Abstract

SDS-PAGE에 의한 분자량이 약 57,000~120,000이고 양쪽성 전해질을 사용하는 등전점 전기영동법에 의한 등전점 (pI)이 약 3.8~5.1인 효소는 말토오스를 트레할로오스로, 트레할로오스를 말토오스로 전환시킨다. 이 효소는 피멜로박터속(Pimelobater 屬), 슈우도모나스속(Pseudomonad 屬) 및 테르무스속(Thermus 屬)의 미생물로 부터 분리하였다. 이 효소를 사용하여 시판품인 말토오스로 부터 트레할로오스를 공업적인 규모로 저렴하게 생성시킬 수 있다. 트레할로오스와 이것을 함유하는 당질 조성물은 모두가 이 효소로 제조되는 것들인데 음식물, 화장품 조성물 및 의약품 조성물에 적절히 사용할 수 있다.

Claims (24)

  1. 말토오스를 트레할로오스로, 그리고 트레할로오스를 말토오스로 전환시키는 효소.
  2. 제1항에 있어서, 미생물 유래의 것인 효소.
  3. 제2항에 있어서, 상기 미생물이 피멜로박터속, 슈우도모나속 또는 테르무스속에 속하는 것인 효소.
  4. 제2항에 있어서, 상기 미생물이 피멜로박터 sp.R48(FERM BP-4315), 슈우모나스 푸티다 H262(FERM BP-4579) 또는 테르무스 아쿠아티쿠스(ATCC 33923)인 효소.
  5. 제1항에 있어서, 다음의 물리화학적 성질을 가진 효소.
    (1) 분자량 : 소디움 도데실술페이드 폴리아크릴아미드 겔 전기 영동법 (SDS-PAGE)에서 약 57,000~120,000돌턴; (2) 등전접(pI) 양쪽성 전해질을 사용하는 등전점 전기영동법에서 약 3.8~5.1; (3) 활성억제 1mM Cu++, 50mM Tris-HCl 완충액에 의하여 억제됨.
  6. 제1항에 있어서, 아래와 같이 된 군으로 부터 선택되는 부분 아미노산 서열을 가진 효소.
    (1)Trp-X1-Arg-X2-Ala-X3-Phe
    (위에서 "X1"은 "Phe" 또는 "Pro"를 뜻하고, "X2"는 "Thr" 또는 "Pro"를 뜻하며, "X3"는 "Val" 또는 "Ala"를 뜻함)
    (2)Ala-Val-X4-Tyr
    (위에서, "X4"는 "Phe" 또는 "Ile"를 뜻함)
  7. (가) 영양 배지에서 효소 생성능이 있는 미생물을 배양하여 효소를 생성시키고, (나)는 수득한 배양액으로 부터 효소를 회수함을 특징으로 하는 제1항의 효소의 제조방법.
  8. 피멜로박터 sp. R48(FERM BP-4315)와 슈우도모나스 푸티다 H262(FERM BP-4579)로 된 군으로 부터 선택되는 제1항의 효소 생성능이 있는 미생물.
  9. 제1항의 효소를 용액중에서 말토오스에 작용시키는 단계를 포함하는 말토오스의 환원력 감소 방법.
  10. (가)제1항의 효소를 용액중에서 말토오스에 작용시켜 트레할로오스를 생성시키고, (나)수득한 트레할로오스 함유 당질 조성물을 회수함을 특징으로 하는 트레할로오스 함유 당질 조성물의 제조 방법.
  11. 제10항에 있어서, 상기 말토오스는 β-아밀라아제를 전분 지절 효소 존재하 또는 부재하에 전분물질의 용액에 작용시켜 제조되는 것인 제조 방법.
  12. 제11항에 있어서, 상기 트레할로오스는 트레할로오스 함수 결정 또는 트레할로오스 무수 결정인 제조 방법.
  13. (가) 제1항의 효소를 용액중에서 말토오스에 작용시켜 트레할로오스를 생성시키고, (나)는 수득한 용액중의 트레할로오스를 정제하여, (다)정제된 트레할로오스를 회수함을 특징으로 하는 트레할로오스의 제조 방법.
  14. 제13항에 있어서, 상기 (나)단계에는 용액에 글루코아밀라아제를 작용시켜 트레할로오스의 함유량을 증가시키고, 수득한 트레할로오스 용액을 강산성 양이온 교환 수지의 칼럼을 사용하는 컬럼 크로마토그래피 처리하는 단계를 포함하는 제조 방법.
  15. 제13항에 있어서, 상기 말토오스는 β-아밀라아제를 전분 지절 효소 존재하 또는 부재하에 전분 물질의 용액에 작용시켜 제조되는 것인 제조 방법.
  16. 제13항에 있어서, 상기 트레할로오스는 트레할로오스 함수 결정 또는 트레할로오스 무수 결정인 제조 방법.
  17. 제10항의 제조 방법으로 제조된 당질 조성물을 함유하는 음식물.
  18. 제10항의 제조 방법으로 제조된 당질 조성물을 함유하는 화장품 조성물.
  19. 제10항의 제조 방법으로 제조된 당질 조성물을 함유하는 의약품 조성물.
  20. 제19항에 있어서, 생체에 에너지를 공급하기 위한 의약품인 조성물.
  21. 제13항의 제조 방법으로 제조된 트레할로오스를 함유하는 음식물.
  22. 제13항의 제조 방법으로 제조된 트레할로오스를 함유하는 화장품 조성물.
  23. 제13항의 제조 방법으로 제조된 트레할로오스를 함유하는 의약품 조성물.
