Description
- 1. Menschliches Gewebe, beispielsweise Milzgewebe oder periphere Blutlymphozyten werden mit Mitogenen gezüchtet, um die Bildung von Immuninterferon zu stimu lieren.
- 2. Zellpellets aus diesen Zellkulturen wurden in Gegenwart von Ribonuclease-Inhibitor extrahiert, um sämtliche zytoplasmatische RNA zu isolieren.
- 3. Eine Oligo-dT-Säule isolierte die gesamte meßenger-RNA (mRNA) in polyadenylierter Form. Diese mRNA wurde unter Verwendung eines Saccharose-Dichtegradienten und der Säure-Harnstoff-Gelelektrophorese einer Größenfraktionierung unterworfen.
- 4. Die entsprechende mRNA (12 bis 18 S) wurde in die entsprechende einzelsträngige komplementäre DNA (cDNA) umgewandelt, aus der doppelsträngige cDNA gebildet wurde. Nach poly-dC-tailing wurde sie in einen Vektor, z. B. ein Plasmid mit einem oder mehreren phänotypischen Markern, eingesetzt.
- 5. Die auf diese Weise hergestellten Vektoren wurden zur Transformation von Bakterienzellen unter Bildung einer Koloniebibliothek verwendet. Sowohl aus induzierter als auch aus nicht-induzierter mRNA (auf die vorstehende Weise erhalten) hergestellte radioaktiv markierte cDNA wurde zur getrennten Ermittlung von doppelten Koloniebibliotheken verwendet. Der Überschuß an cDNA wurde sodann entfernt. Die Kolonien wurden auf Röntgenfilme aufgebracht, um die induzierten cDNA-Klone zu identifizieren.
- 6. Von den induzierten cDNA-Klonen wurde die entsprechende Plasmid-DNA isoliert und sequenziert.
- 7. In einer ersten Ausführungsform wurde die sequenzierte DNA sodann in vitro zur Einfügung in einen entsprechenden Expressionsträger zugeschnitten, der zur Transformierung einer E. coli-Wirtszelle verwendet wurde, die wiederum in einem Medium gezüchtet wurde und die Expression des gewünschten Human-Immuninterferonprodukts durchführte.
- 8. Das auf diese Weise experimierte Human-Immuninterferon weist in seiner reifen Form zweifelsfrei 146 Aminosäuren auf, wobei am Beginn Cystein steht, und besitzt einen sehr stark basischen Charakter. Sein monomeres Molekulargewicht wurde mit 17 140 berechnet. Das Molekül kann sich zu oligomeren Formen, z. B. zur dimeren, trimeren oder tetrameren Form, assoziieren, was möglicherweise auf die Anwesenheit von zahlreichen basischen Resten, hydrophobe Eigenschaften, Salzbrückenbildung und dergleichen zurückzuführen ist. Die hohen Molekulargewichte, die früher bei natürlichem Material festgestellt wurden (6), die auf der Basis der Aminosäuresequenz allein nicht erklärbar sind, mögen auf derartige oligomeren Formen sowie auf den Kohlenhydratbeitrag aufgrund von Glycoxylierung nach der Translation zurückzuführen sein.
- 9. In bestimmten Wirtszellsystemen, insbesondere bei Ligatur in einem Expressionsträger, so daß eine Expression zusammen mit dem Signalpeptid stattfindet, wird die reife Form von Human-Immuninterferon in das Zellkulturmedium ausgebracht, was eine unermeßliche Hilfe bei Gewinnungs- und Reinigungsverfahren bedeutet.
JA300 (thr leuB6 thi thyA trpC1117 hsdm- hsdR- strR (20)
wurde zur Selektierung von Plasmiden mit einem Gehalt an funktionellem TRPI-Gen verwendet. Der Hefestamm RH2 18 mit dem Genotyp (trp1 ga12 SUC2 mal CUPI) (18) wurde als Hefetransformationswirt verwendet. RH2 18 ist ohne Einschränkungen bei der American Type Culture Collection unter der Nr. ATCC 44076 hinterlegt. M9 (Minimalmedium) mit 0,25 Prozent Casaminsäuren (CAA) und LB (rich medium) entsprechen den Angaben von Miller (58), wobei die Medien nach Autoklavisierung und Kühlung mit 20 µg/ml Ampicillin versetzt wurden. Hefe wurde auf folgenden Medien gezüchtet: YEPD mit einem Gehalt an 1 Prozent Hefeextrakt, 2 Prozent Pepton und 2 Prozent Glucose± 3 Prozent Difco-Agar. YNB+CAA enthielt 6,7 g Hefe-Stickstoffbase (ohne Aminosäuren (YNB), 10 mg Adenin, 10 mg Uracil, 5 g CAA, 20 g Glucose und±30 g Agar pro Liter.
- 1. Extraktion der Zellen in einem Lysepuffer (etwa pH 8) von hoher Leitfähigkeit mittels Passage durch einen Homogenisator bei hohem Druck, Abkühlen des Abstroms in einem Eisbad.
- 2. Ausfällung der DNA durch Zugabe von Polyethylenimin unter Rühren z. B. bei 4°C.
- 3. pH-Ausfällung der bakteriellen Proteine, wobei wiederum IFN-γ in Lösung bleibt.
- 4. Abzentrifugieren der Feststoffe bei 4°C.
- 5. Konzentrieren des Überstands (nach erneuter Einstellung des pH) z. B. durch Ultrafiltration.
- 6. Dialyse des Konzentrats gegen einen Puffer von niedriger Leitfähigkeit.
- 7. Abzentrifugieren der Feststoffe, wobei IFN-γ in Lösung bleibt.
- 8. Ionenaustauschchromatographie an Carboxymethylcellulose, Elution mit einem Gradienten an zunehmender Ionenstärke.
- 9. Chromatographie an Calciumphosphat-Gel und Elution mit einem Gradienten an zunehmender Ionenstärke.
- 10. Ionenaustauschchromatographie an Carboxymethylcellulose unter schwach denaturierenden Bedingungen und Elution mit einem Gradienten an zunehmender Ionenstärke.
