Claims (12)

- 哺乳動物細胞に投与されると、ウラシル、シトシン、アデニン、およびグアニンを含むヌクレオチドからなるオープンリーディングを含む対応するポリヌクレオチドと比較して、前記ポリヌクレオチドは、前記コードされたポリペプチドの増加された発現を有する、請求項2に記載の組成物。
- 哺乳動物細胞に投与されると、ウラシル、シトシン、アデニン、およびグアニンを含むヌクレオチドからなるオープンリーディングを含む対応するポリヌクレオチドと比較して、前記ポリヌクレオチドは、より長い半減期、または、タンパク質発現のより広い曲線下面積を有する、請求項2または3に記載の組成物。
- 末梢血単核細胞に投与されると、ウラシル、シトシン、アデニン、およびグアニンを含むヌクレオチドからなるオープンリーディングを含む対応するポリヌクレオチドと比較して、前記ポリヌクレオチドは、より低いレベルの検出可能なIFN−αおよびTNF−αをもたらす、請求項2〜4のいずれか一項に記載の組成物。
- 前記複数の脂質ナノ粒子が、80nm〜160nmの平均粒径、0.02〜0.20の平均PDI、および10〜20の脂質:ポリヌクレオチドの平均比(重量/重量)を有す
る、請求項2〜5のいずれか一項に記載の組成物。 - 前記脂質ナノ粒子が、DLin−DMA、DLin−K−DMA、DLin−KC2−DMA、98N12−5、C12−200、DLin−MC3−DMA、DODMA、DSDMA、DLenDMA、reLNP、PLGA、またはPEG化脂質を含む、請求項2〜6のいずれか一項に記載の組成物。
- 前記分泌タンパク質がサイトカインである、請求項2〜7のいずれか一項に記載の組成物。
- 前記結合ヌクレオシドの3’尾部配列が、約160個のヌクレオチドのポリA尾部およびポリA−G四重鎖からなる群から選択される、請求項2〜8のいずれか一項に記載の組成物。
- 前記少なくとも1つの5’末端キャップが、キャップ0、キャップ1、ARCA、イノシン、N1−メチル−グアノシン、2’フルオロ−グアノシン、7−デアザ−グアノシン、8−オキソ−グアノシン、2−アミノ−グアノシン、LNA−グアノシン、および2−アジド−グアノシンからなる群から選択される、請求項2〜9のいずれか一項に記載の組成物。
- 前記オープンリーディングフレームが、コドン最適化されてGC含量を偏らせている、請求項2〜10のいずれか一項に記載の組成物。
- 哺乳動物細胞内でポリペプチドを発現させる方法であって、
請求項1に記載の単離されたポリペプチド、または請求項2〜11のいずれか一項に記載の組成物を細胞に接触させることを含む、方法。
