Den foreliggende opfindelse angår en farmaceutisk eller veterinært acceptabel, amphipatisk, ikke-tværbundet, lineær, forgrenet eller podeblokcopolymer og fremgangsmåde til fremstilling af denne, matrix til brug i et lægemiddel til frigivelse 5 af syrestabile polypeptider, som omfatter en sådan copolymer, samt et farmaceutisk middel til parenteral administration, som omfatter et farmakologisk nyttigt polypeptid og en sådan copolymer. De farmaceutiske midler omfattende farmakologisk aktive polypeptider giver kontinuerlig frigivelse af polypeptiderne 10 over et forlænget tidsrum, når midlerne anbringes i et miljø af vandig fysiologisk type.
Man har længe været klar over, at kontinuerlig eller løbende frigørelse af visse lægemidler over et forlænget tidsrum efter 15 en enkelt indgivelse kan frembyde væsentlige praktiske fordele ved klinisk brug, og der er allerede blevet udviklet produkter, som muliggør forlænget frigørelse af en række klinisk nyttige lægemidler efter oral dosering (se f.eks. Remington's Pharmaceutical Sciences, publiseret af Mack Publishing Compa-20 ny, Easton, Pennsylvania, U.S.A., 15. udgave, 1975, side 1618-1631) efter parenteral administration (ibidem, side 1631 - 1643) og efter topisk administration) (se f.eks. UK patentskrift nr. 1.351.409). En egnet fremgangsmåde til parenteral administration er den subdermale injektion eller implantering 25 af et fast legeme, f.eks. en pille eller en film, indeholdende lægemidlet, og der er blevet beskrevet en række sådanne im-planterbare emner. Det er specielt kendt, at der for mange lægemidlers vedkommende kan opnås velegnede implanterbare emner til forlænget lægemiddelfrigørelse ved at indkapsle lægemidlet 20 i en biologisk nedbrydelig polymer eller ved at dispergere lægemidlet i en grundmasse, såsom en polymer, således at lægemidlet frigøres, efterhånden som nedbrydningen af polymer-grundmassen skrider frem.
35 Egnede biologisk nedbrydelige polymere til anvendelse i sådanne præparater med forlænget frigørelse er velkendte og omfatter polyestere, som nedbrydes lidt efter lidt ved hydrolyse, når de anbringes i et miljø af vandig fysiologisk type. som er afledt af hydroxycarboxylsyrer, og der findes en omfattende kendt teknik rettet mod polymere afledt af ct-hydroxy= carboxylsyrer, specielt mælkesyrer, i både racemiske og op-• tisk aktiye former, og glydolsyre og copolymere deraf ^ se 5 f,eks. US patentskrifterne nr. 3.773.919 og 3.887.699, Jackanicz et al, Contraception, 1973, 8, 227-234; Anderson et al., 'ibidem, 1976, 11, 375-384; Wise et al, Life Sciences, 1976, 19, 867-874; Woodland et al., Journal of Medicinal Chemistry, 1973, 16, 897-901; Yolles et al., Bulletin of the Parenteral Drug Association, 1976, 30, 306^-312; Wise et al., Journal ’of Pharmacy and Pharmacology, 1978, 30, 686-689 og 1979, 31, 1201-204.
Britisk patentskrift nr. 1.325.209 (svarende til US patent-'skrift nr. 3.773.919) og US patentskrift nr. 3.887.669 refe-'rerer til forlænget eller langvarig frigørelse af polypepti-der. Sidstnævnte nævner kun insulin, men indeholder intet sær-‘ ligt eksempel på en sådan formulering, og henvisningen til polypeptider er tilsyneladende helt spekulativ, da det frem- 20 går, at der kun er en omfattende opregning af mange forskellige lægemiddelklasser, som man hævder kan inkorporeres i formuleringer af den heri beskrevne art. I virkeligheden er i alt væsentligt alle de andre lægemiddeltyper, som der henvises til i det nævnte skrift, bortset fra polypeptider, af re- 2 5 lativ hydrofob karakter og af relativ lav molekylvægt, og den nævnte tekst erkender på ingen måde de vanskeligheder, som vi har konstateret i forbindelse med forsøget på opnåelse af tilfredsstillende formuleringer af polypeptider med langvarig frigørelse, hvorhos mange af polypeptiderne er forholds- 3 0 vis hydrofile og forholdsvis højmolekylære.
Det må forstås, at "langvarig" eller "forlænget" frigørelse af et lægemiddel kan være enten løbende eller diskontinuert.
Det har nu vist sig, at i mange tilfælde, når man anvender 3 fi den kendte teknik, og specielt hvad der kendes fra UK patentskrift nr. 1.325.209, til fremstillingen af en formulering af et polypeptid, kan frigørelsen af polypeptidet fra formule- 3

ringen, selvom den optræder over et forlænget tidsrum, også være diskontinuert. Frigørelsen af et polypeptid fra en polylactidpolymer,som beskrevet i nævnte tekst, sker ofte ef-5 ter en væsentlig induktionsperiode, hvor der ikke frigøres polypeptid,eller er flerfaset og omfatter en begyndelsesperiode, hvori der frigøres noget polypeptid, en anden periode, hvor der kun frigøres lidt eller slet intet polypeptid, og en tredje periode, hvori størstedelen af det resterende poly= peptid frigøres. I modsætning hertil er det et formål med den foreliggende opfindelse at tilvejebringe præparater af polypeptider, hvorfra der, bortset fra muligvis en forholdsvis kort induktionsperiode i begyndelsen, sker løbende frigørelse af polypeptidet og ikke eksisterer perioder, hvor der 15 kun frigøres lidt eller slet intet polypeptid. Ordene "løbende frigørelse" anvendes i den foreliggende tekst udelukkende til at beskrive et frigørelsesprofil, som er i alt væsentligt enfaset, selvom det kan have et vendepunkt, men helt klart ikke har nogen "plateau"-fase.
20 UK patentskrift nr. 1.388.580 beskriver insulinholdige formuleringer med langvarig frigørelse, som er baseret på hydroge-1 er dannet ved at omsætte en vandopløselig polymer med et che-1 ateringsmiddel, derpå tværbinde polymer-chelateringsmiddelkæ-25 derne ved reaktion med en polyvalent metalion i vandig opløsning til dannelse af en hydrogel. Insulin blev inkorporeret i en præformet hydrogel i vandig opløsning, og det hele blev homogeniseret og indsprøjtet subkutant eller intramuskulært.
30 US-patentskrift nr. 4.202.880 omhandler amphipatiske, ikke-tværbundne blokcopolymerer med en middelmolekylvægt på mere end 5.000 til anvendelse i lægemidler med forlænget frigivelse. Midler, hvori der anvendes sådanne polymerer, som er omhandlet i det nævnte US-patentskri ft, er imidlertid beskre-35 vet som værende "i alt væsentligt uopløselige og ikke-nedbry-delige i legemsvæsker" (spalte 2, linie 46-47) og er beregnet til at blive fjernet fra legemet, når lægemiddel indholdet er blevet udtømt.

I modsætning hertil inkorporeres i midlerne ifølge den foreliggende opfindelse en amphipatisk copolymer omfattende en hydrofob komponent, som er bionedbrydelig, og en hydrofil komponent, som både kan være bionedbrydelig og ikke-bionedbrydelig.
5 Når et sådant middel implanteres i en dyrekrop, nedbrydes copo-lymerens hydrofobe andel eventuelt til lavmolekylære lipidop-løselige materialer, som transporteres fra implanteringsstedet og derefter elimineres fra legemet på normal måde. Copolyme-rens hydrofile komponent transporteres også, uanset om den er 10 bionedbrydelig eller ej, væk fra implanteringsstedet, idet den er vandopløselig, og elimineres fra legemet gennem nyrerne på samme måde som andre vandopløselige materialer. Når således midlet ifølge opfindelsen først er blevet implanteret i et dyr, frigiver det dets lægemiddel indhold og forsvinder eventu-15 elt fuldstændigt uden noget behov for fjernelse af det udtømte middel fra legemet. I modsætning hertil bibeholder de ikke-bionedbrydel ige midler ifølge ovennævnte US-patentskrift deres integritet og forbliver på ubestemt tid på det sted i legemet, hvor de er anbragt, medmindre de positivt fysisk fjernes.

Det er et formål med den foreliggende opfindelse at tilvejebringe en implanterbar eller injicerbar farmaceutisk eller veterinær formulering for farmakologisk nyttige polypepti der, som har fast form, og som absorberer vand fra det animalske 25 legeme efter implantering, til dannelse af en hydrogel, hvorfra polypeptidet frigøres løbende over et længere tidsrum.
Ifølge den foreliggende opfindelse tilvejebringes således en farmaceutisk eller veterinært acceptabel amphipatisk ikke-30 tværbundet lineær, forgrenet eller podeblokcopolymer, som er ejendommelig ved, at den har en minimal gennemsnitsmolekylvægt på 5.000, at den hydrofobe komponent er biologisk nedbrydelig eller hydrolytisk ustabil under normale fysiologiske betingelser, og at den hydrofile komponent kan være biologisk ned-35 brydelig eller ikke-biologisk nedbrydelig, og at copolymeren er i stand til at absorbere vand til dannelse af en hydrogel, når den anbringes i vand eller et miljø af vandig fysiologisk 5

Når den tørre amphipatiske copolymer indeholdende et polypep-tid i den foreliggende opfindelse neddyppes i vand eller anbringes i et vandigt fysiologisk miljø i et animalsk legeme, er vandoptagelsen en funktion af de hydrofile eller vand- 20 reagerbare dele af den copolymere, og materialet kvælder op.

Et sådant ideelt frigørelsesprofil for det aktive materiale 10 kan også opnås ved at blande forskellige copolymere hver med egne definerede egenskaber (f.eks. molekylvægt, molekylvægts-• fordeling, blokstruktur, hydrofiliskhed, nedbrydningsegenskaber, diffusionsegenskaberi,og ved passende kombination af forskellige sådanne materialer kan frigørelseshastigheden 15 og varigheden af frigørelsen af et aktivt materiale varieres efter ønske.
Ved passende valg af de ovennævnte parametre og/eller passende blanding kan der også opnås et copolymermateriale, som til-20 lader forarbejdning til implanterbare emner ved forholdsvis lave temperaturer, helt sikkert under 100°C, og i nogle tilfælde endog ved stuetemperatur, og således velegnet til fabri-i kationen af implanterbare emner, der omfatter varmefølsomme i eller opløsningsmiddelfølsomme polypeptidaktive materialer.
25 1 Blokcopolymere af polyethylenglycol og amorfe hydrofobe poly- i mere med en glasovergangstemperatur over 37°C er f.eks. særligt nyttige på grund af, at polyethylenglycolblokken blødgør den hydrofobe blok og giver et materiale, som let forarbejdes ved en forholdsvis lav temperatur, ja endog stue-30 temperatur, medens polyethylenglycolblokkene med efterfølgende henstand krystalliserer til dannelse af et sejt, hårdt produkt, som let kan håndteres.

Ifølge endnu en udførelsesform for opfindelsen tilvejebringes således et farmaceutisk eller veterinært produkt, der omfatter en ikke-peptidholdig farmakologisk aktiv forbindelse og en blokcopolymer som defineret ovenfor, og anvendelsen af sådanne blokcopolymere til den løbende frigørelse af en sådan farmakologisk aktiv forbindelse uden peptid.
Ifølge endnu en udførelsesform for den foreliggende opfindel-15 sen tilvejebringes en fremgangsmåde til fremstillingen af en farmaceutisk eller veterinært acceptabel amphipatisk lineær, forgrenet eller podet blokcopolymer som defineret ovenfor, hvilken fremgangsmåde omfatter copolymerisering af mo-2Q norner A og monomer B ved hjælp af sædvanlige fremgangsmåder, såsom podecopolymerisation, polykondensation og polyaddition, eventuelt med en passende katalysator, f.eks. zinkoxid, zink-carbonat, basisk zinkcarbonat, diethylzink, organotinforbindel-ser, f.eks. stannooctoat, stanno-2-ethylhexanoat), tributyl= aluminium, titanium-, magnesium- eller bariumforbindelser eller blyironooxid, og af disse foretrækkes almindeligvis stannooctoat.
Copolymerisationerne gennemføres ellers på sædvanlig måde, således som det er sædvanligt på polymerområdet med hensyn 30 til tid og temperatur.

Eksempel 1
Polyethylenglycol med molekylvægt 20.000 (30 g) blev omrørt og opvarmet under vakuum (< 0,1 mm Hg) ved 120°C i 3 timer. D,L-lactid (15 g) og glycolid (15 g) blev tilsat, og blan-5 dingen blev omrørt under en nitr-ogenatmosfære indtil smeltning af det faste stof. Temperaturen blev hævet til 160°C, og stannooctoat (stanno-2-ethylhexanoat) (0,1 ml) blev tilsat. Blandingen blev holdt ved 160°C i 3 timer, hvorefter den var blevet kraftig viskos, og den blev derefter afkølet og opløst 10 i acetone (200 ml). Denne acetoneopløsning blev langsomt sat til kraftigt omrørt ethanol (1500 ml), og det således bundfald blev filtreret fra og tørret i en vakuumovn i 3 timer ved stuetemperatur og derpå natten over ved 40°C.
Det kernemagnetiske resonansspektrum af denne copolymer i 15 deuteriochloroform viste, at den havde sammensætningen oxy= ethyl en: mælkesyre :glycqls.yre på 2:1:1.
Denne copolymer blev formet ved ca. 60°C til en blød, plastisk transparent film. En prøve (39 mg) kvældede hurtigt op efter placering i vand og optog 135 mg vand i løbet af 4 timer 20 til dannelse af en transparent hydrogel, som derpå blev nedbrudt i løbet i to uger ved 37°C.
Eksempel 2
Den i eksempel 1 beskrevne polymer (20,2 mg) og bovinvæksthormon (BGH) (5,1 mg) blev sammenblandet ved ca. 40°C til 25 dannelse af en uigennemsigtig blanding, som blev formet til 1 mm tyk plade. Denne plade blev neddyppet i en pufferopløsning ved pH-værdi 8,6 (M/15 puffer, pH 8,6, indeholdende 0,01% natriumazid) og frigjorde et materiale med en molekylvægt på ca. 22.000, som havde den samme retentionstid ved høj-30 tryksvæskekromatografi som BGH i løbet af en periode på mindst 12 dage.

Under anvendelse af den i eksempel 2 beskrevne fremgangsmåde blev copolymer/BGH-blandingen omdannet til skiver, der vejede ca. 45 mg og indeholdt ca. 20% BGH. Når sådanne skiver 5 blev implanteret, hver i en hypofysetomiseret rotte, steg dyrenes vægt med gennemsnitligt 25% i løbet af 7 dage, hvorimod vægten af kontroldyrene, som hver fik en placebo-implan-tering, forblev i alt væsentligt uændret.
Eksempel 4 10 Den i eksempel 1 beskrevne copolymer (13,5 mg) og monoklont muserimmunoglobulin A (IgA) med en defineret antigen-specificitet og en molekylvægt på > 180.000 (1,5 mg) blev blandet ved 50°C til dannelse af en homogen blanding af IgA i den copolymere, og protein/copolymer-^blandingen blev formet til 15 en kugle med en diameter på omkring 2 mm. In vitro frigørelsen af IgA blev vurderet ved at neddyppe proteincopolymeren i puffer (phosphatpufret saltvand, pH 7,2) ved 37°C. Ved anvendelse af en enzymbundet immunoprøyeteknik blev det vandige medium undersøgt for aktiyt IgA, og frigivelse af det biolo-20 gisk aktive protein viste sig at starte efter 2 dages forløb og fortsætte i mindst 9 dage.
Eksempel 5
Polyethylenglycol med en molekylvægt på 20.000 (50 g) blev opløst i chloroform (150 mg) og vasket seks gange med destil-25 leret vand (~ 300 ml), idet de vandige vaskevæsker blev ka-seret. Chloroformet blev afdampet under reduceret tryk, og det rensede polyethylenglycol blev tørret ved 160°C/0,05 mm Hg i 1 time.
Stannooctoat (stanno-2-ethylhexanoat) (~ 5 g) bley renset 30 yed opvarmning til 140°C/0,055 mmHg for at fjerne urenheder.
Den rensede polyethylenglycol (14,3 g) blev opvarmet til 160°C

under vakuum (0,05 mmHg) i en 100 ml rundbundet koble i 1 time. Frisk fremstillet rent D,L-lactid (42,9 g) blev tilsat under nitrogen og smeltet ved 160°C. Stannooctoat (0,2 ml) blev tilsat, og blandingen blev omrørt,indtil viskositeten 5 hindrede yderligere omrøring. Efter 3 timers forløb var der opnået et højviskost produkt. Blandingen fik lov til at afkøle, koblen blev åbnet, kolbeindhold blev opløst i acetone (~ 300 ml), og opløsningen blev filtreret. Filtratet blev langsomt sat til ethanol (- 1000 ml) med kraftig omrøring 10 til dannelse af et fibrøst bundfald, som blev opsamlet og tørret ved 30°C i en vakuumovn natten over. Analyse af produktet ved hjælp af kernemagnetisk resonans spek tro skopi viste, at produktet havde en sammensætning af oxyethylen:mælkesyre på 1:3, og strukturviskositeten i chloroform var 1,055.
15 Produktet blev formet til en tynd (0,2 mm) blød, plastisk transparent film. Ved neddypning i vand Øgedes filmens vægt (0,54 g) til 0,95 g på 1 dage ved 37°C. Den hydratiserede transparente film havde en bedre stivhed og styrke end den tørre copolymer i begyndelsen. Efter 35 dages forløb var fil-20 men intakt og bevarede mekaniske egenskaber, hvilket viser, at den copolymere var blevet nedbrudt ganske langsomt, således som det ved hjælp af kernemagnetisk resonansspektroskopi vises ved ændring af sammensætning.
Når bovinvæksthormon inkorporeres i den tørre copolymer ved 25 60°C, frigør den resulterende polypeptid/polymer-blanding et produkt med en molekylvægt på 22.000 i puffer (M/15 phosphatpuffer, pH 8,6) i løbet af mindst 7 dage.
Eksempel 6
Polyethylenglycol med en molekylvægt på 6.000 (50 g) blev 30 renset under anvendelse af den ovenfor i eksempel 5 beskrevne fremgangsmåde.

Den rensede tørre polyethylenglycol (7,5 g) og stannochlorid= dihydrat (15 mg) blev sammenblandet ved stuetemperatur og derpå opvarmet under omrøring til 155°C under kraftigt vakuum (0,1-0,01 mmHg) og holdt ved denne temperatur i 2 timer, 5 medens frisk fremstillet tørt D,L-lactid (22,5 g) blev sat til blandingen under nitrogen og smeltet. Reaktionsblandingen blev holdt ved 155-KL60°C i 3 timer til opnåelse af et yiskost produkt, som blev hældt på poly-tetrafluorethylen-film og fik lov til at afkøle. Det polymere produkt blev opløst i acetone 10 (70 ml) under opvarmning, og den polymere blev isoleret ved at hælde acetoneopløsningen i ethanol (600 ml). Bundfaldet blev tørret i en vakuumovn natten over ved 60°C. Den polymere havde en strukturviskositet i chloroform på 0,41. Efter presning til en tynd film (0,2 mm) og neddypning i vand optager 15 den polymere ca. sin egen vægt af vand ved 37°C i løbet af 24 timer til dannelse af en sej hydrogel.
Eksempel 7 99 mg af blokcopolymer fremstillet på den i eksempel 6 beskrevne måde indeholdende 25 dele polyethylenglycol (molekylvægt 20 6.000) og 75 dele poly(D,L-lactid) blev opløst i 4,5 ml vand fri fri iseddikesyre og 0,5 ml destilleret vand. 200 ·μ1 af en opløsning indeholdende 1,1 mg muse-epiderm vækstfaktor (EGF) blev sat til polymeropløsningen, og blandingen blev homogeniseret. Den homogeniserede opløsning blev frosset og 25 derpå frysetørret i 18 timer, produktet blev formet ved 50°C til opnåelse af et implanterbart emne, der vejer 40 mg (- 8mmx4mmxl mm). Det implanterbare emne blev anbragt i 1 mm humant serum ved 37°C, og frigørelsen af EGF blev målt på serumprøver ved hjælp af radioimmunoprøve. Resultaterne 30 viste en løbende frigørelse af peptid i løbet af mindst 3 dage.

25 g af en copolymer indeholdende ækvimolære mængder af D,L-lactid og glycolid og med en strukturviskositet i chloroform på 0,20 blev opløst i 50 ml tørt ethylacetat, og opløsningen 5 blev opvarmet under tilbagesvaling og omrøring under nitrogen. 0,25 g lauroylperoxid blev opløst i frisk destilleret vinylpyrrolidon (25 ml). Blandingen blev dråbevis sat til den tilbagesvalende polymer i løbet af 2 timer, og blandingen blev opvarmet under tilbagesvaling i yderligere 6 timer. Ved afkø-10 ling gelerede blandingen. Rensning af den amphipatiske blok= podecopolymer ved fjernelse af homopolymer af polyvinylpyrro-lidon under anvendelse af udfældningsteknik var vanskelig, da udfældning ofte resulterede i kolloide suspensioner, og dette indicerede, at der var sket podning af polyvinylpyrrolidon 15. til lactid/glycolidr'copolymeren.
Ethylacetatblandingen blev derfor opvarmet til 70°C, og 50 ml ethanol blev tilsat til opnåelse af en kolloid suspension, hvorfra den polymere blev isoleret ved udfældning i n-hexan (2 liter). Den således opnåede polymer blev tørret ved 90°C 20 natten over under vakuum, hvor ved afkøling opnåedes et skørt produkt bestående af podecopolymer og homopolyvinylpyrrolidon. Produktet havde en strukturviskositet på 0,29 i chloroform, og ca. 50% polyvinylpyrrolidon som homocopolymer og pode-b lokcopo lymer.

Produktet blev formet ved 110°C i 20 sek. til opnåelse af en plade (~ 0,8 cm x 1,2 mm x 2 mm, vægt 30 mg)., som efter 3Q neddypning i vandig pH 7,4 puffer ved 37°C frigjorde pepti-det i løbet af en periode på adskillige dage.

Eksempel 9 21 50 g polyvinylalkohol med en molekylvægt på 14.000 blev opløst i 500 mg kommercielt D,L-mælkesyre (indeholdende ~ 12% vand) med omrøring under nitrogen. Blandingen blev opvarmet til 5 140°C, og vand blev afdestilléret i løbet af 8 timer, i løbet
af hvilket tidsrum blandingen i voksende grad blev mere yiskos, og dets temperatur steg til 190°C. Da der ikke længere destil-lerede vand over, blev trykket sænket til " 25 craHg, og blandingen blev opvarmet i yderligere 8 timer. Til slut blev tryk-10 ket sænket il 0,1 mmHg, og blandingen blev opvarmet til 200°C

Den polymere fik lov til at afkøle, og kolben blev åbnet. Produktet blev opdelt i små stykker og opløst i methanol (1,5 liter), og produktet blev isoleret ved udfældning i 15 10 liter destilleret vand. Bundfaldet blev vasket med yder ligere 5 liter vand og tørret under vakuum ved stuetemperatur i 8 timer og til slut ved 100°C i 16 timer til opnåelse af et ravfarvet glasagtigt produkt, som bestod af et poly= vinylalkoholskelet indeholdende tilhørende polymælkesyre-20 kæder af lav molekylvægt, strukturviskositet = 0,65 i chloroform. Produktet indeholdt ca. 85% polymælkesyre, og tilsvarende polymælkesyre-gennemsnitskædelængde var ca. 3,5.
Den polymere blev formet ved 100°C til opnåelse af en plade på 1 cm x 0,2 cm x 0,2, som blev neddyppet i vand ved 37°C.
25 Produktet absorberede vand og blev bøjeligt og eroderede til dannelse af opløselige produkter i løbet af en periode på 2 måneder.
Eksempel 10
Muse-epiderm vækstfaktor (285 pi) af 21 mg/ml opløsning i 30 destilleret vand blev sat til en opløsning af en 20/80 poly(D,L-lactid)/PEG 6.000 copolymer med strukturviskositet 0,36 i chloroform (45 mg) i 2,5 ml 90% vandig eddikesyre.

Opløsningen af peptid og polymer blev frosset og derpå frysetørret ved ~ 0,01 mmHg i 24 timer til opnåelse af et tørret produkt. Disse blev implanteret subcutant i 2 katte, hver med en gastrisk fistel. Der blev taget blodprøver, og mayesyre-resultat som reaktion på en histaminstimulering blev målt.

Eksempel 11
Muse-epiderm vækstfaktor (120 pi af 10,5 mg peptid i 320 liter 15 destilleret vand blev sat til en opløsning af en 85/15 poly-(D,L-lactid)/PEG 6.000 copolymer med strukturviskositet 0,39 i chloroform (36 mg) i 1,8 ml 90% vandig eddikesyre. Den resulterende opløsning blev frosset og frysetørret natten over. Det frysetørrede materiale blev formet ved 70°C til dannelse 20 af en implanterbar enhed, der vejede 16,9 mg (dimensioner ca.
1 mm x 1 mm x 5 mm) .
Peptidet blev frigjort fra denne implanterbare enhed løbende over mindst 15 dage til 10% humant serum i yand indeholdende 0,1% natriumazid.
25 Eksempel 12
For at vise virkningen af sammensætningen og hydrofiliteten af den blokcopolymere på frigørelse af polypeptid fra implanterbare enheder blev der gennemført følgende sammenligning.
I særskilte forsøg blev der fremstillet implanterbare enheder 30 under anyendelse af 23
DK 165458 B
(a) en blokcopolymer med strukturviskositet 0,39 i chloroform indeholdende 25 vægt/vægt polyethylenglycol med en molekylvægt på 6.000 og 75% vægt/vægt poly(D,L-lactid^ (b) en blokcopolymer med strukturviskositet 0,79 i chloform 5 indeholdende 5% vægt/vægt polyethylenglycol med en mole kylvægt på 6.000 og 95 vægt/vægt poly(D,L-lactid).
76,2 mg polymer og ICI 118.630 (23,8 mg som acetatsaltet svarende til 20 mg rent peptid) blev opløst i anhydridfri iseddikesyre (1,5 ml). Opløsningen blev frosset og fryse-10 tørret i 18 timer, og det frysetørrede produkt blev formet ved ~ 70°C til opnåelse af implanterbare enheder, der vejede ~ 45-50 mg (dimensioner ca. 0,2 cm x 0,2 cm x 1 cm).
De implanterbare enheder blev neddyppet i 1 ml Hcllvains pH 7,4 puffer ved 37°C, og 1 ml prøver af det vandige medium 15 blev udtaget på givne tidspunkter og undersøgt for lægemiddelindhold ved hjælp af højtryksvæskekromatografi. Det fjernede vandige medium blev hver gang erstattet med 1 ml frisk puffer.
Disse frigørelsesforsøg viste, at implanterbare enheder frem-20 stillet under anvendelse af den mere hydrofile polymere indeholdende 25% polyethylenglycol frigjorde forbindelser i ~ 18 dage.
I modsætning hertil frigjorde det implanterbare emne, fremstillet under anvendelse af den mindre hydrofile copolymer, inde-25 holdende 5% polyethylenglycol,forbindelse løbende i mindst 250 dage.

Poly(ethylenglycolmethylether) med en molekylvægt på 5.000 blev renset som i eksempel 5.

20 g af den rensede poly(ethylenglycolmethylether) blev tørret ved 160°C/0f01 mmHg i 1 time. 80 g tørt frisk fremstillet D,L-lactid blev tilsat, og blandingen blev omrørt under en nitrogenatmosfære ved 160°C. Efter smeltning af alt det D,L-5 lactidet blev der tilsat 0,15 ml stannooctoat (stanno-2-ethyl= hexanoat, og blandingen blev holdt ved 160°C i 6 timer, i hvilket tidsrum der blev dannet er højviskost produkt. Blandingen fik lov til at afkøle, kolben blev åbnet, og indholdet blev opløst i 200 ml acetone. Acetoneopløsningen af polymer 10 blev tilsat under kraftig omrøring til 2000 ml hexan til udfældning af den polymere. Den udfælde polymer blev tørret ved 70°C under reduceret tryk i 24 timer til opnåelse af en blokcopolymer med en AB-struktur, hvor A er polyactidet, og B er poly(ethylenglycolmethylether).
15 Denne copolymer er særlig nyttig til fremstillingen af vand-i-olie-dispersioner, der kan anvendes til fremstillingen af mikrokapsler eller ved mikroindkapslingsmetoder.
5 g af den copolymere blev f.eks. opløst i 200 ml methylen= chlorid og 1 ml af en vandig opløsning af ICI 118.630 inde-20 holdende 20 mg forbindelse blev tilsat under kraftig omrøring til fremstilling af en stabil vand-r-i-olie-dispersion.
Vand-i-olie-emulsionen blev omrørt kraftigt, og et ikke-opløs= ningsmiddel, såsom hexan (2000 ml) blev langsomt tilsat til dannelse af mikrokapsler, som blev isoleret ved filtrering 25 og tørring til opnåelse af en lægemiddel/polymer-blanding, som selv i denne mikrokapsel^· eller mikroindkapslede form giver forlænget frigørelse over en periode på adskillige dage.
Den ved ovennævnte fremgangsmåde anvendte poly(ethylenglycol= 30 methylether) blev erstattet med andre derivater af poly(ethylen= glycol) til fremstilling af lignende blokdopolymere, og egnede eksempler er monocetyletherne (cetomerogoler) og stearatestere.
Claims (6)
1. Farmaceutisk eller veterinært acceptabel amphipatisk ikke-5 tværbundet lineær, forgrenet eller podeblokcopolymer, kendetegnet ved, at den har en minimal gennemsnitsmolekylvægt på 5.000, at den hydrofobe komponent er biologisk nedbrydelig eller hydrolytisk ustabil under normale fysiologiske betingelser, og at den hydrofile komponent kan være biologisk 10 nedbrydelig eller ikke-biologisk nedbrydelig, og at copolyme-ren er i stand til at absorbere vand til dannelse af en hydrogel, når den anbringes i vand eller et miljø af vandig fysiologisk type i et animalsk legeme, og at den amphipatiske copolymer er en lineær blokcopolymer med formlen Affl(BA)n eller 15 Bm(AB)n, hvori m er 0 eller 1, n er et helt tal, A er en farmaceutisk eller veterinært acceptabel hydrofob polymer, og B er en farmaceutisk eller veterinært acceptabel hydrofil polymer, eller den amphipatiske copolymer er en pode- eller forgrenet blokpolymer med formlen ABn eller BAn, hvori A, B og n 20 har de ovenfor anførte betydninger, og hvori henholdsvis A eller B er en skeletpolymer med n enheder af henholdsvis en monomer eller polymer B eller A podet derpå, og hvor A er valgt blandt poly-(D-, L- og DL-mælkesyrer}, poly(D-, L- og DL-lactider), polyglycolsyre, polyglycolid, poly-e-caprolac-25 ton, poly(3-hydroxybutansyre), ikke-terapeutiske hydrofobe po-lypeptider, polyacetaler med formlen \ 30. n irxr" .V / > n v - -' 35 DK 165458 B 5 hvori R er et hydrocarbonradikal, n har den ovenfor anførte betydning, og den stiplede linie betyder en eventuel polyme-thylenkæde, polycarbonater eller polyorthoestere med formlen: 1° . i i —r><r0— i i /
