Description

本明細書で使用される用語「変異体」は、親の少なくとも1つのアミノ酸修飾と比べて、親抗体配列と異なる抗体配列を含む。具体的な実施形態において、本明細書における変異体抗体配列は、親抗体配列とは、少なくとも約80%のアミノ酸配列同一性、好ましくは、少なくとも約90%のアミノ酸配列同一性、より好ましくは、少なくとも約95%のアミノ酸配列同一性、より好ましくは、少なくとも約97%のアミノ酸配列同一性、より好ましくは、少なくとも約98%のアミノ酸配列同一性、最も好ましくは、少なくとも約99%のアミノ酸配列同一性を有する。抗体変異体は、抗体自体を指すことができ、前記親抗体の組成物、またはそれをコードするアミノ酸配列を含む。
本明細書で使用される用語「CDC」または「補体依存性細胞毒性」とは、1種または多種の補体タンパク質成分が標的細胞上に結合した抗体を識別してから、標的細胞を切断する反応を含む。
本発明に係るヒト化抗体親抗体は、HD37であり、これはマウスIgG1である(Leukocyte Typing II、pp 391−402)。
ヒトCD19のヒト化抗体のような抗体の結合能力を評価するための標準的なアッセイは、当技術分野で公知であり、例えば、ELISA、Westernブロット及びフローサイトメトリーを含む。適切なアッセイは、実施例に詳細に記載されている。結合を評価するために、CD19を安定的にトランスフェクションしたK562細胞を使用することができ、フローサイトメトリーでEC50を測定することができる。具体的な実施形態において、本発明のヒト化抗体のヒトCD19を安定にトランスフェクトしたK562細胞に対する相対的な結合親和力(EC50)は、100nM未満であり、好ましくは、10nM未満であり、より好ましくは、1〜10nMの間である。
本発明はさらに、ヒトCD19に結合するヒト化抗体及びそのフラグメントの単離をコードする核酸、ベクター及び前記核酸またはベクターを含む宿主細胞を提供する。核酸は、インタクトな細胞中、細胞溶解液中または部分的に精製されたもしくは実質的に精製された形態で位置することができる。
一態様において、本発明は、CD19タンパク質への結合を増強するように操作された抗体または抗体フラグメントを含む多種のキメラ抗原受容体(CAR)を提供する。一態様において、本発明は、CARを発現するように操作された細胞(例えば、T細胞)を提供し、ここで当該CAR T細胞(「CART」)は、抗腫瘍特性を示す。一態様において、細胞をCARで形質転換し、CARを細胞表面に発現させる。ある実施例において、細胞(例えば、T細胞)は、CARをコードするウイルスベクターで形質導入される。ある実施例において、ウイルスベクターは、レンチウイルスベクターである。ある実施形態において、細胞は、CARを安定的に発現することができる。
1.本発明の抗体は、マウス抗に比べて、抗−抗体反応(AAR)とヒト−抗−マウス抗体反応(HAMA)を生じない。

本実施例は、親抗体としてマウス抗HD37(J Immunol.1987 May 1; 138(9):2793−9)を用い、マウス抗HD37は、SEQ ID NO:19に示された軽鎖の可変領域及びSEQ ID NO:20に示された重鎖の可変領域を有し、Kabat、Chothia及びIMGTの3つの抗体CDR領域の命名スキームを組み合わせて、抗体軽鎖及び重鎖の6つのCDR領域配列を決定する。

IMGTデータベースからのgermline配列IGHV1−69*01(SEQ ID NO:43)+IGHJ6*01(SEQ ID NO:44)を、HD37重鎖の抗体テンプレートとして選択し、IMGTデータベースからのgermline配列IGKV7−3*01(SEQ ID NO:45)+IGKJ1*01(SEQ ID NO:46)を、HD37軽鎖の抗体テンプレートとして選択する。
HD37抗体の軽鎖CDR領域を抗体テンプレートIGKV7−3*01+IGKJ1*01のCDR領域で置換して、ヒト化抗体huHD37の軽鎖の可変領域(アミノ酸配列は、SEQ ID NO:1に示される)を構成した。HD37抗体の重鎖CDR領域を抗体テンプレートIGHV1−69*01+IGHJ6*01のCDR領域で置換し、IGHV1−69*01(SEQ ID NO:43)中の第27位グリシンをチロシンに変異させて、ヒト化抗体huHD37の重鎖の可変領域(アミノ酸配列は、SEQ ID NO:3に示される)を構成した。
huHD37軽鎖の可変領域と、HD37、VK7−3*01、VK7−3*01/J1*01の配列とを比較して、及びhuHD37重鎖の可変領域と、HD37、VH1_69*01、VH1_69*01/J1*01の配列とを比較した結果、図1に示された通りである。
ヒト化の抗体huHD37の軽鎖の可変領域(SEQ ID NO:1)、重鎖の可変領域(SEQ ID NO:3)によって、軽鎖の可変領域ヌクレオチド配列(SEQ ID NO:2)及び重鎖の可変領域ヌクレオチド配列(SEQ ID NO:4)を設計及び合成する。
軽鎖ヌクレオチド配列(SEQ ID NO:2)及び重鎖ヌクレオチド配列(SEQ ID NO:4)のそれぞれに対してプライマーを設計し、15つの柔軟なアミノ酸GGGGSGGGGSGGGGS(SEQ ID NO:42)からなるlinker連結を導入してscFv(SEQ ID NO:21)を生成し、ここで、1−124位は、重鎖の可変領域であり、140−251位は、軽鎖の可変領域であり、VHの上流に適切な酵素切断断部位および保護塩基を導入し、VLの下流に適切な酵素切断断部位および保護塩基を導入する。PCR産物を1%アガロースゲル電気泳動で分析し、精製して回收した。酵素切断後、ヒトFc断片の真核発現ベクターV152に連結された(Shanghai Ruijin Biotechnology Co.,Ltd.より購入)。
1)トランスフェクションの1日前に、6−7×105/mlの293F細胞を125ml培養フラスコに接種し、トランスフェクション当日に、3×107の細胞を28mlのFreeStyleTM 293 expression mediumで調整する。
2)Opti−MEM Iで30ugのDNAを希釈し、最終容量1mlであり、充分に混ぜ、Opti−MEM Iで60ulの293fectinTMを希釈し、最終容量1mlであり、充分に混ぜて、室温で5分間インキュベートし、希釈済のDNAを293fectinTMと混合させ、室温で20分間インキュベートし、2mlのDNA−293fectin複合体を28mlの細胞に加えて、37度、8%のCO2、125rpmで5〜7日培養し、上清を回収してlipid−DNA複合体を獲得する。
4)SECにより抗体の凝集状況を分析し、結果は、図2に示された通りであり、モノマー形態的の抗体の割合は、94.3%であり、モノマー形態の抗体の割合は、従来のヒト化抗体より有意に優れて、マウス抗HD37と比較して、凝集度も有意に低下した。
ヒトCD19を安定に発現するK562細胞(K562−CD19)とK562を使用し、細胞を収集し、完全増殖培地で細胞を洗浄し、そして約1−5×105個の細胞/ウェルでU型底マイクロタイタープレートにプレーティングした。勾配希釈したhuHD37のscFv_Fc融合抗体とK562−CD19/K562を氷上で30分間インキュベートし、続いて第2の抗体としてのFITC標識の抗ヒトFcとインキュベートした。2回の洗浄ステップの後、Guava easyCyteTM HT System装置を用いて分析を行い、そしてGraphPad Prismを用いて処理して実験データを処理して、EC50を得た。図4は、HD37及びhuHD37のK562−CD19及びK562細胞への結合の状況を示した。結果は、ヒト化のhuHD37と親抗体HD37の特異性的結合は、K562細胞に結合することなく、CD19のK562細胞を安定にトランスフェクションすることを示した。図5は、HD37及びヒト化のhuHD37のScFvがヒトIgG1 Fcに部分的にキメラ化後に、ヒトCD19を安定にトランスフェクトしたHEK293細胞との相対的な結合親和力(EC50)を示した。親HD37と比較して、ヒト化の抗体huHD37の親和力は向上された。
本実施例は、huHD37を親抗体とし、ファージディスプレイ法によりhuHD37を改造した。ヒト化抗体huHD37に基づくファージライブラリーの構築は、軽鎖及び重鎖のCDR3領域を保持し、縮重プライマーによって、それぞれ軽鎖のCDR1及びCDR2または重鎖のCDR1及びCDR2をランダム化することによって、2つのファージライブラリーを構築した。プライマー情報は、下表に示された通りである。
まず、抗体huHD37(scFv)(アミノ酸配列は、SEQ ID NO:21を参照し、ヌクレオチド配列は、SEQ ID NO:22を参照する)に基づいて、テンプレートプラスミドを構築する。軽鎖CDR1とCDR2のランダムされたファージライブラリーに対して、プライマーLMFとC37L1R、PCR増幅フラグメント1を使用し、プライマーC37L2FとFdR、PCR増幅フラグメント2を使用し、そしてブリッジPCRによってフラグメント1とフラグメント2を連結することにより、ランダム化された配列のscFv全長を得、そしてNcoI及びNotIを使用して全長のフラグメントを酵素切断し、T4連結酵素により、同じく酵素切断されたテンプレートプラスミドに連結した。TG1コンピテントセルへエレクトロポレーションし、記憶容量は、1.76E+9である。
実施例1に示されたように、VHの上流に適切な酵素切断断部位及び保護塩基を導入し、VLの下流に適切な酵素切断断部位及び保護塩基を導入した。PCR産物を1%アガロースゲル電気泳動で分析し、精製して回収した。酵素切断後、ヒトFc断片の真核発現ベクターV152に連結された(Shanghai Ruijin Biotechnology Co.,Ltd.より購入)。293Fectinにより293F細胞に一過性にトランスフェクトし、発現された。rProtein Aカラムによりアフィニティークロマトグラフィーした。
ヒトCD19を安定に発現するK562細胞(K562−CD19)とK562を使用し、細胞を収集し、完全増殖培地で細胞を洗浄し、そして約1−5×105個の細胞/ウェルでU型底マイクロタイタープレートにプレーティングした。勾配希釈したscFv_Fc融合抗体とK562−CD19/K562を氷上で30分間インキュベートし、続いて第2の抗体としてのFITC標識の抗ヒトFcとインキュベートした。2回の洗浄ステップの後、Guava easyCyteTM HT System装置を用いて分析を行い、そしてGraphPad Prismを用いて処理して実験データを処理して、EC50を得た。
3.1ヒト化抗体キメラ抗原受容体プラスミド(CAR)の構築
ベクターとしてPRRLSIN−cPPT.EF−1αを用いて、PRRLSIN−cPPT.EF−1α−huHD37−28Z、PRRLSIN−cPPT.EF−1α−huHD37−BBZ、PRRLSIN−cPPT.EF−1α−huHD37−28Z&IFNb及びPRRLSIN−cPPT.EF−1α−huHD37−BBZ&IFNb(図10)を含むヒト化抗体huHD37の第2世代、第4世代キメラ抗原受容体を発現するレンチウイルスプラスミドを構築した。
1)Tリンパ球活性化:約1×106/mLの密度でリンパ球培養基液で培養し、磁性ビーズ:細胞比2:1で同時に抗CD3及びCD28抗体で被覆された磁性ビーズ(Invitrogen)と最終濃度が500U/mLである組換えヒトIL−2(上海華新バイオハイテク株式会社)に加え、48hで刺激培養する。
2)Retronectin被覆24ウェルプレート:1ウェルあたりに380μl5μg/mlのretronectin溶液(PBS)を加え、4度で一晩インキュベートする。
3)24ウェルプレート中のretronectin溶液(PBS)を捨てて、1mlのPBSで2回洗浄する。
5)MOI=10に従って、PBMC細胞に濃縮後のレンチウイルスを加え、32度、1800rpmで、遠心分離40min後、細胞培養インキュベーターに移す。
6)増幅培養:感染後の細胞を、5×105/mLの密度で一日おきに継代し、同時にリンパ球培養液に最終濃度が500U/mLである組換えヒトIL−2を補充する。
1)レンチウイルス感染Tリンパ球の培養してから第7日目に、1×106のT細胞を採取し、2mlの遠心分離管内に等分する。
2)4度、5000rpmで、遠心分離5minし、上清を捨て、PBSで2回洗浄する。
3)対照群細胞を50μlのPE−SA(1:200で希釈)抗体に加え、氷上で45minインキュベートし、PBS(2%のNBS)で2回洗浄し、再懸濁して対照とする。
5)2mlのPBS(2%のNBS)再懸濁細胞を加え、4℃、5000rpm/minで、5分間遠心分離して、上清を捨て、2回繰り返す。
6)500μlのPBS(2%のNBS)を加え、フローチューブに移す。フローサイトメトリーによってPEチャネルを検出し、CAR陽性のT細胞の割合を決定した。

なお、対照群Mock+T細胞陽性率は90%である。
1)10cmプレートでK562、K562−CD19、Daudi及びRaji細胞のような腫瘍細胞を培養する。
2)1×106の前記細胞を採取し、二等分し、4℃、5000rpm/minで、5分間遠心分離する。
3)一群の細胞に500μlのPBS(1%NBS)を直接に加え、再懸濁後に対照とする。
5)2mlのPBS(1%のNBS)再懸濁細胞を加え、4℃、5000rpm/minで、5分間遠心分離し、上清を捨て、2回繰り返す。
6)500μlのPBS(1%のNBS)再懸濁細胞を加え、細胞をフローチューブに移す。
7)フローサイトメトリーによってPerCPチャネルを検出する。
8)フロー検出結果、K562細胞は、CD19タンパク質を発現せず、K562−CD19、Daudi及びRajiはすべてCD19陽性細胞であることを示した(図12)。
1)標的細胞:75μLの2×105/mLのK562、K562−CD19、Daudi及びRajiを96ウェルプレートにそれぞれ接種する。
2)エフェクター細胞:3:1、1:1または1:3のエフェクター:標的比にT−Mock及び異なるキメラ抗原受容体を発現するCAR T細胞を加える。
3)各群にそれぞれ4つの複製ウェルを設け、4つの複製ウェルの平均値を取る。検出時間は18hである。

[5]培養基背景対照:培養基の血清によって生じるLDH活性及びフェノールレッドによる背景吸収を補正する。
6)細胞毒性の計算式は以下の通りである。%細胞毒性=[(実験群−エフェクター細胞自発群−標的細胞自発群)/(標的細胞最大−標的細胞自発)]×100。
計算の前に、培養基対照をエフェクター細胞対照、標的細胞対照、実験群から差し引き、標的細胞最大溶解量から体積を差し引いた。
出願人は、実施例2で得られたクローン6B3、8E3を用いて実施例3を繰り返し、結果は、6B3及び8E3がhuHD37と同様の効果を生じることを示した。

- 請求項13〜19のいずれか一項に記載のキメラ抗原受容体、または請求項20に記載のヌクレオチド配列、または請求項21に記載の発現ベクター、または請求項22に記載のウイルスの、CD19を発現する腫瘍細胞を標的とする遺伝子修飾された免疫細胞を製造するための使用。
- 請求項20に記載のヌクレオチド配列、または請求項21に記載の発現ベクターもしくは請求項22に記載のウイルスが形質導入されるか;または請求項13〜19のいずれか一項に記載のキメラ抗原受容体を発現する、遺伝子修飾された免疫細胞。
- キメラ抗原受容体以外のサイトカイン、他のキメラ抗原受容体、ケモカイン受容体、PD−1発現を低下させるsiRNAまたはPD−L1を遮断するタンパク質、TCR、または安全スイッチを含む他の配列をさらに発現し;
好ましくは、前記サイトカインには、IL−12、IL−15、IL−21、またはI型インターフェロンが含まれ;
好ましくは、前記ケモカイン受容体には、CCR2、CCR5、CXCR2、またはCXCR4が含まれ;
好ましくは、前記安全スイッチには、iCaspase−9、Truancated EGFRまたはRQR8が含まれることを特徴とする、
請求項24に記載の遺伝子修飾された免疫細胞。 - CD19を発現する腫瘍を阻害するための薬物を製造するための、請求項24または25に記載の遺伝子修飾された免疫細胞の使用。
- 請求項1〜8のいずれか一項に記載の抗体;及び
それに結合された機能性分子を含み;前記機能性分子は、CD19以外の他の腫瘍表面マーカーを標的とする分子、腫瘍を阻害する分子、免疫細胞の表面マーカーを標的とする分子または検出可能な標識から選択される、多機能免疫複合体。 - 前記腫瘍表面マーカーを標的とする分子が、腫瘍表面マーカーに結合する抗体またはリガンドであるか;または
前記腫瘍を阻害する分子が、抗腫瘍のサイトカインまたは抗腫瘍の毒素であり;好ましくは、前記サイトカインは、IL−12、IL−15、IFN−beta、TNF−alphaを含むことを特徴とする、
請求項27に記載の多機能免疫複合体。 - 前記免疫細胞の表面マーカーを標的とする分子が、T細胞表面マーカーに結合する抗体であり、それは請求項1〜8のいずれか一項に記載の抗体とともに、T細胞の関与する二機能性抗体を形成し、好ましくは、前記T細胞表面マーカーに結合する抗体は、抗CD3抗体であることを特徴とする、
請求項27に記載の多機能免疫複合体。 - 融合ポリペプチドであり、リンカーペプチドを請求項1〜8のいずれか一項に記載の抗体とそれに結合された機能性分子との間にさらに含むことを特徴とする、
請求項27に記載の多機能免疫複合体。 - 請求項27〜30のいずれか一項に記載の多機能免疫複合体をコードするヌクレオチド配列。
- 抗腫瘍薬物を製造するための、または
CD19を発現する腫瘍の診断用試薬を製造するための;または
キメラ抗原受容体修飾の免疫細胞を製造するための請求項27〜30のいずれか一項に記載の多機能免疫複合体の使用。 - 請求項1〜8のいずれか一項に記載の抗体または当該抗体をコードするヌクレオチド配列;または
請求項13〜19のいずれか一項に記載のキメラ抗原受容体または当該キメラ抗原受容体をコードするヌクレオチド配列;または
請求項24または25に記載の遺伝子修飾された免疫細胞;または
請求項27〜30のいずれか一項に記載の免疫複合体または当該複合体をコードするヌクレオチド配列
を含む、薬物組成物。
