도 3은 dsRNA로 처리한 팔머 아마란쓰 식물의 이환률(morbidity)을 나타낸다. 도 3a는 글리포세이트 처리 후 7일째의 식물을 묘사한다. 도 3b는 긴 dsRNA 용액으로 처리한 후 72시간 후에 글리포세이트 처리한 표면활성제 처리된 식물을 묘사한다. 도 3c는 짧은 dsRNA 용액으로 처리한 후 72시간 후 글리포세이트 처리한 표면활성제 처리된 식물을 묘사한다.

Claims (16)
- 성장하는 식물 또는 식물 기관에서 표적 내인성 유전자의 발현을 조절하는 방법으로서,
표적 내인성 유전자 또는 당해 표적 내인성 유전자로부터 전사된 전령 RNA 내의 18개 이상의 인접 뉴클레오타이드의 서열과 본질적으로 동일하거나 그 서열에 대해 본질적으로 상보적인 서열을 갖는 적어도 하나의 폴리뉴클레오타이드, 및 상기 적어도 하나의 폴리뉴클레오타이드가 성장하는 식물 또는 식물 기관의 내부에 침투하도록 하는 유효량의 전달제로 구성된 폴리뉴클레오타이드 조성물을 성장하는 식물 또는 식물 기관 위에 국소 피복함으로써 상기 적어도 하나의 폴리뉴클레오타이드가 상기 표적 내인성 유전자의 발현을 조절함을 포함하는 방법. - 성장하는 식물에서 제초제의 활성을 강화시키는 방법으로서,
(a) (i) 제초제와 상호작용하는 단백질을 발현하는, 성장하는 식물의 내인성 유전자 내의 18개 이상의 인접 뉴클레오타이드의 서열과 본질적으로 동일하거나 이에 대해 본질적으로 상보적인 서열을 갖는 적어도 하나의 폴리뉴클레오타이드, 및
(ⅱ) 상기 폴리뉴클레오타이드가 상기 성장하는 식물의 내부에 침투하도록 하는 유효량의 전달제로 구성된 폴리뉴클레오타이드 조성물을 상기 성장하는 식물의 표면에 국소 피복시키는 단계; 및
(b) 상기 제초제를 상기 성장하는 식물에 적용시키는 단계를 포함함으로써, 상기 적어도 하나의 폴리뉴클레오타이드가 상기 성장하는 식물의 내부로 침투하여 상기 내인성 유전자의 억제를 유도함으로써 상기 성장하는 식물에서 상기 제초제의 활성을 강화시키는 방법. - 표면활성제 및 적어도 하나의 식물 치사제를 포함하는, 성장하는 식물의 외부 표면 위에 국소 피복하도록 조정된 액상의 제초제 조성물로서, 당해 식물 치사제가, 식물 유전자의 서열 또는 식물 유전자의 전사된 RNA의 서열과 본질적으로 동일하거나 이에 대해 본질적으로 상보적인 서열을 갖는 폴리뉴클레오타이드를 포함하며, 상기 폴리뉴클레오타이드가 상기 식물 유전자에 대한 프로브(probe)에 하이브리드화하는 작은 RNA의 전신계적 생산을 수행하는 것을 개선점으로 하는 액상의 제초제 조성물.
- 비-뉴클레오타이드 제초제를 포함하는 액상의 제초제 조성물로서, 당해 조성물이, 식물 유전자의 서열 또는 식물 유전자의 전사된 RNA의 서열과 본질적으로 동일하거나 이에 대해 본질적으로 상보적인 서열을 갖는 폴리뉴클레오타이드를 추가로 포함하는 것을 개선점으로 하는 액상의 제초제 조성물.
- (a) 식물의 내인성 유전자 서열 또는 당해 내인성 유전자로부터 전사된 RNA의 서열과 본질적으로 동일하거나 또는 이에 대해 본질적으로 상보적인 서열을 갖는 전사가능하지 않은 이중가닥 DNA 폴리뉴클레오타이드의 용액; 여기서 상기 이중 가닥 DNA 폴리뉴클레오타이드는 상기 내인성 유전자의 사일런싱을 수행하기 위하여 상기 식물의 세포 내 생리학적 조건하에서 상기 내인성 유전자 또는 상기 내인성 유전자로부터 전사된 상기 RNA로 하이브리드화 할 수 있다; 및
(b) 상기 식물의 표면으로부터 상기 식물의 세포내로 상기 이중가닥 DNA 폴리뉴클레오타이드의 전달을 촉진하는데 효과적인 전달제로 본질적으로 이루어진 조성물. - (a) 식물의 세포내에서 생리학적 조건하에 내인성 유전자 또는 당해 내인성 유전자로부터 전사된 RNA에 하이브리드화하여 상기 내인성 유전자의 사일런싱을 수행할 수 있는, 식물의 내인성 유전자의 서열 또는 당해 내인성 유전자로부터 전사된 RNA의 서열과 본질적으로 동일하거나 또는 이에 대해 본질적으로 상보적인 서열을 갖는 전사가능하지 않은 단일가닥 DNA 폴리뉴클레오타이드의 용액; 및
(b) 상기 식물의 표면으로부터 상기 식물의 세포내로 상기 단일가닥 DNA 폴리뉴클레오타이드의 전달을 촉진하는데 효과적인 전달제로 본질적으로 이루어진 조성물. - (a) 식물의 세포내에서 생리학적 조건하에 내인성 유전자 또는 당해 내인성 유전자로부터 전사된 RNA에 하이브리드화하여 상기 내인성 유전자의 사일런싱을 수행할 수 있는, 식물의 내인성 유전자의 서열 또는 당해 내인성 유전자로부터 전사된 RNA의 서열과 본질적으로 동일하거나 또는 이에 대해 본질적으로 상보적인 서열을 갖는 전사가능하지 않은 이중가닥 RNA 폴리뉴클레오타이드의 용액; 및
(b) 상기 식물의 표면으로부터 상기 식물의 세포내로 상기 이중가닥 RNA 폴리뉴클레오타이드의 전달을 촉진하는데 효과적인 전달제로 본질적으로 이루어진 조성물. - (a) 식물의 세포내에서 생리학적 조건하에 내인성 유전자 또는 당해 내인성 유전자로부터 전사된 RNA에 하이브리드화하여 상기 내인성 유전자의 사일런싱을 수행할 수 있는, 식물의 내인성 유전자의 서열 또는 당해 내인성 유전자로부터 전사된 RNA의 서열과 본질적으로 동일하거나 또는 이에 대해 본질적으로 상보적인 서열을 갖는 전사가능하지 않은 단일가닥 RNA 폴리뉴클레오타이드의 용액; 및
(b) 상기 식물의 표면으로부터 상기 식물의 세포내로 상기 단일가닥 RNA 폴리뉴클레오타이드의 전달을 촉진하는데 효과적인 전달제로 본질적으로 이루어진 조성물. - (a) 식물의 세포내에서 생리학적 조건하에 내인성 유전자 또는 당해 내인성 유전자로부터 전사된 RNA에 하이브리드화하여 상기 내인성 유전자의 사일런싱을 수행할 수 있는, 식물의 내인성 유전자의 서열 또는 당해 내인성 유전자로부터 전사된 RNA의 서열과 본질적으로 동일하거나 또는 이에 대해 본질적으로 상보적인 서열을 갖는 전사가능하지 않은 이중가닥 DNA/RNA 하이브리드 폴리뉴클레오타이드의 용액; 및
(b) 상기 식물의 표면으로부터 상기 식물의 세포내로 상기 이중가닥 DNA/RNA 하이브리드 폴리뉴클레오타이드의 전달을 촉진하는데 효과적인 전달제로 본질적으로 이루어진 조성물. - 자생 식물의 세포내에서 생리학적 조건하에 (i) 당해 자생 식물의 성장 또는 생애를 유지시키기 위한 필수 유전자이거나, (ⅱ) 상기 자생 식물에 대해 내성인 제초제를 제공하는 단백질을 암호화하거나, (ⅲ) RNA 조절제로 전사하는 내인성 유전자로부터 전사되는 전령 RNA에 하이브리드화할 수 있는 전신계적인 단일가닥 RNA를 상기 자생 식물의 세포내에 제공하는 폴리뉴클레오타이드 올리고머를 포함하는 조성물을 상기 자생 식물의 잎 위에 적용함을 포함하여, 제초제 내성 작물의 현지(field)에서 성장하는 표적화된 제초제 내성 자생 식물을 선택적으로 방제하기 위한 방법.
- 청구항 1에 따르는 조성물을 살아있는 식물의 조직 위에 국소 적용시킴을 포함하여, 식물내의 내인성 유전자를 조절함으로써 역유전학(reverse genetics)을 조사하는 방법.
- (a) 폴리뉴클레오타이드 올리고머에 의한 침투에 대해 식물을 컨디셔닝하기 위한 제제, 및 (b) 상기 식물에 화학적 제초제에 내한 내성을 제공하는 단백질을 암호화하거나 필수 유전자인, 안티-센스 또는 센스 배향의 상기 식물의 내인성 유전자의 18개 이상의 인접 뉴클레오타이드의 적어도 하나의 분절을 포함하는 적어도 하나의 폴리뉴클레오타이드 쇄를 포함하는 폴리뉴클레오타이드 올리고머의 수용액을 포함하는 제초제 조성물.
- 내인성 유전자를 억제하기 위한 외인성 DNA 또는 RNA를 포함하는 식물로서,
상기 외인성 DNA는 상기 식물의 염색체 내로 통합되지 않고 외인성 RNA는 상기 식물의 염색체로 통합된 DNA로부터 전사되지 않으며, 상기 내인성 유전자는 식물이 종자로부터 생긴 후 당해 식물에 대한 폴리뉴클레오타이드의 국소 적용에 의해 억제되는 식물. - 식물 세포의 외부에 상기 폴리뉴클레오타이드를 국소 적용함으로써 식물 세포의 내부로 폴리뉴클레오타이드를 전달하기 위한 방법으로서, 폴리뉴클레오타이드를 오가노실리콘 표면활성제와 조합함을 포함하는 방법.
- 청구항 1, 2, 10, 11 및 14 중의 어느 한 항에 있어서,
상기 표적 내인성 유전자의 발현이 국소 피복된 것 이외의 식물 세포내에서 조절되고,
상기 표적 내인성 유전자가 적어도 하나의 식물 기관 내에서 전신계적으로 조절되거나;
상기 내인성 표적 유전자가 5-에놀피루빌쉬키메이트-3-포스페이트 신타제 (EPSPS), 아세토하이드록시산 신타제 또는 아세토락테이트 신타제(ALS), 아세틸-보조효소 A 카복실라제(ACCase), 디하이드로프테로에이트 신타제, 파이토엔 데사투라제(PDS), 프로토포르피린 IX 옥시게나제(PPO), 하이드록시페닐피루베이트 디옥시게나제(HPPD), 파라-아미노벤조에이트 신타제, 글루타민 신타제(GS), 1-데옥시-D-크실룰로즈 5-포스페이트(DOXP) 신타제, 디하이드로프테로에이트(DHP) 신타제, 페닐알라닌 암노니아 리아제(PAL), 글루타티온 S-트랜스퍼라제(GST), 광시스템 Ⅱ의 D1 단백질, 모노-옥시게나제, 시토크롬 P450, 셀룰로즈 신타제, 베타-투불린, RUBISCO, 해독 개시 인자, 파이토엔 데사투라제 및 이중가닥 DNA 아데노신 트리폴리포스파타제(ddATP)로 이루어진 그룹 중에서 선택된 단백질을 암호화하거나;
폴리뉴클레오타이드가 내인성 유전자 또는 상이한 내인성 유전자의 상이한 분절을 표적화하거나;
식물의 표면으로부터 세포내로의 상기 폴리뉴클레오타이드의 침투가 오가노실리콘을 포함하는 전달제를 사용하거나;
식물의 표면으로부터 세포내로 상기 폴리뉴클레오타이드의 침투가 암모늄 염을 포함하는 전달제를 사용하거나;
상기 식물이 개방된 현지에서 성장하거나;
상기 식물이 온실에서 성장하거나;
상기 식물이 또한 비-폴리뉴클레오타이드 제조제로 분무되거나;
상기한 것들 중의 하나 이상의 조합인 방법. - 청구항 3, 4, 5, 6, 7, 8, 9 및 12 중의 어느 한 항에 있어서,
상기 전달제가 표면활성제 및 염으로 이루어진 그룹 중에서 선택되거나;
상기 표면활성제가 오가노실리콘 표면활성제이거나;
상기 오가노실리콘 표면활성제가 실리콘 폴리에테르 공중합체이거나;
상기 실리콘 폴리에테르 공중합체가 폴리알킬렌 옥사이드 변형된 헵타메틸 트리실록산 및 알릴옥시폴리프로필렌 글리콜 메틸에테르(Silwet? L-77 표면활성제로서 구입가능함)의 공중합체이거나;
비-폴리뉴클레오타이드 제초제 분자를 추가로 포함하거나;
