Description
Der Erfindung lag daher die Aufgabe zugrunde, ein Verfahren zur Herstellung von einen Wirkstoff enthaltenden Mikrokapseln vorzuschlagen, mit dem Mikrokapseln hoher Beladung und verbesserter Qualität erhalten werden können.
Erfindungsgemäss wird diese Aufgabe dadurch gelöst, dass
(a) ein Wirkstoff in einem Lösungsmittel gelöst oder dispergiert und in diesem Lösungsmittel ein eine Umhüllung bildendes Material gelöst wird,
(b) das Lösungsmittel, das den Wirdstoff und das eine Umhüllung bildende Material enthält, in einem eine geschlossene Phase bildenden Behandlungsmedium dispergiert wird,
(c) ein Teil des Lösungsmittels der durch die Massnahme (b) gebildeten Dispersion verdampft wird, um dadurch Mikrokapseln zu bilden, die den Wirkstoff in Suspension enthalten und
(d) das restliche Lösungsmittel aus den Mikrokapseln extrahiert wird.
Das zentrale Kennzeichen des erfindungsgemässen Verfahrens besteht darin, dass während der Herstellung das Lösungsmittel von den in einem flüssigen Medium suspendierten Mikrokapseln in zwei unterschiedlichen Verfahrensstufen, demzufolge anders als bei den einstufig geführten Verfahren, entfernt wird. Diese Technik der Entfernung des Lösungsmittels in zwei Verfahrensstufen führt zu einem Produkt von Mikrokapseln verbesserter Qualität und eines höheren Gehalts an Wirkstoff. Der besondere Vorteil des erfindungsgemässen Verfahrens gegenüber den herkömmlichen Verfahren ist darin zu sehen, dass dieses zweistufige Vorgehen zur Entfernung des Lösungsmittels zu Mikrokapseln mit

einer unerwartet hohen Beladung an Wirkstoff und einer überraschend hohen Qualität führt, wenn mit den herkömmlichen Verfahren zur Herstellung von Mikrokapseln, bei denen das Lösungsmittel in einer einzigen Stufe entfernt wird, verglichen wird.
Die den Mikrokapseln einzuverleibende Substanz wird in einem Lösungsmittel gelöst. Zu den geeigneten Wirkstoffen, die erfindungsgemäss den Mikrokapseln einverleibt werden können, zählen Agrochemikalien, Duftstoffe, Härtungsmittel, Farbstoffe, oxidierende Mittel und biologisch aktive Mittel bzw. biologische Wirkstoffe. Die Menge des in dem Lösungsmittel gelösten oder dispergierten Wirkstoffes ist nicht kritisch, obwohl das Verhältnis des Wirkstoffes zu dem eine Umhüllung bildenden Material insofern bedeutsam ist, als eine zu hohe Menge an Wirkstoff (gegenüber dem eine Umhüllung bildenden Material) Mikrokapseln nicht entstehen lässt. Unter praktischen Gesichtspunkten kann die Menge des mit dem eine Umhüllung bildenden Materials kombinierten Wirkstoffes bis zu etwa 80 Gewichtsteilen Wirkstoff pro 20 Gewichtsteile des eine Umhüllung bildenden Materials ausmachen. Für niedrigere Verhältnisse gibt es keine Beschränkung, bei der der Wirkstoff mit dem eine Umhüllung bildenden Material kombiniert werden kann, wenngleich bei sehr niedriger Beladung der Mikrokapseln mit dem Wirkstoff der praktische Nutzen entfallen kann. Es ist jedoch ersichtlich, dass bei einer stärker verdünnten Lösung die vorliegende Lösungsmittelmenge grösser ist und diese grössere Lösungsmittelmenge schliesslich entfernt werden muss, um Mikrokapseln eines gegebenen Gehalts an Wirkstoff zu erhalten. Gewöhnlich wird der Wirksoff in dem Lösungsmittel in einer Konzentration von 5 bis 30 Gewichtsprozent gelöst oder suspendiert.
Das bei der Herstellung der Lösung eingesetzte und den Wirkstoff enthaltende Lösungsmittel kann unter einer Vielzahl von üblichen organischen Lösungsmittel ausgewählt werden. Hierzu zählen halogenierte aliphatische Kohlenwasserstoffe, wie Chloroform, Methylenchlorid, Methylchloroform und dergleichen, aromatische Kohlenwasserstoffverbindungen, halogenierte aromatische Kohlenwasserstoffverbindungen, cyclische Ether, wie Tetrahydrofuran und dergleichen, Alkohole, Wasser, Aceton und dergleichen. Bei dem Lösungsmittel muss es sich um ein Material handeln, das das Umhüllungsmaterial löst, gegenüber dem Wirkstoff chemisch inert ist, mit dem eine zusammenhängende (geschlossene) Phase bildenden Behandlungsmedium (Dispersionsphase) nicht mischbar ist und das einen Siedepunkt aufweist, der unter dem des eine geschlossene Phase bildenden Behandlungsmediums liegen sollte. Die Menge des in dem Lösungsmittel zu lösenden und eine Umhüllung bildenden Materials hängt von der Löslichkeit des Polymerisats in dem Lösungsmittel und der erhaltenen Viskosität der polymeren Lösung ab. Die Viskosität bezieht sich auf die endgültige Grösse der Mikrokapseln. Mischungen der vorgenannten Lösungsmittel können als geeignete Lösungsmittel des Wirkstoffes ebenfalls verwendet werden. Es ist von Vorteil, Lösungsmittel niedrigeren Siedepunktes zu verwenden, da sie schliesslich leichter durch einfachere Verdampfungstechniken entfernbar sind.
Zu der den Wirkstoff enthaltenden Lösung wird das die Umhüllung bildende Material hinzugegeben, das den Wirkstoff in den Mikrokapseln umgibt und begrenzt. Die Menge des der Lösung zuzufügenden, die Umhüllung bildenden Materials wird so gewählt, dass sich die Mikrokapseln ohne weiteres bilden, wenn die Lösung in dem eine geschlossene Phase bildenden Behandlungsmedium dispergiert wird. Vor dem Dispergieren der Lösung in diesem Medium sollten sowohl der Wirkstoff als auch das Umhüllungsmaterial in dem Lösungsmittel vorliegen. Normalerweise wird das die Um3

Zu den geeigneten Materialien, die eine Umhüllung bilden, zählen Polystyrol, Ethylcellulose, Celluloseacetat, Hy-droxypropylmethylcellulosephthalat, Celluloseacetat, Dibu-tylaminohydroxypropylether, Polyvinylbutyral, Polyvinyl-formal, Poly(meth)acrylsäureester, Polyvinylacetal-diethyl-amino-acetat, 2-Methyl-5-vinylpyridinmethacrylat-Meth-acrylsäure-Mischpolymerisat, Polycarbonat, Polyester, Polypropylen, Vinylchlorid-Vinylacetat-Mischpolymerisat, Polysaccharide, Glycerindistearat und dergleichen. Zu der bevorzugten Gruppe polymerer, die Umhüllung bildenden Materialien zählen biologisch abbaubare Materialien, wie aliphatische Polyester, einschliesslich der Polylactide, Polyglycoli-de, Polycaprolactone und deren Mischpolymerisate. Nach der Herstellung der Lösungsmittelphase, die den Wirkstoff und das Material zur Ausbildung der Umhüllung enthält, werden die Mikrotröpfchen des die Umhüllung bildenden Materials, das den Wirkstoff einschliesst, dadurch gebildet, indem die Lösung, die den Wirkstoff und das die Umhüllung bildende Material enthält, in einem Behandlungsmedium mit geschlossener Phase dispergiert wird. Dieses Behandlungsmedium darf mit dem Lösungsmittel nicht mischbar sein und muss einen höheren Siedepunkt als das Lösungsmittel aufweisen. In vielen Fällen wird das Behandlungsmedium zwar Wasser sein. Es gibt jedoch auch Beispiele von geeigneten nicht-wässrigen Behandlungsmedien mit geschlossener Phase, wozu z.B. organische Flüssigkeiten, wie Xylol oder Toluol, synthetische Öle, wie Silikonöle, und natürliche Öle, wie Pflanzenöle, einschliesslich Erdnussöl, zählen. Gewöhnlich wird ein grenzflächenaktives Mittel (emulgierendes Mittel) zu dem eine geschlossene Phase bildenden Behandlungsmedium gegeben, um zu verhindern, dass die Mikrokapseln agglomerieren, und um die Grösse der Lösungsmittelmikrotröpfchen in der Emulsion einzuregeln. Ein gutes grenzflächenaktives Mittel für Wasser als Behandlungsmedium mit geschlossener Phase ist Polyvinylalkohol (PVA) in einer Konzentration von 1 bis 10%. Die Dispersion kann durch mechanisches Rühren des Behandlungsmediums mit geschlossener Phase durch eine Einrichtung, wie eine Kolloidmühle, einen Homogenisator oder dergleichen gebildet werden. Eine Emulsion kann auch dadurch gebildet werden, indem kleine Tropfen der Lösung, die den Wirkstoff und das die Umhüllung bildende Material enthält, zu dem eine geschlossene Phase aufweisenden Behandlungsmedium gegeben werden. Bei der Durchführung der Despersionsmass-nahmen wird es bevorzugt, dass die Lösung mit dem organischen Lösungsmittel in einer den Polyvinylalkohol enthaltenden wässrigen Lösung dispergiert wird. Die Temperatur während der Bildung der Emulsion ist nicht besonders kritisch. Sie kann jedoch Grösse und Qualität der Mikrokapseln beeinflussen. In Abhängigkeit von dem Lösungsmittel und dem verwendeten Behandlungsmedium mit geschlossener Phase sollte die Temperatur darüber hinaus nicht zu niedrig gewählt werden, da sich sonst das Lösungsmittel und das Behandlungsmedium verfestigen oder das Behandlungsmedium unter praktischen Gesichtspunkten zu viskos wird. Auch sollte sie nicht zu hoch gewählt werden, da sonst das Behandlungsmedium verdampfen würde und das flüssige Behandlungsmedium somit nicht aufrechterhalten bleibt. Darüber hinaus sollte die Temperatur des Behandlungsmediums nicht zu hoch sein, da sonst die Stabilität des besonderen Wirkstoffes, der den Mikrokapseln einzuverleiben ist, nachteilig beeinflusst würde. Demzufolge kann das Disper-gieren bei jeder beliebigen Temperatur durchgeführt werden, die stabile Betriebsbedingungen einhält.
Die gebildete Dispersion ist eine stabile Emulsion. Daraus wird das organische Lösungsmittel in den Mikrotröpfchen (der mit dem organischen Lösungsmittel nicht mischbaren Flüssigkeit) teilweise in der ersten Stufe der Entfernung des Lösungsmittels entfernt. Das Lösungsmittel kann leicht anhand üblicher Techniken entfernt werden, wie durch Erhitzen oder durch Auferlegung eines verminderten Drucks oder auch eine Kombination dieser beiden Möglichkeiten. Die zum Verdampfen des Lösungsmittels von den Mikrotröpfchen gewählte Temperatur ist nicht kritisch. Sie sollte jedoch nicht so hoch sein, dass ein bei der Herstellung gegebener Mikrokapseln eingesetztes temperaturempfindliches Material abgebaut wird. Auch sollte sie nicht so hoch sein, dass das Lösungsmittel mit einer solchen Geschwindigkeit abgedampft wird, dass sich Fehler in dem eine Umhüllung bildenden Material einstellen. Im allgemeinen werden 10 bis 90 %, vorzugsweise 40 bis 60 % des Lösungsmittels in der ersten Stufe der Lösungsmittelentfernung entzogen.
Nach dieser ersten Entfernung des Lösungsmittels werden die in dem mit dem flüssigen Medium nicht mischbaren Lösungsmittel dispergierten Mikrokapseln aus dem flüssigen Medium in herkömmlicher Weise abgetrennt bzw. isoliert. So kann z. B. die Flüssigkeit von den Mikrokapseln dekantiert werden. Auch können die Mikrokapseln aus der flüssigen Suspension abfiltriert werden. Jedoch können verschiedene andere Kombinationen von Abtrennungsmassnahmen, sofern zweckmässig, herangezogen werden.
Nach der Isolierung der Mikrokapseln aus dem Behandlungsmedium mit geschlossener Phase wird das restliche Lösungsmittel in den Mikrokapseln durch Extraktion entfernt. In dieser zweiten Stufe können die Mikrokapseln in dem gleichen, eine geschlossene Phase aufweisenden Behandlungsmedium, das bei der Stufe 1 herangezogen wurde, mit oder ohne Verwendung eines grenzflächenaktiven Mittels oder in einer anderen Flüssigkeit suspendiert werden. Das Extraktionsmedium extrahiert das Lösungsmittel aus den Mikrokapseln, ohne dass die Mikrokapseln selbst gelöst werden. Während der Extraktion muss das Extraktionsmedium mit dem gelösten Lösungsmittel entfernt und durch frisches Extraktionsmedium ersetzt werden. Dieses erfolgt am besten kontinuierlich, wobei die Geschwindigkeit des Ersatzes des Extraktionsmediums nicht kritisch ist. Wenn die Geschwindigkeit zu niedrig ist, dann treten Kristalle des Mittels aus den Mikrokapseln heraus oder wachsen in das Extraktionsmedium hinein. Offensichtlich ist die Geschwindigkeit des Ersatzes des Extraktionsmediums bei einem gegebenen Verfahren variabel und kann ohne weiteres während der Durchführung des Verfahrens festgelegt werden. Genaue Begrenzungen dieser Geschwindigkeit müssen daher nicht vorher festgelegt werden. Nach der Entfernung des restlichen Lösungsmittels von den Mikrokapseln werden diese dadurch getrocknet, indem sie Luft ausgesetzt oder anderen herkömmlichen Trocknungsmassnahmen unterzogen werden, wie z. B. dem Vakuumtrocknen, Trocknen über einem Trocknungsmittel und dergleichen.
Die erfindungsgemäss erhältlichen Mikrokapseln bestehen im allgemeinen aus Teilchen kugeliger Gestalt, obwohl sie gelegentlich auch ungleichmässige Form aufweisen können. Sie können bezüglich ihrer Grösse schwanken, wobei es sich um einen Submikron- (50-2000 Â) bis mm-Durchmesser handeln kann. Vorzugsweise sind Durchmesser von 1 Submikron bis 250 um für pharmazeutische Zubereitungen zweckmässig, was die Verabreichung der Mikrokapseln über eine Standardspritze und -nadel ermöglicht. Die Mikrokapseln sind in einem weiten Anwendungsbereich brauchbar, was von der Art der überführbaren und darin einverleibten Substanz abhängt. Die erfindungsgemäss erhältlichen Mikrokapseln sind besonders zur Verabreichung einer Vielzahl biologisch aktiver Mittel bzw. Wirkstoffe an menschliche und tierische Lebewesen nützlich. verhütendes Mittel enthalten, dann können sie z.B. dem jeweiligen Patienten durch Injektion oder durch transzervikale Wanderung und Transport zu den inneren weiblichen Fortpflanzungsorganen zwecks Geburtenkontrolle verabreicht werden. Zu den geeigneten empfängnisverhütenden Mitteln zählen Östrogene, wie Diethylstilböstrol, 17-beta-Estradiol, Östron, Ethinylöstradiol, Mestranol und dergleichen, Proge-stine, wie Norethindron, Norgestryl, Norgestrel, Ethyno-dioldiacetat, Lynestrenol, Medroxyprogesteronacetat, Di-methisteron, Megestrolacetat, Chlormadinonacetat, Norge-stimat, Norethisteron, Ethisteron, Melengestrol, Norethyno-drel und dergleichen, und Spermizide Verbindungen, wie No-nylphenoxypolyoxyethylenglycol, Benzethoniumchlorid, Chlorindanol und dergleichen. Zu anderen biologischen Wirkstoffen, die den erfindungsgemäss erhältlichen Mikrokapseln einverleibt werden können, zählen gastrointestinale Therapeutika, wie Aluminiumhydroxid, Calciumcarbonat, Magnesiumcarbonat, Natriumcarbonat und dergleichen, nicht-steroidale Antifertilitätsmittel bzw. Ovulationshemmer, parasympathikomimetische Mittel, psychotherapeutisch wirkende Mittel, Neuroleptika, wie Chloropromazin HCl, Clozapin, Mesoridazin, Metiapin, Reserpin, Thiorida-zin und dergleichen, Tranquilizer, wie Chlordiazepoxid, Diazepam, Meprobamat, Temazepam und dergleichen, rhinolo-gische Abschwellungsmittel, Schlafmittel bzw. Nachfolgend wird die Erfindung noch weiter anhand von erläuternden Beispielen beschrieben.
Beispiel 1
2,5 g Progesteron und 10 g Poly (DL-lactid) wurden in 38 g Methylenchlorid gelöst. Die organische Lösung wurde in Form von Mikrotröpfchen in 120 g eines wässrigen Mediums, das 5 Gew.% Polyvinylalkohol enthielt, dispergiert. Die Dispersion wurde durch Zugabe der organischen Lösung zu der gerührten wässrigen Polyvinylalkohollösung, die in einem 200 ml-Harzkessel enthalten war, erhalten. Zum Rühren der Emulsion wurde ein «Teflon»-Turbinenschnell-rührer verwendet, der durch einen «Fisher-Stedi-Speed-Motor» angetrieben wurde.
Nachdem eine stabile Öl-in-Wasser-Emulsion hergestellt worden war, wurde der Druck oberhalb der Emulsion kontinuierlich vermindert und gerade oberhalb des Punktes gehalten, bei dem ein Schäumen auftrat. Das Abdampfen des Lösungsmittels wurde so lange fortgeführt, bis 40 bis 60 % des Methylenchlorids entfernt worden waren. Die Suspension der Mikrokapseln wurde bei 41 g 10 min lang zentrifugiert. Die überstehende Flüssigkeit wurde darauf dekantiert. Die Mikrokapseln wurden dann erneut in 50 bis 100 ml entioni-siertem Wasser suspendiert. Die Hälfte dieser Suspension wurde über einen feinen (4-5,5 (im) Frittenglastrichter gegossen. Unter Absaugen wurden die Mikrokapseln langsam filtriert. Während der Filtration wurden die Mikrokapseln durch gelindes Rühren und durch kontinuierliche Zugabe weiteren entionisierten Wassers in Suspension gehalten. Die Mikrokapseln wurden so lange in dieser Weise gewaschen, bis 400 bis 500 ml entionisierten Wassers verbraucht waren. Dieses Waschen extrahierte restliches Methylenchlorid, das in den Mikrokapseln verblieb und härtete die Mikrokapseln.
Die Mikrokapseln setzten sich ab, während die letzten 10 ml Wasser entfernt worden waren. Nachdem das Wasser etwa 1 min lang von dem Mikrokapselkuchen abgelaufen war, wurde der Kuchen schnell mit einem Spatel in kleinere Stücke zerbrochen. Unter weiterem Ausüben einer Saugwirkung auf den Filtertrichter wurden die Mikrokapseln kontinuierlich in dem Trichter 30 min lang gerührt. Die Mikrokapseln waren dann hinreichend trocken. Sie wurden über ein nicht-rostendes 250-|xm-Stahlsieb unter Verwendung ei5

ner Kamelhaarbürste gesiebt. Das Sieben unterstützte das Trocknen der Mikrokapseln. Nach diesem Sieben konnten die Mikrokapseln leicht über ein Sieb mit kleineren Siebmaschen gesiebt oder über Nacht zum Trocknen stehengelassen werden. Das Fertigprodukt bestand aus freifliessenden kugelförmigen Teilchen, die 20 Gewichtsprozent Progesteron im Poly (DL-lactid) enthielten.
Beispiel 2
3 g Norgestimat und 3 g Poly(lactid-co-glycolid) wurden in 18 g Methylenchlorid gelöst. Die organische Lösung wurde in Form von Mikrotröpfchen in 58 g eines wässrigen Mediums mit 5 Gewichtsprozent Polyvinylalkohol dispergiert. Die Emulsion wurde durch Zugabe der organischen Lösung zu einer gerührten wässrigen Polyvinylalkohollösung, die in einem Harzkessel enthalten war, gewonnen. Mittels eines «Teflon»-Turbinenschnellrührers, der mit einem «Fisher-Steadi-Speed-Motor» angetrieben wurde, wurde die Emulsion gerührt.
Nach Einstellung einer stabilen Öl-in-Wasser-Emulsion wurde der Druck oberhalb der Emulsion kontinuierlich herabgesetzt und gerade auf dem Punkt gehalten, bei dem kein Schäumen auftrat. Das Verdampfen des Lösungsmittels wurde so lange fortgeführt, bis 40 bis 60 % des Methylenchlorids entfernt worden waren. Die Suspension der Mikrokapseln wurde bei 27 g 3 min lang zentrifugiert. Die überstehende Flüssigkeit wurde dann dekantiert. Die Mikrokapseln wurden dann erneut in 50-100 ml entionisiertem Wasser suspendiert und über einen feinen (4-5,5 um) Frittenglastrichter gegossen. Unter Ansaugen wurden die Mikrokapseln langsam abfiltriert. Während der Filtration wurden die Kapseln durch gelindes Rühren und durch kontinuierliche Zugabe weiteren entionisierten Wassers in Suspension gehalten. Die Mikrokapseln wurden in dieser Weise so lange gewaschen, bis etwa 2000 ml entionisierten Wassers verwendet worden waren. Dieses Waschen extrahierte restliches Methylenchlorid, das in den Mikrokapseln verblieben war, und härtete die Mikrokapseln.
Die Mikrokapseln wurden während des Entfernens der letzten 10 ml Wasser absetzen gelassen. Nachdem das Wasser während etwa 1 min von dem Mikrokapselkuchen abgetopft war, wurde der Kuchen schnell mit einem Spatel in kleinere Stücke zerbrochen. Unter weiterem Absaugen des Filtertrichters wurden die Mikrokapseln kontinuierlich 30 min lang in dem Trichter gerührt. Die Mikrokapseln waren dann hinlänglich trocken. Sie wurden unverzüglich über ein nicht-rostendes Stahlsieb unter Verwendung einer Kamelhaarbürste gesiebt. Das Fertigprodukt bestand aus freifliessenden kugelförmigen Teilchen mit einem 50gewichtsprozen-tualen Gehalt an Norgestimat in Poly(lactid-co-glycolid).
Die beschriebene Erfindung kann ersichtlich vielfaltigen Veränderungen bzw. Modifikationen unterliegen, ohne dass man sich von deren Wesen oder aus deren Rahmen löst.

- 6484942PATENTANSPRÜCHE1. Verfahren zur Herstellung von einen Wirkstoff enthaltenden Mikrokapseln, dadurch gekennzeichnet, dass(a) ein Wirkstoff in einem Lösungsmittel gelöst oder disper-giert und in diesem Lösungsmittel ein eine Umhüllung bildendes Material gelöst wird,(b) das Lösungsmittel, das den Wirkstoff und das eine Umhüllung bildende Material enthält, in einem eine geschlossene Phase bildenden Behandlungsmedium dispergiert wird,(c) ein Teil des Lösungsmittels der durch die Massnahme (b) gebildeten Dispersion verdampft wird, um dadurch Mikrokapseln zu bilden, die den Wirkstoff in Suspension enthalten, und(d) das restliche Lösungsmittel aus den Mikrokapseln extrahiert wird.
- 2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass die durch die Massnahme (c) gebildeten Mikrokapseln vor der vollständigen Entfernung des Lösungsmittels durch die Massnahme (d) aus dem eine geschlossene Phase bildenden Behandlungsmedium abgetrennt werden.
- 3. Verfahren nach Anspruch 1 oder 2, dadurch gekennzeichnet, dass Wasser, Xylol, Toluol, ein synthetisches Öl oder ein natürliches Öl als das eine geschlossene Phase bildende Behandlungsmedium verwendet wird.
- 4. Verfahren nach einem der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass das eine geschlossene Phase bildende Behandlungsmedium des weiteren ein grenzflächenaktives Mittel enthält.
- 5. Verfahren nach Anspruch 4, dadurch gekennzeichnet, dass als ein eine geschlossene Phase bildendes Behandlungsmedium Wasser, das 1 bis 10 Gew. % Polyvinylalkohol enthält, verwendet wird.
- 6. Verfahren nach einem der Ansprüche 1 bis 5, dadurch gekennzeichnet, dass bei der Massnahme (b) so vorgegangen wird, dass ein flüssiges Produkt in Form einer stabilen Was-ser-in-Öl- oder Öl-in-Wasser-Emulsion entsteht.
- 7. Verfahren nach einem der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass als Lösungsmittel halogenierte aliphatische Verbindungen, halogenierte aromatische Kohlenwasserstoffverbindungen, aromatische Kohlenwasserstoffverbindungen, cyclische Äther, Alkohole, Wasser oder Aceton oder Gemische davon verwendet werden.
- 8. Verfahren nach Anspruch 7, dadurch gekennzeichnet, dass als halogenierte aliphatische Verbindung Methylenchlorid verwendet wird.
- 9. Verfahren nach einem der Ansprüche 1 bis 8, dadurch gekennzeichnet, dass das Lösungsmittel aus der Dispersion durch Herabsetzung des Drucks oberhalb der Dispersion verdampft wird.
- 10. Verfahren nach einem der Ansprüche 1 bis 9, dadurch gekennzeichnet, dass bei der Massnahme (c) 10 bis 90 Gewichtsprozent des Lösungsmittels von den Mikrokapseln abgedampft werden.
- 11. Verfahren nach einem der Ansprüche 1 bis 10, dadurch gekennzeichnet, dass das restliche Lösungsmittel von den Mikrokapseln bei der Massnahme (d) durch Waschen der Mikrokapseln mit ein grenzflächenaktives Mittel enthaltendem Wasser unter Vakuumfiltration entfernt wird.
- 12. Verfahren nach Anspruch 11, dadurch gekennzeichnet, dass die Mikrokapseln durch Einwirken eines Vakuums getrocknet und danach gesiebt werden.
- 13. Verfahren nach einem der Ansprüche 1 bis 12, dadurch gekennzeichnet, dass Polystyrol, Ethylcellulose, Celluloseacetat, Hydroxypropylmethylcellulosephthalat, Celluloseacetat, Dibutylaminohydroxypropylether, Polyvinylbuty-ral, Polyvinylformal, Poly(meth)acrylsäureester, Polyvinyl-acetal-diethylaminoacetat, ein 2-Methyl-5-vinylpyridin-methacrylat-Methacrylsäure-Mischpolymerisat, ein Polycar-bonat, ein Polyester, Polypropylen, ein Vinylchlorid-Vinyl-acetat-Mischpolymerisat, ein Polysaccharid oder Glycerindi-stearat oder ein Gemisch davon als ein eine Umhüllung bildendes Material verwendet wird.
- 14. Verfahren nach Anspruch 13, dadurch gekennzeichnet, dass als Polyester Polylactid, Polyglycolid oder Polyca-prolacton oder ein Mischpolymerisat davon verwendet wird.
- 15. Verfahren nach einem der Ansprüche 1 bis 14, dadurch gekennzeichnet, dass Östrogene, Progestine, gastrointestinale therapeutische Mittel, nicht-steroidale Antifertili-tätsmittel, parasympathikomimetische Mittel, psychotherapeutische Mittel, Neuroleptika, Tranquilizer, rhinologisch abschwellende Mittel, Schlafmittel, nicht-estrogene Steroide und nicht den Gelbkörpern zuzuordnende Steroide, Sulfonamide, sympathikomimetische Mittel, Vakzine, Vitamine, Kräftigungsmittel, Mittel gegen Malaria, Mittel gegen Migräne, Mittel gegen die Parkinson'sche Krankheit, Antispa-stika, Anticholinergika, hustenlindernde Mittel, Bronchien-dilatoren, Mittel gegen Herz- und Kreislauferkrankungen, Elektrolytersatzmittel, Mutterkornalkaloide, Analgetika, nicht-narkotische Mittel, Antibiotika, Antikrebsmittel, krampflösende Mittel, Antiemetika, Antihistamine, entzündungshemmende Mittel, Prostaglandine, zytotoxische Arzneimittel, Antigene oder Antikörper oder Gemische davon als biologischer Wirkstoff verwendet werden.
- 16. Verfahren nach einem der Ansprüche 1-15, dadurch gekennzeichnet, dass der Wirkstoff in einer Konzentration von 5-30 Gewichtsprozent in dem Lösungsmittel gelöst oder dispergiert wird.
- 17. Verfahren nach einem der Ansprüche 1 - 16, dadurch gekennzeichnet, dass das eine Umhüllung bildende Material in einer Konzentration von 5-35 Gewichtsprozent in dem Lösungsmittel gelöst wird.
- 18. Verfahren nach einem der Ansprüche 1-17, dadurch gekennzeichnet, dass das Gewichtsverhältnis des Wirkstoffs zu dem eine Umhüllung bildenden Material in dem Lösungsmittel bis zu 80 Gewichtsprozent Wirkstoff zu 20 Gewichtsprozent des eine Umhüllung bildenden Materials beträgt.
- 19. Nach dem Verfahren nach Anspruch 1 hergestellte Mikrokapseln.Es gibt eine Vielzahl von Verfahren, nach denen jeder beliebige Verbindungstyp in Mikrokapseln eingeschlossen werden kann. Dabei wird das einzukapselnde Material im allgemeinen in einem Lösungsmittel dispergiert, das ein eine Umhüllung bildendes Material enthält. Bei einem einstufig geführten Verfahren wird das Lösungsmittel von den Mikrokapseln entfernt und danach das eigentliche Mikrokapsel-produkt erhalten. Zum Beispiel wird ein herkömmliches Verfahren zur Herstellung von Mikrokapseln in der US-PS 3 737 337 beschrieben, bei dem eine Lösung eines eine Wandoder Umhüllung bildenden polymeren Materials in einem Lösungsmittel hergestellt wird. Das Lösungsmittel ist lediglich teilweise in Wasser löslich. Ein festes Material oder Kernmaterial wird in dem die Lösung enthaltenden Polymerisat gelöst oder dispergiert. Danach wird in einer einzigen Stufe das die Lösung enthaltende Kernmaterial in einer wässrigen Flüssigkeit, die mit dem organischen Lösungsmittel nicht mischbar ist, dispergiert, um das Lösungsmittel von den Mikrokapseln zu entfernen. Ein anderes Beispiel eines Verfahrens, bei dem das Lösungsmittel von den eine Substanz enthaltenden Mikrokapseln in einer einzigen Stufe entfernt wird, ergibt sich aus der US-PS 3 523 906. Bei diesem5101520253035404550556065Verfahren wird ein einzukapselndes Material in einer Lösung eines polymeren Materials in einem Lösungsmittel, das mit Wasser nicht mischbar ist, emulgiert. Dann wird die Emulsion in einer ein hydrophiles Kolloid enthaltenden wässrigen Lösung emulgiert. Das Entfernen des Lösungsmittels von den Mikrokapseln wird dann in einer einzigen Stufe durch Verdampfen vollzogen. Das angestrebte Produkt wird dann erhalten. Bei einem weiteren Verfahren, das in der US-PS 3 691 090 beschrieben wird, wird das organische Lösungsmittel aus der Dispersion der Mikrokapseln in einem wässrigen Medium in einer einzigen Stufe abgedampft, vorzugsweise unter vermindertem Druck. Ein ähnliches Verfahren wird in der US-PS 3 891 570 beschrieben, wonach das Lösungsmittel aus einer Dispersion von Mikrokapseln in einem Medium aus mehrwertigem Alkohol dadurch von den Mikrokapseln abgedampft wird, indem Hitze zur Einwirkung gebracht oder die Mikrokapseln unter verminderten Druck gesetzt werden. Ein weiteres Beispiel eines Verfahrens zur Entfernung des Lösungsmittels in einer Stufe ergibt sich aus der US-PS 3 960 757. Weitere Hinweise zum Stand der Technik ergeben sich aus den GB-PS 1 394 780,1 048 696/7 und den US-PS 3 657 144, 3 576 758 und 3 773 919.Die mit dem herkömmlichen Einkapselungsverfahren verbundenen Probleme bestehen insbesondere darin, dass der Arzneimittelbeladung eine Grenze gesetzt ist, was insbesondere dann gilt, wenn dieses Verfahren zur Mikroeinkap-selung eines pharmazeutischen Mittels, wie Progesteron oder Norgestimat, herangezogen wird. In Abhängigkeit von den physikalischen und chemischen Eigenschaften des Arzneimittelträgers und des mikroeinzukapselnden Mittels führt ein Versuch zur Herstellung von Mikrokapseln mit hoher Beladung zu einer unvollständigen Einkapselung, d.h. die Kristalle des Mittels ragen aus den Mikrokapseln heraus oder wachsen in das Behandlungsmedium. Es bestand daher das Bedürfnis, eine Technik zu entwickeln, mit der Mikro-teilchen verbesserter Eigenschaft und hoher Qualität herstellbar sind.
