Устройство для сбора образцов биологических жидкостей для сбора естественным образом выделяемых биологических жидкостей, причем данное устройство содержит:колпачок, содержащий:внешнюю стенку, имеющую зацепляющий элемент,внутреннюю камеру для содержания жидкости, причем:данная камера содержит внутренние стенки, определяющие внутреннее пространство, и отверстие,данное отверстие конфигурируют для закупоривания с помощью съемного блокирующего элемента, иданный блокирующий элемент включает первый соединительный элемент для зацепления за соответствующий второй соединительный элемент в пробирке, чем вызывается удаление данного блокирующего элемента и открывание данного отверстия, когда данный колпачок соединен с данной пробиркой; ипробирку, содержащую:изолирующую стенку, определяющую резервуар для сбора образцов биологических жидкостей, зацепляющий элемент, комплементарный зацепляющему элементу колпачка, и второй соединительный элемент.2. Устройство для сбора образцов по п. 1, в котором съемный блокирующий элемент представляет собой элемент в форме диска, который с помощью резьбы зацепляется за отверстие.3. Устройство для сбора образцов по п. 2, в котором первый соединительный элемент содержит углубление, расположенное по центру в нижней части блокирующего элемента, и второй соединительный элемент расположен по центру пробирки.4. Устройство для сбора образцов по п. 2, в котором первый соединительный элемент содержит выемку, расположенную несимметрично относительно центра в нижней части блокирующего элемента, и второй соединительный элемент расположен в пробирке несимметрично относительно центр
Claims (55)
1. Устройство для сбора образцов биологических жидкостей для сбора естественным образом выделяемых биологических жидкостей, причем данное устройство содержит:
колпачок, содержащий:
внешнюю стенку, имеющую зацепляющий элемент,
внутреннюю камеру для содержания жидкости, причем:
данная камера содержит внутренние стенки, определяющие внутреннее пространство, и отверстие,
данное отверстие конфигурируют для закупоривания с помощью съемного блокирующего элемента, и
данный блокирующий элемент включает первый соединительный элемент для зацепления за соответствующий второй соединительный элемент в пробирке, чем вызывается удаление данного блокирующего элемента и открывание данного отверстия, когда данный колпачок соединен с данной пробиркой; и
пробирку, содержащую:
изолирующую стенку, определяющую резервуар для сбора образцов биологических жидкостей, зацепляющий элемент, комплементарный зацепляющему элементу колпачка, и второй соединительный элемент.
2. Устройство для сбора образцов по п. 1, в котором съемный блокирующий элемент представляет собой элемент в форме диска, который с помощью резьбы зацепляется за отверстие.
3. Устройство для сбора образцов по п. 2, в котором первый соединительный элемент содержит углубление, расположенное по центру в нижней части блокирующего элемента, и второй соединительный элемент расположен по центру пробирки.
4. Устройство для сбора образцов по п. 2, в котором первый соединительный элемент содержит выемку, расположенную несимметрично относительно центра в нижней части блокирующего элемента, и второй соединительный элемент расположен в пробирке несимметрично относительно центра.
5. Устройство для сбора образцов по п. 1, в котором съемный блокирующий элемент содержит кольцеобразный элемент, имеющий витки резьбы, расположенные на нем, причем данный кольцеобразный блокирующий элемент по существу покрывает отверстие, и причем внутренняя стенка колпачка включает в себя комплементарные витки резьбы, так что данный кольцеобразный элемент можно ввинчивать во внутреннее пространство, для того чтобы открывать отверстие.
6. Устройство для сбора образцов по любому из пп. 1-5, причем данное устройство дополнительно содержит фиксирующий механизм.
7. Устройство для сбора образцов по п. 6, в котором фиксирующий механизм содержит клин и комплементарный выступ.
8. Устройство для сбора образцов по п. 1, причем данное устройство дополнительно содержит закупоривающий механизм.
9. Устройство для сбора образцов по п. 8, в котором закупоривающий механизм содержит закупоривающее вещество, связанное с зацепляющим элементом колпачка, причем после соединения колпачка с пробиркой данное закупоривающее вещество втекает по меньшей мере в данный зацепляющий элемент колпачка.
10. Устройство для сбора образцов по п. 1, дополнительно содержащее обеспечивающее контроль вскрытия средство для определения того, был ли колпачок открыт.
11. Устройство для сбора образцов по п. 10, в котором обеспечивающее контроль вскрытия средство содержит кольцо, первая часть которого соединена с открытым концом колпачка, причем после присоединения колпачка к пробирке данное кольцо прилегает к пробирке.
12. Устройство для сбора образцов по п. 12, в котором при отсоединении колпачка от пробирки первая часть разламывается, и кольцо остается по существу прилегающим к пробирке.
13. Устройство для сбора образцов по п. 1, в котором жидкость содержит раствор для консервации клеток.
14. Устройство для сбора жидких образцов, содержащее:
колпачок, имеющий внутреннюю камеру для содержания жидкости и первый зацепляющий элемент; и
пробирку, содержащую изолирующую стенку, определяющую резервуар для сбора образцов, и второй зацепляющий элемент для зацепления за первый зацепляющий элемент.
15. Устройство для сбора образцов по п. 14, в котором:
колпачок содержит внешнюю стенку, имеющую первый зацепляющий элемент,
камера содержит внутренние стенки, определяющие внутреннее пространство, которое содержит жидкость, и
отверстие, причем данное отверстие сконфигурировано для закупоривания с помощью съемного блокирующего элемента.
16. Устройство для сбора образцов по п. 15, в котором блокирующий элемент включает первый соединительный элемент для зацепления за соответствующий второй соединительный элемент пробирки, и в котором после присоединения колпачка к пробирке данный блокирующий элемент сдвигается, и отверстие открывается.
17. Способ собирания образца естественным образом выделяемой биологической жидкости, причем данный способ включает:
предоставление устройства для сбора биологической жидкости, содержащего:
колпачок, содержащий:
внешнюю стенку, имеющую первый зацепляющий элемент,
внутреннюю камеру для содержания жидкости для консервации, причем:
данная камера содержит внутренние стенки, определяющие внутреннее пространство, и отверстие,
данное отверстие конфигурируют для закупоривания с помощью съемного блокирующего элемента, и
данный блокирующий элемент включает в себя первый соединительный элемент для зацепления за соответствующий второй соединительный элемент в пробирке, чем вызывается движение данного блокирующего элемента и открывание данного отверстия, когда данный колпачок соединен с данной пробиркой; и
пробирку, содержащую изолирующую стенку, определяющую резервуар для сбора образцов биологических жидкостей, второй зацепляющий элемент, комплементарный первому зацепляющему элементу колпачка, и второй соединительный элемент;
помещение биологической жидкости в данную камеру; и
соединение данного колпачка и пробирки вместе, так что соответствующие зацепляющие элементы зацепляются, причем блокирующий элемент сдвигается, и жидкость для консервации втекает в резервуар, содержащий биологическую жидкость, так что клетки, содержащиеся в биологической жидкости, консервируются для анализа.
18. Способ собирания образца жидкости, включающий:
предоставление устройства для сбора жидкости, содержащего:
колпачок, имеющий внутреннюю камеру для содержания заключенной жидкости и первый зацепляющий элемент, и пробирку, содержащую изолирующую стенку, определяющую резервуар для сбора образцов, и второй зацепляющий элемент для зацепления за первый зацепляющий элемент.
помещение жидкости в данную камеру; и
соединение данного колпачка и пробирки вместе, причем соответствующие зацепляющие элементы зацепляются, и жидкость выливается в резервуар для консервации по меньшей мере чего-то одного из жидкости и тех и/или иных компонентов в жидкости для анализа.
19. Способ по п. 17 или 18, дополнительно включающий выделение одного или нескольких типов клеток из множества типов клеток в собранной биологической жидкости.
20. Способ по п. 19, дополнительно включающий анализ собранных клеток.
21. Способ по п. 20, в котором анализ включает экстрагирование по меньшей мере чего-то одного из РНК, ДНК и одного или нескольких белков из тех и/или иных клеток.
22. Набор для сбора естественным образом выделяемой биологической жидкости, содержащий множество устройств для сбора образцов по любому из пп. 1-16.
23. Раствор для консервации клеток в биологической жидкости для дальнейшего разделения по типам клеток и последующего эпигенетического анализа, который позволяет при хранении клеток в биологической жидкости сохранять их антигенность и клеточное строение, причем данный раствор содержит по меньшей мере одно химическое фиксирующее средство и по меньшей мере один ингибитор протеазы и буферизирован при pH от приблизительно 6,4 до приблизительно 8,4.
24. Раствор по п. 23, дополнительно содержащий одно или несколько из по меньшей мере одного противомикробного средства и белков сыворотки человека и/или других видов животных.
25. Раствор по п. 23, в котором химическое фиксирующее средство выбирают из группы, состоящей из альдегидов.
26. Раствор по п. 25, в котором химическое фиксирующее средство представляет собой параформальдегид.
27. Раствор по п. 23, в котором химическое фиксирующее средство присутствует в концентрации, равной приблизительно 1% (об./об.).
28. Раствор по п. 23, в котором противомикробное средство выбирают из группы, состоящей из антибактериальных и противогрибковых антибиотиков.
29. Раствор по п. 23, в котором ингибитор протеазы выбирают из группы, состоящей из: ингибиторов аспарагиновых протеаз, ингибиторов цистеиновых протеаз, ингибиторов металлопротеаз, ингибиторов сериновых протеаз, ингибиторов треониновых протеаз, ингибиторов трипсина, ингибитора протеазы Kunitz STI и комбинации всего вышеперечисленного.
30. Раствор по п. 23, в котором ингибитор протеазы выбирают из группы, состоящей из: азида натрия, ФМСФ, апротинина, леупептина, пепстатина, натуральных или синтетических ингибиторов протеиназ, смесей ингибиторов протеаз, как натуральных, так и синтетических, и любых комбинаций вышеперечисленного.
31. Раствор по п. 23, в котором ингибитор протеазы представляет собой азид натрия.
32. Раствор по п. 23, причем данный раствор буферизирован при pH, составляющем от приблизительно 7,2 до приблизительно 7,6.
33. Раствор по п. 23 или 32, в котором буфер выбирают из группы, состоящей из: барбитала, трифосфата, цитрата, какодилата, других нефосфатных буферов и любых комбинаций вышеперечисленного.
34. Раствор по п. 23 или 32, в котором буфер представляет собой фосфатный буфер.
35. Способ консервации клеток в естественным образом выделяемой биологической жидкости, включающий приведение биологической жидкости в контакт с раствором для консервации по любому из пп. 23-34.
36. Способ выделения типа белых кровяных клеток из естественным образом выделяемой биологической жидкости, включающий
предоставление образца биологической жидкости, содержащего химически зафиксированные клетки;
центрифугирование образца биологической жидкости для получения осадка, содержащего клетки;
ресуспендирование данного осадка в буфере;
подвергание ресуспендированного осадка разделению в градиенте плотности для получения слоя из множества типов белых кровяных клеток; и
выделение первого типа белых кровяных клеток из данного множества белых кровяных клеток.
37. Способ по п. 36, в котором выделение включает:
приведение смеси типов клеток в контакт с раствором, содержащим магнитные гранулы, связанные с антителами, специфичными к эпитопу, обнаруживаемому на первом типе белых кровяных клеток, и
магнитное выделение данного первого типа белых кровяных клеток из множества типов белых кровяных клеток.
38. Способ по п. 36, в котором биологическую жидкость выбирают из группы, состоящей из: слюны, мочи и их комбинации.
39. Способ по п. 36, в котором буфер содержит азид натрия.
40. Способ по п. 36, в котором буфер содержит фосфатный буферный солевой раствор и азид натрия.
41. Способ по п. 36, в котором буфер дополнительно содержит фетальную бычью сыворотку.
42. Способ по п. 36, в котором буфер имеет pH от приблизительно 7,2 до приблизительно 7,6.
43. Способ по п. 36, в котором смесь типов белых кровяных клеток промывают один или несколько раз в буфере перед выделением.
44. Способ по п. 36, в котором первым типом белых кровяных клеток является лимфоцит.
45. Способ по п. 36, в котором первым типом лимфоцитарной клетки является T-клетка.
46. Способ по п. 37, в котором антитела являются специфичными к антигену, характерному для T-клеток.
47. Способ по п. 46, в котором антиген представляет собой по меньшей мере один из CD4 или CD3.
48. Способ выделения белых кровяных клеток из биологической жидкости, включающий:
предоставление образца биологической жидкости, содержащего химически фиксированные клетки;
подвергание данного образца разделению в градиенте плотности для получения слоя из множества типов белых кровяных клеток; и
выделение первого типа белых кровяных клеток из данного множества типов белых кровяных клеток.
49. Способ по п. 48, в котором выделение включает:
приведение множества типов клеток в контакт с раствором, содержащим магнитные гранулы, связанные с антителами, специфичными к эпитопу, обнаруживаемому на первом типе белых кровяных клеток; и
магнитное выделение данного первого типа белых кровяных клеток из множества типов белых кровяных клеток.
50. Способ по п. 48, дополнительно включающий центрифугирование образца биологической жидкости для получения осадка, содержащего клетки, и ресуспендирование данного осадка в буфере перед подверганием данного образца разделению в градиенте плотности.
51. Способ выделения конкретного типа белых кровяных клеток, включая, но без ограничения, лимфоциты, из биологической жидкости, включающий:
предоставление образца слюны, содержащего химически зафиксированные клетки;
подвергание образца разделению в градиенте плотности для получения слоя из смеси типов белых кровяных клеток, включая лимфоциты; и
выделение первого типа белых кровяных клеток из множества белых кровяных клеток.
52. Способ по п. 51, в котором выделение включает:
приведение смеси типов клеток в контакт с раствором, содержащим специфически связывающие средства для эпитопа, обнаруживаемого на конкретном типе белых кровяных клеток; и
выделение данного конкретного типа белых кровяных клеток из данной смеси типов белых кровяных клеток.
53. Способ по п. 51, дополнительно включающий центрифугирование образца биологической жидкости для получения осадка, содержащего клетки, и ресуспендирование данного осадка в буфере перед подверганием данного образца разделению в градиенте плотности.
54. Система для выделения типа белых кровяных клеток из образца биологической жидкости, содержащая:
центрифугу для центрифугирования образца биологической жидкости для получения осадка, содержащего клетки;
ресуспендирующее средство для ресуспендирования данного осадка в буфере;
средство для подвергания данного ресуспендированного осадка разделению в градиенте плотности для получения слоя из множества типов белых кровяных клеток; и
выделяющее средство для выделения первого типа белых кровяных клеток из множества белых кровяных клеток.
55. Система по п. 54, в которой выделяющее средство содержит:
средство для приведения смеси типов клеток в контакт с раствором, содержащим магнитные гранулы, связанные с антителами, специфичными к эпитопу, обнаруживаемому на первом типе белых кровяных клеток, и
магнитное выделяющее средство для магнитного выделения данного первого типа белых кровяных клеток из множества типов белых кровяных клеток.
