Claims (8)
- 流体試料から所望の分析物を分離すると共に、上記分析物を或る量の溶離流体に濃縮するための方法において、
a)上記試料をカートリッジに導入するステップを備え、
上記カートリッジは
i)試料孔と、
ii)上記試料孔から伸びる試料流路と、
iii)上記試料流路内に抽出チャンバとを有し、上記抽出チャンバは上記試料から上記所望の分析物を捕獲するために少なくとも1つの固体支持体を含有し、
iv)上記分析物を上記抽出チャンバから溶離させるために溶離流路を有し、上記溶離流路は上記チャンバを通過し、上記チャンバを通過した後に上記試料流路から分岐し、
b)上記試料流路を通って上記試料を強制的に流すステップを備え、これによって上記試料が上記チャンバを通って流れるときに上記固体支持体で上記分析物を捕獲し、上記チャンバを通って強制的に流される試料の容積と上記チャンバの収容容積との比は少なくとも2:1であり、上記チャンバを通って強制的に流される試料の容積は少なくとも0.1ミリリットルであり、
c)溶離流路を通って溶離流体を強制的に流すステップを備え、これによって上記捕獲された分析物を上記固体支持体から上記溶離流体の中に放出することを特徴とする方法。 - 請求項1に記載の方法において、
抽出チャンバを通って上記溶離流体を流す間に上記固体支持体を加熱するステップを更に備えていることを特徴とする方法。 - 請求項2に記載の方法において、
固体支持体は60〜95℃の温度範囲に加熱されることを特徴とする方法。 - 請求項1に記載の方法において、
上記抽出チャンバを通って上記試料を強制的に流すステップの後で、かつ、上記抽出チャンバを通って上記溶離流体を強制的に流すステップの前に、上記抽出チャンバを通ってガスを強制的に流すステップを更に備えていることを特徴とする方法。 - 請求項1に記載の方法において、
上記抽出チャンバを通って流す試料の容積と上記チャンバの収容容積の比は、少なくとも10:1であることを特徴とする方法。 - 請求項1に記載の方法において、
上記チャンバを通って流す試料の容積は、少なくとも1ミリリットルであることを特徴とする方法。 - 請求項1に記載の方法において、
上記カートリッジは試料流路の中に溶解チャンバを更に含み、上記溶解チャンバは上記試料中の細胞または胞子または微生物を捕獲するためにフィルタを含有し、上記方法は、
a)上記試料が上記溶解チャンバを通って流れるときに上記細胞または胞子または微生物を捕獲するステップと、
b)上記溶解チャンバの壁に超音波変換器を連結するステップと、
c)上記捕獲された細胞または胞子または微生物を溶解するために、超音波エネルギーを変換器から上記溶解チャンバに伝達するステップとを
備えていることを特徴とする方法。 - 請求項7に記載の方法において、
溶解チャンバ内のビーズを攪拌させるステップを更に備えていることを特徴とする方法。
