Claims (43)
1. Способ борьбы с заражением растения видами рода Leptinotarsa, включающий:
(а) контакт вышеуказанных видов рода Leptinotarsa с полинуклеотидом, содержащим нуклеотидную последовательность, которая является комплементарной по меньшей мере 21 смежному нуклеотиду целевого гена, имеющего нуклеотидную последовательность, выбранную из группы, состоящей из SEQ ID №:730, SEQ ID №:807, SEQ ID №:1-725, SEQ ID №:726-729, SEQ ID №:731-806, SEQ ID №:808-830 и SEQ ID №:1087-1094, или РНК, транскрибируемой с указанного целевого гена; или
(б) обеспечение в рационе питания вышеуказанных видов рода Leptinotarsa полинуклеотида, содержащего нуклеотидную последовательность, которая является комплементарной по меньшей мере 21 смежному нуклеотиду целевого гена, имеющего нуклеотидную последовательность, выбранную из группы, состоящей из SEQ ID №:730, SEQ ID №: 807, SEQ ID №:1-725, SEQ ID №:726-729, SEQ ID №:731-806, SEQ ID №:808-830 и SEQ ID №:1087-1094, или РНК, транскрибируемой с указанного целевого гена; или
(в) вызывание смертности или задержки роста у личинок вышеуказанных видов рода Leptinotarsa путем обеспечения рациона питания указанных личинок по меньшей мере одним полинуклеотидом, содержащим по меньшей мере один элемент сайленсинга, содержащий 21 смежный нуклеотид, комплементарный целевому гену, имеющему нуклеотидную последовательность, выбранную из группы, состоящей из SEQ ID №:730, SEQ ID №:807, SEQ ID №:1-725, SEQ ID №:726-729, SEQ ID №:731-806, SEQ ID №:808-830 и SEQ ID №:1087-1094, или РНК, транскрибируемой с указанного целевого гена; или
(г) поверхностное применение к вышеуказанному растению композиции, содержащей по меньшей мере один полинуклеотид, содержащий нуклеотидную последовательность, которая является комплементарной по меньшей мере 21 смежному нуклеотиду целевого гена, имеющего нуклеотидную последовательность, выбранную из группы, состоящей из SEQ ID №:730, SEQ ID №:807, SEQ ID №:1-725, SEQ ID №:726-729, SEQ ID №:731-806, SEQ ID №:808-830 и SEQ ID №:1087-1094, или РНК, транскрибируемой с указанного целевого гена; или
(д) поверхностное применение к вышеуказанному растению композиции, содержащей по меньшей мере один полинуклеотид, таким образом, что эффективное количество указанного полинуклеотида проглатывается видами рода Leptinotarsa, питающимися на указанном растении; вышеуказанный полинуклеотид содержит по меньшей мере 21 смежный нуклеотид, комплементарный целевому гену, имеющему нуклеотидную последовательность, выбранную из группы, состоящей из SEQ ID №:730, SEQ ID №:807, SEQ ID №:1-725, SEQ ID №:726-729, SEQ ID №:731-806, SEQ ID №:808-830 и SEQ ID №:1087-1094, или РНК, транскрибируемой с указанного целевого гена; или
(е) экспрессию в вышеуказанном растении по меньшей мере одного полинуклеотида, содержащего по меньшей мере один сегмент, который является идентичным или комплементарным по меньшей мере 21 смежному нуклеотиду ДНК, имеющей последовательность, выбранную из группы, состоящей из SEQ ID №:730, SEQ ID №:807, SEQ ID №:1-725, SEQ ID №:726-729, SEQ ID №:731-806, SEQ ID №:808-830 и SEQ ID №:1087-1094; или
(ж) обеспечение вышеуказанного растения по меньшей мере одним полинуклеотидом, содержащим по меньшей мере один сегмент, который является идентичным или комплементарным по меньшей мере 21 смежному нуклеотиду целевого гена или РНК, транскрибируемой с указанного целевого гена, причем указанный целевой ген выбран из группы, состоящей из генов, идентифицированных в группе последовательностей целевого гена, или РНК, транскрибируемой с указанного целевого гена; или
(з) контакт вышеуказанных видов рода Leptinotarsa с эффективным количеством двухцепочечной РНК, одна цепь которой комплементарна по меньшей мере 21 смежному нуклеотиду гена, который кодирует рибосомный белок, при этом индуцируется РНК-интерференция и наблюдается смертность; или
(и) контакт вышеуказанных видов рода Leptinotarsa с полинуклеотидом, содержащим по меньшей мере один сегмент, идентичный или комплементарный по меньшей мере 21 смежному нуклеотиду целевого гена, выбранного из группы, состоящей из генов, идентифицированных в группе последовательностей целевого гена, или РНК, транскрибируемой с указанного целевого гена.
2. Способ по п. 1, отличающийся тем, что вышеуказанный полинуклеотид представляет собой двухцепочечную РНК.
3. Способ по п. 2, отличающийся тем, что вышеуказанная двухцепочечная РНК является химически синтезированной, или полученной путем экспрессии в микроорганизме, или путем экспрессии в клетке растения.
4. Способ по п. 2, отличающийся тем, что вышеуказанная двухцепочечная РНК содержит цепь, содержащую последовательность, выбранную из группы, состоящей из SEQ ID №:989, 1049, 831, 842, 849, 898, 910, 925, 928, 931, 932, 937, 938, 940, 941, 942, 943, 944, 945, 947, 948, 949, 950, 951, 952, 955, 956, 957, 958, 960, 961, 964, 966, 967, 968, 969, 970, 971, 973, 976, 978, 979, 982, 983, 985, 987, 988, 991, 992, 994, 995, 996, 997, 999, 1006, 1007, 1008, 1009, 1010, 1013, 1018, 1019, 1020, 1022, 1025, 1029, 1030, 1033, 1035, 1036, 1037, 1038, 1039, 1040, 1041, 1042, 1043, 1045, 1046, 1047, 1050, 1053, 1054, 1058, 1060, 1061, 1064, 1065, 1066, 1067, 1068, 1070, 1073, 1074, 1075, 1077, 1078, 1080, 1081, 1082, 1084, 1085, 1095, 1096, 1097, 1098, 1099, 1100, 1101, 1102, 1103, 1104, 1105, 1110, 1111, 1112, 1113 и 1114.

Способ по п. 1, отличающийся тем, что указанный способ включает поверхностное применение к указанному растению композиции, содержащей по меньшей мере один полинуклеотид, содержащий нуклеотидную последовательность, которая является комплементарной по меньшей мере 21 смежному нуклеотиду целевого гена или РНК, транскрибируемой с указанного целевого гена, причем указанный целевой ген имеет нуклеотидную последовательность, выбранную из группы, состоящей из SEQ ID №:730, SEQ ID №:807, SEQ ID №:1-725, SEQ ID №:726-729, SEQ ID №:731-806, SEQ ID №:808-830 и SEQ ID №:1087-1094; и причем указанная композиция дополнительно содержит один или более компонентов, выбранных из группы, состоящей из агента-носителя, поверхностно-активного вещества, катионного липида, кремнийорганического материала, кремнийорганического поверхностно-активного вещества, полинуклеотидной гербицидной молекулы, неполинуклеотидной гербицидной молекулы, неполинуклеотидного пестицида, антидота и регулятора роста насекомых.

Способ по п. 1, отличающийся тем, что указанный способ включает контакт вышеуказанных видов рода Leptinotarsa с эффективным количеством раствора, содержащего двухцепочечную РНК, причем по меньшей мере одна цепь двухцепочечной РНК является комплементарной по меньшей мере 21 смежному нуклеотиду гена, который кодирует рибосомный белок, или РНК, транскрибируемой с указанного гена, причем указанный вид рода Leptinotarsa представляет собой Leptinotarsa decemlineata, в котором индуцируется РНК-интерференция и наблюдается смертность Leptinotarsa decemlineata, и причем указанный рибосомный белок представляет собой рибосомный белок L7, или белок, который кодируется SEQ ID №:730, или в котором указанная двухцепочечная РНК содержит последовательность, выбранную из группы, состоящей из SEQ ID №:989, 988, 1104 или 1105.
7. Способ по п. 6, отличающийся тем, что вышеуказанный раствор дополнительно содержит один или более компонентов, выбранных из группы, состоящей из кремнийорганического поверхностно-активного вещества или катионного липида.
8. Способ по п. 1, отличающийся тем, что указанный способ включает поверхностное применение к вышеуказанному растению композиции, содержащей по меньшей мере один полинуклеотид, таким образом, что эффективное количество указанного полинуклеотида проглатывается видами рода Leptinotarsa, питающимися на указанном растении, причем вышеуказанный полинуклеотид содержит нуклеотидную последовательность, комплементарную по меньшей мере 21 смежному нуклеотиду целевого гена, имеющего нуклеотидную последовательность, выбранную из группы, состоящей из SEQ ID №:730, SEQ ID №:807, SEQ ID №:1-725, SEQ ID №:726-729, SEQ ID №:731-806, SEQ ID №:808-830 и SEQ ID №:1087-1094, или РНК, транскрибируемой с указанного целевого гена; причем указанный вид рода Leptinotarsa представляет собой Leptinotarsa decemlineata; и причем указанный целевой ген имеет последовательность SEQ ID №:730, или отличающийся тем, что указанный полинуклеотид представляет собой двухцепочечную РНК, имеющую цепь с последовательностью, выбранной из группы, состоящей из SEQ ID №:989, 988, 1104 или 1105.
9. Способ по п. 1, отличающийся тем, что вышеуказанный вид рода Leptinotarsa выбран из группы, состоящей из Leptinotarsa behrensi, Leptinotarsa collinsi, Leptinotarsa decemlineata (колорадского жука), Leptinotarsa defecta, Leptinotarsa haldemani (зеленого картофельного жука), Leptinotarsa heydeni, Leptinotarsa juncta (ложного картофельного жука), Leptinotarsa lineolata, Leptinotarsa peninsularis, Leptinotarsa rubiginosa, Leptinotarsa texana, Leptinotarsa tlascalana, Leptinotarsa tumamoca и Leptinotarsa typographica.
10. Растение, имеющее повышенную устойчивость к заражению видами рода Leptinotarsa, представленное способом по п. 1, или плод, семя или пропагативная часть указанного растения.
11. Растение по п. 10, отличающееся тем, что указанное растение выбрано из группы, состоящей из картофеля, томата и баклажана.
12. Инсектицидная композиция для борьбы с видами рода Leptinotarsa, включающая:
(а) эффективное для инсектицидного действия количество полинуклеотида, содержащего по меньшей мере 21 смежный нуклеотид, комплементарный целевому гену, имеющему нуклеотидную последовательность, выбранную из группы, состоящей из SEQ ID №:730, SEQ ID №:807, SEQ ID №:1-725, SEQ ID №:726-729, SEQ ID №:731-806, SEQ ID №:808-830 и SEQ ID №:1087-1094, или РНК, транскрибируемой с указанного целевого гена; или
(б) эффективное для инсектицидного действия количество по меньшей мере одного полинуклеотида, содержащего по меньшей мере один элемент сайленсинга, который является комплементарным по меньшей мере 21 смежному нуклеотиду целевого гена или РНК, транскрибируемой с указанного целевого гена, причем указанный целевой ген имеет нуклеотидную последовательность, выбранную из группы, состоящей из SEQ ID №:730, SEQ ID №:807, SEQ ID №:1-725, SEQ ID №:726-729, SEQ ID №:731-806, SEQ ID №:808-830 и SEQ ID №:1087-1094; или
(в) эффективное для инсектицидного действия количество по меньшей мере одной РНК, содержащей по меньшей мере один сегмент, который является идентичным или комплементарным по меньшей мере 21 смежному нуклеотиду целевого гена, имеющего нуклеотидную последовательность, выбранную из группы, состоящей из SEQ ID №:730, SEQ ID №:807, SEQ ID №:1-725, SEQ ID №:726-729, SEQ ID №:731-806, SEQ ID №:808-830 и SEQ ID №:1087-1094, или РНК, транскрибируемой с указанного целевого гена; или
(г) молекулу РНК, которая вызывает смертность или задержку роста у видов рода Leptinotarsa при поглощении или контакте с указанными видами рода Leptinotarsa, причем указанная молекула РНК содержит по меньшей мере 21 смежный нуклеотид, который является комплементарным целевому гену, имеющему нуклеотидную последовательность, выбранную из группы, состоящей из SEQ ID №:730, SEQ ID №:807, SEQ ID №:1-725, SEQ ID №:726-729, SEQ ID №:731-806, SEQ ID №:808-830 и SEQ ID №:1087-1094, или РНК, транскрибируемой с указанного целевого гена; или
(д) инсектицидную двухцепочечную молекулу РНК, которая вызывает смертность или задержку роста у видов рода Leptinotarsa при поглощении или контакте с указанными видами рода Leptinotarsa, причем по меньшей мере одна нить указанной инсектицидной двухцепочечной молекулы РНК состоит из 21 смежного нуклеотида, который является комплементарным целевому гену или РНК, транскрибируемой с указанного целевого гена, причем указанный целевой ген имеет последовательность, выбранную из группы, состоящей из SEQ ID №:730, SEQ ID №:807, SEQ ID №:1-725, SEQ ID №S: 726-729, SEQ ID №:731-806, SEQ ID №:808-830 и SEQ ID №:1087-1094; или
(е) эффективное для инсектицидного действия количество по меньшей мере одной двухцепочечной РНК, содержащей последовательность, выбранную из группы, состоящей из SEQ ID №:989, 1049, 831, 842, 849, 898, 910, 925, 928, 931, 932, 937, 938, 940, 941, 942, 943, 944, 945, 947, 948, 949, 950, 951, 952, 955, 956, 957, 958, 960, 961, 964, 966, 967, 968, 969, 970, 971, 973, 976, 978, 979, 982, 983, 985, 987, 988, 991, 992, 994, 995, 996, 997, 999, 1006, 1007, 1008, 1009, 1010, 1013, 1018, 1019, 1020, 1022, 1025, 1029, 1030, 1033, 1035, 1036, 1037, 1038, 1039, 1040, 1041, 1042, 1043, 1045, 1046, 1047, 1050, 1053, 1054, 1058, 1060, 1061, 1064, 1065, 1066, 1067, 1068, 1070, 1073, 1074, 1075, 1077, 1078, 1080, 1081, 1082, 1084, 1085, 1095, 1096, 1097, 1098, 1099, 1100, 1101, 1102, 1103, 1104, 1105, 1110, 1111, 1112, 1113 и 1114.

Инсектицидная композиция по п. 12, отличающаяся тем, что вышеуказанная инсектицидная композиция находится по меньшей мере в одной форме, выбранной из группы, состоящей из твердого вещества, жидкости, порошка, суспензии, эмульсии, спрея, инкапсулированного препарата, микрогранул, частиц носителей, пленки, матрикса, препарата для обработки семян, препарата для пропитывания почвы, имплантируемого препарата или препарата для борозды.

Инсектицидная композиция по п. 12, дополнительно содержащая по меньшей мере один компонент, выбранный из группы, состоящей из агента-носителя, поверхностно-активного вещества, катионного липида, кремнийорганического материала, кремнийорганического поверхностно-активного вещества, полинуклеотидной гербицидной молекулы, неполинуклеотидной гербицидной молекулы, неполинуклеотидного пестицида, антидота и регулятора роста насекомых.

Инсектицидная композиция по п. 12, отличающаяся тем, что вышеуказанная инсектицидная композиция содержит инсектицидную двухцепочечную молекулу РНК, которая вызывает смертность или задержку роста у видов рода Leptinotarsa при поглощении или контакте с указанными видами рода Leptinotarsa, причем указанная инсектицидная двухцепочечная молекула РНК содержит по меньшей мере один сегмент, который является комплементарным 21 смежному нуклеотиду ДНК, имеющей последовательность, выбранную из группы, состоящей из SEQ ID №:730, SEQ ID №:807, SEQ ID №:1-725, SEQ ID №:726-729, SEQ ID №:731-806, SEQ ID №:808-830 и SEQ ID №:1087-1094, или РНК, транскрибируемой с указанной ДНК, и причем указанная двухцепочечная молекула РНК имеет длину по меньшей мере 50 пар оснований или длину в пределах от около 100 до около 500 пар оснований.

(б) ДНК, содержащей 21 или более смежных нуклеотидов, имеющих 100% идентичность с фрагментом ДНК эквивалентной длины, имеющей последовательность, выбранную из группы, состоящей из SEQ ID №:730, SEQ ID №:807, SEQ ID №:1-725, SEQ ID №:726-729, SEQ ID №:731-806, SEQ ID №:808-830 и SEQ ID №:1087-1094, или комплементарной им ДНК; или
(в) ДНК, кодирующей по меньшей мере один элемент сайленсинга, который является комплементарным по меньшей мере 21 смежному нуклеотиду целевого гена или РНК, транскрибируемой с указанного целевого гена, причем указанный целевой ген имеет последовательность, выбранную из группы, состоящей из SEQ ID №:730, SEQ ID №:807, SEQ ID №:1-725, SEQ ID №:726-729, SEQ ID №:731-806, SEQ ID №:808-830 и SEQ ID №:1087-1094; или
(г) ДНК, кодирующей по меньшей мере один элемент сайленсинга, содержащий по меньшей мере 21 смежный нуклеотид, который является комплементарным целевому гену, выбранному из группы, состоящей из группы последовательностей целевого гена, или РНК, транскрибируемой с указанного целевого гена; или

Трансгенная клетка пасленового растения по п. 18, отличающаяся тем, что вышеуказанная трансгенная клетка пасленового растения дополнительно имеет в своем геноме ДНК, кодирующую по меньшей мере один пестицидный агент, выбранный из группы, состоящей из пататина, растительного лектина, фитоэкдистероида, инсектицидного белка Bacillus thuringiensis, инсектицидного белка Xenorhabdus, инсектицидного белка Photorhabdus, инсектицидного белка Bacillus laterosporous и инсектицидного белка Bacillus sphaericus.
20. Трансгенное пасленовое растение, содержащее трансгенную клетку пасленового растения по п. 18, или плод, семя или пропагативную часть указанного трансгенного пасленового растения.
