Claims (21)
1. Instrument til detektion af mikrokolonier af målceller i en prøve, hvilke instrument omfatter: (a) en fotoelektrisk array-detektor med samleoptik til detektion af et detektions-område med i det mindste én dimension, som er > 1 mm, uden forstørrelse af detek-tionsområdet med mere end 5 gange; (b) en belysningskilde, som belyser detektionsområdet; og (c) en computer, som er programmeret til at modtage data opsamlet af den fotoelektriske array-detektor, og som er programmeret til billedanalyse, omfattende analyse af de nævnte data for at detektere én eller flere mikrokolonier, som har et mål på mindre end 50 mikrometer i i det mindste to orthogonale dimensioner, og som er programmeret til at kvantificere antallet af mikrokolonier, som er detekteret i detektionsområdet, hvor instrumentet detekterer en intrinsisk optisk egenskab for mikrokolonierne og hvor cellerne i den ene eller flere mikrokolonier forbliver kompetente til at replikere efterfølgende detektion.
2. Instrument ifølge krav 1, som yderligere omfatter en inkubator til mikrobiel replikation.
3. Instrument ifølge krav 2, hvor instrumentet er i stand til at detektere detektionsområdet på adskillige tidspunkter under inkubering.
4. Instrument ifølge krav 1, som yderligere omfatter en automatisk fokus til at fokusere på detektionsområdet.
5. Instrument ifølge krav 1, hvor samleoptikken ikke forstørrer.
6. Instrument ifølge krav 1, hvor billedanalysen yderligere omfatter detektion af voksende mikrokolonier.
7. Instrument ifølge krav 1, hvor instrumentet er i stand til automatisk at indføre prøven.
8. Instrument ifølge krav 1, hvor detektionen detekterer lys udsendt, spredt, reflekteret eller absorberet som et resultat af belysning af den ene eller flere mikrokolonier.
9. Instrument ifølge krav 1, hvor detektionen detekterer autofluorescens udsendt af mikrokolonierne.
10. Instrument ifølge krav 1, hvor belysningskilden anvenden én eller flere lasere.
11. Instrument ifølge krav 1, som yderligere omfatter ét eller flere optiske filtre, som alene lader udvalgte bølgelængder af lys passere.
12. Instrument ifølge krav 1, hvor instrumentet er i stand til at oprette adskillige billeder af detektionsområdet via registreringsmarkeringer.
13. Instrument ifølge krav 1, hvor den fotoelektriske array-detektor omfatter en CCD-detektor, fotomultiplikatorrør-detektor, eller en fotodiode-detektor, især en kølet CCD-detektor.
14. Instrument ifølge krav 1, hvor detektionen omfatter bestemmelse af lokationerne i detektionsområdet af den nævnte ene eller flere mikrokolonier.
15. Instrument ifølge krav 1, hvor billedanalysen skelner mellem objekter, som ændrer størrelse med tiden og objekter, som ikke ændrer størrelse med tiden.
16. Instrument ifølge krav 1, som yderligere omfatter et automatiseret X-Y trin til positionering af prøven i forhold til belysningskilden og detektoren.
17. Instrument ifølge krav 1, hvor computeren er i stand til automatisk at gemme udgangsdata for søgning.
18. Instrument ifølge krav 1, hvor computeren er programmeret til at detektere de nævnte mikrokolonier via et objekt-findende program ("object-finding utility"), som forbinder sammenhængende pixler, som har en værdi over en automatisk eller brugerdefineret tærskel, for etablering af en konturlinie omkring omkredsen af objektet, hvor omkreds-pixler og pixler indvendigt er definerede som objektet.
19. Instrument ifølge krav 1, hvor samleoptikken omfatter en samlelinse og en fokuseringslinse.
20. Instrument ifølge krav 1, hvor instrumentet automatisk sporer prøven via en stregkode eller tilsvarende markering.
21. Instrument ifølge krav 1, hvor instrumentet belyser detektionsområdet med synligt lys.
