(iii)膜貫通ドメインが
(a)配列番号12のアミノ酸配列の少なくとも1、2または3の修飾を有するが、修飾が20、10または5を超えないアミノ酸配列または配列番号12のアミノ酸配列と95〜99%同一性を有する配列;または
(b)配列番号12の配列を含む;
(iv)抗原結合ドメインがヒンジ領域により膜貫通ドメインに接続され、ここで、該ヒンジ領域が配列番号2または配列番号6またはそれと95〜99%同一性を有する配列を含む;および/または

(v)細胞内ドメインが配列番号14または配列番号16の配列および配列番号18または配列番号20の配列を含み、ここで、細胞内シグナル伝達ドメインを含む配列が同じフレーム内に、かつ単一ポリペプチド鎖として発現される、
請求項17または18に記載の単離CARポリペプチド分子。 - (i)請求項11〜15のいずれかに記載の核酸分子;
(ii)請求項17〜19のいずれかに記載のCARポリペプチド分子;または
(iii)請求項16に記載のベクター
を含む、免疫エフェクター細胞(例えば、免疫エフェクター細胞の集団)。 - (i)第二CAR分子をコードする第二核酸分子、
(ii)第二CARポリペプチド分子または
(iii)第二CAR分子をコードする核酸分子を含む第二ベクター;任意に
(a)第二CAR分子が第一CAR分子と同じ抗原を標的とし、ここで、第二CAR分子の抗原結合ドメインが第一CAR分子の抗原結合ドメインと異なる;または
(b)第二CAR分子の抗原結合ドメインが第一CAR分子の抗原結合ドメインと異なる抗原を標的とする、
をさらに含む、請求項20に記載の細胞。 - (i)第一CAR分子が共刺激シグナル伝達ドメインを含む細胞内シグナル伝達ドメインを含むが、一次シグナル伝達ドメインを含まず、第二CAR分子が一次シグナル伝達ドメインを含む細胞内シグナル伝達ドメインを含むが、共刺激シグナル伝達ドメインを含まない、
(ii)第一CAR分子が4−1BB、CD28、CD27またはOX−40からなる群から選択されるタンパク質の機能的シグナル伝達を含む共刺激シグナル伝達ドメインを含み、第二CAR分子がCD3ゼータの機能的シグナル伝達ドメインを含む一次シグナル伝達ドメインを含む;
(iii)第二CARが阻害性CARであり、ここで、阻害性CARが抗原結合ドメイン、膜貫通ドメインおよび阻害分子の細胞内ドメインを含み、ここで、阻害分子がPD1、PD−L1、CTLA4、TIM3、LAG3、VISTA、BTLA、TIGIT、LAIR1、CD160、2B4、TGFRベータ、CEACAM−1、CEACAM−3およびCEACAM−5からなる群から選択される;
(iv)第二CAR分子が一次シグナル伝達ドメインを含む細胞内シグナル伝達ドメインおよび/または細胞内シグナル伝達ドメインをさらに含み、ここで、細胞内シグナル伝達ドメインがCD3ゼータの機能的ドメインを含む一次シグナル伝達ドメインおよび4−1BBの機能的ドメインを含む共刺激シグナル伝達ドメインを含む;および/または
(v)第二CAR分子が配列番号26のアミノ酸配列を含む、
請求項20または21に記載の細胞。 - 免疫エフェクター細胞がヒトT細胞(例えば、CD8+ T細胞)またはヒトNK細胞であり、所望により、ここで、T細胞がジアシルグリセロールキナーゼ(DGK)および/またはイカロス欠損である、請求項20〜22のいずれかに記載の細胞。
- CAR分子が
(i)請求項11〜15のいずれかに記載の核酸分子によりコードされる;
(ii)請求項17〜19のいずれかに記載のCARポリペプチド分子;または
(iii)請求項16に記載のベクターによりコードされる、
請求項1〜10のいずれかに記載の使用のための組成物。 - 使用のための免疫エフェクター細胞が請求項20〜23のいずれかに記載のものである、請求項1〜10のいずれかに記載の使用のための組成物。
- CAR発現免疫エフェクター細胞(例えば、CAR発現免疫エフェクター細胞の集団)を製造する方法であって、請求項16に記載のベクターまたは請求項11〜15のいずれかに記載のCARをコードする核酸分子を、CAR分子が発現される条件下に、免疫エフェクター細胞に導入することを含む、方法。
- a)免疫エフェクター細胞の集団(例えば、T細胞またはNK細胞)を提供し;そして
b)集団からT制御性細胞を除去し、それによりT制御性枯渇細胞の集団を提供することを含み;
ここで、工程a)およびb)を、CARをコードする核酸を集団に導入する前に行う、任意に、T制御性細胞を、抗CD25抗体または抗GITR抗体を使用して細胞集団から除去する、
