- Die Anmeldung ist eine „Continuation-in-part"-Anmeldung der U.S.-Patentanmeldung 08/956,323, eingereicht am 23. Oktober 1998, der U.S.-Patentanmeldung 08/956,564, eingereicht am 23. Oktober 1998 und der U.S.-Patentanmeldung 08/956,324, eingereicht am 23. Oktober 1998, die allesamt hiermit in ihrer Gänze in diese Unterlagen aufgenommen werden.
- Hintergrund der Erfindung
- Serinproteasen sind eine Untergruppe der Carbonylhydrolasen. Sie umfassen verschiedene Klassen von Enzymen mit einem breiten Spektrum von Spezifitäten und biologischen Funktionen. Stroud, R., Sci. Amer., 131:74-88. Trotz ihrer funktionalen Verschiedenartigkeit haben sich der katalytischen Maschinerie von Serinproteasen wenigstens zwei genetisch unterschiedliche Familien von Enzymen bedient: 1) die Subtilisine und 2) die aus Säugetieren stammenden, mit Chymotrypsin verwandten und homologen bakteriellen Serinproteasen (z.B. Trypsin und S. gresius-Trypsin). Diese beiden Familien von Serinproteasen zeigen bemerkenswert ähnliche Katalysemechanismen. Kraut, J. (1977), Annu. Rev. Biochem., 46:331-358. Darüber hinaus bringt, obwohl die Primärstruktur keine Verwandtschaft aufweist, die Tertiärstruktur von diesen zwei Enzymfamilien eine konservierte katalytische Triade von Aminosäuren, welche aus Serin, Histidin und Asparat besteht, zusammen.
- Subtilisine sind Serinproteasen (MG ungefähr 27.500), die in großen Mengen durch eine große Vielzahl von Bacillus-Spezies und anderer Mikroorganismen sekretiert werden. Die Proteinsequenz von Subtilisin ist aus wenigstens neun unterschiedlichen Bacillus-Spezies bestimmt worden. Markland, F.S. et al. (1983), Hoppe Seyler's Z. Physiol. Chem., 364:1537-1540. Es ist über die dreidimensionale kristallographische Struktur von Subtilisinen aus Bacillus amyloliquefaciens, Bacillus licheniformis und mehreren natürlichen Varianten von B. lentus berichtet worden. Diese Untersuchungen zeigen, dass obwohl Subtilisin mit den Serinproteasen aus Säugetieren genetisch nicht verwandt ist, es eine ähnliche Struktur im Bereich des aktiven Zentrums aufweist. Die Röntgen-Kristallstrukturen von Subtilisin, das kovalent gebundene Peptidinhibitoren (Robertus, J.D. et al. (1972), Biochemistry, 11:2439-2449) oder Produktkomplexe (Robertus, J.D. et al., (1976), J. Biol. Chem., 251:1097-1103) enthält, haben ebenfalls Informationen über das aktive Zentrum und die mutmaßliche Substratbindungungsspalte von Subtilisin geliefert. Zusätzlich ist eine große Anzahl von kinetischen und chemischen, Biol. Chem., 244:5333-5338) und eine umfassende ortsspezifische Mutagenese ausgeführt worden (Wells und Estell (1988) TIBS 13:291-297).
- Zusammenfassung der Erfindung

- Obwohl eine jegliche Kombination der oben aufgelisteten Aminosäuresubstitutionen eingesetzt werden kann, umfassen die bevorzugten Proteasevariante-Enzyme, die für die Erfindung nützlich sind, die Substitution von Aminosäureresten in den folgenden Kombinationen von Positionen. Alle der Positionen von Resten entsprechen Positionen von Bacillus amyloliquefaciens-Subtilisin:
- (1) eine Proteasevariante, welche Substitutionen der Aminosäurereste an Position 236 umfasst;
- (2) eine Proteasevariante, die Substitutionen der Aminosäurereste an den Positionen 103 und 245 und an einer oder mehreren der folgenden Positionen 1, 9, 12, 61, 62, 68, 76, 97, 98, 101, 102, 104, 109, 130, 131, 159, 170, 183, 185, 205, 209, 210, 211, 212, 213, 215, 217, 222, 230, 232, 248, 252, 257, 260, 261, 270 und 275 umfasst; oder
- (3) eine Proteasevariante, die Substitutionen der Aminosäurereste an den Positionen 103, 236 und 245 und an einer oder mehreren der folgenden Positionen 1, 9, 12, 61, 62, 68, 76, 97, 98, 101, 102, 104, 109, 130, 131, 159, 183, 185, 205, 209, 210, 211, 212, 213, 215, 217, 230, 232, 243, 248, 252, 257, 260, 270 und 275 umfasst.

- Die am meisten bevorzugten Proteasevarianten sind diejenigen, die in Tabelle 3 gezeigt sind.
- Es ist ein weiterer Gegenstand, DNA-Sequenzen, die solche Proteasevarianten kodieren, wie auch Expressionsvektoren, die solche abgewandelten DNA-Sequenzen enthalten, bereitzustellen.
- Noch weiter besteht ein anderer Gegenstand der Erfindung darin, Wirtszellen, die mit solchen Vektoren transformiert sind, wie auch Wirtszellen, die in der Lage sind, eine derartige DNA zu exprimieren, um Proteasevarianten entweder intrazellulär oder extrazellulär herzustellen, bereitzustellen.
- Es wird des weiteren eine Reinigungszusammensetzung bereitgestellt, welche eine Proteasevariante der Erfindung umfasst.
- Zusätzlich wird ein Tierfutter bereitgestellt, welches eine Proteasevariante der Erfindung umfasst.
- Ebenfalls bereitgestellt wird eine Zusammensetzung für die Behandlung eines Gewebes, welche eine Proteasevariante der Erfindung umfasst.
- KURZE BESCHREIBUNG DER ZEICHNUNGEN
- Die
1A -1C zeigen die DNA- und Aminosäuresequenz für Bacillus amyloliquefaciens-Subtilisin und eine partielle Restriktionskarte dieses Gens. -
2 zeigt die konservierten Aminosäurereste zwischen den Subtilisinen aus Bacillus amyloliquefaciens (BPN)' und Bacillus lentus (Wildtyp). - Die
3A und3B zeigen die Aminosäuresequenz von vier Subtilisinen. Die obere Zeile repräsentiert die Aminosäuresequenz von Subtilisin aus Bacillus amyloliquefaciens-Subtilisin (manchmal auch als Subtilisin BPN' bezeichnet). Die zweite Zeile zeigt die Aminosäuresequenz von Subtilisin aus Bacillus subtilis. Die dritte Zeile zeigt die Aminosäuresequenz von Subtilisin aus Bacillus licheniformis. Das Symbol * bezeichnet das Fehlen von speziellen Aminosäureresten verglichen mit dem Subtilisin BPN'. - Detaillierte Beschreibung der Erfindung
- Proteasen sind Carbonylhydrolasen, die allgemein wirksam werden, um Peptidbindungen von Proteinen oder Peptiden zu spalten. Wie hier verwendet, bedeutet „Protease" eine in der Natur vorkommende Protease oder eine rekombinante Protease. In der Natur vorkommende Proteasen umfassen α-Aminoacylpeptidhydrolase, Peptidylaminosäurehydrolase, Acylaminohydrolase, Serincarboxypeptidase, Metallocarboxypeptidase, Thioproteinase, Carboxylproteinase und Metalloproteinase. Es werden Serin-, Metallo-, Thiol- und saure Proteasen mit umfasst wie auch Endo- und Exoproteasen.
- Die Erfindung umfasst Proteaseenzyme, die in der Natur nicht vorkommende Carbonylhydrolasevarianten (Proteasevarianten) mit einer unterschiedlichen proteolytischen Aktivität, Stabilität, Substratspezifität, pH-Profil und/oder Leistungseigenschaft verglichen mit der Vorläufer-Carbonylhydrolase, von welcher die Aminosäuresequenz der Variante abgeleitet ist, sind. Speziell weisen solche Proteasevarianten eine Aminosäuresequenz auf, die in der Natur nicht gefunden wird, die durch Substitution einer Mehrzahl von Aminosäureresten einer Vorläufer-Protease durch unterschiedliche Aminosäuren abgeleitet ist. Die Vorläufer-Protease kann eine in der Natur vorkommende Protease oder rekombinante Protease sein.
- Die Proteasevarianten, die hier nützlich sind, umfassen die Substitution einer jeglichen der neunzehn, in der Natur vorkommenden L-Aminosäuren an den bezeichneten Aminosäurerestpositionen. Solche Substitutionen können bei einem jeglichen Vorläufer-Subtilisin (prokaryotisch, eukaryotisch, aus Säugetieren u.s.w.) vorgenommen werden. In dieser gesamten Anmeldung wird auf verschiedene Aminosäuren mittels der üblichen Ein- und Drei-Buchstaben-Codes Bezug genommen. Solche Codes werden in Dale, M.W. (1989), Molecular Genetics of Bacteria, John Wiley & Sons, Ltd., Anhang B, identifiziert.
- Die Proteasevarianten, die hier nützlich sind, sind vorzugsweise von einem Bacillus-Subtilisin abgeleitet. Die Proteasevarianten sind von Bacillus lentus-Subtilisin und/oder Subtilisin 309 abgeleitet.
- Subtilisine sind bakterielle oder pilzliche Proteasen, die allgemein wirksam werden, um Peptidbindungen von Proteinen oder Peptiden zu spalten. Wie hier verwendet, bedeutet „Subtilisin" ein in der Natur vorkommendes Subtilisin oder ein rekombinantes Subtilisin. Eine Reihe von in der Natur vorkommenden Subtilisinen wird bekanntermaßen von verschiedenen mikrobiellen Spezies produziert und oftmals sekretiert. Aminosäuresequenzen von den Mitgliedern dieser Reihe sind nicht vollständig homolog. Jedoch zeigen die Subtilisine in dieser Reihe die gleiche oder eine ähnliche Art von proteolytischer Aktivität. Diese Klasse von Serinproteasen weist eine gemeinsame Aminosäuresequenz auf, die eine katalytische Triade definiert, die diese von der mit Chymotrypsin verwandten Klasse von Serinproteasen unterscheidet. Die Subtilisine und mit Chymotrypsin verwandten Serinproteasen weisen beide eine katalytische Triade auf, die Aspartat, Histidin und Serin umfasst. Bei den mit Subtilisin verwandten Proteasen ist die relative Reihenfolge von diesen Aminosäuren, gelesen vom Amino- zum Carboxyterminus, Aspartat-Histidin-Serin. Bei den mit Chymotrypsin verwandten Proteasen ist die relative Reihenfolge jedoch Histidin-Aspartat-Serin. So bezieht sich Subtilisin hier auf eine Serinprotease, die die katalytische Triade von mit Subtilisin verwandten Proteasen aufweist. Beispiele umfassen, sind aber nicht beschränkt auf die hier in
3 identifizierten Subtilisine. Im Allgemeinen und für die Zwecke der Erfindung entspricht die Nummerierung der Aminosäuren in Proteasen den Nummern, die der in1 aufgeführten Sequenz von reifem Bacillus amyloliquefaciens-Subtilisin zugewiesen wurden. - „Rekombinantes Subtilisin" oder „rekombinante Protease" beziehen sich auf ein Subtilisin oder eine Protease, bei dem bzw. der die das Subtilisin oder die Protease kodierende DNA-Sequenz so modifiziert ist, dass eine abgewandelte (oder mutierte) DNA-Sequenz hergestellt wird, die die Substitution, Deletion oder Insertion von einer oder mehreren Aminosäuren in der in der Natur vorkommenden Aminosäurese quenz kodiert. Geeignete Verfahren, um solche Modifikationen herzustellen, und die mit jenen, die hier offenbart werden, kombiniert werden können, umfassen jene, die in dem U.S.-Patent RE 34,606, U.S.-Patent 5,204,015 und U.S.-Patent 5,185,258, U.S.-Patent 5,700,676, U.S.-Patent 5,801,038 und U.S.-Patent 5,763,257 offenbart werden.
- „Nicht-humane Subtilisine" und die DNA, die diese kodiert, können aus vielen prokaryotischen und eukaryotischen Organismen erhalten werden. Geeignete Beispiele von prokaryotischen Organismen umfassen gramnegative Organismen, wie E. coli oder Pseudomonas, und grampositive Bakterien, wie Micrococcus oder Bacillus. Beispiele von eukaryotischen Organismen, aus welchen Subtilisin und deren Gene erhalten werden können, umfassen Hefe, wie Saccharomyces cerevisiae, Pilze, wie Aspergillus-Spezies.
- Eine „Proteasevariante" hat eine Aminosäuresequenz, die von der Aminosäuresequenz einer „Vorläufer-Protease" abgeleitet ist. Die Vorläufer-Proteasen umfassen in der Natur vorkommende Proteasen und rekombinante Proteasen. Die Aminosäuresequenz der Proteasevariante ist von der Aminosäuresequenz der Vorläufer-Protease „abgeleitet" durch die Substitution, Deletion oder Insertion von einer oder mehreren Aminosäuren der Vorläufer-Aminosäuresequenz. Solche Modifikationen erfolgen an der „Vorläufer-DNA-Sequenz", welche die Aminosäuresequenz der Vorläufer-Protease kodiert, anstelle einer Manipulation des Vorläufer-Proteaseenzyms per se. Geeignete Methoden für eine derartige Manipulation der Vorläufer-DNA-Sequenz umfassen Methoden, die hier offenbart werden, wie auch Verfahren, die den Fachleuten auf diesem Gebiet bekannt sind (siehe beispielsweise

- Bevorzugte Varianten sind diejenigen, die Kombinationen von Substitutionen an Positionen von Resten, die Positionen von Bacillus amyloliquefaciens-Subtilisin in Tabelle 1 entsprechen, aufweisen. Mehr bevorzugte Varianten sind diejenigen, die Kombinationen von Substitutionen an Positionen von Resten, welche Positionen von Bacillus amyloliquefaciens-Subtilisin in Tabelle 3 entsprechen, aufweisen.
- Weitere bevorzugte Varianten sind jene, die Kombinationen von Substitutionen an Positionen von Resten, die Positionen von Bacillus amyloliquefaciens-Subtilisin in Tabelle 2 entsprechen, aufweisen.
- Diese Aminosäurepositionsnummern beziehen sich auf jene, die der in
1 aufgeführten Sequenz von reifem Bacillus amyloliquefaciens-Subtilisin zugewiesen worden sind. Die Erfindung ist jedoch nicht auf die Mutation dieses speziellen Subtilisins beschränkt, sondern erstreckt sich auf Vorläufer-Proteasen, die Aminosäurereste an Positionen, die zu den jeweiligen identifizierten Resten in Bacillus amyloliquefaciens-Subtilisin „äquivalent" sind, enthalten. In einer bevorzugten Ausführungsform der Erfindung ist die Vorläufer-Protease Bacillus lentos-Subtilisin und die Substitutionen werden an den äquivalenten Positionen von Aminosäureresten in B. lentus, welche jenen, die oben aufgelistet worden sind, entsprechen, vorgenommen. - Eine Position eines Rests (einer Aminosäure) einer Vorläufer-Protease ist äquivalent zu einem Rest von Bacillus amyloliquefacies-Subtilisin, wenn sie entweder homolog (d.h. entsprechend hinsichtlich der Position entweder in der Primär- oder Tertiärstruktur) oder analog zu einem bestimmten Rest oder Abschnitt jenes Rests in Bacillus amyloliquefaciens-Subtilisin ist (d.h. mit dem gleichen oder einem ähnlichen funktionalen Vermögen, eine Bindung oder Assoziation einzugehen, zu reagieren oder chemisch zu wechselwirken).
- Um Homologie zu einer Primärstruktur zu ermitteln, wird die Aminosäuresequenz einer Vorläufer-Protease direkt mit der Primärsequenz des Bacillus amyloliquefaciens-Subtilisins und insbesondere mit einem Satz von Resten, der bekanntermaßen in Subtilisinen, für welche die Sequenz bekannt ist, unveränderlich ist, verglichen. Beispielsweise zeigt
2 hier die konservierten Reste, wie sie zwischen B. amyloliquefaciens-Subtilisin und B. lentus-Subtilisin bestehen. Nach gegenseitiger Ausrichtung anhand von Homologie („Alignment") der konservierten Reste, wobei erforderliche Insertionen und Deletionen, um die gegenseitige Ausrichtung anhand von Homologie („Alignment") aufrecht zu erhalten (d.h. Vermeiden der Beseitigung von konservierten Resten durch willkürliche De Deletion und Insertion), zugelassen werden, werden die Reste, die zu bestimmten Aminosäuren in der Primärsequenz von Bacillus amyloliquefaciens-Subtilisin äquivalent sind, definiert. Eine Ausrichtung anhand von Homologie („Alignment") von konservierten Resten sollte vorzugsweise 100% von solchen Resten bewahren. Jedoch ist auch eine Ausrichtung anhand von Homologie („A lignment") von mehr als 75% oder von so wenig wie 50% der konservierten Reste adäquat, um äquivalente Reste zu definieren. Die Konservierung der katalytischen Triade, Asp32/His64/Ser221, sollte beibehalten werden. Siezen et al. (1991) Protein Eng. 4(7):719-737, zeigen die Ausrichtung anhand von Homologie („Alignment") einer großen Anzahl von Serinproteasen. Siezen et al. beziehen sich auf die Einstufung als Subtilasen oder Subtilisin-artige Serinproteasen. - Beispielsweise sind in
3 die Aminosäuresequenz von Subtilisin aus Bacillus amyloliquefaciens, Bacillus subtilis, Bacillus licheniformis (carlsbergensis) und Bacillus lentus gegenseitig anhand von Homologie ausgerichtet („Alignment), um das maximale Ausmaß von Homologie zwischen Aminosäuresequenzen zu ermöglichen. Ein Vergleich von diesen Sequenzen zeigt, dass es eine Anzahl von konservierten Resten, die in jeder Sequenz enthalten sind, gibt. Diese konservierten Reste (wie beispielsweise zwischen BPN' und B. lentus) sind in2 angegeben. - Diese konservierten Reste können dementsprechend verwendet werden, um die entsprechenden äquivalenten Aminosäurereste von Bacillus amyloliquefaciens-Subtilisin in anderen Subtilisinen, wie beispielsweise Subtilisin aus Bacillus lentus (PCT-Veröffentlichung Nr. WO 89/06279, veröffentlich am 13. Juli 1989), das hier bevorzugte Protease-Vorläufer-Enzym, oder dem Subtilisin, das als PB92 bezeichnet wird (
EP 0 328 299 ), das zu dem bevorzugten Bacillus lentus-Subtilisin hochgradig homolog ist, zu definieren. Die Aminosäuresequenz von bestimmten von diesen Subtilisinen sind in den3A und3B gegenüber der Sequenz von Bacillus amyloliquefaciens-Subtilisin anhand von Homologie ausgerichtet („Alignment"), um die maximale Homologie von konservierten Resten herzustellen. Wie ersehen werden kann, gibt es eine Anzahl von Deletionen in der Sequenz von Bacillus lentus verglichen mit dem Bacillus amyloliquefaciens-Subtilisin. So ist beispielsweise die äquivalente Aminosäure für Val165 in Bacillus amyloliquefaciens-Subtilisin in den anderen Subtilisinen Isoleucin für B. lentus und B. licheni formis. - „Äquivalente Reste" können auch durch Bestimmung der Homologie auf der Ebene der Tertiärstruktur für eine Vorläufer-Protease, deren Tertiärstruktur durch Röntgenkristallographie bestimmt worden ist, definiert werden. Äquivalente Reste werden als diejenigen definiert, bei denen die Atomkoordinaten von zwei oder mehreren der Hauptkettenatome eines bestimmten Aminosäurerests der Vorläufer-Protease und von Bacillus amyloliquefaciens-Subtilisin (N gegenüber N, CA gegenüber CA, C gegenüber C und O gegenüber O) nach der gegenseitigen Ausrichtung anhand von Homologie („Alignment") innerhalb von 0,13 nm und vorzugsweise 0,1 nm liegen. Die gegenseitige Ausrichtung anhand von Homologie („Alignment") wird erreicht, nachdem das beste Modell so orientiert und positioniert worden ist, dass die maximale Überlappung der Atomkoordinaten von Nicht-Wasserstoff-Proteinatomen der fraglichen Protease mit dem Bacillus amyloliquefaciens-Subtilisin erhalten wird. Das beste Modell ist das kristallographische Modell, das den niedrigsten R-Faktor für experimentelle Beugungsdaten bei der höchsten verfügbaren Auflösung liefert.
- Äquivalente Reste, die funktional analog zu einem bestimmten Rest von Bacillus amyloliquefaciens-Subtilisin sind, werden als jene Rminosäuren der Vorläufer-Protease definiert, die eine solche Konformation annehmen können, dass sie die Proteinstruktur, die Substratbindung oder die Katalyse in einer definierten Weise und einer Weise, die einem bestimmten Rest des Bacillus amyloliquefaciens-Subtilisin zugeschrieben wird, entweder verändern, modifizieren oder dazu beitragen. Ferner sind sie jene Reste der Vorläufer-Protease (für welche eine Tertiärstruktur durch Röntgenkristallographie erhalten worden ist), die eine analoge Position einnehmen in dem Ausmaß, dass, obwohl die Hauptkettenatome des gegebenen Rests die Äquivalenzkriterien auf der Grundlage der Einnahme einer homologen Position möglicherweise nicht erfüllen, die Atomkoordinaten von wenigstens zwei der Seitenkettenatome des Rests innerhalb von 0,13 nm der entsprechenden Seitenkettenatome von Bacillus amyloliquefaciens-Subtilisin liegen. Die Koordinaten der dreidimensionalen Struktur von Bacillus amyloliquefaciens-Subtilisin sind in der EPA-Veröffentlichung Nr. 0 251 446 (äquivalent zu dem U.S.-Patent 5,182,204, dessen Offenbarung in diese Unterlagen unter Bezugnahme aufgenommen wird) angegeben und können, wie oben erläutert, verwendet werden, um äquivalente Reste auf der Ebene der Tertiärstruktur zu bestimmen.
- Einige der hinsichtlich einer Substitution identifizierten Reste sind konservierte Reste, wohingegen andere dies nicht sind. In dem Falle von Resten, die nicht konserviert sind, ist die Substitution von einer oder mehreren Aminosäuren auf Substitutionen beschränkt, die eine Variante erzeugen, die eine Aminosäuresequenz aufweist, die keiner, die in der Natur gefunden wird, entspricht. In dem Falle von konservierten Resten sollten solche Substitutionen nicht zu einer in der Natur vorkommenden Sequenz führen. Die Proteasevarianten der Erfindung umfassen die reifen Formen von Proteasevarianten wie auch die Pro- und Präpro-Formen von solchen Proteasevarianten. Die Präpro-Formen sind die bevorzugten Konstruktion, da diese die Expression, Sekretion und Reifung der Proteasevarianten vereinfacht.
- „Prosequenz" bezieht sich auf eine Sequenz von Aminosäuren, die an den N-terminalen Abschnitt der reifen Form einer Protease gebunden ist, die, wenn sie entfernt wird, zu dem Auftreten der „reifen" Form der Protease führt. Viele proteolytische Enzyme werden in der Natur als translationale Proenzym-Produkte gefunden und werden bei Fehlen einer posttranslationalen Prozessierung auf diese Weise exprimiert. Eine bevorzugte Prosequenz für die Herstellung von Proteasevarianten ist die mutmaßliche Prosequenz von Bacillus amyloliquefaciens-Subtilisin, obwohl andere Protease-Prosequenzen verwendet werden können.
- Eine „Signalsequenz" oder „Präsequenz" bezieht sich auf eine jegliche Sequenz von Aminosäuren, die an den N-terminalen Abschnitt einer Protease oder an den N-terminalen Abschnitt einer Proprotease gebunden ist und die an der Sekretion der reifen Form oder Pro-Formen der Protease teilnehmen kann. Die Definition der Signalsequenz ist eine funktionale, was bedeutet, dass diese alle jene durch den N-terminalen Abschnitt des Proteasegens kodierten Aminosäuresequenzen umfasst, die an der Bewirkung der Sekretion der Protease unter nativen Bedingungen teilnehmen. Die Erfindung setzt solche Sequenzen ein, um die Sekretion der Proteasevarianten, wie hier definiert, zu bewirken. Eine mögliche Signalsequenz umfasst die ersten sieben Aminosäu rereste der Signalsequenz aus Bacillus subtilis-Subtilisin, fusioniert mit dem Rest der Signalsequenz des Subtilisins aus Bacillus lentus (ATCC 21536).
- Eine „Präpro"-Form einer Proteasevariante besteht aus der reifen Form der Protease, die eine Prosequenz, die funktional oder operativ mit dem Aminoterminus der Protease verknüpft ist, und eine „Prä"- oder „Signal"sequenz, die funktional oder operativ mit dem Aminoterminus der Prosequenz verknüpft ist, aufweist.
- „Expressionsvektor" bezieht sich auf ein DNA-Konstrukt, welches eine DNA-Sequenz enthält, die funktional oder operativ mit einer geeigneten Kontrollsequenz, die in der Lage ist, die Expression der DNA in einem geeigneten Wirt zu bewirken, verknüpft ist. Solche Kontrollsequenzen umfassen einen Promotor, um die Transkription zu bewirken, eine optionale Operatorsequenz, um eine solche Transkription zu kontrollieren, eine Sequenz, welche geeignete mRNA-Ribosomenbindungsstellen kodiert, und Sequenzen, die die Termination von Transkription und Translation kontrollieren. Ein Vektor kann ein Plasmid, ein Phagenpartikel oder einfach ein potentielles genomisches Insert sein. Ist damit einmal ein geeigneter Wirt transformiert, kann der Vektor replizieren und unabhängig von dem Wirtsgenom funktionieren oder kann in einigen Fällen sich in das Genom selbst integrieren. In den vorliegenden Unterlagen werden „Plasmid" und „Vektor" manchmal gegeneinander austauschbar verwendet, da das Plasmid gegenwärtig die am häufigsten verwendete Vektorform ist. Die Erfindung soll jedoch solche anderen Formen von Expressionsvektoren, die äquivalenten Funktionen dienen und die in diesem Fachgebiet bekannt sind oder werden, mit umfassen.
- Die „Wirtszellen", die im Rahmen der Erfindung verwendet werden, sind allgemein prokaryotische oder eukaroytische Wirtsorganismen, die vorzugsweise durch die in dem U.S.-Patent RE 34,606 offenbarten Methoden manipuliert worden sind, um diese unfähig zu machen, enzymatisch aktive Endoprotease zu sekretieren. Eine bevorzugte Wirtszelle für die Expression einer Protease ist der Bacillus-Stamm BG2036, der hinsichtlich der enzymatisch aktiven neutralen Protease und der alkalischen Protease (Subtilisin) einen Defekt aufweist. Die Konstruktion des Stamms BG2036 wird detailliert in dem U.S.-Patent 5,264,366 beschrieben. Andere Wirtszellen für die Expression von Protease umfassen Bacillus subtilis I168 (auch in dem U.S.-Patent RE 34,606 und dem U.S.-Patent 5,264,366, deren Offenbarung in diese Unterlagen unter Bezugnahme aufgenommen wird, beschrieben) wie auch einen jeglichen geeigneten Bacillus-Stamm, wie z.B. B. licheniformis, B. lentus u.s.w.
- Wirtszellen werden mit Vektoren, die unter Verwendung von Techniken der in vitro-DNA-Rekombination konstruiert worden sind, transformiert oder transfiziert. Solche transformierten Wirtszellen sind in der Lage, entweder Vektoren, welche die Proteasevarianten kodieren, zu replizieren oder die gewünschte Proteasevariante zu exprimieren. In dem Falle von Vektoren, welche die Prä- oder Präpro-Form der Proteasevariante kodieren, werden solche Varianten, wenn sie exprimiert werden, typischerweise aus der Wirtszelle in das Wirtszellmedium sekrektiert.
- „Funktional verknüpft" oder „operativ verknüpft", wenn dies die Beziehung zwischen zwei DNA-Regionen beschreibt, bedeutet einfach, dass sie in funktionaler Beziehung zueinander stehen. Beispielsweise ist eine Präsequenz funktional mit einem Peptid verknüpft, wenn sie als Signalsequenz wirkt, indem sie an der Sekretion der reifen Form des Proteins, welche höchstwahrscheinlich eine Abspaltung der Signalsequenz mit umfasst, teilnimmt. Ein Promotor ist funktional oder operativ mit einer kodierenden Sequenz verknüpft, wenn er die Transkription der Sequenz kontrolliert; eine Ribosomenbindungsstelle ist funktional oder operativ mit einer kodierenden Sequenz verknüpft, wenn sie so positioniert ist, dass sie eine Translation erlaubt.
- Die die in der Natur vorkommende Vorläufer-Protease kodierenden Gene können gemäß den allgemeinen Methoden, die den Fachleuten auf diesem Gebiet bekannt sind, erhalten werden. Die Methoden umfassen im allgemeinen das Synthetisieren von markierten Sonden, die mutmaßliche Sequenzen aufweisen, die Regionen der Protease von Interesse kodieren, Herstellen von genomischen Bibliotheken aus Organismen, die die Protease exprimieren, und Screenen der Bibliotheken auf das Gen von Interesse durch Hybridisierung mit den Sonden. Positiv hybridisierte Klone werden dann kartiert und sequenziert.
- Die klonierte Protease wird dann verwendet, um eine Wirtszelle zu transformieren, um die Protease zu exprimieren. Das Proteasegen wird dann in ein Plasmid mit hoher Kopienzahl („high copy number"-Plasmid) kloniert. Dieses Plasmid repliziert in Wirten in dem Sinne, dass es die wohlbekannten Elemente enthält, die für die Plasmidreplikation erforderlich sind: einen Promotor, der funktional oder operativ mit dem fraglichen Gen verknüpft ist (der als der eigene homologe Promotor des Gens bereitgestellt werden kann, sofern er durch den Wirt erkannt, d.h. transkribiert wird), eine Transkriptionsterminations- und Polyadenylierungsregion (notwendig für die Stabilität der durch den Wirt von dem Proteasegen in bestimmten eukaryotischen Wirtszellen transkribierten mRNA), die exogen ist oder durch die endogene Terminatorregion des Proteasegens bereitgestellt wird, und wünschenswerterweise ein der Selektion dienendes Gen, wie ein Antibiotikumresistenzgen, das ein kontinuierliches Halten von durch das Plasmid infizierten Wirtszellen in Kultur durch Kultivierung in Antibiotikum enthaltenden Medium ermöglicht. Plasmide mit hohen Kopienzahlen enthalten auch einen Replikationsstartpunkt für den Wirt, wodurch ermöglicht wird, dass große Anzahlen von Plasmiden in dem Zytoplasma ohne chromosomale Einschränkungen erzeugt werden. Es liegt jedoch innerhalb des hiesigen Umfangs, eine Mehrzahl von Kopien des Proteasegens in ein Wirtsgenom zu integrieren. Dies wird durch prokaryotische und eukaryotische Organismen, die gegenüber einer homologen Rekombination besonders empfänglich sind, vereinfacht.
- Das Gen kann ein natürliches B. lentus-Gen sein. Alternativ kann ein synthetisches Gen, welches eine in der Natur vorkommende oder mutierte Vorläufer-Protease kodiert, hergestellt werden. Bei einem solchen Ansatz wird die DNA- und/oder Aminosäuresequenz der Vorläufer-Protease bestimmt. Danach wird eine Mehrzahl von überlappenden synthetischen einzelsträngigen DNA-Fragmenten synthetisiert, welche nach Hybridisierung und Ligation eine synthetische DNA, die die Vorläufer-Protease kodiert, produzieren. Ein Beispiel für die Konstruktion eines synthetischen Gens wird in Beispiel 3 des U.S.-Patents 5,204,015, dessen Offenbarung in diese Unterlagen unter Bezugnahme aufgenommen wird, erläutert.
- Ist einmal das in der Natur vorkommende oder synthetische Vorläufer-Protease-Gen kloniert, wird eine Anzahl von Modifikationen vorgenommen, um die Verwendung des Gens über die Synthese der in der Natur vorkommenden Vorläufer-Protease hinaus zu verbessern. Solche Modifikationen umfassen die Herstellung von rekombinanten Proteasen, wie in dem U.S.-Patent RE 34,606 und der EPA-Veröffentlichung Nr. 0 251 446 offenbart, und die Produktion von Proteasevarianten, die hier beschrieben wird.
- Das folgende Kassetten-Mutagenese-Verfahren kann verwendet werden, um die Konstruktion der Proteasevarianten der Erfindung zu vereinfachen, obwohl andere Methoden verwendet werden können. Zuerst wird das in der Natur vorkommende Gen, welches die Protease kodiert, erhalten und vollständig oder teilweise sequenziert. Dann wird die Sequenz hinsichtlich einer Stelle, an welcher gewünscht wird, eine Mutation (Deletion, Insertion oder Substution) von einer oder mehreren Aminosäuren in dem kodierten Enzym vorzunehmen, gescreent. Die diese Stelle flankierenden Sequenzen werden auf die Anwesenheit von Restriktionsstellen für das Ersetzen eines kurzen Abschnitts des Gens durch einen Oligonukleotidpool, der bei einer Expression verschiedene Mutanten kodieren wird, hin ausgewertet. Solche Restriktionstellen sind vorzugsweise einmalig vorkommende Stellen innerhalb des Proteasegens, um das Ersetzen des Genabschnitts zu erleichtern. Es kann jedoch eine jegliche geeignete Restriktionsstelle, die in dem Proteasegen nicht übermäßig redundant ist, verwendet werden, vorausgesetzt, dass die durch Restriktionsverdau erzeugten Genfragmente in der korrekten Abfolge erneut zusammengefügt werden können. Wenn Restriktionsstellen nicht an Stellen innerhalb eines geeigneten Abstands von der ausgewählten Stelle (10 bis 15 Nukleotide) vorhanden sind, werden solche Stellen durch Ersetzen von Nukleotiden in dem Gen derart, dass weder das Leseraster noch die kodierten Aminosäuren in dem endgültigen Konstrukt verändert werden, erzeugt. Eine Mutation des Gens, um dessen Sequenz zu verändern, damit diese mit der gewünschten Sequenz übereinstimmt, erfolgt durch M13-Primer-Verlängerung gemäß allgemein bekannten Methoden. Die Aufgabe, geeig nete flankierende Regionen zu lokalisieren und die benötigten Änderungen auszuwerten, um zu zwei geeigneten Restriktionsstellensequenzen zu gelangen, wird durch die Redundanz des genetischen Codes, eine Restriktionsenzymkarte des Gens und die große Anzahl von unterschiedlichen Restriktionsenzymen zur Routine. Es ist anzumerken, dass, wenn eine geeignete flankierende Restriktionsstelle zur Verfügung steht, die obige Methode nur in Verbindung mit der flankierenden Region, die keine Stelle enthält, verwendet werden muss.
- Ist die in der Natur vorkommende DNA oder synthetische DNA einmal kloniert, werden die Restriktionsstellen, die die zu mutierenden Positionen flankieren, mit den diese erkennenden Restriktionsenzymen verdaut und eine Mehrzahl von zu den End-Termini komplementären Oligonukleotidkassetten wird an das Gen ligiert. Die Mutagenese wird durch diese Methode vereinfacht, da alle Oligonukleotide so synthetisiert werden können, dass sie die gleichen Restriktionsstellen aufweisen und keine synthetischen Linker erforderlich sind, um die Restriktionsstellen zu erzeugen.
- Wie hier verwendet, wird proteolytische Aktivität als die Hydrolysegeschwindigkeit von Peptidbindungen pro Milligramm aktives Enzym definiert. Viele wohlbekannte Verfahren existieren für die Messung von proteolytischer Aktivität (K.M. Kalisz, „Microbial Proteinases", Advances in Biochemical Engineering/Biotechnology, A. Fiechter, Hrsg., 1988). Zusätzlich zu oder als Alternative zu modifizierter proteolytischer Aktivität können die Enzymvarianten der Erfindung andere modifizierte Eigenschaften aufweisen, wie Km, Kcat, Kcat/Km-Verhältnis und/oder modifizierte Substratspezifität und/oder modifiziertes pH-Aktivitätsprofil. Diese Enzyme können für das jeweilige Substrat, von dem antizipiert wird, dass es beispielsweise in dem Präparat von Peptiden vorhanden ist, oder für hydrolytische Verfahren, wie Waschanwendungen, maßgeschneidert werden.
- Unter einem Aspekt der Erfindung besteht das Ziel darin, eine Proteasevariante mit veränderten, vorzugsweise verbesserten Wascheigenschaften verglichen mit einer Vorläufer-Protease in wenigstens einer Waschmittelformulierung und/oder unter wenigstens einem Satz von Waschbedingungen zu erzielen.
- Es gibt verschiedene Waschbedingungen, einschließlich unterschiedlicher Waschmittelformulierungen, Waschwasservolumen, Waschwassertemperatur und Länge der Waschdauer, denen eine Proteasevariante möglicherweise ausgesetzt ist. Beispielsweise haben Waschmittelformulierungen, die in verschiedenen Gebieten verwendet werden, unterschiedliche Konzentrationen an ihren relevanten Bestandteilen, die in dem Waschwasser vorhanden sind. Beispielsweise weist ein europäisches Waschmittel typischerweise ungefähr 4500-5000 ppm Waschmittelbestandteile in dem Waschwasser auf, während ein japanisches Waschmittel typischerweise ungefähr 667 ppm Waschmittelbestandteile im Waschwasser aufweist. In Nordamerika, insbesondere in den Vereinigten Staaten, weist ein Waschmittel typischerweise ungefähr 975 ppm Waschmittelbestandteile auf, die in dem Waschwasser vorhanden sind.
- Ein System mit niedriger Waschmittelkonzentration umfasst Waschmittel, wo weniger als ungefähr 800 ppm Waschmittelbestandteile in dem Waschwasser vorhanden sind. Japanische Waschmittel werden typischerweise als System mit niedriger Waschmittelkonzentration angesehen, da bei diesen ungefähr 667 ppm Waschmittelbestandteile im Waschwasser vorhanden sind.
- Eine mittlere Waschmittelkonzentration umfasst Waschmittel, wo zwischen 800 ppm und ungefähr 2000 ppm Waschmittelbestandteile in dem Waschwasser vorhanden sind. Nordamerikanische Waschmittel werden im allgemeinen als Systeme mit mittlerer Waschmittelkonzentration angesehen, da bei diesen ungefähr 975 ppm Waschmittelbestandteile im Waschwasser vorhanden sind. In Brasilien sind ungefähr 1500 ppm Waschmittelbestandteile im Waschwasser vorhanden.
- Ein System mit hoher Waschmittelkonzentration umfasst Waschmittel, wo mehr als ungefähr 2000 ppm Waschmittelbestandteile in dem Waschwasser vorhanden sind. Europäische Waschmittel werden im allgemeinen als Systeme mit hoher Waschmittelkonzentration angesehen, da bei diesen ungefähr 4500-5000 ppm Waschmittelbestandteile im Waschwasser vorhanden sind.
- Die lateinamerikanischen Waschmittel sind im allgemeinen hochkonzentrierte Seifenlauge-Phosphat-Builder-Waschmittel und der Bereich von Waschmitteln, die in Lateinamerika verwendet werden, kann sowohl unter die mittleren als auch hohen Waschmittelkonzentrationen fallen, da sie von 1500 ppm bis 6000 ppm Waschmittelbestandteile im Waschwasser reichen. Wie oben erwähnt, sind in Brasilien typischerweise ungefähr 1500 ppm Waschmittelbestandteile im Waschwasser vorhanden. Jedoch kann es in anderen Gegenden mit hochkonzentrierten Seifenlauge-Phosphat-Builder-Waschmitteln, die nicht auf andere lateinamerikanische Länder beschränkt sind, Systeme mit hoher Waschmittelkonzentration bis zu ungefähr 6000 ppm Waschmittelbestandteilen im Waschwasser geben.
- Im Lichte des Vorangegangen ist offensichtlich, dass Konzentrationen von Waschmittelzusammensetzungen in typischen Waschlösungen in der ganzen Welt von weniger als ungefähr 800 ppm Waschmittelzusammensetzung („Gegenden mit niedrigen Waschmittelkonzentrationen"), beispielsweise ungefähr 667 ppm in Japan, bis zwischen ungefähr 800 ppm bis ungefähr 2000 ppm („Gegenden mit mittleren Waschmittelkonzentrationen"), beispielsweise ungefähr 975 ppm in den Vereinigten Staaten und ungefähr 1500 ppm in Brasilien, bis mehr als ungefähr 2000 ppm („Gegenden mit hohen Waschmittelkonzentrationen"), beispielsweise ungefähr 4500 ppm bis ungefähr 5000 ppm in Europa und ungefähr 6000 ppm in Ländern mit hochkonzentrierte Seifenlauge-Phosphat-Builder-Zusammensetzungen variieren.
- Die Konzentrationen der typischen Waschlösungen werden empirisch bestimmt. Beispielsweise enthält in den Vereinigten Staaten eine typische Waschmaschine ein Volumen von ungefähr 64,4 1 Waschlösung. Dementsprechend müssen, um eine Konzentration von ungefähr 975 ppm Waschmittel innerhalb der Waschlösung zu erhalten, ungefähr 62,79 g Waschmittelzusammensetzung zu den 64,4 l Waschlösung hinzugegeben werden. Diese Menge ist die typische Menge, die in das Waschwasser durch den Verbraucher unter Verwendung des Messbechers, der mit dem Waschmittel bereitgestellt wird, abgemessen wird.
- Als ein weiteres Beispiel verwenden unterschiedliche Gegenden unterschiedliche Waschtemperaturen. Die Temperatur des Waschwassers in Japan ist typischerweise niedriger als jene, die in Europa verwendet wird.
- Dementsprechend umfasst ein Aspekt der Erfindung eine Proteasevariante, die unter wenigstens einem Satz von Waschbedingungen verbesserte Wascheigenschaften zeigt.
- Unter einem anderen Aspekt der Erfindung ist festgestellt worden, dass Substitutionen an Positionen, die 103 und 245 entsprechen, in Kombination mit einer oder mehreren Substitutionen, ausgewählt aus der Gruppe bestehend aus den Positionen entsprechend 1, 3, 4, 8, 10, 12, 13, 16, 17, 18, 19, 20, 21, 22, 24, 27, 33, 37, 38, 42, 43, 48, 55, 57, 58, 61, 62, 68, 72, 75, 76, 77, 78, 79, 86, 87, 89, 97, 98, 99, 101, 102, 104, 106, 107, 109, 111, 114, 116, 117, 119, 121, 123, 126, 128, 130, 131, 133, 134, 137, 140, 141, 142, 146, 147, 158, 159, 160, 166, 167, 170, 173, 174, 177, 181, 182, 183, 184, 185, 188, 192, 194, 198, 203, 204, 205, 206, 209, 210, 211, 212, 213, 214, 215, 216, 217, 218, 222, 224, 227, 228, 230, 232, 236, 237, 238, 240, 242, 243, 244, 246, 247, 248, 249, 251, 252, 253, 254, 255, 256, 257, 258, 259, 260, 261, 262, 263, 265, 268, 269, 270, 271, 272, 274 und 275 von Bacillus amyloliquefaciens-Subtilisin, bei der Verbesserung der Wascheigenschaften des Enzyms von Bedeutung sind.
- Diese Substitutionen werden vorzugsweise in Bacillus lentus (rekombinant oder vom nativen Typ)-Subtilisin vorgenommen, obwohl die Substitutionen in einer jeglichen Bacillus-Protease vorgenommen werden können.
- Basierend auf den Screening-Ergebnissen, die mit den Proteasevarianten erhalten wurden, sind die angegebenen Mutationen in Bacillus amyloliguefaciens-Subtilisin wichtig für die proteolytische Aktivität, Leistungseigenschaften und/oder Stabilität von diesen Enzymen und die Reinigungs- oder Wascheigenschaften von solchen Enzymvarianten.
- Viele der Proteasevarianten der Erfindung sind nützlich bei der Formulierung von verschiedenen Waschmittelzusammensetzungen oder Kör perpflegeformulierungen, wie Shampoos oder Lotionen. Eine Anzahl von bekannten Verbindungen sind geeignete grenzflächenaktive Mittel oder Tenside, die in Zusammensetzungen, welche die Proteasemutanten der Erfindung umfassen, nützlich sind. Diese umfassen nichtionische, anionische, kationische oder zwitterionische Detergentien, wie in
US 4,404,128 von Barry J. Anderson und U.S. 4,261,868 von Jiri Flora et al. offenbart. Eine geeignete Waschmittelformulierung ist jene, die in Beispiel 7 des U.S.-Patents 5,204,015 (das zuvor unter Bezugnahme aufgenommen wurde) beschrieben wird. Dieses Fachgebiet ist vertraut mit den verschiedenen Formulierungen, die als Reinigungs- oder Waschzusammensetzungen verwendet worden können. Es versteht sich natürlich, dass zusätzlich zu typischen Reinigungs- oder Waschzusammensetzungen die Proteasevarianten der Erfindung für einen jeglichen Zweck, zu dem native oder Wildtyp-Proteasen verwendet werden, verwendet werden können. So können diese Varianten beispielsweise in Seifenstück- oder Flüssigseife-Anwendungen, Geschirrspülformulierungen, Kontaktlinsenreinigungslösungen oder -produkten, Peptidhydrolyse, Abfall/Abwasserbehandlung, Textilanwendungen, als Fusion-Spaltungs-Enzyme bei der Proteinherstellung u.s.w. verwendet werden. Die Varianten der Erfindung können verbesserte Leistungseigenschaften in einer Waschmittelzusammensetzung (verglichen mit dem Vorläufer) umfassen. Wie hier verwendet, werden verbesserte Leistungseigenschaften in einem Waschmittel als eine verbesserte Reinigung von bestimmten Enzym-empfindlichen Flecken, wie Gras oder Blut, definiert, wie durch die übliche Auswertung nach einem Standard-Waschzyklus bestimmt. - Proteasen der Erfindung können zu bekannten pulverförmigen und flüssigen Waschmitteln mit einem pH zwischen 6,5 und 12,0 mit Konzentrationen von ungefähr 0,01 bis ungefähr 5% (vorzugsweise 0,1% bis 0,5%), bezogen auf das Gewicht, formuliert werden. Diese Detergenzien enthaltenden Reinigungszusammensetzungen können auch andere Enzyme, wie bekannte Proteasen, Amylasen, Cellulasen, Lipasen oder Endoglycosidasen, wie auch Builder und Stabilisatoren umfassen.
- Die Zugabe von Proteasen der Erfindung zu herkömmlichen Reinigungszusammensetzungen bewirkt keinerlei spezielle Einschränkung hinsichtlich der Verwendung. In anderen Worten ist eine jegliche Tempe ratur und ein jeglicher pH, der für das Waschmittel geeignet ist, auch für die vorliegenden Zusammensetzungen geeignet, solange der pH innerhalb des obigen Bereichs liegt und die Temperatur unterhalb der Denaturierungstemperatur der beschriebenen Protease liegt. Zusätzlich können Proteasen der Erfindung in einer Reinigungszusammensetzung ohne Detergenzien, wieder entweder allein oder in Kombination mit Buildern und Stabilisatoren verwendet werden.
- Die Erfindung betrifft auch Reinigungszusammensetzungen, die die Proteasevarianten der Erfindung enthalten. Die Reinigungszusammensetzungen können zusätzlich Additive oder Zusatzstoffe enthalten, die in Reinigungszusammensetzungen üblicherweise verwendet werden. Diese können ausgewählt werden aus, sind aber nicht beschränkt auf Bleichen, grenzflächenaktive(n) Substanzen oder Tenside, Builder, Enzyme und Bleichkatalysatoren. Für die Fachleute auf diesem Gebiet wäre leicht ersichtlich, welche Additive oder Zusatzstoffe für eine Aufnahme in die Zusammensetzungen geeignet sind. Die hier bereitgestellte Liste ist keinesfalls erschöpfend und sollte nur als Beispiele für geeignete Additive oder Zusatzstoffe aufgefasst werden. Für die Fachleute auf diesem Gebiet ist ebenfalls leicht ersichtlich, nur jene Additive oder Zusatzstoffe zu verwenden, die mit den Enzymen und anderen Bestandteilen in der Zusammensetzung, beispielsweise den Tensiden, verträglich sind.
- Wenn vorhanden, beträgt die Menge von vorhandenen Zusatzstoffen oder Additiven in der Reinigungszusammensetzung ungefähr 0,01% bis ungefähr 99,9%, vorzugsweise ungefähr 1% bis ungefähr 95%, mehr bevorzugt ungefähr 1% bis ungefähr 80%.
- Die Proteasevarianten der Erfindung können in Tierfutter Eingang finden, wie als Teil von Tierfutterzusätzen, wie beispielsweise in U.S. 5,612,055, U.S. 5,314,692 und U.S. 5,147,642 beschrieben.
- Ein Aspekt der Erfindung ist eine Zusammensetzungen für die Behandlung eines Gewebes, die Proteasevarianten der Erfindung umfasst. Die Zusammensetzung kann verwendet werden, um beispielsweise Seide oder Wolle zu behandeln, wie in Veröffentlichungen, wie RD 216,034,
EP 134,267 EP 344,259 - Das Folgende wird beispielhaft aufgeführt und soll nicht als eine Beschränkung hinsichtlich des Umfangs der Ansprüche verstanden werden.
- Beispiel 1
- Eine große Anzahl von Proteasevarianten wurde unter Verwendung von Methoden, die in diesem Fachgebiet wohlbekannt sind, hergestellt und gereinigt. Alle Mutationen erfolgten in Bacillus lentus GG36-Subtilisin. Die Varianten sind in Tabelle 3 gezeigt.
- Beispiel 2
- Eine große Anzahl von Proteasevarianten, die in Beispiel 1 hergestellt worden waren, wurde auf Leistungseigenschaften bei zwei Arten von Waschmittel- und Waschbedingungen unter Verwendung eines Mikrostoffproben („microswatch")-Assay, der in „An improved method of assaying for a preferred enzyme and/or preferred detergent composition", U.S. Serial Nr. 60/068,796 beschrieben wird, getestet.
- Tabelle 4 listet die dem Assay unterzogenen Proteasevarianten und die Ergebnisse der Untersuchung in zwei unterschiedlichen Waschmitteln auf. Bei Spalte A war das Waschmittel 0,67 g/l filtriertes Ariel Ultra (Procter & Gamble, Cincinnati, OH, USA) in einer Lösung, welche 3 Grain pro Gallone gemischte Ca2+/Mg2+-Härte enthielt, und es wurde in jeder Vertiefung 0,3 ppm Enzym bei 20°C verwendet. Bei Spalte B war das Waschmittel 3,38 g/l filtriertes Ariel Futur (Procter & Gamble, Cincinnati, OH, USA) in einer Lösung, welche 15 Grain pro Gallone gemischte Ca1+/Mg2+-Härte enthielt, und es wurde in jeder Vertiefung 0,3 ppm Enzym bei 40°C verwendet.
- Beispiel 3
- Tabelle 5 listet die dem Assay unterzogenen Proteasevarianten aus Beispiel 1 und die Ergebnisse der Untersuchung in vier verschiedenen Waschmitteln auf. Es wurden an den aufgeführten Proteasevarianten die gleichen Leistungseigenschaftstests wie in Beispiel 2 ausgeführt mit den folgenden Waschmitteln. Für Spalte A war das Waschmittel 0,67 g/l filtriertes Ariel Ultra (Procter & Gamble, Cincinnati, OH, USA) in einer Lösung, welche 3 Grain pro Gallone gemischte Ca1+/Mg2+-Härte enthielt, und es wurde in jeder Vertiefung 0,3 ppm Enzym bei 20°C verwendet. Für Spalte B war das Waschmittel 3,38 g/l filtriertes Ariel Futur (Procter & Gamble, Cincinnati, OH, USA) in einer Lösung, welche 15 Grain pro Gallone gemischte Ca2+/Mg2+-Härte enthielt, und es wurde in jeder Vertiefung 0,3 ppm Enzym bei 40°C verwendet. Für Spalte C 3,5 g/l HSP 1-Waschmittel (Procter & Gamble, Cincinnati, OH, USA) in einer Lösung, welche 8 Grain pro Gallone gemischte Ca1+/Mg2+-Härte enthielt, und es wurde in jeder Vertiefung 0,3 ppm Enzym bei 20°C verwendet. Für Spalte D 1,5 ml/l Tide KT-Waschmittel (Procter & Gamble, Cincinnati, OH, USA) in einer Lösung, welche 3 Grain pro Gallone gemischte Ca1+/Mg2+-Härte enthielt, und es wurde in jeder Vertiefung 0,3 ppm Enzym bei 20°C verwendet.
