Description
本发明涉及一种具活性的重组类似胰蛋白酶的蛋白酶,一种包含该类似胰蛋白酶的蛋白酶的蛋白水解组合物,一种包含编码所说类似胰蛋白酶的蛋白酶DNA序列的DNA构建物,以及包含该DNA构建物的载体和细胞。并且本发明还涉及一种通过使用重组DNA技术制备该类似胰蛋白酶的蛋白酶的方法。
蛋白酶被广泛地用作商业洗涤剂的成份,以提高对蛋白质组成的污垢的去污力。多年来,具有广范围专一性的蛋白酶,如枯草杆菌蛋白酶(芽孢杆菌丝氨酸蛋白酶)是使用最广泛的洗涤剂蛋白酶。然而,近年来具有较窄范围专一性的蛋白酶在洗涤剂中的应用已引起日益增多的关注。
WO89/06270公开了一种洗涤剂组合物,其含有窄范围底物专一性的蛋白酶,即类似胰蛋白酶的蛋白酶,其能够断裂赖氨酸或精氨酸C末端的钛键。公开于所述参考文献中的类似胰蛋白酶的蛋白酶通过常规的镰孢霉属(Fusarium sp.)菌株尖孢镰孢(F.oxysporum)DSM2672的发酵来生产,并且不可避免地与另一种蛋白酶(蛋白酶1)一同产生,而在洗涤剂组合物中不需要蛋白酶1的存在。因此,来自尖孢镰孢菌株的类似胰蛋白酶的蛋白酶的生产包含从类似胰蛋白酶的蛋白酶中分离出不需要的蛋白酶这一步骤,这样就使得类似胰蛋白酶的蛋白酶的大规模生产成本比所希望的要高。所以需要使所述镰孢霉属的类似胰蛋白酶的蛋白酶的生产变得容易方便,尤其是避免上述不需要的蛋白酶1的同时产生,以能够生产出更多产量的酶及能以比现有技术更为经济的方式进行生产。
本发明人现已成功地实现了编码镰孢霉属类似胰蛋白酶的蛋白酶的DNA序列的克隆,并从所述DNA序列中得到了具活性的类似胰蛋白酶的蛋白酶的表达。
因此,本发明的一方面涉及一种具有活性的重组类似胰蛋白酶的蛋白酶,其包含有氨基酸序列的1—224氨基酸残基,氨基酸序列示于所附SEQ ID No.2。
本发明中的术语“类似胰蛋白酶的蛋白酶”是指一种具有类似胰蛋白酶活性的酶,即能够切断赖氨酸或精氨酸C末端的肽键。该类似胰蛋白酶的蛋白酶活性的测定可通过基于胰蛋白酶底物如N—苯甲酰基—L—精氨酸—对—硝基苯胺盐酸化物(L—BAPA或L—BApNA)的测定来进行,例如描述于下面的“材料和方法”部分中。
有关本发明的类似胰蛋白酶的蛋白酶中所用的术语“重组”是指通过以编码蛋白酶的DNA序列转化的细胞来生产蛋白酶。因此,重组类似胰蛋白酶的蛋白酶是通过另一种微生物而非其亲本微生物来生产的,所以该蛋白酶基本上不含所述亲本微生物产生的组分,即由尖孢镰孢株DSM 2672生产的组分。这些组分可使蛋白酶具有不需要的性质。如产生不需要的酶活力。在这一方面,可以容易地生产本发明的重组类似胰蛋白酶的蛋白酶而不含有蛋白酶1,并且也不需要类似于WO 89/06270中所进行的昂贵的分离步骤。
在导致本发明的研究过程中,即下述实施例所详细描述的本发明,非常意外地发现,通过对SEQ ID No.1所示的DNA序列转化的微生物(米曲霉Aspergillus oryzae)这一常规使用的生产菌株进行培养,很难得到活性类似胰蛋白酶的蛋白酶的任一基本产品。在对该DNA序列和表达产品进行分析的基础上发现,以该DNA序列编码的酶被表达为非活性的酶原,并且有大量的这种酶原被表达。因此,即使可获得大量的活性酶的前体形式,也得不到一点具有活性的成熟酶,而且已得出结论,酶原在米曲霉发酵液中不稳定。
令人惊奇地发现,如果对包含编码酶的酶原形式的DNA序列的宿主细胞在培养基中进行培养,同时培养基中也含能将该酶原原位转化为活性酶的蛋白水解酶,则活性重组类似胰蛋白酶的蛋白酶的产量能大大提高。我们认为,当其它蛋白水解酶不存在时表达的低酶活力可归因于非活性的类似胰蛋白酶的夤白酶的酶原形式很不稳定,并很快地由宿主微生物表达的蛋白水解酶降解,并且当加入一种蛋白水解酶,其比降解酶原的蛋白水解酶反应要快,则其与酶原反应导致成熟的活性类似胰蛋白酶的蛋白酶的产生。本发明的另一方面涉及含有编码上述本发明活性重组类似胰蛋白酶的蛋白酶DNA序列的DNA构建物。
本发明的另一方面涉及含有本发明DNA构建物的重组表达载体,涉及含有本发明DNA构建物或表达载体的细胞,并涉及本发明类似胰蛋白酶的蛋白酶的生产工艺,其中对如上所述的本发明细胞在有助于蛋白酶生产的培养条件下培养,并随后从培养物中回收该蛋白酶。
最后,本发明涉及含有本发明活性类似胰蛋白酶的蛋白酶的洗涤剂添加剂及洗涤剂组合物。
从菌株尖孢镰孢DSM 2672中分离出的本发明重组类似胰蛋白酶的蛋白酶的氨基酸序列与其它来源的类似胰蛋白酶的蛋白酶的氨基酸序列排列在一起,发现其与牛胰蛋白酶(GenBank Acc.No.P00760)具有约43%的同源性,与灰色链霉菌(Streptomycesgriseus)(GenBank Acc.No.P00776)具有44%的同源性。已发现本发明类似胰蛋白酶的蛋白酶与果蝇的蛋白酶(GenBank Acc.No.A23493)具有最高的同源性,即48%。
这些同源性表明在类似胰蛋白酶的蛋白酶间存在着某种进化关系,但本发明的镰孢霉属类似胰蛋白酶的蛋白酶也代表了一类独特的类似胰蛋白酶的蛋白酶。人们期望本发明的类似胰蛋白酶的蛋白酶或编码该蛋白酶的DNA可以从其它微生物中分离出来,包括动物,尤其是哺乳动物,昆虫,植物或微生物。本文中尤其令人感兴趣的来源是细菌或真菌,包括酵母和丝状真菌。
含有序列SEOID No.2的1—224氨基酸残基的本发明活性重组类似胰蛋白酶的蛋白酶具有许多特征。例如,与牛胰蛋白酶相比,该蛋白酶令人惊讶地显示出逆反的Arg/Lys专一性,即本发明的类似胰蛋白酶的蛋白酶对Arg具有比对Lys更强的活性。
本发明的类似胰蛋白酶的蛋白酶的pH活性曲线表明在pH8至11具有广范围的活性最佳值,特别是在25℃下使用H—D—Val—Leu—Lys—pNA为底物时最佳pH约为10。pH9.5以H—D—Val—Leu—Lys—pNA为底物时最佳温度值约为40℃。成熟的活性酶由224个氨基酸残基组成,且分子量为22190。其以前酶原(pre—pro—enzyme)形式表达,其中根据Von Heijne规则(1986)认为-24至-8位的氨基酸残基为信号肽,前肽氨基酸残基为-7至-1。酶的比活力至少约为30个酪蛋白蛋白酶单位(CPU)/g并优选高于35CPU/g。蛋白酶活力可由WO89/06270的方法进行测定,其内容在此引为参考文献。
本发明DNA构建物中编码上述重组蛋白酶的DNA序列优选示于所附序列SEQ ID No.1。所述序列的类似物(有1个或多个不同密码子但编码该重组蛋白酶),也在本发明范围之内。
本发明DNA构建物的DNA序列可以用熟知的方法进行分离。因此,DNA序列如可通过建立cDNA或基因组文库从认为包含该序列的微生物中分离出来,并通过常规程序进行阳性克隆筛选。这一程序的例子为与寡核苷酸探针杂交,该探针是根据标准技术(参见Sambrook等,1989)以序列SEQ ID No.2所示的全部氨基酸序列或其亚序列的基础上合成的,和/或选择表达上述类似胰蛋白酶的蛋白酶活性的克隆,和/或选择产生蛋白质的克隆,此蛋白质可与类似胰蛋白酶的蛋白酶的抗体反应,类似胰蛋白酶的蛋白酶含有SEQ ID No.2所示的氨基酸序列,尤其是序列的1—224氨基酸残基。
从cDNA或基因组文库中分离本发明DNA构建物的优选方法是使用聚合酶链反应(PCR),该反应使用简并寡核苷酸探针,该探针是在含有SEQ ID No.2的1—224氨基酸残基的本发明类似胰蛋白酶的蛋白酶的氨基酸序列的基础上制备的。例如,可用US专利No.4,683,202描述的技术或由R.K.Saiki等(1988)描述的技术进行PCR。
可选择地,可以使用已建立的标准方法对本发明DNA构建物的DNA序列进行合成制备,如Beaucage和Caruthers(1981)描述的磷酰胺(phosphoamidite)法或Matthes等(1984)描述的方法。根据磷酰胺法,例如在自动DNA合成仪上合成寡肽,纯化,退火,连接及在合适的载体中克隆。
最终,DNA构建物可以是混合的基因组基因和合成基因的构建物,混合的合成基因和cDNA的构建物或混合的基因组基因和cDNA源的构建物,其是根据标准技术通过连接合成片断,基因组片断或cDNA源(合适的话)片断来制备的,这些片断相应于整个重组DNA分子的各个部分。
携带本发明DNA构建物的重组表达载体可以是能方便地用于重组DNA过程的任何载体,并且载体的选择常常依赖于所要导入的宿主细胞。因此,载体可以是能自主复制的载体,即作为染色体外实体存在的载体,其复制独立于染色体复制之外,如质粒,噬菌体,或染色体外遗传因子,微型染色体或人工染色体。可选择地,当被导入宿主细胞时,载体可以被整合入宿主细胞基因组中并与其已整合入的染色体一起复制。
在载体中,DNA序列应可操纵地与合适的启动子序列连接。启动子可以是任一在所选择的宿主细胞中显示有转录活性的DNA序列,并且可以来自编码宿主细胞的同源或异源蛋白质的基因。特别是在细菌宿主中引导本发明DNA构建物转录的合适启动子的例子有大肠肝菌(E.Coli)的乳糖操纵子的启动子,天蓝色链霉菌(Streptomyces Coelicolor)琼脂酶基因dagA启动子,地衣形芽孢杆菌(Bacillus licheniformis)α—淀粉酶基因(amyL)启动子,嗜热脂肪芽孢杆菌(Bacillus stearothermophilus)生麦芽糖淀粉酶基因(amyM)启动子,解淀粉芽孢杆菌(BacillusAmyloliquefaciens)a—淀粉酶(amyQ)启动子,枯草芽孢杆菌(Bacillus subtilis)xylA和xylB基因启动子等等。在真菌宿主中转录时,有用的启动子的例子有那些来自编码米曲霉TAKA淀粉酶,Rhizomucor miehei天冬氨酸蛋白酶,黑曲霉(A.niger)中性a—淀粉酶,黑曲霉酸稳定的a—淀粉酶,黑曲霉葡萄糖淀粉酶,Rhizomucor miehei脂肪酶,米曲霉碱性蛋白酶,米曲霉磷酸丙糖异构酶或构巢曲霉(A.nidulans)乙酰胺酶的基轩。
本发明的表达载体也可含有适宜的转录终止子和在真核生物中的聚腺苷酸化序列,这一序列可操纵地与编码本发明重组蛋白酶的DNA序列连接。终止子和聚腺苷酸化序列可从与启动子相同的来源得到。
载体可进一步包含能使载体在所述宿主细胞中进行复制的DNA序列。该序列的例子有质粒pUC19,pACYC177,pUB110,pE194,pAMB1和pIJ702的复制起点。
载体也可包含一选择性标记,如弥补宿主细胞中缺失位点的基因产品,象来自枯草芽孢杆菌或地衣形芽孢杆菌的dal基因,或赋予抗生素抗性的基因,诸如氨苄青霉素,卡那霉素,氯霉素或四环素抗性。曲霉的选择性标记的例子包括amdS,argB,niaD和sC,给予潮霉素抗性的标记。并且,可通过共转化来完成选择,如描述于WO91/17243。
虽然在某些方面胞内表达是有利的,如当使用某些细菌作为宿主细胞时,但通常优选细胞外表达。如上所述,包含SEQ IDNo.2所示的氨基酸序列的本发明类似胰蛋白酶的蛋白酶包含一个前区(preregion),该区允许被表达的蛋白酶在培养基中分泌。如果需要,该前区可用一个不同的前区或信号序列代替,可通过编码各个前区的DNA序列的替换方便地完成。
连接本发明DNA构建物,启动子,终止子和其它因子及将其插入包含复制必需信息的适当载体中,这些程序对本领域技术人员而言是熟知的(参见,如Sambrook等(1989))。
包含上述本发明DNA构建物或表达载体的本发明细胞利于用作本发明多肽重组产品的宿主细胞。该细胞可以由本发明的DNA构建物转化,即通过方便地将DNA构建物整合入宿主染色体。由于DNA序列很可能在细胞中稳定地存在,所以这一整合通常被认为是有利的。可根据常规手段如同源重组或异源重组进行DNA构建物进入宿主染色体的整合。可选择地,细胞可以用一种上述的与不同类型宿主细胞相关的表达载体转化。
本发明的细胞可以是较高等生物体如哺乳动物或昆虫的细胞,但优选微生物细胞,如细菌或真菌(包括酵母)细胞。
合适的细菌的例子有革兰氏阳性菌,如枯草芽孢杆菌,地衣形芽孢杆菌,缓慢芽孢杆菌(Bacillus lentus),短芽孢杆菌(Bacillus brevis),嗜热脂肪芽孢杆菌(BacillusStearothermophilus),嗜碱芽孢杆菌(Bacillus alkalophilus),解淀粉芽孢杆菌(Bacillus amyloliquefaciens),凝结芽孢杆菌(Bacilluscoagulans),环状芽孢杆菌(Bacillus circalans),Bacillus lautus,巨大芽孢杆菌(Bacillus megaterium),苏云金芽孢杆菌(Bacilusthuringiensis),或变青链霉菌(Streptomyces lividans)或鼠灰链霉菌(Streptomyces murinuis)或革兰氏阴性菌,如大肠杆菌。细菌的转化例如可通过原生质体转化来实现,或通过使用感受态细胞以熟知的方式来实现。

从上述公开的内容明显可知,当本发明的类似胰蛋白酶的蛋白酶被表达为酶原时,当在蛋白水解酶存在时进行培养或可任意选择地有两种或多种能在培养基中将酶原转化为活性酶的蛋白水解酶存在时,可获得显著提高的产量。
蛋白水解酶可被加入到培养产生酶原的细胞的发酵液中。要加入的蛋白水解酶可以是芽孢杆菌金属蛋白酶,商业上可得的酶,如NeutraseR,嗜热菌蛋白酶,或另外一种具有相似活性的酶,如来自嗜热脂肪芽孢杆菌的蛋白水解酶(Zamost等,1990)或来自尖孢镰孢的天冬氨酸蛋白酶,根据本文实施例1描述的方法获得。可选择地,可通过在适当的条件下对编码蛋白水解酶的DNA序列转化的宿主细胞进行培养来生产酶,并从培养基中回收该酶,从而可生产出蛋白水解酶。
进一步地,通过将编码所述酶的DNA序列插入到含本发明DNA构建物的细胞中,以这种方式可使蛋白水解酶得到表达,且产生的量足以活化本发明的蛋白酶,从而可实现蛋白水解酶在发酵液中的存在。
可通过常规程序将产生的类似胰蛋白酶的蛋白酶从培养基中回收,包括离心或过滤以从培养基中分离细胞,细胞破碎后若需要的话,通过盐析法沉淀上清液或滤液中的蛋白质组分,如使用硫酸铵。然后通过各种层析方法如离子交换层析,亲合层析等进行纯化。洗涤剂组合物
根据本发明,类似胰蛋白酶的蛋白酶可典型地作为洗涤剂组合物的组分。这样它可以无尘颗粒形式,稳定化的液体或被保护的酶形式存在于洗涤剂组合物中。例如可根据US4,106,991和4,661,452(均属于Novo Industri A/S)公开的方法生产无尘颗粒,并可任意选择地采用本领域已知方法对颗粒进行包覆。蜡质包覆材料的例子有平均摩尔量为1000至2000的聚(环氧乙烷)产品(聚乙二醇,PEG),具有16至50个环氧乙烷单元的乙氧化壬基酚;有15到80个环氧乙烷单元的乙氧化脂肪醇,其中的醇含12至20个碳原子;脂肪醇;脂肪酸;以及脂肪酸的单酸甘油酯,甘油二酯和甘油三酯。适用的以流化床技术生产的成膜包覆材料的例子公开于专利GB 148 3591。根据已知方法例如可加入多羟基化合物如丙二醇、糖或糖醇、乳酸或硼酸来稳定液体酶制剂。其它酶稳定剂均是本领域所熟知的。根据EP238,216公开的方法可制备保护的酶。
本发明的洗涤剂组合物可以呈任意一种可便捷使用的形式,如粉状,粒状,糊状或液体。液体洗涤剂可以是水溶液,通常含上限至70%的水和0—30%的有机溶剂,或是非水溶液。
洗涤剂组合物含有一种或多种表面活性剂,其可以是阴离子的,非离子的,阳离子的或两性离子的。洗涤剂通常含有0—50%的阴离子表面活性剂,如直链烷基苯磺酸盐(LAS),a—烯烃磺酸盐(AOS),烷基磺酸盐(脂族醇磺酸盐)(AS),脂肪醇乙氧基硫酸盐(AEOS或AES),仲链烷基磺酸盐(SAS),a—磺基脂肪酸甲基酯,烷基或烯基琥珀酸或皂。洗涤剂组合物也可含有0—40%的非离子表面活性剂,如脂肪醇乙氧基化物(AEO或AE),羧基化的脂肪醇乙氧基化物,壬基酚乙氧基化物,烷基多苷(alkylpolyglycoside),烷基二甲基氧化胺,乙氧基化的脂肪酸单乙醇酰胺,脂肪酸单乙醇酰胺,或多羟基烷脂肪酸酰胺(如描述于WO 92/06154中)。
另外洗涤剂组合物还可含有一种或多种其它酶,如淀粉酶,脂肪酶,角质酶(cutinase),另一种蛋白酶,纤维素酶,过氧化物酶和氧化酶。
洗涤剂可含有1—65%的助洗剂或配位剂,如沸石、二磷酸、三磷酸、膦酸、柠檬酸、次氮基三乙酸(NTA),乙二胺四乙酸(EDFA)、二亚乙基三胺五乙酸(DTMPA),烷基或烯基琥珀酸,可溶性硅酸盐或层式硅酸盐(如Hoechst的SKS—6)。洗涤剂也可以是未复配的,即基本上不含洗涤剂助洗剂。
洗涤剂可含一种或多种聚合物。例如有羧甲基纤维素(CMC),聚乙烯基吡咯烷酮(PVP),聚乙二醇(PEG),聚乙烯醇(PVA),聚羧酸酯如聚丙烯酸酯,马来酸/丙烯酸共聚物和十二烷基异丁烯酸/丙烯酸共聚物。
洗涤剂可含有一种漂白体系,该体系可包含有H2O2源,如过硼酸盐或过碳酸盐,其可与如四乙酰乙二胺(TAED)或壬酰氧基苯磺酸盐(NOBS)这样的形成过酸的漂白活化剂结合。可选择地,漂白体系可含有如酰胺,酰亚胺或砜的过氧酸。
本发明洗涤剂组合物中的酶可用常规的稳定剂稳定,例如多元醇,如丙二醇或丙三醇,糖或糖醇,乳酸,硼酸或硼酸衍生物如芳香族硼酸酯。组合物可根据如WO92/19709和WO92/19708所描述的方法进行配制。

本发明的类似胰蛋白酶的蛋白酶可被掺入洗涤剂常规采用的浓缩物中,目前认为在本发明洗涤剂组合物中可加入的类似胰蛋白酶的蛋白酶的量相应地为每升洗涤液体0.001—100毫克酶。

0.01M二甲基戊二酸(Sigma D4379),0.2M硼酸和0.002M氯化钙,以NaOH调节至pH6.5。底物
L—BAPA可从Sigma(B3133)或Merck(Art.10754)获得。制备成0.2M的二甲基亚砜的储备溶液(87mg/ml)(冷冻存储)并以上述的缓冲液稀释至0.004M。测定
类似胰蛋白酶的蛋白酶被稀释至约15μg/ml进行分析测定。在一96孔微量滴定板上100μl已稀释的类似胰蛋白酶的蛋白酶与100μl的0.004ML—BAPA混合。100lμ缓冲液和100lμ0.004ML—BAPA空白液用于较正。
在Elisa读数器上405nm处监测光吸收的变化(ΔOD/分钟),监测时间10分钟,每5分钟读一次数据,温度为25℃或室温。
计算结果为相对于参比镰孢属类似胰蛋白酶的蛋白酶的胰蛋白酶含量。质粒pCDV 1—PL(Noma等,1986)p777(描述于EP0 489 718)pToC90(描述于WO91/17243)实施例实施例1来自镰孢属的类似胰蛋白酶的蛋白酶的纯化
大体上按照WO89/06270所描述方法,通过在含有大豆粗粉的培养基中发酵尖孢镰孢DSM2672,从发酵上清液中分离出尖孢镰孢的粗蛋白酶制品。接着,通过0—1M/0—25%氯化钠/异丙醇梯度洗脱在pH7.0从杆菌肽—多羟基硅亲合柱(Mortensen等,1989)上将尖孢镰孢类似胰蛋白酶的蛋白酶与两种其它的尖孢镰孢蛋白酶分离开来。以这种方法从尖孢镰孢类似胰蛋白酶的蛋白酶分离出的两种其它的蛋白酶为类枯草溶菌素酶和具有活化尖孢镰孢类似胰蛋白酶的蛋白酶前体形式(Ash—Ile断裂)所需特异性的天冬氨酸蛋白酶。
其它纯化步骤均根据众所周知的方法进行,包括在SeparoseCL—bB上进行离子交换层析(在杆菌肽分离之前),凝胶过滤和超滤浓缩。
0.1%纯化的类似胰蛋白酶的尖孢镰孢蛋白酶溶液在280hm的特异吸收测定为1.32。
纯化的蛋白酶随后进行氨基酸序列测定和分析并测定N末端的氨基酸序列。实施例2尖孢镰孢胰蛋白酶基因的克隆mRNA的制备
上述实施例1获得的尖孢镰孢菌株DSM2672的培养物中的菌丝体在液氮条件下被粉碎,重新悬浮于5M硫氰酸胍中,CsCl—肌氨酰梯度分离,由此分离出mRNA。将mRNA以乙醇洗涤,并在oligo—dT柱(Chirgwin等(1978),Truelsen等(1979))上分级分离。在用网织红细胞溶胞产物和35S—蛋氨酸对所得mRNA进行的体外翻译中测定mRNA的质量。SDS—PAGE分析显示出小于20至大于100KDa的区带。cDNA文库的制备和筛选

切裂在异亮氨酸密码子的探针被分入32片滤纸的三个池。筛选了三组滤纸。在42℃时洗涤后,可能的候选组被再次分离,识别出三个阳性克隆。这些阳性克隆中均发现有ATC—异亮氨酸探针。基因的识别和测定
根据Sambrook等1989的方法从三个克隆中制备质粒,限制酶切作图后,发现两个是一样的,第三个要短100bp。使用Maxam和Gilbert(1980)的方法测定长克隆双链序列,得到的DNA序列示于SEQ ID No.1。
由实施例1得到的纯化的尖孢镰孢类似胰蛋白酶的蛋白酶的氨基酸组成与从SEQ ID No.1所示的DNA—序列得到的组成是一致的。
根据Von Heijne(1986)的规则这一序列分析显示出编码具有17个氨基酸信号肽的类似胰蛋白酶的蛋白酶的基因(在Ala—17和Ala—18处切割),并且在信号肽酶切位点和已知的酶N—末端有一7个氨基酸的前肽(Ala—Pro—Gln—Glu—Ile—Pro—Ash)。这一假设前肽的作用还属未知,但蛋白酶通常被制成无活性的酶原。
成熟酶显示出与所有已知的胰蛋白酶具有少于50%的一致性,这些已知酶来自哺乳动物,果蝇或细菌(链霉菌属)。发现成熟的类似胰蛋白酶的蛋白酶与果蝇胰蛋白酶具有最高的一致性(48%)。基因产品的表达
编码上述的类似胰蛋白酶的尖孢镰孢蛋白酶的cDNA被插入Noma等(1986)描述的载体pCDV1—PL,得到质粒pSX180。cDNA的编码区作为Ncol—Xbal片断插入曲霉表达质粒P777(EP 0489748),该质粒被Bam HI切口,并部分被Xbal切口。为将克隆DNA的5’端连接到载体上,合成的接头DNA KFN 709/710(图1所示)被加入到连接反应中。得到的质粒pSX183与携带来自构巢曲霉(A.nidulans)(WO91/17243)的amdS的质粒pToC90共转化入米曲霉(IFO4177)中。转化子被选择出来以便在乙酰胺上生长。当在YPD培养基上生长时,上清液中出现的蛋白质比从尖孢镰孢上清液中分离的类似胰蛋白酶的蛋白酶稍大些。令人惊讶的是由L—苯甲酰—精氨酰—pNA(一种胰蛋白酶底物)的水解测不出基因产品的活性。实施例3酶原的活化
为得到有活性的类似胰蛋白酶的蛋白酶,在含有无活性类似胰蛋白酶的蛋白酶的发酵培养基中加入从尖孢镰孢上清液分离出的天冬氨酰蛋白酶(描述于实施例1中)
通过这样处理发现,从转化的米曲霉得到的蛋白质变得与在SDS凝胶中的标准蛋白质具有同样大小,并且由L—苯甲酰—精氨酰—pNA水解测定(数据未示出),样品显示出胰蛋白酶的活力。这就表明酶是以酶原形式被表达,并且证明了由DNA序列推出的前肽的存在。活性研究表明,前肽的作用是保持酶呈一非活性形式。

Claims (20)
1.一种活性重组类似胰蛋白酶的蛋白酶,其含有所附SEQID No.2所示的氨基酸序列中的第25—224氨基酸残基。
2.一种蛋白水解组合物,其含有如权利要求1所述的重组蛋白酶。
3.一种DNA构建物,其含有编码权利要求1所述的重组蛋白酶的DNA序列。
4.一种根据权利要求3的DNA构建物,其中DNA序列如所附SEQ ID No.1所示。
5.一种根据权利要求3或4任一项的DNA构建物,其中编码重组蛋白酶的DNA序列来自一种微生物。
6.一种根据权利要求5的DNA构建物,其中DNA序列来自细菌或真菌。
7.一种根据权利要求6的DNA构建物,其中DNA序列来自镰孢属的菌株,特别是尖孢镰孢菌株。
8.一种重组表达载体,其含有根据权利要求3—7任一项的DNA构建物。
9.一种细胞,其含有根据权利要求3—7任一项的DNA构建物或根据权利要求8的载体。
10.一种根据权利要求9的细胞,其是微生物细胞。
11.一种根据权利要求10的细胞,其是细菌细胞或真菌细胞。
12.一种根据权利要求11的细胞,其中细菌细胞是革兰氏阳性菌的细胞,如芽孢杆菌属或链霉菌属或者是革兰氏阴性菌的细胞,如大肠杆菌属,真菌细胞是酵母细胞,如糖酵母属,或丝状真菌的细胞,如曲霉属或镰孢属。
13.一种生产权利要求1所述的重组蛋白酶的方法,其中根据权利要求9—12任一项的细胞在有助于蛋白酶生产的条件下培养,随后从培养物中回收蛋白酶。
14.根据权利要求13的方法,其中重组蛋白酶以酶原的形式表达,并且细胞在蛋白水解酶存在的条件下培养,该蛋白水解酶能将蛋白酶的酶原转化为成熟酶。
14.根据权利要求19的方法,其中蛋白水解酶是一种金属蛋白酶,特别是芽孢杆菌金属蛋白酶。
15.一种含有权利要求1的重组蛋白酶的洗涤剂添加剂,可任意选择地呈无粉尘颗粒形式,稳定化的液体或被保护的酶的形式。
16.一种根据权利要求14的洗涤剂添加剂,每克添加剂含0.02—200毫克酶蛋白。
17.一种根据权利要求15或16的洗涤剂添加剂,其另外含有另一种酶,如另一种蛋白酶,淀粉酶,脂肪酶,过氧化物酶和/或纤维素酶。
18.一种含有根据权利要求1的重组蛋白酶的洗涤剂组合物。
19.一种根据权利要求18的洗涤剂组合物,其另外含有另一种酶,如另一种蛋白酶,淀粉酶,脂肪酶,过氧化物酶和/或纤维素酶。
