- Die Erfindung betrifft ein Verfahren zur Herstellung von (1R,2S,4R)-(–)-2-[(2'-{N,N-Dimethylamino}-ethoxy)]-2-[phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan und pharmazeutisch annehmbaren Säureadditionssalzen davon.

- Die Verbindung der Formel I fällt unter die allgemeine Formel I des
Ungarischen Patents Nr. 179 164 Ungarischen Patent Nr. 179 164 - Die Herstellung der Verbindung der Formel I wurde tatsächlich in dem
Ungarischen Patent Nr. 212 574 - Es ist sehr wichtig und es ist zu betonen, dass ein beträchtlicher Teil von (+)-Campher der Formel II, der in der Grignard-Reaktion verwendet wird, nicht reagiert, und dieses Ausgangsmaterial kann technisch nicht von dem gewünschten Produkt entfernt werden aufgrund der physikalischen Eigenschaften von (+)-Campher und der Labilität der gebildeten Verbindung (1R,2S,4R)-(–)-2-[Phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan-2-ol der Formel III, da die Verbindung (1R,2S,4R)-(–)-2-[Phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan-2-ol der Formel III für eine Zersetzung empfänglich ist. Aus diesem Grund findet, gemäß dem in dem
Ungarischen Patent Nr. 212 574 - Das vorstehend Gesagte führt zu den Nachteilen des Verfahrens, das in dem
Ungarischen Patent Nr. 212 574 - Ein weiterer Nachteil der Umkristallisation aus Dimethylformamid ist der, dass die Spuren des Lösungsmittels nicht von dem gewünschten pharmazeutischen aktiven Hauptstoff bis zum erforderlichen Ausmaß entfernt werden können.
- Es ist herausgefunden worden, dass im Fall der Alkylierungsreaktion von (1R,2S,4R)-(–)-2-[Phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan-2-ol der Formel III, die mit (2-{Chlor}-ethyl)-dimethylamin durchgeführt wird, (+)-Campher der Formel II immer vorliegt, zu der Bildung von beträchtlichen Mengen an Nebenprodukten, z. B. (1R,3S,4R)-3-[(2'-{N,N-Dimethylamino}-ethyl)]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan-2-on der Formel führt.
- Das Nebenprodukt (1R,3S,4R)-3-[(2'-{N,N-Dimethylamino}-ethyl)]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan-2-on der Formel V wird wie folgt gebildet: Unter den Bedingungen, die in der Veretherungsreaktion zur Anwendung kommen, bildet (+)-Campher der Formel II ein Alkalisalz in der Position 3, welches seinerseits mit dem (2-{Chlor}-ethyl)-dimethylamin, das als Alkylierungsmittel verwendet wird, reagiert, wobei die Verbindung der Formel V erhalten wird. Die Menge des Nebenproduktes der Formel V kann so hoch wie 1 bis 10% sein. Die Löslichkeit des Fumarats 2-(E)-Butendioat (1:1) der Verbindung von (1R,3S,4R)-3-[(2'-{N,N-Dimethylamino}-ethyl)]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan-2-on der Formel V ist in etwa identisch mit der des Fumarats der gewünschten Verbindung (1R,2S,4R)-(–)-2-[(2'-{N,N-Dimethylamino}-ethoxy)]-2-[phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan der Formel I und kristallisiert deshalb mit dem Fumarat der Verbindung (1R,2S,4R)-(–)-2-[(2'-{N,N-Dimethylamino}-ethoxy)]-2-[phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan der Formel I und kontaminiert das gewünschte Endprodukt. Wenn die Veretherung in Toluol durchgeführt wird, wie in dem
Ungarischen Patent 212 574 - Das Salz ist eine stark unlösliche Verbindung und kann nur aus Dimethylformamid umkristallisiert werden. Gleichwohl ist die Umkristallisation aus Dimethylformamid nicht in der Lage, die Verbindung (1R,2S,4R)-(–)-2-[(2'-{N,N-Dimethylamino}-ethoxy)]-2-[phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan der Formel I in einer Reinheit bereitzustellen, die gemäß den Pharmacopoeia erforderliche ist, und zwar aus den folgenden Gründen:
- a) Das Produkt, welches nach der Umkristallisation aus Dimethylformamid erhalten wird, enthält immer noch die Verbindung (1R,3S,4R)-3-[(2'-{N,N-Dimethylamino}-ethyl)]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan-2-on der Formel V in einer Menge oberhalb des Schwellenwertes, der von der Pharmacopöe erlaubt ist (etwa 0,5%);
- b) Dimethylformamid besitzt einen hohen Siedepunkt und kann nicht aus dem Produkt im erforderlichen Ausmaß entfernt werden, da bei der hohen Temperatur eine Zersetzung des Produktes stattfindet.
- Eine Reinigung zum Erhalt von Produkten mit einer Reinheit, die für Medikamente gemäß den Pharmacopoeias ausreichend ist, könnte nicht durch gängige Reinigungsprozesse erreicht werden, wie durch Umkristallisation aus Lösungsmitteln oder fraktioneller Destillation.
- Wenn man die strengen Anfordernisse der Pharmacopoeia berücksichtigt, könnten Verunreinigungen, die in zu hohen Mengen vorliegen, den Einsatz des aktiven Hauptstoffes für pharmazeutische Zwecke gefährden. Die Verunreinigung (1R,3S,4R)-3-[(2'-{N,N-Dimethylamino}-ethyl)]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan-2-on der Formel V kann deshalb Probleme bei der Verwendung der Verbindung (1R,2S,4R)-(–)-2-[(2'-{N,N-Dimethylamino}-ethoxy)]-2-[phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan der Formel I als aktiven Wirkstoff verursachen.
- Im Ergebnis kann behauptet werden, dass die Reinigung des (1R,2S,4R)-(–)-2-[(2'-{N,N-Dimethylamino}-ethoxy)]-2-[phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan der Formel I, das durch das bekannte Verfahren hergestellt wird, durchgeführt wird, Dimethylformamid das einzig denkbare Lösungsmittel wäre. Gleichwohl ist dieses Umkristallisationsverfahren für die Herstellung eines pharmazeutisch aktiven Bestandteils ungeeignet, der die Anfordernisse der Pharmacopoeia erfüllen soll, da Dimethylformamid einen so hohen Siedepunkt besitzt, dass Spuren davon nicht aus dem Produkt in einem ausreichenden Ausmaß entfernt werden können. Bei der erforderlichen hohen Temperatur erfährt die Verbindung (1R,2S,4R)-(–)-2-[(2'-{N,N-Dimethylamino)-ethoxy)]-2-[phenyl]-1‚7,7-tri-[methyl]-bicyclo[2.2.1]heptan der Formel I eine Zersetzung.
- Wie bereits oben offenbart, liegt (+)-Campher der Formel II während der Alkylierungsreaktion vor. Aus (+)-Campher wird als weitere Verunreinigung (1R,4R)-2-[(2'-{N,N-Dimethylamino}-ethoxy)]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan der Formel gebildet. Wenn ein Alkalimetallhydrid oder Alkalimetallamid als basisches salzbildendes Mittel verwendet wird, beträgt die Menge der Kontamination (1R,4R)-2-[(2'-{N,N-Dimethylamino}-ethoxy)]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan der Formel IV 1 bis 10%. Die Verbindung (1R,4R)-2-[(2'-{N,N-Dimethylamino}-ethoxy)]-1,7,7-tri-[methyl]-bicyclo[2.2.1]-heptan der Formel IV ist vom Stand der Technik her bekannt [Yakugaku Zasshi, 75, 1377, (1955); Chem., Abstr. 9340 (1956)]. Die Verbindung (1R,4R)-2-[(2'-{N,N-Dimethylamino}-ethoxy)]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan der Formel IV wird wie folgt gebildet: Das Alkalimetallhydrid oder Alkalimetallamid, welches zur Natriumsalzbildung in dem Veretherungsschritt verwendet wird, reduziert 1 bis 10% vom (+)-Campher der Formel II zu Borneol, welches unter den angewandten Reaktionsbedingungen zu dem Alkalimetallsalz umgewandelt wird, und das genannte Alkalimetallsalz tritt mit (2-{Chlor}-ethyl)-dimethylamin in eine Alkylierungsreaktion ein. Gleichwohl kann der Borneolether (1R,4R)-2-[(2'-{N,N-Dimethylamino}-ethoxy)]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan der Formel IV aus der gewünschten Verbindung (1R,2S,4R)-(–)-2-[(2'-{N,N-Dimethylamino}-ethoxy)]-2-[phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan der Formel I im Verlauf der Aufarbeitung der Reaktionsmischung getrennt werden.
- Das der Erfindung zugrunde liegende Problem ist es, ein Verfahren zur Herstellung von (1R,2S,4R)-(–)-2-[(2'-{N,N-Dimethylamino}-ethoxy)]-2-[phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan der Formel I und pharmazeutisch annehmbaren Säureadditionssalzen davon mit einem höheren Reinheitsgrad ohne der Notwendigkeit von Umkristallisationsreinigungsschritten bereitzustellen, welche ohnehin nur zu einer unzureichenden Reinigung führen würden und die Ausbeute senken würden, und außerdem den Nachteil aufweisen würden, dass das restliche Lösungsmittel nicht von dem Endprodukt bis zu einem ausreichenden Ausmaß selbst durch komplizierte Verfahren entfernt werden könnte.
- Überraschenderweise wurde das Obenstehende durch die vorliegende Erfindung gelöst.
- Die vorliegende Erfindung basiert auf der überraschenden Erkenntnis, dass, wenn die Reaktion zwischen der Reaktionsmischung, welche die Verbindung (1R,2S,4R)-(–)-2-[Phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan-2-ol der Formel III und (2-{Chlor}-ethyl)-dimethylamin enthält, in Gegenwart eines Alkalimetallhydrids oder Alkalimetallamids in einem Medium, das mindestens 50 Gew.-%, vorzugsweise 75 Gew.-%, an einer oder mehreren Verbindung(en) mit einem 5- oder 6-gliedrigen gesättigten heterocyclischen Ring mit 2 Sauerstoffringsatomen als [einem] Lösungsmittel(n), insbesondere Dioxolan und Dioxan, enthält, durchgeführt wird, die Reaktion weitestgehend in Richtung auf die Bildung des gewünschten (1R,2S,4R)-(–)-2-[(2'-{N,N-Dimethylamino}-ethoxy)]-2-[phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan der Formel I begünstigt wird, und das Nebenprodukt (1R,3S,4R)-3-[(2'-{N,N- Dimethylamino}-ethyl)]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan-2-on der Formel V nur in einer minimalen Menge gebildet wird. Die obenstehende Erkenntnis ermöglichst die Herstellung der gewünschten Verbindung (1R,2S,4R)-(–)-2-[(2'-{N,N-Dimethylamino}-ethoxy)]-2-[phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan der Formel I in höheren Ausbeuten und mit einem höheren Grad der Reinheit, insbesondere unter Berücksichtigung der Verunreinigung (1R,3S,4R)-3-[(2'-{N,N-Dimethylamino}-ethyl)]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan-2-on der Formel V. Dadurch kann die Verbindung (1R,2S,4R)-(–)-2-[(2'-{N,N-Dimethylamino}-ethoxy)]-2-[phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan der Formel I, welche direkt die Anfordernisse der Pharmacopoeia im Hinblick auf die Reinheit und den Gehalt an restlichen Lösungsmitteln erfüllt, erhalten werden.
- Im gesamten Text sind die Prozentangaben im Hinblick auf den Gehalt der Verbindungen der Formeln I und V und von anderen Verbindungen das Ergebnis der Gaschromatographieanalyse, wobei sie das Verhältnis der Fläche unter dem bestimmten Peak und der Gesamtfläche unter allen Peaks darstellen.
- Der Ausdruck "pharmazeutisch annehmbare Säureadditionssalze", der in der vorliegenden Patentschrift verwendet wird, bedeutet Salze, die mit anorganischen Säuren, z. B. Chlorwasserstoffsäure, Bromwasserstoffsäure, Schwefelsäure oder Phosphorsäure, oder organischen Säuren, z. B. Essigsäure, Weinsäure, Bernsteinsäure, Äpfelsäure, Milchsäure, Zitronensäure, Maleinsäure oder Fumarsäure, gebildet werden. Das Salz, welches mit Fumarsäure gebildet wird, besitzt besonders brauchbare Eigenschaften.
- Das (1R,2S,4R)-(–)-2-[(2'-{N,N-Dimethylamino}-ethoxy)]-2-[phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan der Formel I besitzt drei asymmetrische Zentren, nämlich in den Positionen 1, 2 und 4.
- Somit ist der Gegenstand der Erfindung ein Verfahren zur Herstellung von (1R,2S,4R-(–)-2-[(2'-{N,N-Dimethylamino}-ethoxy)]-2-[phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan der Formel I und pharmazeutisch annehmbaren Säureadditionssalzen davon durch die Umwandlung von (+)-1,7,7-Tri-[methyl]-bicyclo[2.2.1]heptan-2-on {(+)-Campher} der Formel II zu (1R,2S,4R)-(–)-2-[Phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan-2-ol der Formel III durch die Umsetzung des ersteren mit einem Phenylmagnesiumhalogenid oder Phenylalkaliverbindungen, sofern erforderlich, Durchführen einer Hydrolyse des Reaktionsproduktes, und Umsetzen des dadurch erhaltenen (1R,2S,4R)-(–)-2-[Phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan-2-ols der Formel III mit einem (2-{Halogen}-ethyl)-dimethylamin in Gegenwart eines Alkalimetallhydrids oder Alkalimetallamids in einem organischen Lösungsmittel, und, sofern erwünscht, Umwandeln der so erhaltenen Base (1R,2S,4R)-(–)-2-[2'-{N,N-Dimethylamino}-ethoxy)]-2-[phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan der Formel I zu einem Salz, dadurch gekennzeichnet, dass die Reaktion von (1R,2S,4R)-(–)-2-[Phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan-2-ol der Formel III und (2-{Halogeno}-ethyl)-dimethylamin in einem Medium, welches mindestens 50 Gew.-%, vorzugsweise 75 Gew.-%, von 1 oder mehreren Verbindung(en) mit einem 5- oder 6-gliedrigen gesättigten heterocyclischen Ring mit 2 Sauerstoffringatomen als [ein] Lösungsmittel enthält, durchgeführt wird.
- Das wesentliche Merkmal des Verfahrens der vorliegenden Erfindung ist das, dass die Alkylierung in einem Lösungsmittel durchgeführt wird, welches die Alkylierungsreaktion in der Position 3 vom (+)-Campher der Formel II in Gegenwart eines basischen salzbildenden Mittels nicht begünstigt. Es wurde herausgefunden, dass Verbindungen mit einem 5- oder 6-gliedrigen gesättigten heterocyclischen Ring mit 2 Sauerstoffringatomen, insbesondere Dioxan und Dioxolan, in vorteilhafter Weise für diesen Zweck verwendet werden können.
- Vorzugsweise wird Phenylmagnesiumbromid verwendet. Phenylmagnesiumchlorid kann ebenfalls verwendet werden.
- Geeigneterweise kann das Verfahren gemäß der Erfindung wie folgt ausgeführt werden:
Im ersten Schritt des Verfahrens der vorliegenden Erfindung wird (+)-Campher der Formel II einer Grignard-Reaktion mit zum Beispiel Phenylmagnesiumbromid unterzogen. Die Reaktion wird in einer per se bekannten Weise durchgeführt. Als Reaktionsmedium kann vorzugsweise Tetrahydrofuran verwendet werden. Phenylmagnesiumbromid kann in einer Menge von 1 bis 3 Mol, vorzugsweise etwa 1,5 Mol, in bezug auf 1 Mol (+)-Campher der Formel II verwendet werden. Man kann vorzugsweise so vorgehen, dass zuerst das Grignard-Reagens aus Magnesium und Brombenzol in dem verwendeten Lösungsmittel hergestellt wird und danach die Lösung des (+)-Camphers der Formel II in einem organischen Lösungsmittel am Siedepunkt der Reaktionsmischung hinzugesetzt wird. Es ist bevorzugt, das gleiche Lösungsmittel zur Herstellung des Grignard-Reagenses und zur Auflösung des (+)-Camphers der Formel II zu verwenden. Als ein Lösungsmittel kann zweckmäßigerweise Tetrahydrofuran verwendet werden. Vorteilhafterweise wird die Reaktion am Siedepunkt der Reaktionsmischung durchgeführt. - Die Reaktionsmischung wird dann gekühlt und das erhaltene Addukt wird hydrolysiert. Die Hydrolyse kann in einer bekannten Weise, vorzugsweise in saurem Medium, durchgeführt werden. Es ist bevorzugt, Chlorwasserstoffsäure für diesen Zweck zu verwenden.
- Das (1R,2S,4R)-(–)-2-[Phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan-2-ol der Formel III, welches nach der Zersetzung des Grignard-Komplexes erhalten wird, kann einer Alkylierung ohne Reinigung der Reaktionsmischung, die dasselbe enthält, unterzogen werden. Die Reaktion kann in Gegenwart von nicht umgesetztem (+)-Campher der Formel II durchgeführt werden. Gleichwohl führt dies nur zur Bildung einer kleineren Menge von alkylierten Nebenprodukten, da gemäß dem Verfahren der vorliegenden Erfindung die Bildung von (1R,3S,4R)-3-[(2'-{N,N-Dimethylamino}-ethyl)]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan-2-on der Formel V unterdrückt wird.
- Wie oben bereits erwähnt, wird die Alkylierung in einem Lösungsmittel durchgeführt, welches die Alkylierungsreaktion in der Position 3 von (+)-Campher der Formel II nicht begünstigt, d. h. in der (+)-Campher der Formel II in der Position 3 höchstens nur zu einem sehr geringen Ausmaß alkyliert wird. Es ist besonders bevorzugt, Dioxan und Dioxolan als organische Lösungsmittel zu verwenden, da in einem Medium, welches Dioxan und Dioxolan enthält, die Alkylierung vom (+)-Campher der Formel II höchstens nur zu einem sehr geringen Ausmaß stattfindet und folglich die Menge des unerwünschten (1R,3S,4R)-3-[(2'-{N,N-Dimethylamino}-ethyl)]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan-2-on der Formel V in dem Endprodukt (1R,2S,4R)-(–)-2-[(2'-{N,N-Dimethylamino}-ethoxy)]-2-[phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan der Formel I niedrig ist.
- Die asymmetrischen Zentren von (1R,2S,4R)-(–)-2-[(2'-{N,N-Dimethylamino}-ethoxy)]-2-[phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan der Formel I in den Positionen 1 und 4 sind von dem (+)-1,7,7-Tri-[methyl]-bicyclo[2.2.1]heptan-2-on {(+)-Campher} der Formel II abgeleitet.
- Die Alkylierung wird in Gegenwart eines Alkalimetallhydrids, z. B. Natriumhydrids, oder Alkalimetallamids, z. B. Natriumamid, durchgeführt. Diese wandeln die Hydroxidgruppe in ein Salz um. Es ist bevorzugt, Natriumamid zu verwenden.
- Vorzugsweise wird als ein (2-{Halogen}-ethyl)-dimethylamin (2-{Chlor}-ethyl)-dimethylamin verwendet.
- Geeigneterweise wird das Alkalimetallhydrid oder Alkalimetallamid in einer Menge von 1 bis 3 Mol, vorzugsweise 1,5 bis 2 Mol, in bezug auf 1 Mol von dem (1R,2S,4R)-(–)-2-[Phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan-2-ol der Formel III verwendet. Die Menge des Alkylierungsmittels liegt vorteilhafterweise bei 1,0 bis 2,5 Mol, vorzugsweise bei 1 bis 1,1 Mol, in bezug auf das Alkalimetallhydrid oder Alkalimetallamid. Vorzugsweise wird die Alkylierungsreaktion von (1R,2S,4R)-(–)-2-[Phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan-2-ol der Formel III mit dem (2-{Halogen}-ethyl)-dimethylamin unter Erhitzen, insbesondere am Siedepunkt der Reaktionsmischung, durchgeführt. Geeigneterweise findet die Reaktion innerhalb von etwa 3 bis 5 Stunden statt. Eine vorteilhafte Reaktionszeit liegt bei etwa 4 Stunden.
- Das (1R,2S,4R)-(–)-2-[(2'-{N,N-Dimethylamino}-ethoxy)]-2-[phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan der Formel I kann zu einem pharmazeutisch annehmbaren Salz, vorzugsweise dem Fumarat, gegebenenfalls ohne Isolierung des ersteren, umgewandelt werden. Man kann vorzugsweise wie folgt vorgehen: Aus der Reaktionsmischung, die nach der Alkylierung erhalten wird, werden die anorganischen Salze mittels Filtration bei 0 bis 30°C, vorzugsweise bei 20°C, entfernt, woraufhin die entsprechende pharmazeutisch annehmbare Säure, vorzugsweise Fumarsäure, dem Filtrat in einer in etwa equimolaren Menge (1,0 bis 1,5 Mol) hinzugesetzt wird. Das kristalline Produkt, welches aus dem Medium, wie dem Dioxan-Medium, präzipitiert, wird abfiltriert.
- Da das (1R,2S,4R)-(–)-2-[(2'-{N,N-Dimethylamino}-ethoxy)]-2-[phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan der Formel I oder pharmazeutisch annehmbare Säureadditionssalze davon, insbesondere das (1R,2S,4R)-(–)-2-[(2'-{N,N-Dimethylamino}-ethoxy)]-2-[phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan-Fumarat (1:1), welches durch das Verfahren gemäß der Erfindung erhalten wird, höchstens niedrige Mengen an (1R,3S,4R)-3-[(2'-{N,N- Dimethylamino}-ethyl)]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan-2-on der Formel V bzw. von dem pharmazeutisch annehmbaren Säureadditionssalzen davon enthalten [enthält], können sie [kann es] durch das Verfahren gemäß der Erfindung ohne Umkristallisation aus Dimethylformamid aufgearbeitet werden, was ohnehin nur zu einer unzureichenden Reinigung mittels angewandter bekannter Verfahren führt, und folglich ohne Entfernung von Spuren an Dimethylformamid von dem pharmazeutisch aktiven Wirkstoff mittels Verfahren, die ohnehin für den vorgegebenen Zweck ungeeignet sind. Ebenfalls ist dies letztere ein signifikanter Fortschritt im Hinblick auf die Unmöglichkeit, Dimethylformamid bis zum notwendigen Ausmaß zu entfernen, und zwar aufgrund seiner hohen Siedetemperatur, bei der (1R,2S,4R)-(–)-2-[(2'-{N,N-Dimethylamino}-ethoxy)]-2-[phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan der Formel I sich zersetzen würde.
- Der Vorteil des Verfahrens der vorliegenden Erfindung ist der, dass es mit ausgezeichneten Ausbeuten durchgeführt werden kann und ein hochreines Produkt erhalten wird. Somit ist die Ausbeute von etwa 46%, die in den Beispielen gezeigt wird, beträchtlich höher als die Ausbeuten, welche im Stand der Technik offenbart sind, welche 25% nicht überschreiten, selbst wenn (+)-Campher mehrere Male rückzirkuliert wird.
- Die Erfindung wird weiter durch die folgenden Beispiele veranschaulicht. Die in den Beispielen angegebenen Schmelzpunkte sind unkorrigierte Werte.
- Beispiel 1
- (1R,2S,4R)-(–)-2-[(2'-N,N-Dimethylamino}-ethoxy)]-2-[phenyl]-1,7,7-tri-{methyl]-bicyclo[2.2.1]heptan-Fumarat (1:1) [Formel I]
- Grignard-Reaktion
- Zu einer Suspension von 48,6 g (1,5 g-Atom) Magnesiumspänen und 600 ml an wasserfreiem Tetrahydrofuran wurde eine Portion von 20 ml einer Mischung von 236 g (1,5 Mol) Brombenzol und 200 ml wasserfreiem Tetrahydrofuran am Siedepunkt hinzugesetzt. Sobald die Grignard-Reaktion begonnen hat, wird der restliche Teil der Brombenzolmischung zu der Suspension tropfenweise innerhalb einer Stunde hinzugesetzt. Die Reaktionsmischung wird bis zum Sieden erhitzt, bis sich das Magnesium vollständig gelöst hat. Zu der Grignard-Verbindung wird eine Lösung von 152,2 g (1,0 Mol) (+)-Campher der Formel II und 300 ml wasserfreies Tetrahydrofuran unter konstantem Erhitzen bis zum Sieden innerhalb von etwa einer halben Stunde hinzugesetzt, und die Reaktionsmischung wird bis zum Sieden für eine weitere Zeitdauer von 5 Stunden erhitzt.
- Hydrolyse
- Die Reaktionsmischung wird auf 25°C gekühlt und auf einer Mischung von 500 ml Heptan, 400 g Eis, 30 g Natriumchlorid und 150 ml konzentrierter Chlorwasserstoffsäure unter Rühren bei 0°C gegossen. Die organische Phase wird abgetrennt und auf einen pH-Wert von 10 durch Zugabe einer 25%igen w/v wässrigen Ammoniumhydroxidlösung alkalisch gemacht. Nach wiederholter Abtrennung wird die Lösung getrocknet und im Vakuum abgedampft. Dadurch werden 220 g eines farblosen Öls erhalten.
- Analyse auf GC-Basis
-
- – Der Test wird auf einem Perkin Elmer-Autosystem-Gaschromatographen durchgeführt.
- – Länge: 10 m (0,25 mm).
- – Es wird eine 14% Cyanopropyl – 14% Methylpolysiloxan – Festphasen (CPSil-19CB, Chrompack[Handelsprodukt]-Kapillarsäule verwendet.
- – Die Injektion wird bei 200°C durchgeführt.
- – Heizgeschwindigkeit: 10°C/Minute.
- – Trägergas: Helium.
- – Detektor: FID, Injektionstemperatur: 200°C, Endtemperatur: 250°C, Gasdruck: 40 kPa. Gehalt an (1R,2S,4R)-(–)-2-[Phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan-2-ol: 66,5% (+)-Campher-Gehalt: 25%.
- Veretherung
- Zu einer Suspension von 45,5 g (1,05 Mol) Natriumamid (Gehalt: 90% w/w) und 500 ml wasserfreiem Dioxan wird eine Mischung von 220 g des farblosen Öls, welches durch die Hydrolyse erhalten wurde, enthaltend (1R,2S,4R)-(–)-2-[Phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan-2-ol, und 100 ml an wasserfreiem Dioxan am Siedepunkt innerhalb von einer halben Stunde hinzugesetzt. Die Mischung wird zum Sieden 2 Stunden lang erhitzt, woraufhin 113,0 g (1,05 Mol) (2-{Chlor}-ethyl)-dimethylamin hinzugesetzt werden und die Reaktionsmischung bis zum Sieden für eine Zeitdauer von 4 Stunden erhitzt wird.
- Bildung des Fumaratsalzes
- Die Suspension wird auf 20°C gekühlt, filtriert, zu dem klaren Filtrat werden 121,9 g (1,05 Mol) Fumarsäure unter kräftigem Rühren hinzugesetzt. Die Reaktionsmischung wird bis zum Sieden 10 Minuten lang erhitzt, auf 15°C gekühlt, für eine weitere Zeitdauer von einer Stunde gerührt und filtriert. Der Filterkuchen wird mit Dioxan, Wasser und Ethanol gewaschen und bei 80°C so lange getrocknet, bis er frei von Lösungsmittel war. Auf diese Weise werden 190,5 g (0,456 Mol) an weißen Kristallen erhalten, Ausbeute: 45,6% [basierend auf (+)-Campher]. Der Schmelzpunkt der weißen Kristalle beläuft sich auf 214 bis 216°C.
Analyse für die Formel C20H31NO.C4H4O4 (417,55)
berechnet: C% = 69,03%; H% = 8,45%; N% = 3,35%;
gefunden: C% = 69,06%, H% = 8,42%; N% = 3,39%.
[α] 20 / D = –92,5° (c = 0,4, Dimethylsulfoxid, 435 nm). - Das Produkt enthält weniger als 0,05% an (1R,3S,4R)-3-[(2'-{N,N-Dimethylamino}-ethyl)]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan-2-on-Fumarat (1:1)-Kontamination.
- Beispiel 2 (Vergleichsbeispiel)
- (1R,2S,4R)-(–)-2-[(2'-{N,N-Dimethylamino}-ethoxy)]-2-[phenyl]-1,7,7-tri-{methyl]-bicyclo[2.2.1]heptan-Fumarat (1:1) [Formel I]
- Reproduktion des im
Ungarischen Patent Nr. 212 574 - Grignard-Reaktion
- Zu einer Grignard-Verbindung, die aus 5,52 g (0,23 g-Atom) an Magnesiumspänen und 36,1 g (0,23 Mol) Brombenzol in 200 ml wasserfreiem Diethylether hergestellt worden war, wurde eine Lösung von 30,4 g (0,20 Mol) (+)-Campher und 50 ml wasserfreiem Diethylether gegeben. Die Reaktionsmischung wird bis zum Sieden 5 Stunden lang erhitzt. Der Grignard-Komplex wird durch Zugaber einer eiskalten wässrigen Lösung von 20 g Ammoniumchlorid zersetzt, die Mischung wird mit jeweils 30 ml Wasser dreimal gewaschen, abgetrennt, über wasserfreiem Magnesiumsulfat getrocknet, und das Lösungsmittel wird durch Abdampfung entfernt. Auf diese Weise werden 40,5 g eines farblosen Öls erhalten, welches gemäß GC folgendes enthält:
57,5% (+)-Campher der Formel II 5,8% 1,7,7-Tri-[methyl]-bicyclo[2.2.1]heptan-2-ol [Borneol]; 34,5% (1R,2S,4R)-(–)-2-[Phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan-2-ol der Formel III; und 2,2% an weiteren Kontaminationen in geringem Mengen. - Veretherung
- Zu einer Suspension von 3,4 g (67 mMol) an Natriumhydrid (47,5%ige w/w Dispersion) und 50 ml wasserfreiem Toluol wird eine Lösung von 40,0 g des Öls, das in der Grignard-Reaktion erhalten wurde, enthaltend (1R,2S,4R)-(–)-2-[Phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan-2-ol der Formel III, und 30 ml wasserfreies Toluol hinzugesetzt. Die Reaktionsmischung wird eine Stunde lang bis zum Sieden erhitzt, woraufhin eine Lösung von 6,85 g (67 mMol) an (2-{Chlor}-ethyl-)-dimethylamin und 10 ml Toluol am Siedepunkt hinzugesetzt werden. Die Reaktionsmischung wird bis zum Sieden für eine weitere Zeitdauer von 4 Stunden erhitzt.
- Abtrennung
- Die Reaktionsmischung wird dreimal mit jeweils 25 ml Wasser gewaschen. Das Produkt wird mit drei gleichen Portionen einer Lösung von 18 g (0,12 Mol) Weinsäure und 40 ml Wasser extrahiert. Die Phasen werden abgetrennt, die wässrigen Schichten werden vereinigt, auf einen pH-Wert von 10 mit einer konzentrierten Ammoniumhydroxidlösung alkalisch gemacht, dreimal mit jeweils 20 ml Dichlorethan extrahiert, über Magnesiumsulfat getrocknet, und das Lösungsmittel wird im Vakuum entfernt. Auf diese Weise werden 14,5 g eines farblosen Öls erhalten, welches gemäß GC-Analyse folgendes enthält:
74,2% von (1R,2S,4R)-(–)-2-[(2'-{N,N-Dimethylamino}-ethoxy)]-2-[phenyl]- 1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan der Formel I; 16,5% von (1R,4R)-2-[(2'-{N,N-Dimethylamino}-ethoxy)]-1,7,7-tri-[methyl]- bicyclo[2.2.1]heptan der Formel IV 6,5% von (1R,3S,4R)-3-[(2'-{N,N-Dimethylamino}-ethyl)]-1,7,7-tri-[methyl]- bicyclo[2.2.1]heptan-2-on der Formel V; und - Bildung des Fumaratsalzes
- Zu einer Lösung von 14,0 g der Base (1R,2S,4R)-(–)-2-[(2'-{N,N-Dimethylamino}-ethoxy)]-2-[phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan der Formel I, welche von dem Tartratsalz freigesetzt wurde, und 150 ml Ethanol werden 5,07 g (43,6 mMol) an Fumarsäure bei 70°C hinzugegeben. Das Produkt wird bei 0°C filtriert und aus 50 ml Dimethylformamid umkristallisiert.
- Auf diese Weise werden 13,5 g des gewünschten Produktes (1R,2S,4R)-(–)-2-[(2'-{N,N-Dimethylamino)-ethoxy)]-2-[phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan-Fumarat (1:1) der Formel I in Form von weißen Kristallen erhalten. Ausbeute: 16,2% [basierende auf (+)-Campher]. Gemäß der GC-Analyse können im Produkt 0,5% an (1R,3S,4R)-3-[(2'-{N,N-Dimethylamino}-ethyl)]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan-2-on nachgewiesen werden. Schmp.: 214 bis 216°C.
Analyse für die Formel C20H31NO.C4H4O4 (417,55)

gefunden: C% = 69,16%, H% = 8,52%; N% = 3,32%.
Claims (10)
- Verfahren zur Herstellung von (1R,2S,4R-(–)-2-[(2'-{N,N-Dimethylamino}-ethoxy)]-2-[phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan der Formel und pharmazeutisch annehmbaren Säureadditionssalzen davon durch die Umwandlung von (+)-1,7,7-Tri-[methyl]-bicyclo[2.2.1]heptan-2-on {(+)-Campher} der Formel II zu (1R,2S,4R)-(–)-2-[Phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan-2-ol der Formel III durch die Umsetzung des ersteren mit einem Phenylmagnesiumhalogenid oder Phenylalkaliverbindungen, sofern erforderlich, Durchführen einer Hydrolyse des Reaktionsproduktes, und Umsetzen des dadurch erhaltenen (1R,2S,4R)-(–)-2-[Phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan-2-ols der Formel III mit einem (2-{Halogeno}-ethyl)-dimethylamin in Gegenwart eines Alkalimetallhydrids oder Alkalimetallamids in einem organischen Lösungsmittel, und, sofern erwünscht, Umwandeln der so erhaltenen Base (1R,2S,4R)-(–)-2-[2'-{N,N-Dimethylamino}-ethoxy)]-2-[phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan der Formel I zu einem Salz, dadurch gekennzeichnet, dass die Reaktion von (1R,2S,4R)-(–)-2-[Phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan-2-ol der Formel III und (2-{Halogeno}-ethyl)-dimethylamin in einem Medium, welches mindestens 50 Gew.-% von einer oder mehreren Verbindung(en) mit einem 5- oder 6-gliedrigen gesättigten heterocyclischen Ring mit 2 Sauerstoffringatomen als [ein] Lösungsmittel enthält, durchgeführt wird.
- Verfahren gemäß Anspruch 1, dadurch gekennzeichnet, dass als ein Lösungsmittel Dioxolan verwendet wird.
- Verfahren gemäß Anspruch 1 oder 2, dadurch gekennzeichnet, dass das (1R,2S,4R)-(–)-2-[Phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan-2-ol der Formel III, welches nach der Zersetzung des Grignard-Komplexes erhalten wird, einer Alkylierung ohne Reinigung der Reaktionsmischung, die dieses enthält, unterzogen wird.
- Verfahren gemäß Anspruch 1 oder 2, dadurch gekennzeichnet, dass Phenylmagnesiumbromid verwendet wird.
- Verfahren gemäß mindestens einem der Ansprüche 1 oder 2, dadurch gekennzeichnet, dass Phenylmagnesiumchlorid verwendet wird.
- Verfahren gemäß mindestens einem der Ansprüche 1 bis 5, dadurch gekennzeichnet, dass ein 2-(2'-{Halogen}-ethyl)-dimthylamin-2-(2'-{chlor}-ethyl)-dimethylamin verwendet wird.
- Verfahren gemäß mindestens einem der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass ein Alkalimetallamidonatriumamid verwendet wird.
- Verfahren gemäß mindestens einem der Ansprüche 1 bis 7, gekennzeichnet durch die Durchführung der Alkylierungsreaktion von (1R,2S,4R)-(–)-2-[Phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan-2-ol der Formel III mit dem 2-(2'-{Halogen}-ethyl)-dimethylamin unter Erhitzen, insbesondere am Siedepunkt der Reaktionsmischung.
- Verfahren gemäß mindestens einem der Ansprüche 1 bis 8, dadurch gekennzeichnet, dass durch die Umwandlung der Base (1R,2S,4R)-(–)-2-[(2'-{N,N-Dimethylamino}-ethoxy)]-2-[phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan der Formel I zu einem Salz das 2-(E)-Butendioat (1:1)-Salz (Fumarat) hergestellt wird.
- Verfahren gemäß mindestens einem der Ansprüche 1 bis 9, dadurch gekennzeichnet, dass die Umwandlung des (1R,2S,4R)-(–)-2-[(2'-{N,N-Dimethylamino}-ethoxy)]-2-[phenyl]-1,7,7-tri-[methyl]-bicyclo[2.2.1]heptan der Formel I zu dem 2-(E)-Butandioat (1:1)-Salz (Fumarat) ohne Isolierung des ersteren durchgeführt wird.
