Korte aanduiding: Werkwijze voor de bereiding van microcapsules.
De uitvinding heeft betrekking op een werkwijze voor de bereiding van microcapsules, die een actief middel bevatten. Meer in het bijzonder 5 heeft de uitvinding betrekking op een werkwijze voor het bereiden van micro-capsules, die een biologisch actief middel bevatten.
Er zijn een aantal werkwijzen bekend, waarbij een bepaalde verbinding kan worden omkapseld in de vorm van microcapsules. Bij deze werkwijzen wordt het materiaal dat moet worden omkapseld in het algemeen gedispergeerd 10 in een oplosmiddel dat een wandvormend materiaal bevat. Door een enkele be-werkingstrap wordt het oplosmiddel verwijderd uit de microcapsules én daarna wordt het produkt bestaande uit microcapsules verkregen. Een voorbeeld van een bekende werkwijze voor het verkrijgen van een micro-omkapseling is beschreven in het Amerikaanse octrooischrift 3.737.337, waarbij een oplossing 15 van een wand- of schilvormend polymeer materiaal in een oplosmiddel wordt bereid. Een vast materiaal of kernmateriaal wordt opgelost of gedispergeerd in de polymeerhoudende oplossing en daarna in een enkele trap wordt de kernmateriaal bevattende oplossing gedispergeerd in een waterige vloeistof die niet mengbaar is met het organisch oplosmiddel teneinde het oplos-20 middel te verwijderen uit dé microcapsules.
Een andere werkwijze waarbij oplosmiddel wordt verwijderd uit micro-capsules die een stof bevatten, volgens een enkele trapsbewerking, is vermeld in het Amerikaanse octrooischrift 3.523.906. Bij deze werkwijze wordt een materiaal, dat moet worden omkapseld geëmulgeerd in een oplossing van een 25 polymeer materiaal in een oplosmiddel, dat niet mengbaar is met water en vervolgens wordt de emulsie geëmulgeerd in een waterige oplossing die een hydrofiel colloid bevat. Het verwijderen van het oplosmiddel uit de microcapsules zoals vervolgens bewerkstelligd in een enkele trap door verdamping en zo wordt het produkt verkregen. Volgens een andere werkwijze, zoals vermeld in 30 het Amerikaanse octrooischrift 3.691.090 wordt een organisch oplosmiddel verdampt uit een dispersie van microcapsules in een waterig medium in een enkele trap, bij voorkeur onder verlaagde druk. Op dezelfde wijze geeft het Amerikaanse octrooischrift 3.891.570 een werkwijze weer, waarbij een oplosmiddel wordt afgedampt uit een dispersie van microcapsules in een '-35 meerwaardige alcohol uit de microcapsules door toepassing van warmte of door de microcapsules onder verlaagde druk te brengen. Een andere werkwijze volgens een één-traps procédé ter verwijdering van het oplosmiddel is vermeld in het Amerikaanse octrooischrift 3-960.757.

Het probleem met betrekking tot de conventionele omkapseling is dat speciaal wannéér deze worden gebruikt om een farmaceutisch middel te voorzien van een micro-omkapseling, zoals progesteron of norgestimaat, de hoeveelheid toe te passen geneesmiddel een beperkende factor is. In af-5 hankelijkhéid van de fysische en chemische eigenschappen van de drager en het middel dat moet worden omkapsel^ hebben pogingen ter bereiding van micro-capsules met een te hoge- belasting geresulteerd in een onvolledige omkapseling, waardoor kristallen van het te omkapselen materiaal uitsteken uit de mieroeapsules of terechtkomen in het procesmedium. Daarom is het verder 10 noodzakelijk gebleken om een werkwijze te vinden ter bereiding van micro-deeltjes met verbeterde eigenschappen en van een betere kwaliteit.
Zodoende is een van de doelstellingen volgens de uitvinding het verkrijgen van een werkwijze ter bereiding van mieroeapsules, die een actief middel bevatten en bestaan uit een verbeterd materiaal, waardoor de capsules meer 15 middel kunnen bevatten en van een betere kwaliteit zijn.
Deze doelstelling lean worden bewerkstelligd door de uitvoering van een werkwijze volgens de uitvinding voor de bereiding van mieroeapsules en deze wordt hierdoor gekenmerkt, dat a) een actief middel wordt opgelost of gedispergeerd in een oplos-20 middel en een wandvormend materiaal wordt opgelost in het oplosmiddel, b) het oplosmiddel met het actieve middel en het wandvormend middel worden gedispergeerd in een continue fase-bewerkingsmedium, c) esn deel van het oplosmiddel wordt verdampt uit de dispersie van trap b) waarbij mieroeapsules worden gevormd met het actieve middel in de 25suspensie, en d) de rest van het oplosmiddel wordt uit de mieroeapsules geëxtraheerd .
Een van de belangrijke aspecten volgens de onderhavige werkwijze ter verkrijging van mieroeapsules met verbeterde eigenschappen berust op 30 het feit, dat tijdens de bereiding oplosmiddel wordt verwijderd uit de micro-capsules die zijn gesuspendeerd in een vloeibaar medium in twee afzonderlijke trappen in plaats van in een enkele bewerkingstrap. De techniek om het oplosmiddel te verwijderen in twee trappen resulteert in een microcapsule-produkt van een verbeterde kwaliteit en bevat een hoger gehalte aan actieve 35stof. Het voordeel volgens de uitvinding ten opzichte van bekende procédés is, dat de onderhavige techniek waarbij het oplosmiddel in twee trappen wordt verwijderd, resulteert in een microcapsule-produkt met een verrassend hoge hoeveelheid actieve stof en van een verrassend goede kwaliteit in tegenstel- 8104507 ** ' * -3- 22062/Vk/ts ling tot de conventionele microcapsule-produkten, bereid door een werkwijze waarbij het oplosmiddel wordt verwijderd in een enkelvoudige stap.
Een stof, die wordt verwerkt in de microcapsules wordt opgelost in een oplosmiddel. Geschikte actieve stoffen, die kunnen worden verwerkt in 5 de microcapsules omvatten landbouwkundige chemicaliën, parfums, hardingsmiddelen, kleurstoffen, oxydatiemiddelen en biologisch actieve stoffen.
De hoeveelheid actieve stof, opgelost of gedispergeerd in het oplosmiddel is met name niet kritisch, hoewel de verhouding actieve stof tot wandvor-mend materiaal van belang is in zoverre, dat bij een te hoge verhouding van 10 de hoeveelheid actieve stof tot wandvormend materiaal de microcapsules niet zullen worden gevormd. Met name kan de hoeveelheid actief middel, gecombi -neerd met wandvormend materiaal variëren tot ongeveer 80 gew. delen actieve stof tot 20 gew. delen wandvormend materiaal. Er is geen beperking met betrekking tot de ondergrens, waarbij actieve stof kan worden gecombineerd 15 met wandvormend materiaal, behalve dat bij een zeer lage belasting aan actieve stof in de microcapsules, de microcapsules in de praktijk niet voldoen.
Het zal echter duidelijk zijn, dat hoe verdunder de oplossing is, hoe groter de hoeveelheid oplosmiddel is die daarin aanwezig is en die dan eventueel moet worden verwijderd ter vorming van het microcapsule-produkt met een 20 bepaald gehalte aan actiele stof. Gewoonlijk wordt 5 tot 30 gew. % actieve stof opgelost of gesuspendeerd in het oplosmiddel.
Het oplosmiddel, dat wordt toegepast voor de bereiding van de oplossing die actieve stof bevat, kan worden gekozen uit een groot aantal organische oplosmiddelen met name gehalogeneerde alifatische koolwaterstoffen, 25 zoals chloroform, methyleenchloride, methylchloroform en dergelijke aromatische koolwaterstoffen, gehalogeneerde aromatische koolwaterstoffen, cyclische ethers zoals tetrahydrofuran en dergelijke,alcoholen, water, aceton en dergelijke. Het oplosmiddel moet een stof zijn waarin het wandmateriaal makkelijk oplost, het oplosmiddel moet chemisch inert zijn ten aanzien van 30 het actieve middel, dat niet mengbaar moet zijn met het bewerkingsmedium met name de continue fase hiervan en moet een kookpunt hebben beneden dat van het continue fase-bewerkingsmedium. De hoeveelheid wandmateriaal dat moet worden opgelost in het oplosmiddel is afhankelijk van de oplosbaarheid van het polymeer in het oplosmiddel en de verkregen viscositeit van de polymere 35 oplossing. De viscositeit hangt samen met de uiteindelijke grootte van de microcapsules. Mengsels van bovenvermelde oplosmiddelen kunnen ook worden gebruikt als geschikt oplosmiddel voor de actieve stof. Aan de oplossing die het actieve middel bevat wordt het wandvormen-de materiaal toegevoegd dat het actieve middel omgeeft en opsluit in de 5 microcapsules. De hoeveelheid wandvormend materiaal die wordt toegevoegd aan de oplossing is zodanig, dat de microcapsules makkelijk zullen worden gevormd wanneer de oplossing wordt gedispergeerd in de continue fase van het bewerkingsmedium. Voordat de dispersie van het oplosmiddel in het continue fase-bewerkingsmedium wordt bewerkstelligd, moeten zowel de actieve 10stof als het wandvormende materiaal aanwezig zijn in het oplosmiddel. Meestal bedraagt de hoeveelheid wandvormend materiaal dat wordt opgelost in het oplosmiddel 5- 35 gew. %.
Geschikte wandvormende stoffen omvatten polystyreen, ethylcellulo-se, celluloseacetaat, hydroxypropylmethylcelluloseftalaat, dibjitylaminohydro-15 xypropylether, polyvinylbutyral, polyvinylformal, poly('meth)acrylzure ester, polyvinylacetaal-diethylamino-acetaat, 2-methyl-5-vinylpyridinemethacrylaat-methacrylzuurcopolymeer,?oiycarbonaat, polyesters,polypropyleen, vinylchlo- ride-vinylacetaatcopolymeer»polysaccharide en glyceroldistearaat. Een bij voorkeur toegepaste groep- van polymere, wandvormende stoffen omvatten die 20 stoffen, die bio-afbreekbaar zijn zodals alifatische polyesters met inbegrip, van polylactide, polyglycolide, polycaprolacton en copolymeren hiervan.
Na de bereiding van de oplosmiddelfase, die de actieve stof bevat en het wandvormende materiaal, wofden microdruppels gevormd van het wandvormende materiaal met de actieve stof door de oplossing met de actieve 25 stof en het wandvormende materiaal te dispergeren in een continue fase-, bewerkingsmedium. Het continue fase-bewerkingsmedium moet niet mengbaar zijn met het oplosmiddel en een kookpunt hebben, dat hoger is dan van het oplosmiddel. Hoewel als bewerkingsmedium in veel gevallen water kan worden gebruikt, zijn andere voorbeelden van niet-waterige continue fase-bewerkings-30 media, organische vloeistoffen, zoals xyleen of tolueen, synthetische oliesoorten, zoals siliconolie en natuurlijke oliesoorten, zoals plantaardige olie met inbegrip van aardnotenolie. Gewoonlijk wordt een oppervlakte actief middel, emulgeermiddel, toegevoegd aan het continue fase-bewerkingsmedium o.m te voorkomen, dat de microcapsules samenklonteren en om de grootte van 35 de microdruppels van het oplosmiddel in de emulsie te regelen. Een goed oppervlakte actief middel dat kan worden toegepast wanneer water wordt gebruikt als continue fase-bewerkingsmedium, is poly(vinylalcohol) (PVA) bij een concentratie van 1-10 %. De dispersie kan worden gevormd door mechanisch 8104507 m -5- 22062/Vk/ts het continue i'ase-bewerkingsmediura te roeren met behulp van een roerorgaan zoals een colloldmolen, homogeniseerorgaan of dergelijke. Een emulsie kan ook worden gevormd door het toevoegen van kleine druppels van het actieve raiddel-wandvormende materiaal in oplossing aan het continue fase-bewerkings-5' medium. In een bij voorkeur toe te passen uitvoeringsvorm voor de dispersie wordt de oplossing van organisch oplosmiddel gedispergeerd in een waterige oplossing,die polyvinylalbohol bevat. De temperatuur tijdens de vorming van de emulsie is niet met name kritisch, maar kan van invloed zijn op de grootte en de kwaliteit van de microcapsules. Bovendien moet 10 in afhaneklijkbeid van het oplosmiddel en het continue fase-bewerkings-medium de temperatuur niet te laag zijn of het oplosmiddel en het bewer-kingsmedium zullen stollen of het bewerkingsmedium zal te viskeus worden voor praktische doeleinden of te hoog waardoor het bewerkingsmedium zal verdampen of het vloeibare bewerkingsmedium zal niet worden gehandhaafd.Boven-15 dien moet de temperatuur van het medium niet te hoog zijn, vanwege de stabiliteit van het deeltjesvormige actieve middel dat wordt verwerkt in de microcapsules, dat dan nadelig kan worden beïnvloed. 20 De dispersie die wordt gevormd is een stabiele emulsie en uit deze dispersie wordt het organische oplosmiddel in de microdruppels in het organische oplosmiddel, dat niet mengbaar is met de vloeistof gedeeltelijk verwijderd in de eerste bewerkingstrap voor het verwijderen van oplosmiddel.
Het oplosmiddel kan makkelijk worden verwijderd door het uitvoeren van een 25 bekende techniek, zoals verwarmen, het uitoefenen van een verlaagde druk of een combinatie van beide. De temperatuur die wordt toegepast om oplosmiddel te verdampen uit de microdruppels is niet kritisch maar moet niet zo hoog zijn, dat deze temperatuur van nadelige invloed is op de tempera-tuursgevoelige stoffen, die worden toegepast bij de bereiding van de micor-30 capsules en moet ook niet zo hoog zijn, dat oplosmiddel bij een zodanige hoge snelheid verdampt, dat defecten optreden bij het wandvormende materiaal. In het algemeen wordt 10-90 ?,*bij voorkeur 40-60 % van het oplosmiddel verwijderd in de eerste trap ter verwijdering van oplosmiddel.
Nadat het oplosmiddel in de eerste trap is verwijderd, worden de 35 gedispergeerde microcapsules in het oplosmiddel-niet mengbare vloeistof-medium, afgescheiden uit het vloeistofmedium door het uitvoeren van een hiertoe geschikte scheidingsbewerking. Na het afscheiden van de microcapsules uit de continue fase van het bewerkingsmedium, wordt de rest van het oplosmiddel in de microcapsules 5 verwijderd door extractie.' Bij deze tweede fetap kunnen de microcapsules worden gesuspendeerd in hetzelfde continue fase-bewerkingsmedium, dat wordt toegepast in de eerste trap, al of niet met oplosmiddel of in een andere vloeistof. Het extractieraedium extraheert het oplosmiddel uit de microcapsules en hierin zullen de microcapsules toch niet oplossen. Tijdens de 10 extractie moet het extractiemedium met opgelost oplosmiddel worden verwijderd en vervangen door vers extractiemedium. Dit kan het best worden bewerkstelligd op continue basis, waarbij de mate waarin het extractiemedium wordt aangevuld niet kritisch is. Het zal duidelijk zijn, dat de mate waarin het extractiemedium wordt aangevuld bij een bepaald procédé een variabele is, die makke- · lijk kan worden bepaald op het tijdstip dat de bewerking wordt uitgevoerd en daarom zijn geen bepaalde grenzen aangegeven voor deze mate van aanvulling. Nadat de rest van het oplosmiddel is verwijderd uit de microcap-20 sules, worden de microcapsules gedroogd, door deze bloot te stellen aan lucht of door een andere conventionele droogtechniek, zoals het drogen onder verlaagde druk, drogen boven een droogmiddel ofdergelijke.
Het raicfocapsulepródukt volgens de uitvinding wordt gewoonlijk Samengesteld uit deeltjes met een ronde vorm, hoewel ook soms microcapsules 25 een onregelmatige vorm kunnen hebben. De microcapsules kunnen variëren in grootte en wel van delen van microns tot enkele millimeters. Andere biologisch actieve stoffen, die kunnen worden verwerkt in de onderhavige microcapsu-les omvatten gastrointestinale therapeutische middelen zoals aluminiumhy-droxyde, calciumcarbonaat, magnesiumcarbonaat, natriumcarbonaat en dergelijke, non-steroïdale · geboorteregelende middelen, para-symptomatische 10 stoffen, psychotherapeutische stoffen, kalmerende middelen zoals chloro-promazine-HCl,clozapine, mesoridazine, metiap-ine, reserpine, thioridazine en dergelijke en zwakkere kalmerende middelen zoals chlordiazepoxyde, dizepam, meprobamaat, temazepam en dergelijke; rhinologische decongestanten, sedatieve-hypnotische stoffen zoals codeïne, fenobarbital, natriumpentobarbital, natrium-15 sscobarbital en dergelijke. De uitvinding wordt nadere toegelicht aan de hand van de volgende voorbeelden, die niet als beperkend moeten worden opgevat.
5 VOORBEELD I.
Progesteron werd in een hoeveelheid van 2,5 g en verder 10 g poly (DL-lactide) opgelost in 38 g methyleenchloride. De organische oplossing werd gedispergeerd in de vorm van microdruppels in 120 g 5% waterig poly(vinyl-alcohol). De dispersie werd verkregen door toevoeging van de organische 10 oplossing aan een geroerde, waterige poly(vinylalcohol) oplossing aanwezig in een kunststof vat met een inhoud van 200 ml. De emulsie werd geroerd met behulp van een Teflon turbine-roerder , aangedreven met behulp van een Fisher-Steadi-Speed motor.
Nadat een stabiele olie-in water emulsie was bereid, werd de druk 15 boven de emulsie continu verlaagd en gehouden boven het punt, waarbij schuimvorming zou optreden. De verdamping van het oplosmiddel werd voortgezet totdat 40-60 % van het methyleenchloride was verwijderd. De suspensie van microcapsules werd gecentrifugeerd bij 41 G gedurende 10 minuten. De bovenstaande vloeistof werd vervolgens gedecanteerd, de microcapsules op-20 nieuw gesuspendeerd met 50-100 ml gedeioniseerd water en de helft van deze suspensie werd uitgegoten in een fijn glasfilter (4-5,5 yum). Het afzuigen werd bewerkstelligd en de microcapsules werden langzaam afgefiltreerd. Tijdens de filtratie werden de microcapsules in suspensie gehouden door zacht roeren en door continu meer gedeioniseerd water toe te voegen. De microcap-25 sules werden op deze wijze gewassen totdat ongeveer 400-500 ml gedeioniseerd water was gebruikt. Door deze wasbewerking werd het resterende methyleenchloride geëxtraheerd uit de microcapsules en werden de microcapsules gehard.
De microcapsules werden bewaard terwijl de laatste 100 ml water 30 werd verwijderd. Nadat het water was afgevoerd uit de microcapsule-houdende koek gedurende 1 minuut, werd de koek snel gebroken tot kleinere deeltjes met een spatel. Terwijl de afzuiging werd gehandhaafd over de filtertreehter, werden de microcapsules continu geroerd in de trechter, gedurende 30 minuten. De microcapsules waren toen redelijk droog en werden onmiddellijk 35 gezeefd door een roestvrij stalen zeef van 250 jun onder toepassing van een borstel van kamelenhaar. De zeefbewerking was een ondersteuning voor het drogen van de capsules. Nadat de zeefbewerking was uitgevoerd over een zeef van 250 jum, konden de microcapsules makkelijk door een kleinere zeef wor- 8104507 r -10- 22062/Vk/ts den gezeefd of de microcapsules konden gedurende een nacht worden bewaard om te drogen. Het eindprodukt bestond uit een vrijstromend materiaal van ronde deeltjes, die 20 gew. % progesteron bevatten in poly(DL-lactide). VOORBEELD II.
5 Norgestimaat werd in een hoeveelheid van 3 g en poly(lactide-co-glyco- lide) in een hoeveelheid van 3 g opgelost in .18 g' methyleenchloride. De or- ganische oplossing werd in de vorm van microdruppels gedispergeerd in 58 g van een 5 gew. % waterige oplossing van poly (vinylalcphol). De emulsie werd verkregen door het toevoegen van de organische oplossing aan de ge-10 erde waterige poly(vinylalcohol) oplossing in een kunststof vat. Een Teflon turbineroerder, aangedreven met behulp van een Fisher Steady Speed motor werd gebruikt om de emulsie te roeren.
Nadat een stabiele olie-in-water emulsie was bereid, werd de driik boven de emulsie continue verlaagd en gehouden juist boven het punt waarop 15 schuimvorming optrad. Het afdampen van het oplosmiddel werd voortgezet totdat' 140—60 % van het methyleenchloride was verwijderd. De suspensie met microcapsules werd gecentrifugeerd bij 27 G gedurende 3 minuten. De bovenstaande vloeistof werd gedecanteerd en de microcapsules opnieuw gesuspendeerd met 50-100 ml gedeioniseerd water en werden uitgegoten in een fijne glazen 20 filtertrechter .....__ van 4-5,5 ^im. Het afzuigen werd bewerkstelligd en de micorcapsules werden langzaam afgefiltreerd. T.ijdens de filtratie werden de microcapsules in suspensie gehouden door deze zacht te roeren en door continue meer gedeioniseerd water toe te voegen. De microcapsules werden op deze wijze gewassen, totdat ongeveer 2000 ml gedeioniseerd water was 25 gebruikt. Door deze wasbewerking werd het resterende methyleenchloride geëxtraheerd uit de microcapsules en deze microcapsules werden gehard.
De microcapsules werden bewaard totdat de laatste 10 ml water was verwijderd. Nadat water was afgezogen uit de koek van de capsules gedurende ongeveer 1 minuut, werd de koek snel gebroken tot kleinere stukjes met 30 behulp van een spatel. Tprwijl de afzuiging werd. gehandhaafd od de filtertrechter, werden de microcapsules continu geroerd in de trechter gedurende 30 minuten. De microcapsules waren daarna redelijk droog en werden onmiddellijk gezeefd door een roestvrij stalen zeef onder gebruikmaking van een bor_ stel van kamelenhaar. Het eindprodukt bestond uit een vrij stromend deeltjes-35 vormig rond produkt, met 50 gew. % norgestimaat in poly(lactide-co-glycolide).

Claims (16)
1. Werkwee voor de bereiding van microcapsules die een actief middel bevatten, met het kenmerk, dat 5 a) een actief middel wordt opgelost of gedispergeerd in een oplos middel en een wandvormend materiaal wordt opgelost in het oplosmiddel, b) het oplosmiddel met het actieve middel en het wandvormend middel worden gedispergeerd in een continue fase-bewerkingsmedium, o) een deel van het,oplosmiddel wordt verdampt uit de dispersie van 10 trap b) , waarbij microcapsules worden gevormd met het actieve middel in de suspensie, en d) de rest van het oplosmiddel wordt uit de microcapsules geëxtraheerd.
2. Werkwijze volgens conclusie 1, met het kenmerk, dat de microcapsules gevormd in trap c) worden afgescheiden uit het continue fase-bewerkings- 15 medium voordat het oplosmiddel in trap d) volledig-.:wordt verwijderd.
