Claims (45)
- 인간 CTLA-4에 대해 특이적으로 결합하는 인간 단일클론 항체의 중쇄 및 경쇄를 암호화하는 폴리뉴클레오티드를 포함하는 포유동물 숙주 세포주로서, 상기 항체는 서열 번호 1에 있는 중쇄 CDR1, CDR2 및 CDR3 아미노산 서열 및 서열 번호 14에 있는 경쇄 CDR1, CDR2 및 CDR3 아미노산 서열을 포함하는 것인 포유동물 숙주 세포주.
- 제1항에 있어서, 상기 항체는 서열 번호 1에 있는 중쇄 FR1 내지 FR4 아미노산 서열 및 서열 번호 14에 있는 경쇄 FR1 내지 FR4 아미노산 서열을 포함하는 것인 포유동물 숙주 세포주.
- 제1항에 있어서, 상기 항체는 서열 번호 1의 아미노산 서열을 포함하는 중쇄 및 서열 번호 14의 아미노산 서열을 포함하는 경쇄를 포함하는 것인 포유동물 숙주 세포주.
- 제1항에 있어서, 상기 인간 단일클론 항체는 인간 B7-1 및 인간 B7-2에 대한 인간 CTLA-4의 결합을 저해하며, 상기 인간 단일클론 항체의 중쇄는 인간 DP50 유전자에 의하여 암호화되는 아미노산 서열에 대하여 90% 이상의 서열 동일성을 갖는 아미노산 서열을 포함하며, 경쇄는 인간 A27 Vκ 유전자에 의하여 암호화되는 아미노산 서열에 대하여 90% 이상의 서열 동일성을 가지는 아미노산 서열을 포함하는 것인 포유동물 숙주 세포주.
- 제1항에 있어서, 상기 인간 단일클론 항체는 인간 B7-1 및 인간 B7-2에 대한 인간 CTLA-4의 결합을 저해하며, 상기 인간 단일클론 항체의 중쇄를 암호화하는 폴리뉴클레오티드는 중쇄 FR1 내지 FR4 아미노산 서열을 암호화하는 서열 번호 138에서의 뉴클레오티드 서열에 대하여 90% 이상의 서열 동일성을 갖는 뉴클레오티드 서열을 포함하며, 상기 인간 단일클론 항체의 경쇄를 암호화하는 폴리뉴클레오티드는 경쇄 FR1 내지 FR4 아미노산 서열을 암호화하는 서열 번호 141에서의 뉴클레오티드 서열에 대하여 90% 이상의 서열 동일성을 갖는 뉴클레오티드 서열을 포함하는 것인 포유동물 숙주 세포주.
- 제1항에 있어서, 상기 항체는 인간 A27 Vκ 유전자를 이용하는 경쇄를 포함하고 인간 B7-1 및 인간 B7-2에 대한 인간 CTLA-4의 결합을 저해하고, 추가로 상기 항체의 중쇄를 암호화하는 폴리뉴클레오티드는 인간 DP50 유전자에 의하여 암호화되는 아미노산 서열에 대하여 90% 이상의 서열 동일성을 갖는 아미노산 서열을 암호화하는 것인 포유동물 숙주 세포주.
- 제1항에 있어서, 상기 항체는 인간 CD28, 인간 B7-2, 인간 CD44, 및 hIgG1 에 대하여 100:1 초과의 인간 CTLA-4에 대한 선택성을 가지고, 5 nM 미만의 IC50 으로 인간 CTLA-4와 인간 B7-2 사이의 결합을 저해하며, 추가로 상기 인간 단일클론 항체의 중쇄는 인간 DP50 유전자에 의하여 암호화되는 아미노산 서열에 대하여 90% 이상의 서열 동일성을 갖는 아미노산 서열을 포함하며, 경쇄는 인간 A27 Vκ 유전자에 의하여 암호화되는 아미노산 서열에 대하여 90% 이상의 서열 동일성을 갖는 아미노산 서열을 포함하는 것인 포유동물 숙주 세포주.
- 제4항에 있어서, 상기 항체의 중쇄를 암호화하는 폴리뉴클레오티드가 인간 DP50 유전자의 뉴클레오티드 서열에 대하여 95% 이상의 서열 동일성을 갖는 뉴클레오티드 서열을 포함하는 것인 포유동물 숙주 세포주.
- 제4항에 있어서, 상기 항체의 중쇄를 암호화하는 폴리뉴클레오티드가 인간 DP50 유전자의 뉴클레오티드 서열에 대하여 95% 이상의 서열 동일성을 갖는 뉴클레오티드 서열을 포함하며, 상기 항체의 경쇄를 암호화하는 폴리뉴클레오티드가 인간 A27 Vκ 유전자의 뉴클레오티드 서열에 대하여 95% 이상의 서열 동일성을 갖는 뉴클레오티드 서열을 포함하는 것인 포유동물 숙주 세포주.
- 제5항에 있어서, 상기 항체의 중쇄를 암호화하는 폴리뉴클레오티드가 중쇄 FR1 내지 FR4 아미노산 서열을 암호화하는 서열번호 138에서의 뉴클레오티드 서열에 대하여 95% 이상의 서열 동일성을 갖는 뉴클레오티드 서열을 포함하는 것인 포유동물 숙주 세포주.
- 제5항에 있어서, 상기 항체의 경쇄를 암호화하는 폴리뉴클레오티드가 경쇄 FR1 내지 FR4 아미노산 서열을 암호화하는 서열번호 141에서의 뉴클레오티드 서열에 대하여 95% 이상의 서열 동일성을 갖는 뉴클레오티드 서열을 포함하는 것인 포유동물 숙주 세포주.
- 제5항에 있어서, 상기 항체의 중쇄를 암호화하는 폴리뉴클레오티드가 중쇄 FR1 내지 FR4 아미노산 서열을 암호화하는 서열번호 138에서의 뉴클레오티드 서열에 대하여 95% 이상의 서열 동일성을 갖는 뉴클레오티드 서열을 포함하며, 상기 항체의 경쇄를 암호화하는 폴리뉴클레오티드가 경쇄 FR1 내지 FR4 아미노산 서열을 암호화하는 서열번호 141에서의 뉴클레오티드 서열에 대하여 95% 이상의 서열 동일성을 갖는 뉴클레오티드 서열을 포함하는 것인 포유동물 숙주 세포주.
- 제6항에 있어서, 상기 항체의 경쇄가 서열번호 14에 있는 FR1 내지 FR4 아미노산 서열을 포함하며, 상기 항체의 중쇄가 서열번호 1에 있는 FR1 내지 FR4 아미노산 서열을 포함하는 것인 포유동물 숙주 세포주.
- 제7항에 있어서, 상기 항체의 중쇄를 암호화하는 폴리뉴클레오티드가 중쇄 FR1 내지 FR4 아미노산 서열을 암호화하는 서열번호 138에서의 뉴클레오티드 서열에 대하여 90% 이상의 서열 동일성을 갖는 뉴클레오티드 서열을 포함하며, 상기 항체의 경쇄를 암호화하는 폴리뉴클레오티드가 경쇄 FR1 내지 FR4 아미노산 서열을 암호화하는 서열번호 141에서의 뉴클레오티드 서열에 대하여 90% 이상의 서열 동일성을 갖는 뉴클레오티드 서열을 포함하는 것인 포유동물 숙주 세포주.
- 제7항에 있어서, 상기 항체의 경쇄를 암호화하는 폴리뉴클레오티드가 서열번호 14에 있는 경쇄 FR1 내지 FR4 아미노산 서열에 대하여 90% 이상의 서열 동일성을 갖는 아미노산 서열을 암호화하는 뉴클레오티드 서열을 포함하는 것인 포유동물 숙주 세포주.
- 제7항에 있어서, 상기 항체의 경쇄를 암호화하는 폴리뉴클레오티드가 서열번호 14에 있는 경쇄 FR1 내지 FR4 아미노산 서열에 대하여 95% 이상의 서열 동일성을 갖는 아미노산 서열을 암호화하는 뉴클레오티드 서열을 포함하는 것인 포유동물 숙주 세포주.
- 제1항 내지 제7항 중 어느 한 항에 있어서, 글루타민 합성효소 발현 시스템을 추가로 포함하는 것인 포유동물 숙주 세포주.
- 제1항, 제2항, 제3항, 제4항 및 제7항 중 어느 한 항에 있어서, 상기 인간 단일클론 항체는 100 nM 이하의 IC50으로 인간 B7-1 또는 인간 B7-2에 대한 인간 CTLA-4의 결합을 저해하는 것인 포유동물 숙주 세포주.
- 제1항 내지 제6항 중 어느 한 항에 있어서, 상기 인간 단일클론 항체는 5 nM 이하의 IC50으로 인간 B7-1에 대한 인간 CTLA-4의 결합을 저해하는 것인 포유동물 숙주 세포주.
- 제1항 내지 제7항 중 어느 한 항에 있어서, 상기 인간 단일클론 항체는 2 nM 이하의 IC50으로 인간 B7-1에 대한 인간 CTLA-4의 결합을 저해하는 것인 포유동물 숙주 세포주.
- 제1항 내지 제6항 중 어느 한 항에 있어서, 상기 인간 단일클론 항체는 5 nM 이하의 IC50으로 인간 B7-2에 대한 인간 CTLA-4의 결합을 저해하는 것인 포유동물 숙주 세포주.

- 제2항 또는 제16항에 있어서, 상기 서열번호 1에 있는 중쇄 FR1 내지 FR4 아미노산 서열 및 서열번호 14에 있는 경쇄 FR1 내지 FR4 아미노산 서열을 포함하는 항체가 각각 서열번호 1 및 서열번호 14 에서의 중쇄 및 경쇄 불변부 아미노산 서열을 추가로 포함하는 것인 포유동물 숙주 세포주.
- 제1항 내지 제16항 중의 어느 한 항에 있어서, 포유동물 숙주 세포주가 CHO 세포주인 것인 포유동물 숙주 세포주.
