Abstract
Die Erfindung betrifft ein Verfahren zur Herstellung von neuen 1-Aryl-5H-2,3-benzodiazepin-Derivaten der allgemeinen Formel (I) und deren Saeureadditionssalzen. Die neuen 1-Aryl-5H-2,3-benzodiazepin-Derivate entsprechen der allgemeinen Formel (I), worin R und R1 unabhaengig voneinander ein Wasserstoffatom oder Chloratom, eine Alkylgruppe mit 1 bis 4 C-Atomen oder eine Alkoxygruppe mit 1 bis 4 C-Atomen, R2 ein Wasserstoffatom oder eine Alkylgruppe mit 1 bis 4 C-Atomen, R3 und R4 eine Alkylgruppe mit 1 bis 4 C-Atomen und R3 und R4 gemeinsam eine Methylengruppe bedeuten. Die erfindungsgemaess hergestellten Verbindungen stellen wertvolle ZNS-Stoffe mit antiaggressiver, anxiolytischer, narkotischer und narkosepotenzierender Wirkung dar. Beispielsweise wird 1-(4-Aminophenyl)-4-methyl-7,8-dimethoxy-5H-benzodiazepin hergestellt. Formel I

Verfahren zur Herstellung von neuen 1-Aryl- -5r[-2,3-benzodiazepin-Derivaten
Die Erfindung betrifft ein Verfahren zur Herstellung von neuen l-Aryl-5H_-2,3~benzodiazepin-Derivaten der allgemeinen Formel (I) und deren Säureadditionssalzen.
Die erfindungsgemäß hergestellten l-Aryl-5h[-2,3-benzodiazepin-Derivate entsprechen der allgemeinen Formel (I),
R und R]_ unabhängig voneinander ein Wasserstoff atom oder Chloratom, eine Aikylgruppe mit 1 bis 4 C-Atomen oder eine Alkoxygruppe mit 1 bis
4 C-Atomen, R2 ein Wasserstoffatom oder eine Alkylgruppe mit 1 bis 4 C-Atomen,
Rj und R^ eine Alkylgruppe mit 1 bis 4 C-Atomen und R3 und R^ zusammen eine Methylengruppe bedeuten.
Das pharmakologische Wirkungsspektrum der aus der Fachliteratur bekannten 5H_-2,3-Benzodiazepin-Derivate (US-PS 3 736 315 und 3E-PS 879 404) ist unterschiedlich von dem
der 1,4-Benzodiazepine, und zwar es ist wesentlich günstiger, da die Verbindungen keine spasmolytische Wirkung ausüben und keine Muskelschwäche oder Koordinationsstörungen hervorrufen.
Die neuen 5H_-2,3-Benzodiazepin-Derivate der allgemeinen Formel (I) üben einen starken EinfluB auf das zentrale Nervensystem aus und besitzen hauptsächlich eine antiaggressive und anxiolytische Wirkung. Ihre Wirkungsstärke übertrifft mehrfach die Wirkung des Handelspräparats Tofisopam (Grandaxin = l-(3,4-Dimethoxyphenyl)-4-methyl-5- -äthyl-yjS-dimethoxy-SH-ZjS-benzodiazepin), sowie die des l-(3-Chlorphenyl)-4-methyl-7 ,8-dimethoxy-5H_-2 ,3-1benzodiazepine (US-PS 4 322 346). Während die bekannten Verbindungen neben ihrer anxiolytischen Wirkung grundsätzlich antidepressiven Charakter haben, sind die erfindungsgemäßen Verbindungen der allgemeinen Formel (I) daneben milde Neuroleptika. So sind die letztgenannten Verbindungen als Wirkstoffe* von Tagesberuhigngsmitteln für die Behandlung von anxiolytischen und Streßzuständen geeignet.
Ziel der Erfinduna:
Ziel der Erfindung ist es, neue Wirkstoffe der obigen Art und ein Verfahren zur Herstellung derselben bereitzustellen
Die Verbindungen der allgemeinen Formel (I) werden erfindungsgemäß folgenderweise hergestellt: eins Nitro-Verbindung der allgemeinen Formel (II), worin R, R1, R0, R-, und R λ die obige 3edeutung haben, wird in einem organischen Lösungsmittel reduziert, und falls gewünscht, wird aus dar erhaltenen Base der allgemeinen Formel (I) ein Säureadditionssalz gebildet, gewünschtenfalls kann die freie Base aus ihrem Salz nach einer bekannten Methode freigesetzt
und mittels einer Säure in ein anderes Säureadditionssalz überführt werden.
Als organische Lösungsmittel können Alkohole mit 1 bis 4 C-Atomen, Dioxan, Tetrahydrofuran, Benzol, Toluol, Xylol, Dimethylformamid, Dimethylacetamid oder Gemische derselben eingesetzt werden.
Gemäß einer vorteilhaften Ausführungsform des erfindungsgemäßen Verfahrens wird eine Nitro-Verbindung der allgemeinen Formel (II) in Dimethylformamid gelöst und katalytisch hydriert.
Die Hydrierung kann vorzugsweise bei Raumtemperatur durchgeführt werden. Als Katalysator wird Palladium-Aktivkohle, Platin oder Raney-Nickel angewandt.
Das Reaktionsgemisch w,ird mittels einer bekannten Methode aufgearbeitet. Der Katalysator wird filtriert, das Lösungsmittel im Vakuum eingedampft, und derRückstand mittels Präzipitation, Umkristallisierung und/oder Digerierung gereinigt. Niedere Alkohole sind bevorzugte Umkristallisierungsmittel.
Unter den obigen Umständen wird bei der katalytischen Hydrierung der 5H_-2,3-3enzodiazepin-Derivate der allgemeinen Formel (II) nur die Nitrogruppe reduziert, während in den 1,4-Benzodiazepinen die ähnlich gelagerte C=M-Doppelbindung mit katalytischer Hydrierung gesättigt werden kann (D. Org. Chem. 28, 2456 /1963/; Coll. Czech. Chem. Comm. 31, 1264 /1965/).
Gemäß einer weiteren vorteilhaften Ausführungsform des erfindungsgemäßen Verfahrens wird eine Nitro-Verbindung der allgemeinen Formel (II) in einem niederen Alkohol suspen-

diert und in Gegenwart eines Katalysators mit Hydrazin oder Hydrazinhydrat reduziert. Bei der Reaktion werden vorzugsweise 98 - 100 %iges Hydrazinhydrat, und als Katalysator Palladium-Aktivkohle, Platin oder Raney-Nickel eingesetzt. Die Reduktion wird in einem Temperaturbereich, der zwischen 200C und dem Siedepunkt des Lösungsmittels liegt, vorzugsweise bei Raumtemperatur, durchgeführt. Das Reaktionsgemisch wird in bekannter Weise wie oben aufgearbeitet .
Die mit Hydrazinhydrat durchgeführte Reduktion ist besonders bei jenen Verbindungen günstig, die einen Chlorsub- · stituenten besitzen (Chem. Rev. 5J5, 51 /1965/).
Für die Reduktion der Nitro-Ver'oiridungen der allgemeinen Formel (II) sind alle weiteren Reduktionsmethoden geeignet, die in der Struktur des Diazepinringes selbst leine Änderung verursachen. So können für die Reduktion keine komplexen Metallhydride eingesetzt werden, da diese nicht nur zur Reduktion der Nitro.gruppe , sondern auch noch zur Sättigung der C=N-Doppelbindung im Diazepinring zwischen dem Stickstoffatom in Position 3 und dem Kohlenstoffatom in Position 4 führt (US-PS 4 423 044). Bei der Zinkstaubreduktion in Eisessig wird aus dem 7-gliedrigen Ring der 5H_-2,3-Benzodiazepin-Derivate mittels Freisetzung eines Mols Ammoniaks ein 6-gliedriger Ring gebildet, wordurch Isochinolin-Derivate entstehen (Chem. B'er. 107, 3883 /1974/).
Bei dem erfindungsgemäßen Verfahren werden die Verbindungen der allgemeinen Formel (I) in Form einer Base gewonnen. Für die Herstellung von Säureadditionssalzen der Verbindungen der allgemeinen Formel (I) wird die Base vorzugsweise in einem geeigneten Lösungsmittel, zum Beispiel in Methanol, Äthanol, Isopropanol, Eisessig oder

Wasser gelöst oder suspendiert, dann mit einer entsprechenden Säure, zum 3eispiel Salzsäure, Bromwasserstoffsäure , Phosphorsäure, Schwefelsäure oder mit einer Lösung derselben, versetzt. Die Salze können direkt durch Filtrieren oder durch Abdampfen des Lösungsmittels gewonnen werden.
Einige rohe Basen können nicht durch direktes Umkristallisieren gereinigt werden, diese werden vorzugsweise aus ihren gemäß dem obigen Verfahren gebildeten reinen Säureadditionssalzen freigesetzt.. Für die Freisetzung der Basen der allgemeinen Formel (I) werden ihre- entsprechenden Salze in einem Lösungsmittel, zum Beispiel in Wasser, gelöst, worau hin die Lösung entweder mit einer organischen Base, zum Beispiel mit Triäthylamin oder Pyridin, oder mit einer anorganischen Base, zum Beispiel mit einer wäßrigen Natriumhydroxid- oder Ammoniumhydroxid-Lösung versetzt wird. Die freigesetzte Base wird sodann filtriert. Falls gewünscht, wird aus der gewonnenen reinen Base nach der obigen Methode mittels einer entsprechenden Saure ein Säureadditionssalz gebildet. ·
Die bei dem erfindungsgemäßen Verfahren als'Ausgangsmaterialien benutzten Verbindungen der allgemeinen Formel (II) sind teilweise neue und teilweise bekannte Stoffe. Si können vorzugsweise aus den entsprechenden Pyrylium-Salzen bzw. aus 1,5-Qiketonen mit Hydrazin oder Hydrazinhydrat (BE-PS 879 404) gewonnen werden.
Die erfindungsgemäßen Verbindungen der allgemeinen Formel (I) üben einen wesentlichen Einfluß auf das zentrale Nervensystem aus und besitzen wertvolle antiaggressive, anxiolytische, narkosepotenzierende und hypnotische Wirkung, außerdem hemmen sie die psychomotorische Hyperaktivität.
Die antiaggressive Wirkung wurde mit dem "Fighting behaviour" -Test untersucht (J. Pharm. Exp. Ther. 125, 28 /1959/). Tofisopam und l-(3-Chlorphenyl)-4-methyl-7,8-dimethoxy-5H_-2,3-benzodiazepin dienten als Referenzsub stanzen. Die Testergebnisse sind in Tabelle 1 zusammen gefaßt .

Die anxiolytische Wirkung wurde mittels des "Lick-»conflict'1-Testes nach Vogel untersucht (Psychopharmacol. 21, 1 /1971/), Tofisopam , 1-(3-Chlorphenyl)-4-methyl-7,8-dimethoxy-5Hi-2,3- -benzodiazepin und Chlordiazepoxid (7-Chlor-2-methylamino- -5-phenyl-3hl·-!,4-benzodiazepin-4-oxid) dienten als Referenzsubstanzen. Die Testergebnisse sind in Tabelle 2 zusammengefaßt .

Die spontane motorische Aktivität wurde mit einem Motimeter, Typ Animex, bestimmt. Mach 24 Stündigem Fasten wurden je 3 CFLP (Gewicht 20-23 g) in den Apparat gesetzt, und nach einer oralen Vorbehandlung von 10 Minuten wurde 2 Stunden lang ununterbrochen die Aktivität der Tiere gemessen. Es wurde jene Dosis bestimmt, die die spontane motorische Aktivität der Tiere im Vergleich zu der nur mit Vehikel behandelten Konstrollgruppe auf die Hälfte herabsetzte. Die EOj-n-Werte wurden nach der Methode von Lichtfield und Wilcoxon (O. Pharmacol. Exp. Ther. 96_, 99 /1949/) berechnet und einige in Tabelle 3 zusammengefaßt.

Die Narkosepotenzierungsuntersuchung jeder Verbindung wurde bei Gruppen von Mäusen von je 15-20 Tieren, mit 3 bis 4 peroalen Dosen durchgeführt. Der ED5Q-Wert ist jene- Dosis, die bei der Hälfte der Tiere, die mit einer intravenösen Dosis von 50 mg/kg Hexobarbital-natriurn hervorgerufene Narkosezeit auf das Doppelte verlängert.

Die in den Tabellen zusammengefaßten Versuchsergebnisse beweisen, daß mehrere Verbindungen die Wirksamkeit der Referenzsubstanzen erreichen oder sie wesentlich übertreffen, die wirksamsten von ihnen sind das l-(4-Aminophenyl)- -4-methyl-7,8-dimethoxy-5H;-2,3-benzodiazepin (Verbindung von Beispiel 1) und das l-(3-Chlor-4-aminophenyl)-4-methyl- -7,8-dimethoxy-5H-2,3-benzodiazepin (Verbindung von Beispiel 18) .
Das l-(4-Aminophenyl)-4-methyl-7,8-methylendioxy-5H-2,3-benzodiazepin (Verbindung von Beispiel 8) besitzt eine signifikante hypnotische Aktivität, seine ED_ -Werte bei Mäusen sind 140 mg/kg p.o., 70 mg/kg i,p., 40 mg/kg i.V., und bei Ratten 40 mg/kg' i.V.. Die ED_Q-Werte des Nitrazepam (7-Nitro-5-phenyl-l,3-dihydro-2H-l,4-benzodiazepin-2-on) bei Mäusen sind 185 mg/kg p.o., 150 mg/kg i.p., 76 mg/kg i.v. und bei Ratten 95 mg/kg i.v...

Dene Verbindungen der allgemeinen Formel (I), bei denen R-, und R^ zusammen eine Methylengruppe bedeuten, haben im Gegensatz zu den anderen Verbindungen der allgemeinen Formel (I) auch eine signifikante muskelrelaxan.te und spasmolytische Wirkung. So ist zum Beispiel das l-(4-Arninophenyl)-4-methyl-7 ,8-methylendioxy-5iH-2,3-benzodia~ zepin (Verbindung von Beispiel 8) ein Muskelrelaxant mit zentralem Angriffspunkt wesentlich wirksamer als die Referenzverbindung Zoxazolamine (2-Amino-5-chlor-l,3-benzoxazol). Ihre EDcn-Werte betragen 47 bzw. >1OO mg/kg i,p. im "inclinde screen" test (0- Pharm. Exp. 129, 163 /I960/) und 24 bzw. 74 mg/kg i.p. im Rotarod Test (0. of Am. Pharm. Assoc. 46, 208 /1957/).
Pharmazeutische Kompositionen können als Wirkstoff wenigstens eine Verbindung der allgemeinen Formel (I) zusammen mit einem oder mehreren üblichen Hilfsstoffen, sowie Trägerstoffen, Verdünnungsmitteln und/oder Zusatzstoffen enthalten. Falls gewünscht, können die pharmazeutischen Kompositionen auch noch weitere Wirkstoffe enthalten. Die pharmazeutischen Kompositionen könnennach üblichen, an sich bekannten Methoden hergestellt werden.
Für therapeutische Zwecke wird aus der erfindungsgemäß hergestellten Verbindung eine tägliche" Humandosis von 25 - 75 kg vorgesehen.
Die Erfindung wird an Hand der nachstehenden Beispiele näher erläutert ohne den Schutzumfang auf diese Beispiele einzuschränken.

Seispiel 1
l-( 4-Aminophenyl )-4-methyl-7 ,8-dimethoxy-5H!- -2,3-benzodiazeρin
Eine Suspension von 26,6 g (0,078 M) l-(4-Nitrophenyl)-4- -methyl-7,8-dimethoxy-5H_-2,3-benzodiazepin in 540 ml Dimethylformamid wird mit einer Suspension von 2 g 10 %igem Palladium-Aktivkohle-Katalysator in 60 ml Dimethylformamid versetzt, und das Gemisch wird in einer Wasserstoffatmosphäre bei Raumtemperatur gerührt. Die Reduktion ist innerhalb von ungefähr 15 Stunden vollstäadig durchgeführt. Dann wird der Katalysator filtriert, das Filtrat mit Aktivkohle behandelt und im Vakuum eingedampft. Der Kristallrückstand wird eine Stunde lang mit 250 ml Äthanol unter Rückfluß erhitzt. Nach Abkühlen werden die Kristalle filtriert, zweimal mit je 30 ml Äthanol gewaschen und bei SO-IOO0C getrocknet. Ausbeute: 21,5 g (89 %), Schmp. 225-2270C. C18H19N3O2 = 309,374 Herstellung der Hydrochlorid-Salze:
Eine Lösung von 1 g Base in 5 ml Eisessig wird mit einer berechneten Menge von mit HCl-Gas gesättigtem absolutem Alkohol versetzt. Die ausgeschiedenen Kristalle werden filtriert und mit Äthylacetat gewaschen. Monohydrochlorid: ^"c^H^N^^/Cl = 345.839,

Berechnet: Cl 10,24 %. Gefunden: Cl 10,4 % Dihydrochlorid: Z-C13H21N3O2J7Cl2 = 382,304,

Herstellung des Sulfats:
Die wäßrige Lösung von 1 g Base wird mit der berechneten Menge von cc. Schwefelsäure versetzt, und. nach dem Eindampfen der Lösung wird der Rückstand mit heißem Isopropanoi verarbeitet. Das Rohprodukt wird aus Eisessig umkristalli-

Nach dem Verfahren des Beispiels 1 werden noch die Verbindungen der Beispiele 2 bis 7 hergestellt.

l-(4-Aminophenyl)-4-methyl-7,8-methylendioxy- -5J1-2 ,3-benzodiazepin ,
Eine Suspension von 1,52 g (0,0047 M) l-(4-Nitrophenyl)- -4-methyl-7,8-methylendioxy-5Hi-2,3-benzodiazepin in 45 ml Methanol wird mit 0,2 g. 10 %igem Palladium-Aktivkohle-Katalysator und 0,7 ml (0,014 M) 100 ^igem Hydrszinhydrat versetzt, das Reaktionsgemisch wird 6 Stunden lang bei Raumtemperatur gerührt und nachfolgend auf 50-60OC er-

hitzt. Sodann wird der Katalysator filtriert und dreimal mit je 10 ml Methanol gewaschen. Das Filtrat wird im Vakuum eingedampft, und der Rückstand aus 5 ml 99,5 5oigem Äthanol umkristallisiert. Ausbeute: 1,05 g (76 %), Schmp. 193-1999C.
Nach dem Verfahren des Beispiels 8 werden noch die Verbindungen der Beispiele 9 bis 12 hergestellt. Als Reaktionslösungsmittel werden Methanol, Äthanol oder Dioxan eingesetzt, und die Reduktion wird auch am Siedepunkt der Lösungsmittel durchgeführt.

l-(4-Aminophenyl)-4-methyl-5-äthyl-7, 8-dimethoxy-5Hi-2 ,3-benzodiazepin
Schmp. 166-1680C (aus dem Dihydrochlorid in wäßriger Lösuna freigesetzt).
l-(2-ChloΓ-4-aminophonyl)-4-methyl-7,8-dimethoxy-5HI-2,3-benzodiazepin
Ein Gemisch aus 4,10 g (0,011 M) l-(2-Chlor-4-nitro-phenyl)· ~4-methyl-7 ,8-dimethoxy-5H_-2,3-benzodiazepin, 100 ml Methanol, 0,4 g Raney-Nickel-Katalysator und 1,2 ml (0,025 M) 98 tigern Hydrazinhydrat wird eine Stunde lang gerührt. Das Reaktionsgemisch erwärmt sich erst auf 40-45°C, dann kühlt es sich wieder auf Raumtemperatur ab. Nach Vollendung der Reduktion wird das Gemisch auf 40-45°C erwärmt, der Katalysator wird filtriert und zweimal mit je 10 ml Chloroform gewaschen, dann wird das Filtrat im Vakuum eingedampft. Der Rückstand wird 2 Stunden lang mit 20 ml Äthanol unter Rückfluß erhitzt: erst wird eine Lösung gebildet, dann setzt eine schnelle Kristallisation ein. Nach Abkühlen werden die Kristalle filtriert, dreimal mit je 3 ml Äthanol gewaschen und bei 60-1000C bis zum Erreichen des Konstantgewichtes getrocknet. Ausbeute: 3,3 g (87 %). Schmp. 218-22O°C (Zers.).
Nach dem Verfahren des Beispiels 13 werden noch die Verbindungen der Beispiele 14-18 hergestellt, mit dem Unterschied, daß an Stelle von Methanol Äthanol, Isopropanol oder Dioxan eingesetzt wird, und die Reduktion bei höherer Temperatur durchgeführt wird.

Beispiel 19 Herstellung von Tabletten
Tabletten, die als Wirkstoff 20 mg l-(4-Aminophenyl)-4- -emthyl-7 ,8-dimethoxy-5H-2,3-benzodiazepin enthalten, können mit üblichen, an sich bekannten Methoden hergestellt werden. Ihre Zusammensetzung ist folgende:
Wirkstoff 20,0 mg
- Lactose 122,0 mg
Maisstärke . 20,5 mg Cellulose (mikrokristallin) 10,0 mg
Gelatin 3,5 mg
Talcum 2,0 mg
Stearin 1,0mg
Magnesium-stearat 1,0 mg

Claims (3)
1. Verfahren zur Herstellung von neuen l-Aryl-5H_-2f3~benzodiazepin-Derivaten der allgemeinen Formel (I), worin
R und R, unabhängig voneinander ein Wasserstoffatom oder Chloratom, eine Alkylgruppe mit 1 bis 4 C-Atomen oder eine Alkoxygruppe mit 1 bis 4 C-Atomen,
R2 ein Wasserstoffatom oder eine Alkylgruppe mit 1 bis 4 C-Atomen,
R^ und R, eine Alkylgruppe mit 1 bis 4 C-Atomen und R3 und R4 gemeinsam eine Methylengruppe bedeuten und
von Säureadditionssalzen derselben, gekennzeichnet dadurch, daß eine Nitro-Verbindung der allgemeinen Formel (II), worin R, R1, Rn, R-, und R, die obioe Bedeutuna haben, in einem organischen Lösungsmittel reduziert wird, woraufhin gewünschtenfalls aus der erhaltenen Base der allgemeinen Formel (I) ein Säureadditionssalz gebildet wird und, sofern weiterhin gewünscht, dann die freie Base aus ihrem Salz nach einer bekannten Methode freigesetzt und mittels einer Säure in ein anderes Saureadditionsslz überführt wird.
2. Verfahren nach Anspruch 1, gekennzeichnet dadurch, daß man l-(4-Aminophenyl)-4-methyl-7,8-dimethoxy-5Hi-2,3-benzodiazepin, '
l-(3-Aminophenyl)-4-methy1-7,8-dimethoxy-5Hi-2,3-benzodiazepin,
l-(4-Aminophenyl)-4-methyl-7 >8-methylendioxy-5H>-2,3- -benzodiazepin, ' '
