zs' 30 35 szoiaoiëo om det organiska lösningsmedlet utbyts mot endera av följande dispergeringsmedier: mättade eller omättade animaliska eller vegetabiliska fetter eller oljor, tri-n-butylfosfat, vätske- formig silikon, paraffin olja eller ftalsyradibutylester. Vissa av dessa dispergerinsmedier har använts förut för att framställa sfäriska polymerpartiklar. I US patent 4,l69,804 beskrivs en metod för att framställa magnetiska mikrosfärer genom att dispergera an albuminlösning i vegetabilisk olja och sedan upphetta denna till 14006 eller tvärbinda albuminpolymeren med aldehyder.
Denna teknik kan inte användas för att innsluta känsliga bio- material då antingen den höga temperaturen eller den tvärbindande aldehyden medför inaktivering.
I Biotechnology Letters vol. 3, 65-70, 1981 beskrivs en metod för att framställa sfäriska polyuretanpartiklar genom dispergering av monomererna i paraffinolja. Genom att inkludera bakterier i monomerblandningen erhölls en katalytisk aktiv preparation (huru- vida bakterierna har kvar sin förmåga att föröka sig är tveksamt, dessutom föreligger stor risk att de binds kovalent till bäraren).
Genom att kombinera dessa skonsamma dispergeringsmedier med en lämplig immobiliseringsmetod-bärare kan däremot såsom i de här beskrivna preparationerna en 100 %-ig viabilitet och bibehållen tillväxtförmåga erhållas. I tabell 1 jämförs olika dispergerings- mediers denaturerande verkan på agar-inneslutna växtceller. Av tabellen framgår att dessa dispergeringsmedier ger en bibehållen viabilitet jämfört med gängse standardmetod för framställning av sfäriska polymerpartiklar (svenskt patent 7204481-1).
Känsliga system såsom växtceller, alger eller animala celler innesluts lämpligen i polymerer såsom agar, agarose, karrageenan, gelatin, kollagen eller fibrinogen.
Animala celler förekommer i två principiellt skilda typer, suspensions- celler och så kallade ytberoende celler. De ytberoende cellerna måste vara fästade vid en yta för att kunna överleva och tillväxa.
I denna uppfinning används de bildade fibrerna av kollagen eller fibrin som nödvändig yta, detta medför att de ytberoende animala cellerna kan överleva och tillväxa i inneslutet-tillstånd. .___---_..-p~.__._.... . _. _ _ , 3 azo14o1-o Ytberoende celler kan inte tillväxa inneslutna i agarose, men genom att blanda agarose med fibrinogen eller kollagen kan även denna polymer användas för inneslutning av ytberoende celler.
Animala suspensionsceller har inte detta krav utan kan inneslutas i agarose, kollagen eller fibrin med bibehållen tillväxtförmâga.
Eftersom animala cellers potentiella användningsområde är mycket brett (produktion av vacciner, proteiner eller hormoner, trans- formering av prekursorer mm) är det viktigt att utveckla system där de kan användas i stor skala.
Inneslutna animala celler har stora produktionstekniska fördelar jämfört med fria celler, exempelvis kan de lätt användas i kolonn- processer, även i andra typer av reaktorer erhålls fördelar då de är mycket lättare att anpassa till kontinuerlig produktion.
Storleken av den bildade pärlan med inneslutet biomaterial kan varieras inom vida gränser beroende pâ med vilken kraft polymer- lösningen dispergeras. U szoiäoièo Exempel 1 _ Agar eller agarose löses i vatten (5.6 % w/v) genom uppvärmning.
Polymerlösningen (8 ml) tempereras till 500 C och blandas med växtceller (2 g) varefter dispergering i sojaolja (40 ml) sker. 5 När lagom stora kulor erhållits kyles blandningen till 50 C varefter kulorna tvättas över till vatten.
Exempel 2 Karrageenan löses i 0.9 % NaCl (3,1 % w/v) genom uppvärmning. 10 Kulor tillverkas enligt exempel 1.
Exempel 3 Gelatin löses i vatten genom uppvärmning (15 % w/v). Polymerlösningen (8 ml) tempereras till 370 C och blandas med jästceller (2 ml), 15 varefter blandningen dispergeras i sojaolja (40 ml). Efter av- kylning till 150 C tvättas kulorna över till vatten.
Exempel 4 Fibrinogenlösningen (1 ml, 2 % w/v) blandas med balanseradssàtt- 20 lösning (1 ml) samt fetalt kalvserum (D.3 ml). Till detta sätts animala celler (0,7 ml) samt trombin (1.5 U). Blandningen dispergeras i paraffinolja (40 ml). Efter avkylning tvättas kulorna över till odlingsmedium varefter de inneslutna cellerna odlas vid 370 C. 25 Exempel 5 Agarose löses i balanserad saltlösning (PBS) (5 % w/v) genom uppvärmning. Polymerlösningen (5 ml) tempereras till 37° C och _blandas med animala celler (5 ml), varefter blandningen dispergeras i paraffinolja (40 ml). Efter avkylning tvättas kulorna över till 30 medium och de inneslutna cellerna odlas vid 370 C.
Exempel 6 Kollagen löses i utspädd saltsyra. Polymerlösningen (kyld till 50 C, 2 ml) blandas med 10 ggr koncentrerat medium och natriumhydroxid 35 för att erhålla neutralt pH, samt animala celler (2 ml).

Claims (4)
1. Förfarande för immobilisering av levande animala celler, växtceller, bakterier, alger eller svampar med bibehållen tillväxtförmåga genom inneslutning i polymerpärlor, av att cellerna sätts till en vattenlösning k ä n n e- t e c k n a t av en kolhydratbaserad eller proteinbaserad denna vattenlösning därefter dispergeras i ett vattenolöa- polymer, att ligt dispergeringsmedium, som utgöres av en mättad eller omättad vetetabilisk olja, tri-n-butylfosfat, vätskeformig silikon, paraffinolja eller ftalsyradibutylester, och att polymeren bringas att gela under bildning av polymerpärlor, vari cellerna innesluts,genom avkylning, neutralisering följd av uppvärmning eller enzymatisk påverkan att cellernas tillväxtförmâga icke förstörs. under sådana betingelser,
