(実施例)

トウモロコシプロトプラスト一過性アッセイ:
TraP14 v2キメラ葉緑体輸送ペプチド配列(配列番号8)は、緑色蛍光タンパク質遺伝子の上流にクローニングされ、トウモロコシプロトプラストの一過性in plantaアッセイを介した試験のために構築物pDAB109902(図4)に組み込まれた。得られた構築物は、単一の植物転写単位(PTU)を含有した。第1のPTUは、トウモロコシ(Zea mays)ユビキチン1プロモーター(ZmUbi1プロモーター;Christensen, A., Sharrock R., and Quail P., (1992) Maize polyubiquitin genes: structure, thermal perturbation of expression and transcript splicing, and promoter activity following transfer to protoplasts by electroporation, Plant Molecular Biology, 18: 675-689)、TraP−緑色蛍光タンパク質融合遺伝子(TraP14−GFP;米国特許第7,678,893号)、およびトウモロコシ(Zea mays)ペルオキシダーゼ5 3’非翻訳領域(ZmPer5 3’UTR;米国特許第6384207号)で構成された。制限酵素消化および配列決定を介して構築物を確認した。
大腸菌(Esherichia coli)の5−エノールピルビルシキミ酸3−リン酸シンターゼ酵素(EPSPシンターゼ)における1アミノ酸突然変異(G96A)は、グリホサート非感受性をもたらすことができる(Padgette et al, (1991); Eschenburg et al., (2002); Priestman et al, (2005); Haghani et al, (2008))。この突然変異はグリホサートに対する耐性を付与するが、その天然基質であるホスホエノールピルビン酸(PEP)によるEPSPシンターゼの結合に悪影響を及ぼすことも知られている。基質結合効率において生じた変化は、グリホサートに対するin plantaにおける耐性を提供するために、突然変異した酵素を不適当なものにし得る。
dgt−32導入遺伝子に融合されたTraP14およびdgt−33導入遺伝子に融合されたTraP24を含有するトランスジェニックアラビドプシス属(Arabidopsis)植物は、米国特許出願第11975658号に記載されているように作製された。トランスジェニック植物は、異なる比率のグリホサートで噴霧された。増加した比率を含む様々な濃度のグリホサート比率の分布は、抵抗性の相対レベル(105、420、1,680、または3,360g ae/ha)を決定するために、この研究において適用された。グリホサートの典型的な1×フィールド使用率は1,120g ae/haである。この研究で使用したT1アラビドプシス属(Arabidopsis)植物は、Trap14:dgt−32およびTrap24:dgt−33導入遺伝子のコピー数において変動した。低コピー数のT1アラビドプシス属(Arabidopsis)植物は、分子確認アッセイを用いて同定され、自家受粉し、使用して、T2植物を作製した。表1は、グリホサート除草剤抵抗性遺伝子であるdgt−1および野生型対照を引用したTraP14:dgt−32およびTraP24:dgt−33アラビドプシス属(Arabidopsis)植物の比較を示す。表2は、新規な微生物EPSPシンターゼ酵素と、クラスIのEPSPシンターゼ酵素およびグリホサート比率が1,680g ae/haの対照の比較を示す。事象は、構築物pDAB107532(図6)に含まれるTraP14 v2(配列番号7)と連結しているdgt−32と構築物pDAB107534(図7)に含まれるTraP24 v2(配列番号8)と連結しているdgt−33を含む。T1植物のグリホサート選択からのデータは、dgt−32およびdgt−33がTraP14およびTraP24葉緑体輸送ペプチドと連結された場合、高レベルのグリホサートに対する強固な耐性が提供されたことを実証した。比較的に、非形質転換(または野生型)対照は、類似した比率のグリホサートで処理したとき、高濃度のグリホサートの処理に対して耐性を提供しなかった。
上記のバイナリー形質転換ベクターを含むアグロバクテリウムのグリセロールストックは、適切な抗生物質を含むAB最小培地プレート上に筋状にし、20℃にて3〜4日間で増殖させた。単一のコロニーを採取し、同じ抗生物質を含むYEPプレート上に筋状にし、28℃にて1〜2日間インキュベートした。
配列番号2は、デュナリエラ・サリナ(Dunaliella salina)由来のEPSPSペプチドのアミノ酸配列を示す。
配列番号3は、キメラTraP14融合タンパク質のアミノ酸配列を示す。
配列番号4は、コナミドリムシ(Chlamydomonas reinhardtii)由来のEPSPSペプチドのアミノ酸配列を示す。
配列番号5は、デュナリエラ・サリナ(Dunaliella salina)由来のEPSPSペプチドのアミノ酸配列を示す。
配列番号6は、キメラTraP24融合タンパク質のアミノ酸配列を示す。
配列番号7は、TraP14 v2と命名されたTraP14ペプチドをコードするヌクレオチド配列を示す。
配列番号8は、TraP24 v2と命名されたTraP24ペプチドをコードするヌクレオチド配列を示す。
Claims (52)
- 79個未満のアミノ酸長であるペプチドをコードするヌクレオチド配列を含む単離された核酸分子であって、前記ペプチドは配列番号2と少なくとも80%同一であり、対象とするヌクレオチドコード配列とインフレームで作動可能に連結している、核酸分子。
- 前記ペプチドが71個のアミノ酸長である、請求項1に記載の単離された核酸分子。
- 前記ペプチドが配列番号2と少なくとも85%同一である、請求項1に記載の単離された核酸分子。
- 前記ペプチドが配列番号2と少なくとも90%同一である、請求項1に記載の単離された核酸分子。
- 前記ペプチドが配列番号2と少なくとも95%同一である、請求項1に記載の単離された核酸分子。
- 前記ペプチドが配列番号2と少なくとも98%同一である、請求項1に記載の単離された核酸分子。
- 前記ペプチドが配列番号2である、請求項1に記載の単離された核酸分子。
- 前記対象とするヌクレオチドコード配列が、配列番号4または配列番号6である、請求項1に記載の単離された核酸分子。
- 前記ペプチドをコードする前記ヌクレオチド配列が、配列番号1と特異的にハイブリダイズ可能である、請求項1に記載の単離された核酸分子。
- 前記ペプチドをコードする前記ヌクレオチド配列の少なくとも一部が、原核生物、下等光合成真核生物、および緑色植物からなる群から選択される生物由来である、請求項1に記載の単離された核酸分子。
- 前記ペプチドをコードする前記ヌクレオチド配列の前記少なくとも一部が、デュナリエラ・サリナ(Dunaliella salina)またはコナミドリムシ(Chlamydomonas reinhardtii)由来である、請求項10に記載の単離された核酸分子。
- 前記ペプチドをコードする前記ヌクレオチド配列が、原核生物、下等光合成真核生物および緑藻植物からなる群から選択される生物に由来する、請求項10に記載の単離された核酸分子。
- 前記ペプチドをコードする前記ヌクレオチド配列がデュナリエラ・サリナ(Dunaliella salina)またはコナミドリムシ(Chlamydomonas reinhardtii)由来である、請求項12に記載の単離された核酸分子。
- 配列番号2内で連続アミノ酸配列と少なくとも80%同一である連続アミノ酸配列を含むペプチドをコードするヌクレオチド配列を含む単離された核酸分子であって、前記ペプチドは、さらに、配列番号2以外の葉緑体輸送ペプチド由来の連続アミノ酸配列と少なくとも80%同一である第2の連続アミノ酸配列を含み、前記ペプチドをコードするヌクレオチド配列は、対象とするヌクレオチド配列にインフレームで作動可能に連結している、核酸分子。
- 前記配列番号2以外の葉緑体輸送ペプチド由来の連続アミノ酸配列が、デュナリエラ(Dunailella)、アマランサス(Amaranthus)、またはコナミドリムシ(Chlamydomonas reinhardtii)由来である、請求項14に記載の単離された核酸分子。
- 前記ペプチドが、配列番号4、配列番号6、および配列番号7からなる群から選択されるアミノ酸配列を含む、請求項15に記載の単離された核酸分子。
- 前記ヌクレオチド配列が、1種または複数の調節配列に作動可能に連結している、請求項1に記載の単離された核酸分子。
- 請求項1に記載の核酸分子によってコードされるポリペプチド。
- プラスチド含有細胞中のプラスチドにターゲッティングされる、請求項18に記載のポリペプチド。
- 前記ポリペプチドがプラスチドにターゲッティングされると除去される葉緑体輸送ペプチドを含む、請求項19に記載のポリペプチド。
- 前記ポリペプチドが、葉緑体輸送ペプチドが除去された場合に残存する葉緑体ターゲッティングペプチドを含み、前記葉緑体ターゲッティングペプチドが前記対象とするヌクレオチドコード配列によってコードされている、請求項20に記載のポリペプチド。
- 前記対象とするヌクレオチドコード配列が、生物学的に活性なペプチドをコードする、請求項18に記載のポリペプチド。
- 前記対象とするヌクレオチドコード配列が、蛍光ペプチドをコードする、請求項18に記載のポリペプチド。
- 前記生物学的に活性なペプチドが、酵素である、請求項22に記載のポリペプチド。

- 前記生物学的に活性なペプチドが、ゼアキサンチンエポキシダーゼ、コリンモノオキシゲナーゼ、フェロケラターゼ、オメガ3脂肪酸不飽和化酵素、グルタミンシンセターゼ、プロビタミンA、ホルモン、Bt毒素タンパク質および対象とする形質を含む植物の同定において有用なマーカーからなる群から選択される、請求項22に記載のポリペプチド。

- トランスジェニック植物材料を製造する方法であって、

植物材料を、前記核酸分子を用いて形質転換することと
を含む、方法。 - 前記植物材料が、植物細胞、植物組織、植物組織培養物、カルス培養物、植物の一部および全植物体からなる群から選択される、請求項38に記載の方法。
- 前記植物材料が、全植物体ではない、請求項39に記載の方法。
- 請求項38に記載の方法によって製造されるトランスジェニック植物材料。
- 請求項40に記載の方法によって製造されるトランスジェニック植物材料。
- 請求項42に記載の植物材料から再生されたトランスジェニック植物。
- 請求項38に記載の植物材料から製造されたトランスジェニック植物商品生産物。
- 前記対象とするヌクレオチドコード配列が、生物学的に活性なペプチドをコードする、請求項41に記載のトランスジェニック植物材料。
- 前記生物学的に活性なペプチドが、除草剤抵抗性、ウイルス抵抗性、細菌病原体抵抗性、昆虫抵抗性、線虫抵抗性、真菌抵抗性、植物の勢い、植物の収量、温度耐性、土壌条件耐性、低光量レベル耐性、低水分レベル耐性、高水分レベル耐性、化学的環境耐性、種子の色、デンプン変性、アミノ酸合成、光合成、脂肪酸の合成、油の合成、アロテノイド(arotenoid)の合成、テルペノイドの合成、デンプンの合成および除草剤抵抗性からなる群から選択されるプロセスに関与している、請求項45に記載のトランスジェニック植物材料。

- 同種の野生型植物材料と比較した場合に、増大した除草剤抵抗性または除草剤耐性を示す、請求項48に記載のトランスジェニック植物材料。
- 植物体を生成するよう再生できない植物細胞である、請求項32に記載の植物材料。
- 前記植物材料が、植物体を生成するよう再生できない植物細胞である、請求項39に記載の方法。
