Claims (19)
- AT 401 053 B Beispiel 137 1,2-Bis-(2,6-dimethylbenzoyloxy)-3,5-dinitrobenzol 5 Das Verfahren von Beispiel 134 wird bei einer Temperatur von 134*C wiederholt. Das Produkt wird aus 50 % Äthanol kristallisiert. Ausbeute: 60 % der Titelverbindung vom F. 175 bis 178*C. Beispiel 138 w 1,2-Bis-(3-äthoxycarbonylpropionyloxy)-3,5-dinitrobenzol Eine Lösung von 1 g 3,5-Dinitrobrenzkatechin in 2,5 ml Bernsteinsäureäthylesterchlorid wird 3 Stunden auf 100*C erhitzt. Das erhaltene Produkt wird in einer Säule mit Kieselgel gereinigt. Es wird die Titelverbindung vom F. 60 bis 63 · C erhalten. 15 Patentansprüche 1. Brenzkatechin-Derivate der allgemeinen FormelX (I) 25 in der Ri und R2 unabhängig voneinander ein Wasserstoffatom einen Acyl-, Aroyl- oder Alkylcarba-moylrest bedeuten, X ein Halogenatom, eine Nitro-, Cyano-, Trifluormethyl- oder Aldehydgruppe darstellt und R3 einen durch einen CwAlkoxyrest substituierten Ci-+-Alkoxy-Ci-4-alkylrest bedeutet, 30 oder einen der Reste der folgenden Formein darstellt: R, *4 -CH=C-cs_ ocer 35 wobei Rt eine Cyanogruppe oder einen Acylrest darstellt und R5 eine Cyanogruppe, einen Alkoxycar-bonylrest, einen Acylrest, eine gegebenenfalls durch eine oder mehrere Alkylgruppen oder Hydroxyal-kylgruppen substituierte Carbamoylgruppe oder eine Alkylpiperazinylcarbonylgruppe oder einen gegebenenfalls veresterten Carboxyalkylrest bedeutet oder aber FU und Rs zusammen einen 5-7 gliedrigen 40 Cykloalkanonring, der mit 3,4-Dihydroxy-5-nitrobenzy!iden substituiert ist, bilden; -(CH2)m-COR 45 50 wobei m einen Wert von 1 bis 7 hat und R eine Hydroxylgruppe, einen Alkylrest, eine Alkoxygruppe oder eine, gegebenenfalls durch eine oder mehrere Alkyl-, Alkinyl- oder Adamantylgruppen substituierte, Aminogruppe oder eine durch eine Benzylgruppe substituierte Piperazinylgruppe bedeutet; a wobei Re ein Wasserstoffatom ist und R9 Adamantyl ist oder 1¾ und R9 zusammen eine, gegebenenfalls durch Cyclohexylcarbonyl oder Piperidino substituierte, Piperidylgruppe bilden; und ihre physiolo-55 gisch verträglichen Salze.
- 2. Verbindungen nach Anspruch 1 der allgemeinen Formel I, in denen Ri und R2 beide Wasserstoffatome, Acetyl-, Propionyl-, Butyryl-, Benzoyl-bedeuten. 32 ΑΤ 401 053 Β
- 3. Verbindungen nach Anspruch 2 der allgemeinen Formel I, in denen Ri und R2 beide Wasserstoffatome sind.
- 4. Verbindungen nach einem der Ansprüche 1 bis 3 der allgemeinen Formel I. in denen X eine Nitro-, Chlor-, Cyano-, Trifluormethylgruppe in 5-Stellung bedeutet.
- 5. Verbindung nach Anspruch 4 der allgemeinen Formel I, in denen X eine 5-Nitrogruppe bedeutet.
- 6. Verbindungen nach einem der Ansprüche 1 bis 5 der allgemeinen Formel I, in denen R3 einen Rest -CE<4 bedeutet, wobei R» eine Cyanogruppe oder einen Ci -»-Acylrest darstellt und Rs eine Cyanogruppe, einen Ci-»-Acylrest, Ci-*-Alkoxycarbonyl, Aminocarbonyl, Ci -4.-Alkyl oder einen Ci -»-Dialkylamino-carbonylrest darstellt.
- 7. Verbindungen nach einem der Ansprüche 1 bis 6 der allgemeinen Formel I, in denen Ra den Rest -{CH2)m-COR bedeutet, wobei m einen Wert von 1 - 7 hat und R eine Hydroxyl- oder Aminogruppe oder eine Ci-»-Alkyl- oder Ci -»-Dialkylamino- oder eine Adamantylaminogruppe bedeutet.
- 8. Verbindungen nach einem der Ansprüche 1 bis 7 der allgemeinen Formel I, in denen Ra einen Rest der Formel -CO Νφ —a bedeutet, wobei Rs und R9 zusammengenommen eine Piperidino- oder eine 4-Cyclohexylcarbonylpipe-ridino oder 1-Piperidylpiperidinogruppe bedeuten.
- 9. Verbindungen nach Anspruch 8 der allgemeinen Formel I, in denen Ra eine Gruppe der Formel darstellt, wobei R» eine Cyano- oder Acetylgruppe bedeutet und Rs eine Cyano- oder Acetylgruppe bedeutet.
- 10. Verbindungen nach Anspruch 9 der allgemeinen Formel I, in denen Ra den Rest -(CH2H-COR darstellt, wobei R eine Amino-, Ci -»-Alkylamino-, Ci -»-Dialkylamino- oder Adamantylaminogruppe bedeutet. 11. 3,4-Dihydroxy-5-nitro-0,<d-dicyanostyrol. 12. N-Methyl-N-propargyl-5-(3,4-dihydroxy-5-nitrophenyl)-pentansäuremid. 13. N-(1-Adamantyl)-5-(3,4-dihydroxy-5-nitrophenyl)-pentansäureamid. 14. - 26. Verfahren zur Herstellung der neuen Verbindungen der allgemeinen Formel I nach Anspruch 1, dadurch gekennzeichnet, daß man entweder (a) eine säure- oder basenkatalysierte Kondensationsreaktion einer Verbindung der allgemeinen Formel Ra 0 -» , 1 77Γ\ (iij VW/020 I x in der Ri, R2 und X die in Anspruch 1 angegebenen Bedeutungen haben, mit einer Verbindung der allgemeinen Formel R4 (III) c92"R5 durchführt, die eine aktive Methyl- oder Methylengruppe aufweist und in der R4 und Rs die in Anspruch 1 angegebenen Bedeutungen haben, wobei eine Verbindung der allgemeinen Formel la RjO r2oX CE»C-R- (Xa) erhalten wird, in der die Substituenten wie vorstehend definiert sind, und deren Doppelbindung gegebenenfalls zu einer Einfachbindung reduziert werden kann; oder (b) eine Verbindung der allgemeinen Formel RjOHjO-/ O /—coy (X) in der Ri, R2 und X die im Anspruch 1 angegebenen Bedeutungen haben und Y ein Halogenatom oder einen anderen aktivierten Rest bedeutet, mit einem Amin der allgemeinen Formel 34 (XI] AT 401 053 Bin der Re und R9 die im Anspruch 1 angegebenen Bedeutungen haben, zu einer Verbindung der Formel R.O*_X umsetzt, in der Ri, R2, X, Rs und Rg die vorstehend angegebenen Bedeutungen haben.
- 27. Verbindungen der allgemeinen Formel I nach einem der Ansprüche 1 bis 25 zur Verwendung als Arzneistoff.
- 28. Eine Verbindung der allgemeinen Formel I nach einem der Ansprüche 1 bis 25 zur Verwendung bei der Behandlung des Parkinson-Syndroms.
- 29. Verwendung einer Verbindung der allgemeinen Formel I nach einem der Ansprüche 1 bis 25 zur Herstellung eines Arzneimittels zur Behandlung des Parkinson-Syndroms.
- 30. Arzneimittel, dadurch gekennzeichnet, daß sie mindestens eine Verbindung der allgemeinen Formel I nach einem der Ansprüche 1 bis 25 enthalten.
- 31. Arzneimittel zur Behandlung des Parkinson-Syndroms, dadurch gekennzeichnet, daß sie mindestens eine Verbindung der allgemeinen Formel I nach einem der Ansprüche 1 bis 25 und einen Gehalt an Levodopa enthalten.
- 32. Arzneimittel nach Anspruch 31, dadurch gekennzeichnet, daß es zusätzlich einen peripheren Decar-boxylase-lnhibitor enthält.
- 33. Arzneimittel nach Anspruch 32, dadurch gekennzeichnet, daß der Decarboxylase-Inhibitor Carbidopa ist.
- 34. Arzneimittel nach Anspruch 32, dadurch gekennzeichnet, daß der Decarboxylase-Inhibitor Benzerazid ist. Hiezu 2 Blatt Zeichnungen 35
