Claims (31)
1. Одноцепочечный многофункциональный полипептид, включающий (а) первый домен, содержащий антигенсвязывающий участок цепи иммуноглобулина или антитела, который специфически распознает антиген CD19, и (б) второй домен, содержащий антигенсвязывающий участок цепи иммуноглобулина или антитела, который специфически распознает антиген CD3.
2. Полипептид по п. 1, где эти два домена связаны полипептидным линкером.
3. Полипептид по п. 1 или 2, где первый и/или второй домен имитирует или соответствует VH- и VL-области природного антитела.
4. Полипептид по любому из пп. 1-3, где антитело представляет собой моноклональное антитело, синтетическое антитело или "гуманизированное" антитело.
5. Полипептид по любому из пп. 1-4, где по меньшей мере один из этих доменов представляет собой одноцепочечный фрагмент вариабельной области антитела.
6. Полипептид по любому из пп. 1-5, где домены расположены в следующем порядке: VLCD19-VHCD19-VHCD3-VLCD3.
7. Полипептид по любому из пп. 2-6, где полипептидный линкер включает несколько остатков глицина, аланина и/или серина.
8. Полипептид по любому из пп. 2-7, где полипептидный линкер включает несколько последовательных копий аминокислотной последовательности.
9. Полипептид по любому из пп. 2-8, где полипептидный линкер включает 1-5 аминокислотных остатков.
10. Полипептид по любому из пп. 2-9, где полипептидный линкер включает аминокислотную последовательность Gly Gly Gly GLy Ser.
11. Полипептид по любому из пп. 1-10, где первый домен включает по меньшей мере один CDR (гипервариабельный участок) VH- и VL-области, который имеет аминокислотную последовательность, кодируемую последовательностью ДНК, представленной на фиг. 8, от нуклеотида 82 до 414 (VL) и от нуклеотида 460 до 831 (VH), и/или где второй домен включает по меньшей мере один CDR VH- и VL-области, который имеет аминокислотную последовательность, кодируемую последовательностью ДНК, представленной на фиг. 8, от нуклеотида 847 до 1203 (VH) и от нуклеотида 1258 до 1575 (VL).
12. Полипептид по любому из пп. 1-11, где (а) антигенсвязывающий участок первого домена обладает аффинностью по меньшей мере примерно 10-7М, и/или (б) антигенсвязывающий участок второго домена обладает аффинностью ниже, чем примерно 10-7М.
13. Полипептид по любому из пп. 1-12, который представляет собой биспецифическое одноцепочечное антитело.
14. Полипептид по любому из пп. 1-13, включающий по меньшей мере один дополнительный домен.
15. Полипептид по п. 14, где дополнительный домен связан ковалентными или нековалентными связями.
16. Полипептид по п. 14 или 15, где по меньшей мере один дополнительный домен включает эффекторную молекулу, которая имеет конформацию, пригодную для проявления биологической активности, способную секвестровать ион или избирательно связываться с твердой подложкой или с предварительно выбранной детерминантой.
17. Полинуклеотид, который при экспрессии кодирует полипептид по любому из пп. 1-16.
18. Вектор, включающий полинуклеотид по п. 17.
19. Клетка, трансфектированная полинуклеотидом по п. 17 или вектором по п. 18.
20. Способ получения полипептида по любому из пп. 1-16, предусматривающий культивирование клетки по п. 19 и выделение полипептида из культуры.
21. Композиция, включающая полипептид по любому из пп. 1-16, полинуклеотид по п. 17 или вектор по п. 18.
22. Композиция по. 21, которая представляет собой фармацевтическую композицию, необязательно дополнительно включающую фармацевтически приемлемый носитель.
23. Композиция по п. 21, которая представляет собой диагностическую композицию, необязательно включающую дополнительные пригодные средства для обнаружения.
24. Применение полипептида по любому из пп. 1-16, полинуклеотида по п. 17 или вектора по п. 18 для приготовления фармацевтической композиции, предназначенной для лечения злокачественных заболеваний В-клеток, опосредованных В-клетками аутоиммунных заболеваний или истощения В-клеток.
25. Применение по п. 24, где злокачественное заболевание В-клеток представляет собой не-ходжкинскую лимфому.
26. Применение полинуклеотида по п. 17 или вектора по п. 18 для приготовления композиций для генной терапии.
27. Способ выявления активаторов или ингибиторов Т-клеточной активации или стимуляции, предусматривающий (а) культивирование Т-клеток и СD19-позитивных клеток, предпочтительно В-клеток, в присутствии полипептида по любому из пп. 1-16 и необязательно в присутствии соединения, обладающего способностью обеспечивать обнаруживаемый сигнал в ответ на Т-клеточную активацию соединением, подлежащим скринингу, в условиях, обеспечивающих активацию Т-клетки, и (б) обнаружение присутствия или отсутствия сигнала, полученного в результате взаимодействия соединения с клетками.
28. Способ приготовления фармацевтической композиции, предусматривающий стадии способа по п. 27 и приготовление на основе соединения, выявленного на стадии (б), фармацевтически приемлемой формы.
29. Способ по п. 28 или 29, где соединение, указанное на стадии (б), модифицируют с помощью пептидомиметиков.
30. Способ лечения злокачественных заболеваний В-клеток, опосредрованных В-клетками аутоиммунных заболеваний или истощения В-клеток, предусматривающий введение полипептида по любому из пп. 1-16, полинуклеотида по п. 17 или вектора по п. 18 человеку, страдающему от этих злокачественных или других болезней.
31. Способ замедления развития патологического состояния, которое вызывается В-клеточными нарушениями, предусматривающий введение полипептида по любому из пп. 1-16, полинуклеотида по п. 17 или вектора по п. 18 человеку, страдающему от этого патологического состояния.
