Abstract

(목적) 환원성 전분 부분 분해물로부터 트레할로오스를 제조하기 위한 트레할로오스 방출 효소의 제조방법 및 그 용도의 확립을 목적으로 함. (구성) 본 발명은 신규의 내열성 트레할로오스 방출 효소와 그 제조방법 및 환원성 전분 부분 분해물로부터 비환원성 당질 생성 효소와 트레할로오스 방출 효소를 사용하여 트레할로오스 및 이것을 함유하는 당질과 트레할로오스를 함유시킨 조성물을 구성으로 한다.

내열성 트레할로오스 방출 효소와 그 제조 방법 및 용도
본 내용은 요부공개 건이므로 전문내용을 수록하지 않았음
제1도는 DEAE-TOYOPEARL겔 칼럼으로부터의 본 발명의 내열성 트레할로오스 방출 효소와 비환원성 당질 생성 효소의 용출 패턴을 나타내는 그래프.
제2도는 본 발명의 내열성 트레할로오스 방출 효소의 활성에 미치는 온도의 영향을 나타내는 그래프.
제3도는 본 발명의 내열성 트레할로오스 방출 효소의 활성에 미치는 pH의 영향을 나타내는 그래프.

Claims (16)

  1. 말단에 트레할로오스 구조를 가진 글루코오스 중합도 3이상의 비환원성 당질의 트레할로오스 부분과 그외의 글리코실 부분 사이의 결합을 특이적으로 가수분해하는 내열성 트레할로오스 방출 효소.
  2. 제1항에 있어서, 글루코실 부분이 중합도 1이상의 클루코오스 잔기 하나 이상으로 구성되어 있는 내열성 트레할로오스 방출 효소.
  3. 제1항에 있어서, 내열성이 pH 7.0에서 60분간 유지할 경우 85℃부근까지 일정한 내열성 트레할로오스 방출 효소.
  4. 제1항에 있어서. 미생물 유래의 효소인 것을 특징으로 하는 내열성 트레할로오스 방출 효소.
  5. 제4항에 있어서, 미생물이 술폴로부스속(Sufolobus屬)에 속하는 미생물과 이들의 변이주로 된 군으로부터 선택되는 1종인 내열성 트레할로오스 방출 효소.
  6. 제1항에 있어서, 아래의 물리화학적 성질을 가진 내열성 트레할로오스 방출 효소.
    (1) 작용
    말단에 트레할로오스 구조를 가진 글루코오스 중합도 3이상의 비환원성 당질의 트레할로오스 부분과 글리코실 부분 사이의 결합을 특이적으로 가수분해함.
    (2) 분자량
    소디움 도데실술페이트-폴리아크릴아미드 겔 전기 영동법(SDS-PAGE)에 의하여 약 54,000∼64,000달톤
    (3) 등전점(pI)
    양쪽성 전해질 함유 전기 영동법에서 약 5.6∼6.6
    (4) 최적 온도
    pH 6.0 및 30분간 인큐베이트에서 약 75℃
    (5) 최적 pH
    60℃ 및 30분간 인큐베이트에서 약 5.0∼6.0
    (6)열적 안정성
    pH 7.0 및 60분간 유지하여 85℃부근까지 안정
    (7) pH 안정성
    25℃에서 16시간 유지하여 pH 약 4.0∼9.5에서 안정
  7. 제1항 기재의 효소의 산생능을 가진 미생물을 영양 배지에서 배양하고, 수득되는 배양물로부터 상기 효소를 회수하는 것을 특징으로 하는 제1항 기재의 내열성 트레할로오스 방출 효소의 제조방법.
  8. 제7항에 있어서, 미생물이 술폴로부스속(Sufolobus屬)에 속하는 미생물과 이들의 변이주로 된 군으로부터 선택되는 1종인 제조 방법.
  9. 말단에 트레할로오스 구조를 가진 글루코오스 중합도 3이상의 비환원성 당질을 함유하는 용액에 제1항 기재의 효소를 작용시키는 단계를 포함하는 트레할로오스의 제조방법.
  10. 제9항에 있어서, 환원성 전분 부분 분해물 1종 이상과 제1항 기재의 효소를 함유하는 용액에 비환원성 당질 생성 효소를 작용시키는 제조방법.
  11. 제9항에 있어서, 트레할로오스는 트레할로오스 함수 결정, 트레할로오스 무수 결정 및 이들의 혼합물로 된 군으로부터 선택되는 1종인 제조 방법.
  12. 제9항의 제조방법으로 제조되는 트레할로오스.
  13. 제12항의 트레할로오스를 함유하는 조성물.
  14. 제13항에 있어서, 조성물이 음식물인 조성물.
  15. 제13항에 있어서, 조성물이 화장품인 조성물.
  16. 제13항에 있어서, 조성물이 의약품인 조성물.
