученной молекулы биспецифического антитела из культуры.
29. Молекула биспецифического антитела, которая содержит
(i) первый связывающий домен, связывающий антиген на CD8+ Т-клетках, не существующий в естественных условиях в или на CD8+ Т-клетках; и
(ii) второй связывающий домен, связывающий опухолеспецифический антиген, существующий в естественных условиях на поверхности опухолевых клеток,
причем указанную молекулу антитела получают согласно способу по п. 28.
30. Способ лечения заболевания у субъекта, включающий этапы
(a) выделения у субъекта CD8+ Т-клеток;
(b) трансдукции указанных выделенных CD8+ Т-клеток антигеном, не существующим в естественных условиях в и/или на CD8+ Т-клетках, согласно описанию в любом из пп. 1-6; и
(c) введения указанных трансдуцированных CD8+ Т-клеток указанному субъекту.
31. Способ по п. 30, в котором указанные трансдуцированные CD8+ Т-клетки вводят указанному субъекту путем внутривенной инфузии.
32. Способ по п. 30 или 31, в котором осуществляют ко-трансдукцию трансдуцированных CD8+ Т-клеток Т-клеточным рецептором.
33. Способ по любому из пп. 30-31, в котором экспансию указанных трансдуцированных CD8+ Т-клеток осуществляют с использованием анти-CD3 и анти-CD28 антител.
34. Способ по п. 33, в котором экспансию трансдуцированных CD8+ Т-клеток осуществляют в присутствии цитокинов, предпочтительно интерлейкина-2 (IL-2) и/или интерлейкина-15 (IL-15).
35. Способ по любому из пп. 30-31, дополнительно включающий
(d) введение указанному субъекту молекулы биспецифического антитела, описанной в любом из пп. 1-6 или 29.
36. Способ лечения заболевания по любому из пп. 30-31, в котором указанное биспецифическое антитело вводят перед, одновременно или после введения трансдуцированных CD8+ Т-клеток.
37. Способ по любому из пп. 30-31, в котором указанное заболевание представляет собой онкологическое заболевание.
38. Способ лечения по п. 37, в котором указанное онкологическое заболевание выбрано из злокачественного новообразования эпителиального, эндотелиального или мезотелиального происхождения или злокачественного новообразования кроветворной ткани.
