Description
- Hintergrund der Erfindung
- Serinproteasen sind eine Untergruppe der Carbonylhydrolasen. Sie umfassen eine vielfältige Klasse von Enzymen, die einen weiten Bereich an Spezifitäten und biologischen Funktionen aufweisen. Stroud, R. Sci. Amer., 131: 74–88. Trotz ihrer funktionalen Verschiedenheit wurde die katalytische Maschinerie von Serinproteasen durch mindestens zwei genetisch verschiedene Familien von Enzymen angegangen: 1) die Subtilisine und 2) die aus Säugern stammenden, mit Chymotrypsin verwandten und homologen bakteriellen Serinproteasen (z. B. Trypsin und S. gresius-Trypsin). Diese beiden Familien von Serinproteasen weisen bemerkenswert ähnliche Katalysemechanismen auf. Kraut, J. (1977), Annu. Rev. Biochem., 46: 331–358. Darüber hinaus, obwohl die Primärstruktur in keinem Zusammenhang steht, vereinigt die Tertiärstruktur dieser beiden Enzymfamilien eine konservierte katalytische Triade von Aminosäuren, die aus Serin, Histidin und Aspartat besteht.
- Subtilisine sind Serinproteasen (MG ca. 27 500), die in großen Mengen von einer großen Vielfalt von Bacillus-Spezies und anderen Mikroorganismen sekretiert werden. Die Proteinsequenz von Subtilisin wurde von mindestens neun verschiedenen Spezies von Bacillus bestimmt. Markland, F. S., et al. (1983), Hoppe-Seyler's Z. Physiol. Chem., 364: 1537–1540. Es wurde über die dreidimensionale kristallographische Struktur von Subtilisinen aus Bacillus amyloliquefaciens, Bacillus licheniforimis und mehreren natürlichen Varianten von B. lentus berichtet. Diese Untersuchungen deuten darauf hin, dass, obwohl Subtilisin mit den Säugetier-Serinproteasen genetisch nicht verwandt ist, es eine ähnliche Struktur des aktiven Zentrums aufweist. Die Röntgenkristallstrukturen von Subtilisin, welches kovalent gebundene Peptidinhibitoren (Robertus, J. D., et al., (1972), Biochemistry, 11: 2439–2449) oder Produktkomplexe (Robertus, J. D., et al., (1976), J. Biol. Chem., 251: 1097–1103) enthält, stellten auch eine Information bezüglich der aktiven Stelle und der putativen Substratbindungstasche von Subtilisin zur Verfügung. Darüber hinaus wurde für Subtilisin über eine große Anzahl von kinetischen und chemischen Modifizierungsuntersuchungen berichtet: Svendsen, B. (1976), Carlsberg Res. Commun., 41: 237–291: Markland, F. S. ebd.) sowie mindestens ein Bericht, in welchem die Seitenkette von Methionin beim Rest 222 von Subtilisin durch Wasserstoffperoxid zu Methionin-Sulfoxid umgewandelt wurde (Stauffer, D. C., et al. (1965), J. Biol. Chem., 244: 5333–5338) und es wurde eine umfassende ortsspezifische Mutagenese durchgeführt (Wells und Estell (1988) TIBS 13: 291–297).
- Zusammenfassung der Erfindung
- Es ist ein Ziel hierin, eine Protease zur Verfügung zu stellen, die eine Aminosäuresubstitution an einer Rest-Position umfasst, die der Position des Restes 232 von Bacillus amyloliquefaciens-Subtilisin entspricht, wobei die Austausch-Aminosäure Valin ist. Vorzugsweise ist die Substitution A232V, wobei die Protease eine verbesserte Waschleistung im Vergleich zu einer Vorläuferprotease aufweist.
- Stärker bevorzugte Proteasen sind Substitutionssätze, die aus der Gruppe gewählt sind, die aus Substitutionen an Rest-Positionen, entsprechend den Positionen in der Tabelle 1 von Bacillus amyloliquefaciens-Subtilisin, besteht, wobei der Substitutionssatz die Substitution einschließt, auf welche oben Bezug genommen wird, wobei die Austausch-Aminosäure Valin ist und wobei die Protease eine verbesserte Waschleistung im Vergleich zu einer Vorläuferprotease aufweist.
- Am meisten bevorzugte Varianten sind Substitutionssätze, die aus der Gruppe gewählt werden, welche aus Substitutionen besteht, die jenen in Tabelle 2 von Bacillus amyloliquefaciens-Subtilisin entsprechen; wobei der Substitutionssatz die Substitutionen einschließt, auf welche oben Bezug genommen wird, wobei die Austausch-Aminosäure Valin ist.
- Es ist ein weiteres Ziel, DNA-Sequenzen, die solche Proteasen codieren, sowie Expressionsvektoren, die solche varianten DNA-Sequenzen enthalten, bereit zu stellen.
- Noch ein weiteres, anderes Ziel der Erfindung ist es, Wirtszellen zur Verfügung zu stellen, die mit solchen Vektoren transformiert sind, sowie Wirtszellen, die in der Lage sind, solche DNA zu exprimieren, um Proteasen der Erfindung intrazellulär oder extrazellulär zu erzeugen.
- Es wird ferner eine Reinigungszusammensetzung bereit gestellt, die eine Protease-Variante der vorliegenden Erfindung umfasst.
- Es wird auch eine Zusammensetzung für die Behandlung einer Textilie bereit gestellt, welche eine Protease der vorliegenden Erfindung umfasst.
- KURZE BESCHREIBUNG DER ZEICHNUNGEN
- Die
1A –C zeigen die DNA- und die Aminosäuresequenz für Bacillus amyloliquefaciens-Subtilisin und eine partielle Restriktionskarte dieses Gens. - Die
2 zeigt die konservierten Aminosäurereste unter Subtilisinen aus Bacillus amyloliquefaciens (BPN)' und Bacillus lentus (Wildtyp). - Die
3A und3B zeigen die Aminosäuresequenz von vier Subtilisinen. Die obere Zeile stellt die Aminosäuresequenz von Subtilisin aus Bacillus amyloliquefaciens-Subtilisin (manchmal auch als Subtilisin BPN' bezeichnet) dar. Die zweite Zeile zeigt die Aminosäuresequenz von Subtilisin aus Bacillus subtilis. Die dritte Zeile zeigt die Aminosäuresequenz von Subtilisin aus B. licheniformis. - Proteasen sind Carbonylhydrolasen, die im Allgemeinen so wirken, dass sie Peptidbindungen von Proteinen oder Peptiden spalten. Wie hierin verwendet, bedeutet „Protease" eine in der Natur vorkommende Protease oder eine rekombinante Protease. Natürlich vorkommende Proteasen schließen α-Aminoacylpeptid-Hydrolase, Peptidylaminosäure-Hydrolase, Acylamino-Hydrolase, Serincarboxypeptidase, Metallocarboxypeptidase, Thiolproteinase, Carboxylproteinase und Metalloproteinase ein. Serin-, Metallo-, Thiol- und Säureproteasen sind eingeschlossen, ebenso wie Endo- und Exoproteasen.
- Die vorliegende Erfindung schließt Protease-Enzyme ein, die nicht-natürlich vorkommende Carbonylhydrolase-Varianten (Proteasen der Erfindung) sind, welche ein(e) unterschiedliche(s) proteolytische Aktivität, Stabilität, Substratspezifität, pH-Profil und/oder Leistungscharakteristik verglichen mit der Vorläufer-Carbonylhydrolase, von der die Aminosäuresequenz der Protease der Erfindung abgeleitet ist, aufweisen. Insbesondere weisen solche Proteasen eine Aminosäuresequenz auf, die nicht in der Natur gefunden wird, welche durch die Substitution einer Mehrzahl von Aminosäureresten einer Vorläuferprotease durch verschiedene Aminosäuren abgeleitet ist. Die Vorläuferprotease kann eine natürlich vorkommende Protease oder eine rekombinante Protease sein.
- Die hierin nützlichen Proteasen umfassen die Substitution einer beliebigen der neunzehn in der Natur vorkommenden L-Aminosäuren an den bezeichneten Aminosäurerest-Positionen. Solche Substitutionen können bei einem beliebigen Vorläufer-Subtilisin (prokaryotisch, eukaryotisch, von Säugern stammend etc.) durchgeführt werden. Überall in dieser Anmeldung wird auf verschiedene Aminosäuren mittels der gängigen Ein- und Drei-Buchstaben-Codes Bezug genommen. Solche Codes sind bei Dale, M. W. (1989), Molecular Genetics of Bacteria, John Wiley & Sons, Ltd., Anhang B, bezeichnet.
- Die hierin nützlichen Proteasen sind vorzugsweise von einem Bacillus-Subtilisin abgeleitet. Stärker bevorzugt sind die Proteasen der Erfindung von Bacillus lentus-Subtilisin und/oder Subtilisin 309 abgeleitet.
- Subtilisine sind Bakterien- oder Pilz-Proteasen, die im Allgemeinen so wirken, dass sie Peptidbindungen von Proteinen oder Peptiden spalten. Wie hierin verwendet, bedeutet „Subtilisin" ein natürlich vorkommendes Subtilisin oder ein rekombinantes Subtilisin. Es ist bekannt, dass durch verschiedene mikrobielle Spezies eine Reihe von natürlich vorkommenden Subtilisinen erzeugt und oft sekretiert wird. Aminosäuresequenzen der Mitglieder dieser Reihe sind nicht vollständig homolog. Jedoch zeigen die Subtilisine in dieser Reihe die gleiche oder eine ähnliche Art von proteolytischer Aktivität. Diese Klasse von Serinproteasen hat eine verbreitete Aminosäuresequenz gemein, welche eine katalytische Triade definiert, welche sie von der mit Chymotrypsin verwandten Klasse von Serinproteasen unterscheidet. Die Subtilisine und die mit Chymotrypsin verwandten Serinproteasen weisen beide eine katalytische Triade auf, die Aspartat, Histidin und Serin umfasst. In den mit Subtilisin verwandten Proteasen ist die relative Reihenfolge dieser Aminosäuren, gelesen vom Amino- zum Carboxy-Terminus, Aspartat-Histidin-Serin. In den mit Chymotrypsin verwandten Proteasen ist die relative Reihenfolge jedoch Histidin-Aspartat-Serin. Somit bezieht sich Subtilisin hierin auf eine Serinprotease, welche die katalytische Triade von mit Subtilisin verwandten Proteinen aufweist. Beispiele schließen die in
3 hierin bezeichneten Subtilisine ein, ohne darauf beschränkt zu sein. Allgemein und für die Zwecke der vorliegenden Erfindung entspricht die Nummerierung der Aminosäuren in Proteasen den Nummern, die der Sequenz des reifen Bacillus amyloliquefaciens-Subtilisins, die in1 angegeben ist, zugewiesen wurden. - „Rekombinantes Subtilisin" oder „rekombinante Protease" bezieht sich auf ein Subtilisin oder eine Protease, in welchem bzw. welcher die DNA-Sequenz, die das Subtilisin oder die Protease codiert, so modifiziert ist, dass eine variante (oder mutierte) DNA-Sequenz, die die Substitution, Deletion oder Insertion von einer oder mehreren Aminosäuren in der natürlich vorkommenden Aminosäuresequenz codiert, erzeugt wird. Geeignete Verfahren, um eine solche Modifizierung zu erzeugen, und die mit jenen, die hierin offenbart werden, kombiniert werden können, schließen jene ein, die in dem
US-Patent RE 34 606 , demUS-Patent 5 204 015 und demUS-Patent 5 185 258 , demU.S.-Patent 5 700 676 , demU.S.-Patent 5 801 038 und demU.S.-Patent 5 763 257 offenbart werden. - „Nicht-humane Subtilisine" und die sie codierende DNA können aus vielen prokaryotischen und eukaryotischen Organismen erhalten werden. Geeignete Beispiele von prokaryotischen Organismen schließen gram-negative Organismen wie E. coli oder Pseudomonas und gram-positive Bakterien wie Micrococcus oder Bacillus ein. Beispiele von eukaryotischen Organismen, aus denen Subtilisin und deren Gene erhalten werden können, schließen Hefe wie Saccharomyces cerevisiae, Pilze wie Aspergillus sp. ein.
- Eine Protease der Erfindung weist eine Aminosäuresequenz auf, die von der Aminosäuresequenz einer „Vorläuferprotease" abgeleitet ist. Die Vorläuferproteasen schließen natürlich vorkommende Proteasen und rekombinante Proteasen ein. Die Aminosäuresequenz der Protease der Erfindung ist von der Aminosäuresequenz der Vorläuferprotease durch die Substitution von einer oder mehreren Aminosäuren der Vorläufer-Aminosäuresequenz „abgeleitet". Eine solche Modifikation ist von der „Vorläufer-DNA-Sequenz", die die Aminosäuresequenz der Vorläuferprotease codiert, anstatt einer Manipulation des Vorläuferprotease-Enzyms per se. Geeignete Verfahren für eine solche Manipulation der Vorläufer-DNA-Sequenz schließen Verfahren ein, die hierin offenbart werden, wie auch Verfahren, die den Fachleuten im Fachbereich bekannt sind (siehe zum Beispiel die
EP 0 328 299 ,WO 89/06279 - Spezielle Substitutionen von Aminosäuren an einer oder mehreren Rest-Positionen, die Rest-Positionen entsprechen, gewählt aus der Gruppe, die aus 62, 212, 230, 232, 252 und 257 von Bacillus amyloliquefaciens-Subtilisin besteht, sind hierin bezeichnet.
- Bevorzugte Proteasen der Erfindung sind jene, die Kombinationen von Substitutionen an Rest-Positionen aufweisen, die den Positionen von Bacillus amyloliquefaciens-Subtilisin in Tabelle 1 entsprechen, wobei die Kombinationen von Substitutionen die Substitution einschließt, auf wel che oben Bezug genommen wird, wobei die Austausch-Aminosäure Valin ist, wobei die Protease eine verbesserte Waschleistung verglichen mit einer Vorläuferprotease aufweist.
- Stärker bevorzugte Proteasen sind jene, die Kombinationen von Substitutionen aufweisen, die jenen von Bacillus amyloliquefaciens-Subtilisin in Tabelle 2 entsprechen, wobei die Kombinationen von Substitutionen die Substitution einschließt, auf welche oben Bezug genommen wird, wobei die Austausch-Aminosäure Valin ist, und wobei die Protease eine verbesserte Waschleistung verglichen mit einer Vorläuferprotease aufweist.
- Weiter bevorzugte Proteasen sind jene, die Kombinationen von Substitutionen an Rest-Positionen aufweisen, die den Positionen von Bacillus amyloliquefaciens-Subtilisin in Tabelle 3 entsprechen, wobei die Kombinationen von Substitutionen die Substitution einschließt, auf welche oben Bezug genommen wird, wobei die Austausch-Aminosäure Valin ist, und wobei die Protease eine verbesserte Waschleistung verglichen mit einer Vorläuferprotease aufweist.
- Diese Aminosäurepositionsnummern beziehen sich auf jene, die der Sequenz des reifen Bacillus amyloliquefaciens-Subtilisins, die in
1 aufgeführt ist, zugewiesen sind. Die Erfindung ist jedoch nicht auf die Mutation dieses speziellen Subtilisins beschränkt, sondern erstreckt sich auf Vorläuferproteasen, welche Aminosäurereste an Positionen enthalten, die zu den jeweiligen bezeichneten Resten in Bacillus amyloliquefaciens-Subtilisin „äquivalent" sind. In einer bevorzugten Ausführungsform der vorliegenden Erfindung ist die Vorläuferprotease Bacillus lentus-Subtilisin und die Substitutionen werden an den äquivalenten Aminosäurerestpositionen bei B. lentus vorgenommen, welche jenen, die oben aufgelistet sind, entsprechen. - Eine Position eines Rests (Aminosäure) einer Vorläuferprotease ist äquivalent zu einem Rest von Bacillus amyloliquefaciens-Subtilisin, wenn sie entweder homolog (d. h. entsprechend hinsichtlich der Position in entweder der Primär- oder der Tertiärstruktur) oder analog zu einem speziellen Rest oder einem Teil jenes Rests in Bacillus amyloliquefaciens-Subtilisin ist (d. h. mit dem gleichen oder einem ähnlichen funktionalen Vermögen, zu kombinieren, zu reagieren oder chemisch zu Wechselwirken).
- Um eine Homologie hinsichtlich der Primärstruktur zu ermitteln, wird die Aminosäuresequenz einer Vorläuferprotease direkt mit der Primärsequenz des Bacillus amyloliquefaciens-Subtilisin und insbesondere mit einem Satz von Resten verglichen, welche bekanntermaßen in Subtilisinen, für welche die Sequenz bekannt ist, unveränderlich sind. Zum Beispiel zeigt
2 hierin die konservierten Reste, wie zwischen B. amyloliquefaciens-Subtilisin und B. lentus-Subtilisin. Nach dem Ausrichten (aligning) der konservierten Reste, wobei notwendige Insertionen und Deletionen, um die Ausrichtung beizubehalten, zugelassen werden (d. h. wobei der Ausschluss von konservierten Resten durch eine willkürliche Deletion und Insertion vermieden wird), werden die Reste definiert, die zu bestimmten Aminosäuren in der Primärsequenz von Bacillus amyloliquefaciens-Subtilisin äquivalent sind. Eine Ausrichtung von konservierten Resten sollte vorzugsweise 100% solcher Reste bewahren. Jedoch ist eine Ausrichtung von mehr als 75% oder von nur 50% der konservierten Reste ebenfalls ausreichend, um äquivalente Reste zu definieren. Eine Konservierung der katalytischen Triade Asp32/His64/Ser221 sollte aufrechterhalten werden. Siezen et al. (1991) Protein Eng. 4(7): 719–737 zeigt die Ausrichtung einer großen Anzahl von Serinproteasen. Siezen et al. beziehen sich auf die Eingruppierung als Subtilasen oder Subtilisinartige Serinproteasen. - Zum Beispiel sind in der
3 die Aminosäuresequenzen von Subtilisin aus Bacillus amyloliquefaciens, Bacillus subtilis, Bacillus licheniformis (carlsbergensis) und Bacillus lentus ausgerichtet, um das maximale Maß an Homologie zwischen den Aminosäuresequenzen bereit zu stellen. Ein Vergleich dieser Sequenzen zeigt, dass es eine Anzahl von konservierten Resten gibt, die in jeder Sequenz enthalten sind. Diese konservierten Reste (wie zwischen BPN' und B. lentus) sind in2 gekennzeichnet. - Diese konservierten Reste können somit verwendet werden, um die entsprechenden äquivalenten Aminosäurereste von Bacillus amyloliquefaciens-Subtilisin in anderen Subtilisinen, wie Subtilisin aus Bacillus lentus (PCT-Veröffentlichung Nr.
WO89/06279 EP 0 328 299 ), welches zu dem bevorzugten Bacillus lentus-Subtilisin hochgradig homolog ist, zu definieren. Die Aminosäuresequenzen von bestimmten dieser Subtilisine sind in den3A und3B gegenüber der Sequenz von Bacillus amyloliquefaciens-Subtilisin ausgerichtet, um die maximale Homologie von konservierten Resten zu erzeugen. Wie es erkannt werden kann, gibt es eine Anzahl von Deletionen in der Sequenz von Bacillus lentus, verglichen mit Bacillus amyloliquefaciens-Subtilisin. So ist zum Beispiel die äquivalente Aminosäure für Val165 bei Bacillus amyloliquefaciens-Subtilisin in den anderen Subtilisinen für B. lentus und B. licheniformis Isoleucin. - „Äquivalente Reste" können auch definiert werden, indem eine Homologie auf der Ebene der Tertiärstruktur für eine Vorläuferprotease ermittelt wird, deren Tertiärstruktur durch Röntgenkristallographie bestimmt wurde. Äquivalente Reste sind als jene definiert, bei welchen die Atomkoordinaten von zwei oder mehreren der Hauptkettenatome eines bestimmten Aminosäurerests der Vorläuferprotease und von Bacillus amyloliquefaciens-Subtilisin (N auf N, CA auf CA, C auf C und O auf O) nach dem Ausrichten innerhalb von 0,13 nm und vorzugsweise 0,1 nm liegen. Eine Ausrichtung wird erreicht, nachdem das beste Modell so orientiert und positioniert worden ist, dass es die maximale Überlappung von Atomkoordinaten von Nicht-Wasserstoff-Proteinatomen der fraglichen Protease gegenüber dem Bacillus amyloliquefaciens-Subtilisin ergibt. Das beste Modell ist das kristallographische Modell, welches den niedrigsten R-Faktor für experimentelle Beugungsdaten bei der höchsten verfügbaren Auflösung ergibt.
- Äquivalente Reste, die zu einem speziellen Rest von Bacillus amyloliquefaciens-Subtilisin funktional analog sind, sind als jene Aminosäuren der Vorläuferprotease definiert, die eine solche Konformation annehmen können, dass sie in einer Weise, die definiert ist und einem speziellen Rest von Bacillus amyloliquefaciens-Subtilisin zugeschrieben wird, die Proteinstruktur, Substratbindung oder Katalyse entweder verändern, modifizieren oder einen Beitrag dazu leisten. Ferner sind sie jene Reste der Vorläuferprotease (für welche eine Tertiärstruktur durch Röntgenkristallographie erhalten worden ist), die eine analoge Position in dem Maß innehaben, dass, obwohl die Hauptkettenatome des gegebenen Rests die Kriterien der Äquivalenz auf der Grundlage des Einnehmens einer homologen Position nicht erfüllen können, die Atomkoordinaten von wenigstens zwei der Seitenkettenatome des Rests bei 0,13 nm der entsprechenden Seitenkettenatome von Bacillus amyloliquefaciens-Subtilisin liegen. Die Koordinaten der dreidimensionalen Struktur von Bacillus amyloliquefaciens-Subtilisin sind in der
EPO-Veröffentlichung Nr. 0 251 446 (äquivalent zu demUS-Patent 5 182 204 ) dargelegt und können, wie oben erläutert, verwendet werden, um äquivalente Reste auf der Ebene der Tertiärstruktur zu bestimmen. - Einige der Reste, die für eine Substitution gekennzeichnet sind, sind konservierte Reste, wohingegen andere es nicht sind. Im Fall von Resten, die nicht konserviert sind, ist die Substitution von einer oder mehreren Aminosäuren auf Substitutionen beschränkt, die eine Variante erzeugen, die eine Aminosäuresequenz aufweist, die einer in der Natur gefundenen nicht entspricht. Im Fall von konservierten Resten sollten solche Substitutionen nicht zu einer natürlich vorkommenden Sequenz führen. Die Proteasen der vorliegenden Erfindung schließen die reifen Formen der Proteasen wie auch die Pro- und Präpro-Formen von solchen Proteasen ein. Die Präpro-Formen stellen die bevorzugte Konstruktion dar, da dies die Expression, Sekretion und Reifung der Proteasen erleichtert.
- „Prosequenz" bezieht sich auf eine an den N-terminalen Teil der reifen Form einer Protease gebundene Sequenz von Aminosäuren, die, wenn sie entfernt worden ist, zu dem Auftreten der „reifen" Form der Protease führt. Es werden viele proteolytische Enzyme in der Natur als translationale Proenzym-Produkte gefunden und werden in Abwesenheit einer posttranslationalen Prozessierung auf diese Weise exprimiert. Eine bevorzuge Prosequenz für das Herstellen der Proteasen der Erfindung ist die putative Prosequenz von Bacillus amyloliquefaciens-Subtilisin, obwohl andere Protease-Prosequenzen verwendet werden können.
- Eine „Signalsequenz" oder „Präsequenz" bezieht sich auf eine beliebige Sequenz von Aminosäuren, die an den N-terminalen Teil einer Protease oder an den N-terminalen Teil einer Pro-Protease gebunden ist, welche an der Sekretion der reifen oder Pro-Formen der Protease beteiligt sein können. Diese Definition der Signalsequenz ist eine funktionale, wobei beabsichtigt ist, all jene durch den N-terminalen Teil des Proteasegens codierten Aminosäuresequenzen, die an der Bewirkung der Sekretion von Protease unter nativen Bedingungen beteiligt sind, mit einzuschließen. Die vorliegende Erfindung nutzt solche Sequenzen, um die Sekretion der Proteasen, wie hierin definiert, zu bewirken. Eine mögliche Signalsequenz umfasst die ersten sieben Aminosäurereste der Signalsequenz aus Bacillus subtilis-Subtilisin, welche mit dem Rest der Signalsequenz des Subtilisins aus Bacillus lentus (ATCC 21536) fusioniert sind.
- Eine „Präpro"-Form einer Protease der Erfindung besteht aus der reifen Form der Protease, welche eine Prosequenz aufweist, welche mit dem Amino-Terminus der Protease funktionell verbunden ist, und eine „Prä"- oder „Signal"-Sequenz, welche mit dem Amino-Terminus der Prosequenz funktionell verbunden ist.
- „Expressionsvektor" bezieht sich auf ein DNA-Konstrukt, welches eine DNA-Sequenz enthält, welche mit einer geeigneten Kontrollsequenz, welche in der Lage ist, die Expression der DNA in einem geeigneten Wirt zu bewirken, funktionell verbunden ist. Solche Kontrollsequenzen umfas sen einen Promotor, um eine Transkription zu bewirken, eine optionale Operatorsequenz, um eine solche Transkription zu regulieren, eine Sequenz, die geeignete mRNA-Ribosomenbindungsstellen codiert, und Sequenzen, die die Termination der Transkription und Translation regulieren. Der Vektor kann ein Plasmid, ein Phagenpartikel oder einfach ein potentielles genomisches Insert sein. Sobald er in einen geeigneten Wirt transformiert worden ist, kann sich der Vektor unabhängig von dem Wirtsgenom replizieren und funktionieren oder kann sich, in einigen Fällen, in das Genom selbst integrieren. In der vorliegenden Beschreibung werden „Plasmid" und „Vektor" manchmal austauschbar verwendet, da das Plasmid gegenwärtig die am häufigsten verwendete Form eines Vektors darstellt. Die Erfindung soll jedoch solche anderen Formen von Expressionsvektoren mit einschließen, die äquivalenten Funktionen dienen und die in diesem Fachgebiet bekannt sind oder werden.
- Die in der vorliegenden Erfindung verwendeten „Wirtszellen" sind im Allgemeinen prokaryotische oder eukaryotische Wirte, die vorzugsweise durch die in dem
US-Patent RE 34 606 offenbarten Verfahren manipuliert wurden, um sie unfähig zu machen, enzymatisch aktive Endoprotease zu sekretieren. Eine bevorzugte Wirtszelle für die Expression von Protease ist der Bacillus-Stamm BG2036, dem es an einer enzymatisch aktiven neutralen Protease und alkalischen Protease (Subtilisin) fehlt. Die Konstruktion von Stamm BG2036 wird imUS-Patent 5 264 366 ausführlich beschrieben. Andere Wirtszellen für die Expression einer Protease schließen Bacillus subtilis I168 (auch beschrieben in demUS-Patent RE 34 606 und demUS-Patent 5 264 366 ) wie auch einen beliebigen geeigneten Bacillus-Stamm, wie B. licheniformis, B. lentus usw., mit ein. - Wirtszellen werden mit Vektoren transformiert oder transfiziert, die unter Verwendung von rekombinanten DNA-Techniken konstruiert worden sind. Solche transformierten Wirtszellen sind in der Lage, entweder Vektoren zu replizieren, die die Proteasevarianten codieren, oder die gewünschte Proteasevariante zu exprimieren. Im Fall von Vektoren, die die Prä- oder Präpro-Form der Proteasevariante codieren, werden solche Varianten, wenn sie exprimiert werden, üblicherweise aus der Wirtszelle in das Wirtszellmedium sekretiert.
- „Funktionell verbunden", wenn es die Beziehung zwischen zwei DNA-Regionen beschreibt, bedeutet einfach, dass sie funktionell zueinander in Beziehung stehen. Zum Beispiel ist eine Präsequenz funktionell mit einem Peptid verbunden, wenn sie als eine Signalsequenz fungiert, wobei sie an der Sekretion der reifen Form des Proteins beteiligt ist, was höchstwahrscheinlich mit einer Abspaltung der Signalsequenz verbunden ist. Ein Promotor ist mit einer codierenden Sequenz funktionell verbunden, wenn er die Transkription der Sequenz reguliert; eine Ribosomenbindungsstelle ist mit einer codierenden Sequenz funktionell verbunden, wenn sie so angeordnet ist, dass sie eine Translation erlaubt.
- Die Gene, die die in der Natur vorkommende Vorläuferprotease codieren, können gemäß den allgemeinen Verfahren, die den Fachleuten im Fachbereich bekannt sind, erhalten werden. Die Verfahren umfassen im Allgemeinen das Synthetisieren von markierten Sonden mit putativen Sequenzen, die die Regionen der Protease von Interesse codieren, das Herstellen von genomischen Bibliotheken aus die Protease exprimierenden Organismen und das Screenen der Bibliotheken nach dem Gen von Interesse durch eine Hybridisierung an die Sonden. Positiv hybridisierende Klone werden dann kartiert und sequenziert.
- Die klonierte Protease wird dann dazu verwendet, eine Wirtszelle zu transformieren, um die Protease zu exprimieren. Das Proteasegen wird dann in ein Plasmid mit hoher Kopienzahl ligiert. Dieses Plasmid repliziert sich in den Wirten in dem Sinn, dass es die gut bekannten Elemente enthält, die für die Plasmidreplikation erforderlich sind: einen Promotor, der mit dem fraglichen Gen funktionell verbunden ist (welcher als der eigene homologe Promotor des Gens bereit gestellt werden kann, wenn er durch den Wirt erkannt, d. h. transkribiert wird), eine Transkriptionsterminations- und Polyadenylierungsregion (die für die Stabilität der mRNA erforderlich ist, die durch den Wirt aus dem Proteasegen in bestimmten eukaryotischen Wirtszellen transkribiert wird), die exogen ist oder durch die endogene Terminatorregion des Proteasegens bereit gestellt wird, und wünschenswerterweise ein Selektions-Gen, wie ein Antibiotikumresistenz-Gen, das eine kontinuierliche Erhaltung von mit Plasmid infizierten Wirtszellen in Kultur durch das Wachsen in Antibiotikum enthaltenden Medien ermöglicht. Plasmide mit einer hohen Kopienzahl enthalten auch einen Replikationsstartpunkt für den Wirt, wodurch ermöglicht wird, dass große Zahlen von Plasmiden im Zytoplasma ohne chromosomale Beschränkungen erzeugt werden. Es liegt hierin jedoch innerhalb des Umfangs, vielfache Kopien des Proteasegens in das Wirtsgenom zu integrieren. Dies wird durch prokaryotische und eukaryotische Organismen erleichtert, die für eine homologe Rekombination besonders empfänglich sind.
- Das Gen kann ein natürliches B. lentus-Gen sein. Alternativ kann ein synthetisches Gen, das eine natürlich vorkommende oder mutierte Vorläuferprotease codiert, hergestellt werden. Bei einer solchen Herangehensweise wird die DNA- und/oder die Aminosäuresequenz der Vorläuferprotease bestimmt. Danach werden mehrere überlappende synthetische einzelsträngige DNA-Fragmente synthetisiert, die nach der Hybridisierung und Ligation eine synthetische DNA erzeugen, welche die Vorläuferprotease codiert. Ein Beispiel für die Konstruktion eines synthetischen Gens wird im Beispiel 3 des
US-Patents 5 204 015 dargestellt. - Sobald das natürlich vorkommende oder synthetische Gen der Vorläuferprotease kloniert worden ist, wird eine Zahl von Modifizierungen vorgenommen, um die Verwendung des Gens über die Synthese der natürlich vorkommenden Vorläuferprotease hinaus zu erweitern. Solche Modifizierungen schließen die Herstellung von rekombinanten Proteasen, wie es im
US-Patent RE 34 606 und derEPO-Veröffentlichung Nr. 0 251 446 offenbart ist, und die Herstellung der hierin beschrieben Proteasevarianten ein. - Das folgende Kassetten-Mutagenese-Verfahren kann dazu verwendet werden, die Konstruktion der Proteasevarianten der vorliegenden Erfindung zu erleichtern, obwohl auch andere Verfahren zur Anwendung kommen können. Als erstes wird das natürlich vorkommende, die Protease codierende Gen gewonnen und im Ganzen oder zum Teil sequenziert. Dann wird die Sequenz auf eine Stelle hin untersucht, an welcher es erwünscht ist, eine Mutation (Deletion, Insertion oder Substitution) von einer oder mehreren Aminosäuren in dem codierten Enzym vorzunehmen. Die diese Stelle flankierenden Sequenzen werden hinsichtlich der Gegenwart von Restriktionsstellen für das Ersetzen eines kurzen Segments des Gens mit einem Oligonukleotidpool, welcher, wenn er exprimiert wird, verschiedene Mutanten codieren wird, ausgewertet. Solche Restriktionsstellen sind vorzugsweise einmalige Stellen innerhalb des Proteasegens, um das Ersetzen des Gensegments zu erleichtern. Es kann jedoch eine beliebige zweckdienliche Restriktionsstelle, welche in dem Proteasegen nicht allzu sehr im Überfluss vorhanden ist, verwendet werden, unter der Voraussetzung, dass die durch den Restriktionsverdau erzeugten Genfragmente in der richtigen Reihenfolge erneut zusammengefügt werden können. Wenn Restriktionsstellen an Positionen innerhalb eines geeigneten Abstands von der gewählten Stelle (von 10 bis 15 Nukleotide) nicht vorhanden sind, werden solche Stellen erzeugt, indem Nukleotide in dem Gen in solch einer Art und Weise substituiert werden, dass weder das Leseraster noch die codierten Aminosäuren in der End-Konstruktion verändert werden. Eine Mutation des Gens, um seine Sequenz zu verändern, damit sie mit der gewünschten Sequenz übereinstimmt, wird durch eine M13-Primerverlängerung gemäß allgemein bekannter Verfahren bewerkstelligt. Die Aufgabe, geeignete flankierende Regionen ausfindig zu machen und die notwendigen Veränderungen zu bewerten, um zu zwei geeigneten Restriktionsstellen-Sequenzen zu gelangen, wird routinemäßig über die Redundanz des genetisches Codes, eine Restriktionsenzymkarte des Gens und die große Anzahl von unterschiedlichen Restriktionsenzymen durchgeführt. Es ist anzumerken, dass, wenn eine geeignete flankierende Restriktionsstelle zur Verfügung steht, das obige Verfahren nur in Verbindung mit der flankierenden Region, welche eine solche Stelle nicht enthält, angewendet werden muss.
- Sobald die natürlich vorkommende DNA oder die synthetische DNA kloniert worden ist, werden die Restriktionsstellen, die die zu mutierenden Positionen flankieren, mit den erkennenden Restriktionsenzymen verdaut und eine Vielzahl von zu den Endtermini komplementären Oligonukleotid-Kassetten werden in das Gen ligiert. Die Mutagenese wird durch dieses Verfahren vereinfacht, da alle Oligonukleotide so synthetisiert werden können, dass sie die gleichen Restriktionsstellen aufweisen, und es sind keine synthetischen Linker notwendig, um die Restriktionsstellen zu erzeugen.
- Wie hierin verwendet, ist die proteolytische Aktivität als die Rate der Hydrolyse der Peptidbindungen pro Milligramm an aktivem Enzym definiert. Es existieren zahlreiche gut bekannte Vorgehensweisen für die Messung der proteolytischen Aktivität (K. M. Kalisz, „Microbial Proteina ses." Advances in Biochemical Engineering/Biotechnology, A. Fiechter Hrsg., 1988). Darüber hinaus oder als eine Alternative zu einer modifizierten proteolytischen Aktivität, können die Enzyme der vorliegenden Erfindung andere modifizierte Eigenschaften, wie Km, kkat, kkat/Km-Verhältnis und/oder eine modifizierte Substratspezifität und/oder ein modifiziertes pH-Aktivitätsprofil aufweisen. Diese Enzyme können für das jeweilige Substrat, von dem erwartet wird, dass es vorhanden ist, zum Beispiel bei der Herstellung von Peptiden oder für hydrolytische Verfahren, wie Wäscherei-Anwendungen, maßgeschneidert werden.
- In einem Aspekt der Erfindung besteht das Ziel darin, eine Protease gemäß der Erfindung sicherzustellen, die eine veränderte, vorzugsweise verbesserte Waschleistung im Vergleich mit einer Vorläuferprotease in mindestens einer Waschmittel-Formulierung und oder unter mindestens einem Satz von Waschbedingungen aufweist.
- Es gibt eine Vielzahl von Waschbedingungen, was variierende(s) Waschmittel-Formulierungen, Waschwasservolumen, Waschwassertemperatur und Länge der Waschzeit einschließt, denen eine Proteasevariante ausgesetzt sein könnte. Zum Beispiel weisen Waschmittel-Formulierungen, die in unterschiedlichen Gegenden zum Einsatz kommen, unterschiedliche Konzentrationen ihrer im Waschwasser vorliegenden relevanten Komponenten auf. Zum Beispiel weist ein europäisches Waschmittel üblicherweise etwa 4500–5000 ppm an Waschmittelkomponenten im Waschwasser auf, während ein japanisches Waschmittel üblicherweise etwa 667 ppm an Waschmittelkomponenten im Waschwasser aufweist. In Nordamerika, insbesondere den Vereinigten Staaten weist ein Waschmittel üblicherweise etwa 975 ppm an im Waschwasser vorhandenen Waschmittelkomponenten auf.
- Ein System mit einer niedrigen Waschmittelkonzentration schließt Waschmittel ein, bei denen weniger als etwa 800 ppm an Waschmittelkomponenten im Waschwasser vorhanden sind. Japanische Waschmittel werden üblicherweise als Systeme mit niedriger Waschmittelkonzentration betrachtet, da sie etwa 667 ppm an im Waschwasser vorhandenen Waschmittelkomponenten aufweisen.
- Ein System mit einer mittleren Waschmittelkonzentration schließt Waschmittel ein, bei denen zwischen etwa 800 ppm und etwa 2000 ppm an Waschmittelkomponenten im Waschwasser vorhanden sind. Nordamerikanische Waschmittel werden im Allgemeinen als Systeme mit mittlerer Waschmittelkonzentration betrachtet, da sie etwa 975 ppm an im Waschwasser vorhandenen Waschmittelkomponenten aufweisen. Brasilien weist üblicherweise etwa 1500 ppm an im Waschwasser vorhandenen Waschmittelkomponenten auf.
- Ein System mit einer hoher Waschmittelkonzentration schließt Waschmittel ein, bei denen mehr als etwa 2000 ppm an Waschmittelkomponenten im Waschwasser vorhanden sind. Europäische Waschmittel werden im Allgemeinen als Systeme mit hoher Waschmittelkonzentration betrachtet, da sie etwa 4500–5000 ppm an Waschmittelkomponenten im Waschwasser aufweisen.
- Lateinamerikanische Waschmittel sind im Allgemeinen Waschmittel mit einem hohen Waschlaugen- und Phosphat-Builder-Gehalt und die Spanne von in Lateinamerika verwendeten Waschmitteln kann sowohl in die mittlere als auch in die hohe Waschmittelkonzentration fallen, da sie von 1500 ppm bis 6000 ppm an Waschmittelkomponenten im Waschwasser reichen. Wie oben erwähnt weist Brasilien üblicherweise etwa 1500 ppm an im Waschwasser vorhandenen Waschmittelkomponenten auf. Jedoch können andere Geographien mit Waschmitteln mit hohen Waschlaugen- und Phosphat-Builder-Gehalt, die nicht auf andere lateinamerikanische Länder beschränkt sind, Systeme mit einer hohen Waschmittelkonzentration aufweisen, wobei bis zu etwa 6000 ppm an Waschmittelkomponenten im Waschwasser vorhanden sind.
- Angesichts des Obigen ist es offensichtlich, dass Konzentrationen von Waschmittelzusammensetzungen in typischen Waschlösungen auf der ganzen Welt von weniger als etwa 800 ppm an Waschmittelzusammensetzung („Geographien mit einer niedrigen Waschmittelkonzentration"), zum Beispiel etwa 667 ppm in Japan, bis zu zwischen etwa 800 ppm und etwa 2000 ppm („Geographien mit einer mittleren Waschmittelkonzentration"), zum Beispiel etwa 975 ppm in den Vereinigten Staaten und etwa 1500 ppm in Brasilien, bis zu mehr als etwa 2000 ppm („Geographien mit einer hohen Waschmittelkonzentration"), zum Beispiel etwa 4500 ppm bis etwa 5000 ppm in Europa und etwa 6000 ppm in Geographien mit hohem Waschlaugen- und Phosphat-Builder-Gehalt, variieren.
- Die Konzentrationen der typischen Waschlösungen werden empirisch bestimmt. Zum Beispiel fasst in den Vereinigten Staaten eine typische Waschmaschine ein Volumen von etwa 64,4 l Waschlösung. Dementsprechend müssen, um eine Konzentration von etwa 975 ppm an Waschmittel innerhalb der Waschlösung zu erhalten, etwa 62,79 g der Waschmittelzusammensetzung zu den 64,41 Waschlösung hinzugegeben werden. Diese Menge ist die übliche Menge, die durch den Verbraucher unter Verwendung des mit dem Waschmittel bereit gestellten Messbechers in das Waschwasser abgemessen wird.
- Als ein weiteres Beispiel verwenden unterschiedliche Geographien unterschiedliche Waschtemperaturen. Die Temperatur des Waschwassers ist in Japan üblicherweise geringer als die, die in Europa verwendet wird.
- Dementsprechend schließt ein Aspekt der vorliegenden Erfindung eine Protease ein, die eine verbesserte Waschleistung bei mindestens einem Satz von Waschbedingungen aufweist.
- In einem weiteren Aspekt der Erfindung ist festgestellt worden, dass die Substitution einer Aminosäure an einer Rest-Position, die der Rest-Position 232 von Bacillus amyloliquefaciens- Subtilisin entspricht, bei der Verbesserung der Waschleistung des Enzyms wichtig ist, wobei die Austausch-Aminosäure Valin ist.
- Diese Substitutionen werden vorzugsweise bei Subtilisin von Bacillus lentus (rekombinant oder natürlichen Typs) durchgeführt, obwohl die Substitutionen bei einer beliebigen Bacillus-Protease durchgeführt werden können.
- Auf der Grundlage der Screening-Ergebnisse, die mit den Proteasen der Erfindung erhalten wurden, sind die erwähnten Mutationen bei Bacillus amyloliquefaciens-Subtilisin für die proteolytische Aktivität, die Leistungsfähigkeit und/oder Stabilität dieser Enzyme und die Reinigungs- oder Waschleistung solcher Enzyme wichtig.
- Viele der Proteasen der Erfindung sind bei der Formulierung von verschiedenen Reinigungszusammensetzungen oder Körperpflegeformulierungen wie Haarwaschmittel oder Lotionen nützlich. Eine Anzahl von bekannten Verbindungen sind geeignete grenzflächenaktive Stoffe bzw. Surfaktanzien, die in Zusammensetzungen, welche die Protease-Mutanten der Erfindung umfassen, nützlich sind. Diese schließen nichtionische, anionische, kationische oder zwitterionische Detergenzien ein, wie sie in der
US 4 404 128 von Barry J. Anderson und in derUS 4 261 868 von Jiri Flora, et al. offenbart sind. Eine geeignete Waschmittelformulierung ist jene, die im Beispiel 7 desUS-Patents 5 204 015 beschrieben wird. Der Fachbereich ist mit den verschiedenen Formulierungen vertraut, die als Reinigungszusammensetzungen verwendet werden können. Über die üblichen Reinigungszusammensetzungen hinaus wird es leicht verstanden, dass die Proteasen der vorliegenden Erfindung für einen beliebigen Zweck angewendet werden können, zu dem native oder Wildtyp-Proteasen verwendet werden. Somit können diese Proteasen zum Beispiel bei Seifenanwendungen im Stück oder flüssig, Geschirrpflegeformulierungen, Reinigungs-Lösungen oder -Produkte für Kontaktlinsen, bei der Peptidhydrolyse, Abfallbehandlung, bei Textilanwendungen, als Fusions-Spaltungs-Enzyme bei der Proteinproduktion usw. verwendet werden. Die Proteasen der vorliegenden Erfindung können eine erhöhte Leistungsfähigkeit in einer Reinigungszusammensetzung (verglichen mit dem Vorläufer) umfassen. Wie hierin verwendet, ist eine erhöhte Leistungsfähigkeit in einem Reinigungsmittel dahingehend definiert, dass es die Reinigung von bestimmten, gegenüber Enzymen empfindlichen Flecken, wie Gras oder Blut, erhöht, wie es durch eine übliche Auswertung nach einem Standard-Waschzyklus bestimmt wird. - Die Proteasen der Erfindung können zu bekannten pulverförmigen und flüssigen Waschmitteln mit einem pH-Wert zwischen 6,5 und 12,0 mit Gehalten von etwa 0,01 bis etwa 5 Gew.-% (vorzugsweise 0,1 Gew.-% bis 0,5 Gew.-%) formuliert werden. Diese Waschmittel-Reinigungszusammensetzungen können auch andere Enzyme, wie bekannte Proteasen, Amylasen, Cellulasen, Lipasen oder Endoglycosidasen, ebenso wie Builder und Stabilisatoren einschließen.
- Das Hinzufügen von Proteasen der Erfindung zu herkömmlichen Reinigungszusammensetzungen verursacht keine Beschränkung auf eine spezielle Anwendung. In anderen Worten, eine beliebige Temperatur und ein beliebiger pH-Wert, die für das Waschmittel geeignet sind, sind auch für die vorliegenden Zusammensetzungen geeignet, solange sich der pH-Wert innerhalb des oben genannten Bereichs befindet und die Temperatur unterhalb der Denaturierungstemperatur der beschriebenen Protease liegt. Darüber hinaus können die Proteasen der Erfindung in einer Reinigungszusammensetzung ohne Detergenzien, wieder entweder allein oder in Kombination mit Buildern und Stabilisatoren, verwendet werden.
- Die vorliegende Erfindung betrifft auch Reinigungszusammensetzungen, die die Proteasen der Erfindung enthalten. Die Reinigungszusammensetzungen können darüber hinaus Additive enthalten, die in Reinigungszusammensetzungen üblicherweise zum Einsatz kommen. Diese können aus Bleichmitteln, grenzflächenaktiven Stoffen, Buildern, Enzymen und Bleichkatalysatoren gewählt werden, ohne darauf beschränkt zu sein. Für einen Fachmann im Fachbereich würde es leicht ersichtlich sein, welche Additive für eine Aufnahme in die Zusammensetzungen geeignet sind. Die hierin bereitgestellte Liste ist auf keinen Fall erschöpfend und sollte nur als Beispiele geeigneter Additive genommen werden. Es wird auch für einen Fachmann im Fachbereich leicht ersichtlich sein, nur jene Additive zu verwenden, die mit den Enzymen und anderen Bestandteilen in der Zusammensetzung, zum Beispiel einem grenzflächenaktiven Stoff, verträglich sind.
- Sofern vorhanden, beträgt die Menge an Additiven, die in der Reinigungszusammensetzung vorliegt, von etwa 0,01% bis etwa 99,9%, vorzugsweise von etwa 1% bis etwa 95%, stärker bevorzugt von etwa 1% bis etwa 80%.
- Ein Aspekt der Erfindung ist eine Zusammensetzung für die Behandlung einer Textilie, welche die varianten Proteasen der vorliegenden Erfindung einschließt. Die Zusammensetzung kann dazu verwendet werden, um zum Beispiel Seide oder Wolle zu behandeln, wie es in Veröffentlichungen wie RD 216 034;
EP 134 267 US 4 533 359 undEP 344 259 - Das Folgende wird als Beispiel dargelegt und soll nicht als eine Beschränkung hinsichtlich des Umfangs der Ansprüche ausgelegt werden.
- Beispiel 1
- Es wurde eine große Anzahl an Protease-Varianten unter Verwendung von Verfahren, die im Fachbereich gut bekannt sind, hergestellt und gereinigt. Alle Mutationen wurden bei Bacillus lentus GG36-Subtilisin durchgeführt. Die Varianten sind in der Tabelle 4 gezeigt.
- Beispiel 2
- Eine große Anzahl der in Beispiel 1 hergestellten Protease-Varianten wurden auf die Leistungsfähigkeit bei zwei Arten von Waschmittel- und Waschbedingungen unter Verwendung eines Mikrostoffproben-Assays getestet, der in „An improved method of assaying for a preferred enzyme and/or preferred detergent composition", US-Serien-Nr. 60/068 796 beschrieben ist.
- Tabelle 5 listet die getesteten varianten Proteasen und die Ergebnisse des Testens bei zwei verschiedenen Waschmitteln auf. Für Spalte A war das Waschmittel 0,67 g/l gefiltertes Ariel Ultra (Procter & Gamble, Cincinnati, OH, USA), in einer Lösung, welche 3 Grains pro Gallone gemischter Ca2+/Mg2+-Härte enthielt, und es wurden 0,3 ppm Enzym in jeder Vertiefung bei 20°C verwendet. Für Spalte B war das Waschmittel 3,38 g/l gefiltertes Ariel Futur (Procter & Gamble, Cincinnati, OH, USA), in einer Lösung, welche 15 Grains pro Gallone gemischter Ca2 +/Mg2 +-Härte enthielt, und es wurden 0,3 ppm Enzym in jeder Vertiefung bei 40°C verwendet.
- Beispiel 3
- Tabelle 6 listet die getesteten varianten Proteasen aus Beispiel 1 und die Ergebnisse des Testens bei vier verschiedenen Waschmitteln auf. Die gleichen Leistungsfähigkeitstests wie in Beispiel 2 wurden an den erwähnten varianten Proteasen mit den folgenden Waschmitteln ausgeführt. Für Spalte A war das Waschmittel 0,67 g/l gefiltertes Ariel Ultra (Procter & Gamble, Cincinnati, OH, USA), in einer Lösung, welche 3 Grains pro Gallone gemischter Ca2+/Mg2+-Härte enthielt, und es wurden 0,3 ppm Enzym in jeder Vertiefung bei 20°C verwendet. Für Spalte B war das Waschmittel 3,38 g/l gefiltertes Ariel Futur (Procter & Gamble, Cincinnati, OH, USA), in einer Lösung, welche 15 Grains pro Gallone gemischter Ca2+Mg2+-Härte enthielt, und es wurden 0,3 ppm Enzym in jeder Vertiefung bei 40°C verwendet. Für Spalte C wurden 3,5 g/l HSP1-Waschmittel (Procter & Gamble, Cincinnati, OH, USA), in einer Lösung, welche 8 Grains pro Gallone gemischter Ca2 +/Mg2+-Härte enthielt, und 0,3 ppm Enzym in jeder Vertiefung bei 20°C verwendet. Für Spalte D wurden 1,5 ml/l Tide KT-Waschmittel (Procter & Gamble, Cincinnati, OH, USA), in einer Lösung, welche 3 Grains pro Gallone gemischter Ca2+/Mg2+-Härte enthielt, und 0,3 ppm Enzym in jeder Vertiefung bei 20°C verwendet.
Claims (18)
- Protease, die eine Aminosäuresubstitution an einer Rest-Position umfasst, die der Position des Restes 232 von Bacillus amyloliquefaciens-Subtilisin entspricht, wobei die Austausch-Aminosäure Valin ist und wobei die Protease eine verbesserte Waschleistung im Vergleich zu einer Vorläuferprotease ohne die Substitution aufweist.
- Protease gemäß Anspruch 1, wobei die Substitution A232V ist.
- Protease gemäß Anspruch 1 oder Anspruch 2, welche von einem Bacillus-Subtilisin abgeleitet ist.
- Protease gemäß Anspruch 3, welche von einem Bacillus lentus-Subtilisin abgeleitet ist.
- DNA, welche eine Protease von Anspruch 1 oder Anspruch 2 codiert.
- Expressionsvektor, welcher die DNA von Anspruch 5 codiert.
- Wirtszelle, die mit dem Expressionsvektor von Anspruch 6 transformiert ist.
- Reinigungszusammensetzung, welche die Protease von Anspruch 1 oder Anspruch 2 umfasst.
- Zusammensetzung zur Behandlung einer Textilie, welche die Protease von Anspruch 1 oder Anspruch 2 umfasst.
- Protease gemäß Anspruch 1 oder Anspruch 2, welche einen Substitutionssatz umfast, gewählt aus der Gruppe, bestehend aus Substitutionen an Rest-Positionen, entsprechend den Positionen in Tabelle 1 von Bacillus amyloliquefaciens-Subtilisin, wobei der Substitutionssatz die Substitution einschließt, auf welche in Anspruch 1 Bezug genommen wird, wobei die Austausch-Aminosäure Valin ist.
- Protease gemäß Anspruch 10, welche einen Substitutionssatz umfasst, gewählt aus der Gruppe, bestehend aus Substitutionen, welche denjenigen in Tabelle 2 von Bacillus amyloliquefaciens-Subtilisin entsprechen, wobei der Substitutionssatz die Substitution einschließt, auf welche in Anspruch 1 Bezug genommen wird, wobei die Austausch-Aminosäure Valin ist.
- Protease gemäß Anspruch 10, welche einen Substitutionssatz umfasst, gewählt aus der Gruppe, bestehend aus Substitutionen an Rest-Positionen, entsprechend den Positionen in Tabelle 3 von Bacillus amyloliquefaciens-Subtilisin, wobei der Substitutionssatz die Substitution einschließt, auf welche in Anspruch 1 Bezug genommen wird, wobei die Austausch-Aminosäure Valin ist.
- Protease von Anspruch 1 oder Anspruch 2, welche ferner Substitutionen der Aminosäure-Reste an einer oder mehreren der Positionen 62, 230, 212, 252 oder 257 einschließt.
- Protease von Anspruch 1 oder Anspruch 2, welche einen Substitutionssatz umfasst, gewählt aus der Gruppe, die Substitutionen an jeder einzelnen der Rest-Positionen, entsprechend den Rest-Positionen 68, 103, 104, 159, 232, 236 und 245 von Bacillus amyloliquefaciens-Subtilisin, einschließt, wobei die Austausch-Aminosäure an der Position, die der Position 232 entspricht, Valin ist.
- Protease von Anspruch 1 oder Anspruch 2, welche einen Substitutionssatz umfasst, gewählt aus der Gruppe, die Substitutionen an jeder einzelnen der Rest-Positionen einschließt, entsprechend den Rest-Positionen 103, 104, 159, 232, 236, 245, 248 und 252 von Bacillus amyloliquefaciens-Subtilisin, wobei die Austausch-Aminosäure an der Position, die der Position 232 entspricht, Valin ist.
- Protease von Anspruch 1 oder Anspruch 2, welche einen Substitutionssatz umfasst, gewählt aus der Gruppe, die jede einzelne der Substitutionen V68A, S103A, V104I, G159D, A232V, Q236H und Q245R einschließt.
- Protease von Anspruch 1 oder Anspruch 2, welche einen Substitutionssatz umfasst, gewählt aus der Gruppe, die jede der Substitutionen S103A, V104I, G159D, A232V, Q236H, Q245R, N248D und N252K einschließt.
- Protease von Anspruch 17, wobei der Substitutionssatz ferner die Substitution S101G einschließt.
