Description
A találmány tárgya eljárás humán- vagy állatgyógyászati készítményekben alkalmazható kopolimerek és ilyen kopolimereket tartalmazó humán-, és állatgyógyászati készítmények előállítására. A találmány szerint előállítható készítményekből éle!tani típusú, vizes környezetben a hatóanyag hosszabb idő alatt, folyamatosan szabadul fel.
Már régóta felismerték, hogy a klinikai gyakorlatban igen komoly gyakorlati jeletőségük lehet azoknak a készítményeknek, amelyekből — egyszeri beadás után — a hatóanyag huzamosabb idő alatt, folyamatosan szabadul fel. Eddig már számos olyan készítményt dolgoztak ki, amelyekből egyszeri oráiis (lásd például Remington’s Pharmaceutical Sciences, kiadó: Mack Publíshing Co., Easton, Pennsylvania. Amerikai Egyesült Államok; 15. kiadás, 1975; 1618—1631. oldal), parenterális (idm. mű 1631 — 1643. oldala), vagy helyi kezelés (1 351409 sz. nagy-britanniai szabadalmi leírás) után különféle gyógyhatású anyagok huzamosabb idő alatt szabadulnak fel. A parenterális adagolás célszerű módja az, hogy a hatóanyagot bőr alatti injekció vagy beültethető szilárd készítmény (például pasztilla vagy film) formájában juttatják a szervezetbe; erre a célra eddig már többféle implantációs készítményt ismertettek. Így ismert például az, hogy számos gyógyhatású anyag nyújtott hatású implantációs készítménnyé alakítható úgy, hogy a hatóanyagot biológiailag lebontható polimerkapszulába zárják, vagy ilyen polimer mátrixában diszpergálják; ekkor a hatóanyag a polimer mátrix lebomlásának ütemében szabadul fel.
A fenti típusú, nyújtott hatóanyag-felszabadulást biztosító készítmények előállításához felhasználható biológiailag lebontható polimerek különféle típusai ismertek. Ide tartoznak az élettani típusú, vizes közegben hidrolízis révén iassan Icbomló poliészterek. Az ismert, biológiailag lebontható poliészterek közűi a hidroxi-karbonsav-származékokat említjük meg. Számos közlemény foglalkozik az a-hidroxi-karbonsavakból, elsősorban optikailag aktív, vagy racém tejsavból, továbbá giikolsavból levezethető homo- és kopolimerekkel [lásd például 3 773 919 és 3 887 699 sz. amerikai egyesült államokbeli szabadalmi leírás; Jackanicz és munkatársai: Contraccption 8, 227234 (1973); Anderson és munkatársai: Contiaccption 11, 375—384 (1976); Wise és munkatársai: Life Sciences 19, 867-874 (1976); Woodland és munkatársai: Journal of Medicina! Chemistry 16, 897—901 (1973); Yoiles és munkatársai: Bulletin of tbc Parenteral Drug Association 30, 306-312 (1976); Wise és munkatársai: Journal of Pharmacy and Pharmacology 30, 686-689 (1978) és 31, 201-204 (1979)].
Az 1 325 209 sz. nagy-britanniai (és az annak megfelelő 3 773 919 sz. amerikai egyesült államokbeli) szabadalmi leírás, valamint a 3 887 669 sz. amerikai egyesült államokbeli szabadalmi leírás polipéptideket tartalmazó, nyújtott vagy késleltetett hatóanyagfelszabadulású készítményekről számol be. Az utóbbi közlemény csak az inzulint említi, polipeptideket tartalmazó készítmények előállítására azonban egyetlen példát sem közöl. Nyilvánvaló, hogy ez a szabadalmi leírás csupán elméleti lehetőségként említi a 2 p· ipeptideket a készítményekbe beágyazható hatóan agok között, amit az is alátámaszt, hogy a polipra idekre csak a beágyazható hatóanyagok terjedelme:, rendkívül sokféle vegyületet felölelő listájában hivatkozik. Az idézett szabadalmi leírásban említett, pnlipeptidektől eltérő gyógyhatású anyagok mindegyike viszonylag hídrofób jellegű és viszonylag kis molekulasúlyú anyag, és a leírás nem foglalkozik azzal, milyen nehézségek merülhetnek fel, ha a rendszerint viszonylag hidrofil jellegű, viszonylag nagy molekulasúlyt! polipeptidekbői kívánnak megfelelő minőségű, nyújtott hatóanyag-felszabadulást bizto sí'ó készítményeket előállítani.
Megjegyezzük, hogy a hatóanyagok „nyújtott” vrgy „késleltetett” felszabadulása szakaszosan, és folyamatosan egyaránt végbemehet. Azt tapasztaluk, hogy iia a szakirodalomban — így elsősorban az 1 325 209 sz. nagy-britanniai szabadalmi leírásban — isnertetclt módszereket poiijicptid-íartalmú készítmények előállítására alkalmazzuk, a kapott készítményekből ugyan a polipeptid hosszabb idő alatt szabadul fel, de a hatóanyag felszabadulása sok esetben szakaszosan történik. így például ha az idézett szabadalmi leírásban ismertetett polilakíid-polimerbe ágyazunk be polipeptidet, a hatóanyag-felszabadulását hosszú indukciós szakasz előzi meg, amikor polipeptid egyáltalán nem jut a szervezetbe, vagy a hatóanyag több fázisban szabadul fel, azaz a kezdeti szakaszban bizonyos mennyiségű polipeptid felszabadul, a; ezt követő szakaszban hatóanyag-felszabadulás gyakorlatilag nem észlelhető, majd a harmadik szakaszban szabadul fel a jelenlevő polipeptid főtömege. Érzel szemben a találmány szerint olyan készítményeket állítunk elő, amelyekből — esetenként egy rövid iceig tartó indukciós szakasz után — a polipeptid ff lyamatosan szabadul fel, azaz a hatóanyagleadásnak n ncs olyan időszaka, amikor csak igen kevés hatóanyag jut a szervezetbe, vagy hatóanyag egyáltalán nun szabadul fel. A „folyamatos felszabadulás” megjelölésen a leírásban és az igénypont sorozatban a lényegében egyfázisú hatóanyag-felszabadulást értjük. A hatóanyag-felszabadulás görbéjének inflexiós pontja cíetleg lehet, a görbén azonban telítési szakasz biztost n nem észlelhető.
Az 1 388 580 sz. nagybritanniai szabadalmi leírás inzulint tartalmazó, késleltetett hatóanyag-felszabaduláiú készítményeket ismertet. A készítmények alaparyagaí hidregéiek, amiket úgy állítanak elő, hogy egy vízben oldódó polimert kclátképzö szerrel reagáltatnak, majd a polimer — kelátképzőszer láncokat vites oldatban, többértékű fémionokkal végzett reakcióban egymáshoz kapcsolják. Az utóbbi lépésben képződő hidrogélbe ágyazzák be az inzulint, majd a krpott vizes elegyet homogenizálják, és szubkután vagy intramuszkuláris injekció formájában adják be.
A találmány tárgya eljárás humán- vagy állatgyógyászati szempontból alkalmazható, amfipatikus, Iji.eáris, elágazó vagy ojtott tömbkopolimer előállítására, amelynek tömeg szerinti átlagos molekulatömege legalább 5000. A találmány értelmében úgy jámnk el. hogy DL-laktid. DL-(ejsav vagy (!) kepIc'fí glikoiiü egységeket laríalmazó, biológiailag lebontható ludrofób polimert, vagy az azoknak meg-23 felelő monomereket vagy két, a fentiekben felsorolt biológiailag lebontható liidrofób polimer vagy monomer elegyét ömledékben, vagy a reakció szempontjából közömbös szerves oldószerben, adott esetben az ojtásos kopolimerizációhoz, polikondcnzációhoz vagy poliaddíeíóhoz szokásosan felhasznált katalizátor, előnyösen szerves peroxid, szerves ónvegyület, vagy ásványi savval képezett ón(II)-só jelenlétében, 70— 220 C-on, 2—12 órán át hidrofil polimerre], éspedig po!i(etilén-glikol)-lal, poli(vinil-alkohol)-lal, poli(vinüpirro!idon)-nal vagy po!i(etilén-glikol-inctil-éter)-rcí vagy az azoknak megfelelő monomerekkel reagáltatjuk.
A találmány szerinti eljárással előnyösen Am(BA)n vagy Bm(AB)n általános képletű lineáris tömb-kopolimereket állítunk elő — a képletekben m értéke 0 vagy l, n egész számot jelent, és A a fentiekben meghatározott hidrofób polimert, míg B a fentiekben meghatározott hidrofil polimert jelenti. A találmány szerinti eljárással, továbbá ABn vagy BAn általános képletű ojtott vagy elágazó tömbkopolimereket is kialakíthatunk — a képletekben A, B és n jelentése a fenti —, amelyekben az A vagy B polimer képezi a vázat, és a vázhoz ojtással n számú B vagy A monomer, illetve polimer kapcsolódik.
A találmány tárgya továbbá eljárás a hatóanyagot folyamatosan leadó humán- vagy állatgyógyászati készítmények előállítására. A találmány értelmében úgy járunk el, hogy gyógyhatású anyagot, előnyösen hidrofil polipeptidet, így oxitocint, vazopresszint, adrenokortikotrop hormont (ACTH), epidermisznövekedési faktort (EGF), prolaktint, luliberint, vagy sárgatest-hormont felszabadító hormont (LH-RH), növekedési hormont, növekedési hormont felszabadító faktort, inzulint, szomatosztatint, gliikagónt, interferont, gasztrint, tetragasztrint, pentagasztrínt, urogasztront, szekretint, kalcitonint, enkefaiiuokat, endorflnokat, angiotenzineket, renint, bradikinint, bacitracinokat, polimixineket, kolisztineket, tirocidineket, gramicidineket, a felsorolt polipeptidek szintetikus analógjait, módosulatait vagy gyógyászatilag hatásos fragmenseit, egysarjú antitesteket, vagy oldható vakcinákat a fentiek szerint előállított, humánvágy állatgyógyászati szempontból alkalmazható, amfipatikus, néni térhálós, lineáris, elágazó vagy ojtott tömbkopGiimerekkel, amelyek tömeg szerinti átlagos molekulatömege legalább 5000, és adott esetben egy, vagy több egyéb önmagában ismert, humánvágy állatgyógyászati készítmények előállításában alkalmazható segédanyaggal keverjük össze.
A találmány szerinti eljárással előállítható tömbkopolimerek hidrofób komponense biológiailag lebontható vagy a nonnális élettani körülmények között hidrolitikusan instabil anyag, hidrofil komponense pedig biológiailag lebontható vagy biológiailag le nem bontható anyag- A találmány szerinti eljárással előállítható tömb-kopoliiner, illetve az ezt tartalmazó humán- vagy állatgyógyászati készítmény vízbe, vagy vizes élettani környezetbe helyezve vízfelvétel révén hidrogéllé alakul.
A „vizes élettani környezet” megjelölésen amelegvérűek testét, elsősorban izomzatút vagy keringési szervrendszerét értjük; megjegyezzük azonban, hogy laboratóriumi vizsgálatok céljaira ezt a környezetet adott esetben élettani pH-értékre pufferolt, 35—40 C hőmérsékletű vizes folyadékokkal imitálhatjuk.
Amennyiben a gyógyhatású anyagot — különösen előnyösen hidrofil polipeptidet - tartalmazó száraz, amfipatikus kópiáimért vízbe merítjük, vagy vizes élettani környezetbe (élő szervezetbe) helyezzük, a kopolimer hidrofil része vizet vesz fel, és az anyag megduzzad. A vízfelvétel a kopolimer vízben oldhatatlan részei számára inkompatibilis környezetet teremt, és a kopolimer hidrofób részei olyan kapcsoló pontokként hatnak, amelyek gátolják a további vízfelvételt. Ebben a duzzadt, hidratált állapotban a kopolimer mátrix a beágyazott gyógyhatású anyag - például vízben oldható polipeptid — számára áthatolhatóvá válik, és így a gyógyhatású anyag fokozatosan deszorbeálódik a mátrixból.
A duzzadás folyamán, valamint a némileg egyensúlyi állapotig duzzadt állapotban megkezdődik a kopolimer hidrofób részének hidrolitikus bomlása. A részlegesen lebomlott kopolimer duzzadóképessége fokozódik, így a hidrolízis folyamán nő a kopolimer ví/fclvétclc, fokozódik a mátrix vízálhalolhatósága, ami tovább növeli a gyógyhatású anyag desz.orbeálódását. Ez az utóbbi jelenség ellensúlyozza a gyógyhatású anyag koncentráció-csökkenésének hatását, és biztosítja a hatóanyag folyamatos felszabadulását. A kopolimer összetevőinek megfelelő megválasztásával szabályozhatjuk az egyes fiziko-kémiai átalakulási folyamatokat (duzzadás és hidrogélképződés, a hatóanyag deszorbcálódása, a hidrolitikus bomlás megindulása és fokozódása, a hatóanyagfelszabadulás fokozódása), és így olyan készítményeket alakíthatunk ki, amelyekből a hatóanyag huzamosabb idő alatt folyamatosan szabadul fel.
A hatóanyag ideális felszabadulási ütemét úgy is biztosíthatjuk, hogy különböző tulajdonságokkal (például molekulatömeg, molekulatömeg-eloszlás, tömbszerkezet, hidrofil jelleg, bomlási jellemzők, diffúziós sajátságok) rendelkező kopolimereket keverünk össze. A különböző kopolimerek megfelelő arányú összekeverésével a hatóanyag-felszabadulás ütemét és tartamát tetszés szerint módosíthatjuk.
Λ komponensek fenti tulajdonságainak megfelelő megválasztásával és/vagy megfelelő arányú keverésével olyan kopolimer anyagot is kialakíthatunk, amelyből viszonylag alacsony (100 °C-nál lényegesen alacsonyabb) hőmérsékleteken, egyes esetekben szobahőmérsékleten készíthetők humán- vagy állatgyógyászati készítmények, például implantációs kompozíciók. Ezek a kopolimerek, illetve kopolimer-kombinációk különösen előnyösen alkalmazhatók hőre érzékeny vagy oldószerre érzékeny polipeptid hatóanyagok beágyazására. így például a poli(etilénglikol)-ból, és 37 °C-nál magasabb üvegesedési hőmérsékletű amorf hidrofób polimerekből készített tömbkopolimerck különösen előnyösen alkalmazhatók errg. a célra, mert a poli (etilén-glikol) blokkok lágyítják a hidrofób blokkokat, így a kopolimer viszonylag alacsony hőmérsékleten (esetenként szobahőmérsékleten) is könnyen feldolgozható anyaggá válik. Állás közben azonban a poli(etilén-glikol) blokkok kristályosodnak, és az anyagkeverékből 3
198 093 megfelelően kemény, könnyen kezelhető tennék képződik.
A találmány szerint előállítható humán- vagy állatgyógyászati készítmények például orálisan, intraruminálisan, parenterálisan, okulárisan, rektálisan vagy vaginálisan adagolható kompozíciók lehetnek. Ezeket a kompozíciókat ismert klinikai vagy állatgyógyászati módszerekkel, például intramuszkuláris vagy szubkután injekció vagy bőr alatt implantációs készítmény formájában juttathatják a kezelendő szervezetbe.
A találmány szerint előállítható, a hatóanyagot folyamatosan leadó humán- vagy állatgyógyászati készítmények különösen előnyösen tartalmazhatnak hatóanyagokként hidrofil poli peptideket. Miként már említettük, a szakirodalomban ismertetett módszerekkel hidrofil polipeptid hatóanyag esetén csak olyan gyógyászati készítmények alakíthatók ki, amelyekből a hatóanyag szakaszosan szabadul fel. A találmány szerint előállítható humán- vagy állatgyógyászati kompozíciók azonban pepiidtől eltérő hatóanyagot is tartalmazhatnak, amelyek közül példaként az. ösz.tradiolt említjük meg.
A találmány értelmében a humán- vagy állatgyógyászati szempontból alkalmazható, amfipatikus, nem térhálós kopolimereket az egyedi monomerek, illetve polimerek reakciójával állítjuk elő. A reakciót az. ojtásos kopotimcrizáció, polikondcnzáció vagy poliaddíció önmagában ismert körülményei között végezzük. A reakcióelegyhez adott esetben katalizátort adhatunk. Katalizátorként például cink-oxidot, cinkkarbonátot, bázikus cink-karbonátot, dietil-cinket, szerves ónvegyületeket, így ón(II)-okíoátot (ón /II/2-etiI-hexanoátot), tributil-alumíniumot, titán-, magnézium- és bárium-vegyületeket, ólom-oxidot, és hasonló anyagokat - különösen előnyösen ón(II)oktoátot — használhatunk fel.
Az egyéb reakciókörülmények (például hőmérséklet és reakcióidő) tekintetében a reakciót ismert, hagyományos módon végezzük.
A találmányt az. oltalmi kör korlátozása nélkül az alábbi példákban részletesen ismertetjük.
1. példa g 20 000-es molckulatömegű poli-(etilén-glikol)-t vákuumban (0,1 Hgmin-nál kisebb nyomáson) órán át 120 C-on keverünk. A polimerhez 15 g DL-laktidot és 15 g glikolidot adunk, és az, elegyet a szilárd anyagok megolvadásáig nitrogén-atmoszférában keverjük. Ezután az elegyet 160 °C-ra melegítjük, és 0,1 ml ón(II)-oktoátót (ón/II/-2-eíii-hexanátot) adunk hozzá. Az elegyet 3 órán át 160 °C-on tartjuk, majd a kapott, igen viszkózus anyagot lehűtjük, és 200 ml acetonban oldjuk. Az acetonos oldatot lassú ütemben .erélyes keverés közben 1500 ml etanolba adagoljuk, majd a kivált csapadékot leszűrjük, és vákuumban 3 órán át szobahőmérsékleten, majd éjszakán át 40 °C-on szárítjuk.
A kapott kopolimer deutero-kloroformban felvett NMR-spektruma szerint a termék 2:1:1 arányban tartalmaz oxietilén-, tejsav- és glikoisav-egységeket. A kopoliiner kloroformos oldatának határviszkozitasa 0,58.
A kopolimert körülbelül 60 °C-on lágy, plasztikus, áí átszó filmmé dolgozzuk fel. A film 39 mg tömegű m ntáját vízbe helyezzük. A kopolimer gyorsan meghúzzad, 4 óra alatt 135 mg vizet vesz fel, és átlátszó hidrogéné alakul, amely 2 hét alatt 37 °C-on fokoza'osan Icbomlik.

20,2 mg, az 1. példa szerint előállított kopolimert kh. 40 °C-on 5,1 mg szarvasmarha növekedési hormonnal (EGH) keverünk össze, és a kapott átlátszatlan keverékből 1 mm vastag lemezt készítünk. A lemezt 0,01% nátrium-aziilot tartalmazó. 1/15 mólos vizes pufferoldatba (pH = 8,6) helyezzük. A lemezből legalább 12 napon át körülbelül 22 000-es molekulatömegű anyag szabadul fel, amelynek retenciós ideje (nagynyomású folyadék-kromatográfiával mérve) megegyezik a szan/asmarha növekedési hormon retenciós idejével.
3. példa
A 2. példában közöltek szerint előállított kopolimer/BGH keverékből körülbelül 45 mg tömegű, körülbelül 20% BGli-í tartalmazó korongokat készítünk. A korongokat pajzsmirigyüktől megfosztott patkányokba ültetjük be. A beültetést követően az állatok testtömege 7 nap alatt körülbelül 25%-kal növekszik, míg a placebó készítménnyel „kezelt” kontrollál'atok testtömege változatlan marad.
4. példa
13,5 mg, az 1. példában közöltek szerint előállított kopolimert 50 °C-on 1,5 mgegvsaijú egérimmunoglobin A-val IgA; meghatározott aiitigcn-spccifitású anyag, molekulatömege ISÜOOQ-nél nagyobb) keverünk össze, majd a homogén keverékből öntéssel körülbelül 2 mm átmérőjű gömböt készítünk. A hatóanyag in vitro körülmények között történő felszabadulását úgy vizsgáljuk, hogy a fehérje/kopoiimer készítményt 37 °C-os, foszfátpui'ferrel pufferolt (pH = 7,2) fiziológiás sóoldatba merítjük, és enzimes módszerrel mérjük a vizes közeg IgA-tartalmát. A biológiailag aktív fehérje felszabadulása 2 nap elteltével kezdődik meg, és legalább 9 napon át folytatódik.
5. példa g 20 000-es molekulatömegű poli-(etilén-glik«l)-t 150 mg kloroformban oldunk, és az oldatot körülbelül 300 ml desztillált vízzel hatszor mossuk. A vizes mosófolyadékokat elöntjük. A kloroformot csökkentett nyomáson lepároljuk, és a tisztított poli(eíilcn-g'iikol)-t 1 órán át 160 °C-on, 0,05 Hgmm nyomáson szárítjuk.

A polimerhez nitrogén-atmoszférában 42,9 g frissen készített, tiszta DL-laktidot adunk, és az elegyet 160 °C-on megömlesztjük. Az elegyhez 0,2 ml ón (ll)-oktoátot adunk, és addig keverjük, amíg a viszkozitásnövekedés meg nem akadályozza a további keverést. 3 óra elteltével igen viszkózus terméket kapunk. Az elegyet lehűlni hagyjuk, a lombikot összetörjük, a lombik tartalmát körülbelül 300 ml acetonban feloldjuk, és az oldatot szűrjük. A szűrletet lassú ütemben, erélyes keverés közben körülbelül 1000 ml eíanolba adagoljuk. A kivált rostos csapadekot leszűrjük, és vákuumban éjszakán át 30 °C-on szárítjuk. A termékből öntéssel 0,2 min vastag, lágy, plasztikus, átlátszó filmet készítünk, cs a film 0,54 g tömegű mintáját vízbe merítjük. A 37 °C-os vízben tartott film tömege 1 nap alatt 0,95 g-ra nő. A hidratált, átlátszó film merevsége és szilárdsága jobb a kiindulási száraz kopolímerénél. 35 nap elteltével a film látszólag változatlan marad, és még mindig jó mechanikai tulajdonságokkal rendelkezik, ami azt jelzi, hogy a kopolimer csak lassan bomlik le. Ugyanezt támasztják alá az összetétel elemzése céljából felvett NMR-spektrum adatai.
Ha a fentiek szerint előállított, száraz kopolinterbe 60 °C-on szarvasmarha növekedési hormont ágyazunk be, és a terméket 1/15 mólos vizes foszfátpuffer-oldatba (pH = 8,6) merítjük, a termékből legalább 7 napon keresztül 22 000-es molekulatöinegű anyag szabadul fel.
6. példa g 6000-es molekulatömege poli-(etilén-glikolt)-t az 5. példában ismertetett módszerrel tisztítunk.
7,5 g tisztított, száraz poli-(ctilén-gliko!)-t szobahőmérsékleten 15 mg ón(II)-klorid.-dihidratáttal keverünk össze, az elegyet 0,1-0,01 Hgnmi nyomáson, keverés közben 155 °C-ra melegítjük, majd 2 órán át ezen a hőmérsékleten tartjuk, miközben az elegyhez nitrogén-atmoszférában 22,5 g frissen készített, száraz DL-laktidot adunk, és a laktidot megömlesztjük. Az elegyet 3 órán át 155-160 °C-on tartjuk. A kapott viszkózus terméket poli-(tetrafluoretilén) filmre öntjük, és lehűlni hagyjuk. A polimert melegítés közben 70 ml acetonban oldjuk, majd az acetonos oldatot 600 ml etanolba öntjük. A kivált csapadékot elkülönítjük, és éjszakán ál vákuumban 60 °C-on szántjuk. A polimer kloroformos oldatának határviszkozitása 0,41. A polimerből 0,2 mm vastag filmet készítünk, és a film mintáját 37 °C-os vízbe merítjük. A polimer körülbelül 24 óra alatt saját tömegének megfelelő mennyiségű vizet vesz fel, és ragacsos hidrogélt képez.
7. példa mg, a 6. példában közöltek szerint előállított, 25 rész 6000-es molekulatömegű poli-(etilén-glikolt)-t, és 75 rész poli-(D,L-laktid)-ot tartalmazó tömb-kopolimert 4,5 ml anliidridmcntcs jcgecet és 0,5 ml desztillált víz elegyében oldunk. A polimer oldatához 200pl, 1,1 mg egér epidermisz növekedési faktort (EFG) tartalmazó oldatot adunk, és az elegyet homogenizáljuk. A homogén oldatot fagyasztjuk, majd 18 órán át fagyasztva szárítjuk. A termékből 50 °C-on ömlesztéssel 40 mg tömegű, körülbelül 8 mmX4 mm XI mm méretű implantációs készítményt állítunk elő. A készítményt 1 ml 37 °C-os emberi vérszérumba helyezzük, és a szérumból időről-időre mintát veszünk. A felszabadult EGF mennyiségét radiológiai immunvizsgálattal határozzuk meg. Az eredmények szerint a pepiid folyamatosan, legalább 3 napon át szabadul fel a készítményből.
8. példa g, DL-laktidot és glikolidot ekvimoláris arányban tartalmazó kopolimert (kloroformos oldatban mért határviszkozitás: 0,20) 50 ml vízmentes etilacetátban oldunk, és az oldatot keverés és visszafolyatás közben, nitrogén-atmoszférában forraljuk. 25 ml frissen desztillált vinil-pirrolidonban 0,25 g lauroil-peroxidot oldunk, és az oldatot 2 óra alatt a forrásban levő polimer oldatba csepegtetjük. A kapott reakcióelegyet további 6 órán át forraljuk. Az elegy hűtés hatására kocsonyásodik. A kapott amfipatikus ojlott tömbkopolimerből klcsapásos technikával igen nehezen távolítható el a poli-(vinil-pirroiidon) homopolimer, mert a kicsapás során igen gyakran képződik kolloid szuszpenzió. Ez azt jelzi, hogy a poh-(vinil-pirrolidon) ojtással épült össze a laktid/glikolid kopolimerrel.
Az etil-acetátos elegyet 70 °C-ra melegítjük, cs 50 ml etanolt adunk hozzá. A kapott kolloid szuszpenziót 2 liter n-hexánba öntjük. A kivált polimert elkülönítjük, éjszakán át vákuumban 90 °C-on szárítjuk, majd lehűtjük. Törékeny terméket kapunk, amely ojlott kopolimcrből és bonio-poli-(vinil-pirrolidon)-hói áll. A termék kloroformos oldatának határviszkozitása 0,29. A termék körülbelül 50% honio(poli-vinil-pirrolidon)-ból, és körülbelül 50% ojtott kopolimerböl áll.
0,45 g így kapott polimert és 0,05 g ICI 118 630 jelű polipeptidet 5 ml anhidridmentes jégecetben oldunk, és az oldatot 0,01 Hgmm nyomáson 22 órán át fagyasztva szárítjuk.
A terméket 20 másodpercig 110°C-on megömlesztjük, és az ömledékből 30 mg tömegű, körülbelül 0,8 cmXl,2 mmX2 mm méretű hasábot készítünk. A 37 °C-os vizes pufferoldatba (pH = 7,4) merített hasábból több nap alatt szabadul fe! a polipeptid.
9. példa g, 14 000-cs molekulatöniegű poli-(vinil-alkohoí)-t 500 g kereskedelmi minőségű, körülbel 12% 5 !98 093 vizet tartalmazó DL-tejsavban oldunk keverés közben, nitrogén-atmoszférában. Az oldatot HO °C-ra melegítjük, és a vizet 8 óra alatt kidesztilláljuk az elegyből. Ezalatt az elegy egyre viszkózusabbá válik, és hőmérséklete 190 C-ra emelkedik. Amikor több víz már nem desztillál át, a nyomást körülbelül 25 Hgcm-re csökkentjük, és az elegyet további 8 órán át melegítjük. Végül a nyomást 0,1 Hgmm-re csökkentjük, és az elegyet 8 órán át 200 °C-on tartjuk. Nagyon viszkózus, ámbraszínű terméket kapunk.
A poimert lehűlni hagyjuk, és a lombikot összetörjük. A terméket apró darabokra törjük, 1,5 liter metanolban oldjuk, majd az oldatot 10 liter desztillált vízbe öntjük. A kivált csapadékot elkülönítjük, további 5 liter vízzel mossuk, és vákuumban 8 órán át szobahőmérsékleten, majd 16 órán át 100 °C-on szárítjuk. Ámbraszínű, üvegszerű terméket kapunk, amelyben a poli-(vinil-alkoho!)-vázhoz elagazóan kis molekulatömegű poli-tejsav láncok kapcsolódnak. A termék kloroformos oldatának határviszkozitása 0,65. A termék körülbelül 85% politejsavat tartalmaz, és a politejsav-oldalláncok átlagos lánchossza körülbelül
3,5 egység.
Megismételjük a fenti eljárást azzal a különbséggel, hogy DL-tejsav helyett azonos mennyiségű DL-laktidot használunk. Ámbraszínű, üvegszerű terméket kapunk, amelyben a poli-(vinil-alkohol)-vázhoz elágazóan kis molekulatömegű polilaktid láncok kapcsolódnak, A termék klorofomos oldatának határviszkozisáta 0,66. A termék körülbelül 85% polilaktidot tartalmaz, és a polilaktid oldalíáncok átlagos lánchossza körülbelül 3,5 egység.
Az így kapott polimerekből 100 °C-on öntéssel 1 cm X 0,2 cm X 0,2 cm méretű hasábokat készítünk, és a hasábokat 37 °C-os vízbe merítjük. A termékek vizet kötnek meg, hajlékonyakká válnak, majd bomlásnak indulnak., és 2—2,5 hónapon át oldható anyagokat adnak le.
10. példa mg 80/20 arányú poli-(DL-laktid)/poli (etilénglikol) 6000 kopolimer (a kloroformos oldat határviszkozitása 0,36) 2,5 ml 90%-os vizes ecetsavval készített oldatához 285 jul, 21 mg/ml koncentrációjú, desztillált vízzel készített egér epidermisz növekedési faktor-oldatot adunk. A kapott oldatot fagyasztjuk, majd 24 órán át körülbelül 0,01 Hgmm nyomáson fagyasztva szárítjuk. A fagyasztva szárított anyagból ömlesztéssel, 60 °C-on két implantációs készítményt állítunk elő. Az egyik implantációs készítmény tömege 13,5 mg, és 1,3 mg peptidet tartalmaz, míg a másik készítmény tömege 15,3 mg, és 1,5 mg peptidet tartalmaz.
Az implantációs készítményeket szubkután úton macskákba ültetjük be. A macskák gyomrába előzetesen szondát vezetünk. A macskák véréből mintát veszünk, és vizsgáljuk, hogy a kiválasztott gyomornedv hogyan reagál hisztaminra. A vérmintában a beültetést követő legalább 3 napon át mutatható ki peptid radioiminunológiai méréssel, és a kiválasztott gyoinomcdvben a beültetést követő 3-6 napon át észlelhető gátló hatás.
11. példa mg 85/15 arányú poli-(DL-laktid)/6000-es molekulatömegű poli-(etilén-glikol) kopolimer (a kloroformos oldat határviszkozitása 0,39) 1,8 ml 90%-os vizes acetonnal készített oldatához 120 μΐ desztillált vizes egér epidermisz növekedési faktoroldatot adunk (az oldat 320 liter desztillált vízben 105 mg peptidet tartalmaz). A kapott oldatot fagy isztjuk, ós éjszakán át fagyasztva szárítjuk. A fagyasztva szárított termékből 70 °C-on ömlesztéssel
16.9 mg súlyú, körülbelül 1 mm XI mmX5 mm méretű implantációs készítményt készítünk.
A készítményt 10% emberi vérszérumot és 0,1% nátrium-azidot tartalmazó vízbe merítjük. A peptid legalább 15 napon át folyamatosan szabadul fel.
12. példa
Összehasonlító kísérleteket végeztünk annak megúl’apitására, hogyan befolyásolja a tömb-kopoiiiucr összelétele és hidrofil jellege a polipeptid felszabadulását.
Az implantációs készítmények előállításához a következő kopolimereket használtuk fel:
a) 25 tömeg% 6000-es molekulasúlyú poli-(etilénglikol)-t és 75 tönreg% poli-(DL-laktid)-ot tartalmazó tömbkopolimer (a kloroformos oldat határviszkozitása 0,39): és
b) 5 tömeg % 6000-es molekulasúly ú poli-(etiléngl kol)-t és 95 tömeg % poli-(DL-laktid)-ot tartalmazó tömbkopolimer (a kloroformos oldat határviszkozitása 0,79).
76,2 mg a) vágy b) polimert és 23,8 g JCI 118 630acetátot (20 mg tiszta pcptiddel egyenértékű mennyiségű só) 1,5 ml anhidridmentesjégecetben oldottunk. Az oldatot fagyasztottuk, 18 órán át fagyasztva száritettük, majd a fagyasztva szárított terméket körülbí lül 70 °C-on megömlesz.tve 45—50 mg súlyú implantációs készítményt állítottunk elő (a készítmény közelítő méretei: 0,2 cm^X 0,2 cm X1 cm).
A, készítményeket 1 ml 37 °C-os Mclivatns pufferoidatba (pH = 7,4) merítettük. Λ vizes közegből időről-időre mintát vettünk, és nagynyomású folyadékkromatográfiás módszerrel meghatároztuk a minta pr ptid-tartalmát. Az eltávolított vizes közeget minden esetben azonos térfogatú friss pufferoldattal pótoltuk.
A mérések szerint a nagyobb mértékben hidrofil (25% poli-/etilén-glikol/-t tartalmazó) kopolimer mátrixból körülbelül 18 napon át szabadul fel a hatóanyag. Ezzel szemben a kevésbé hidrofil (5% poli/e tilén-glikoi/-t tartalmazó) mátrixból a hatóanyag legalább 250 napon át szabadul fel folyamatosan.
13. példa
5000-es molekulatömegű poli-(etilén-glikol-meíilérer)-t az 5. példában ismertetett módon tisztítunk.
g tisztított poli-fctilcn-glikol-inctil-étcr)-t 1 órán á’ 160 °C-on, 0,01 llgmin nyomáson szárítunk. A polimerhez 80 g száraz, frissen készített DL-laktidot

198 093 adunk, és az elegyet nitrogén atmoszférában i60°C-on keverjük. Amikor a DL-laktid teljes mértékben megolvadj az elegyhez 0,15 ml ön(Il)-oktoátot (ón/II/2-etil-hexanoátot) adunk, és az elegyet 6 órán át 160 °C-on tartjuk. Ezalatt az elegyből igen viszkózus termék képződik. A terméket lehűtjük, a lombikot összetörjük, majd a terméket 200 ml acetonban oldjuk. A polimer acetonos oldatát erélyes keverés közben 2000 ml hexánhoz adjuk. A kivált polimert elkülönítjük, 24 órán át vákuumban 70 °C-on szárítjuk. AB-szerkezetű tömbkopolimert kapunk, ahol A apolilaktidot, B pedig a poli-(eíiién-ghkol-metil-éter)-t jelenti.
Ez a kopolimer különösen előnyösen használható víz-az-olajban típusú diszperziók előállításához, ami mikrokapszulák készítéséhez, illetve mikrokapszulázási technológiákhoz hasznosítható. A kopolimer hatán iszkozitása (kloroformos oldatban mérve) 0,32.
Megismételjük a fenti eljárást, azzal a különbséggel, hogy DL-laktid helyett azonos mennyiségű DLtejsavat, illetve azonos mennyiségű 75:25 arányú laktid :gliko!id kopolimert használunk. Λ fentiekkel azonos szerkezetű tönib-kopolimcrekct kapunk; n kopolimerek határviszkozitása (kloroformos oldatban mérve) 0,33, illetve 0,32,
5-5 g, a fentiek szerint előállított kopolimert 200-200 ml metilén-kloridban oldunk, és az oldatokhoz erélyes keverés közben 20—20 mg Ghi-HisTrp-Ser-Tyr-D-Ser(0-tBu)-Leu-Arg-Pro-Azg!y-NH2-t (ICI 118 630; a 179 990 sz. magyar szabadalmi leírásban ismertetett vegyület) tartalmazó 1 ml vizes oldatot adunk. Stabil víz-az-oíajban diszperziók képződnek.
A kapott diszperziókhoz erélyes keverés közben a polimert nem oldó anyagot, például 2000 ml hexánt adunk. A képződött mikrokapszulákat kiszűrjük, és szárítjuk. A kapott mikrokapszulákból a hatóanyag késleltetetten és folyamatosan, több napon át szabadul fel.
A fenti eljárásban poli(etilén-glikol-metil-éter) helyett egyéb poíi(ctilén-glikol)-származékokat, így mouocetil-étcreket és sztcarát-észterekct is felhasználhatunk.
14. példa
0,4 g, az 5. példában közöltek szerint előállított kopolimert és 0,1 g ösztradiolt metilén-dikloridban oldunk, és az oldószert Icpároljuk. A kapott anyagot 1 mm vastag filmmé préseljük, és a filmet 0,1 g tömegű hasábokra vágjuk szét. Egy másik kísérletben 0,4 g, az 5. példában közöltek szerint előállított kopolimert finoman elporítunk, a porhoz 0,1 g ösztradiolt adunk, és a szilárd keveréket 4 órán át homogenizáljuk. A homogenizátumot 1 mm vastag filmmé préseljük, és a filmet 0,1 g tömegű hasábokra vágjuk szét.
A hatóanyag-felszabadulás vizsgálata során a hasábokat foszfátpufferrel pH = 7,2 értékre pufferolt fiziológiás sóoldatba merítettük, az elegyet 37 °C-on tartottuk, a sóoldatból időről-időre mintát vettünk, és megvizsgáltuk a minta ösztradiol-tartalmát.
