ein fluorenszierendes Protein oder Chromoprotein kodiert und aus
der Gruppe gewählt
ist, bestehend aus:
a) Nukleinsäure, die ein Protein kodiert, das eine Aminosäure-Sequenz enthält, die in SEQ ID NO: 2, 4, 6, 8, 10, 12, 14, 16, 18, 20 oder 22 gezeigt ist;
b) Nukleinsäure, die eine Nukleotid-Sequenz enthält, die in SEQ ID NO: 1, 3, 5, 7, 9, 11, 13, 15, 17, 19 oder 21 gezeigt ist;
c) Nukleinsäure, die sich unter den Bedingungen der Härte mit der Nukleinsäure (a) oder (b) hybridisiert, die oben angegeben sind;
d) Nukleinsäure, die ein Protein kodiert, das mindestens etwa eine 75%-ige Sequenz-Identität mit der oben angegebenen Aminosäüre-Sequenz (a) aufweist;
e) Nukleinsäure, die mindestens etwa eine 70%-ige Sequenz-Identität mit der oben angegebenen Nukleotid-Sequenz (b) aufweist;
f) Nukleinsäure, die ein Protein kodiert, das mindestens eine Aminosäure-Substitution, Deletion oder Insertion in der Aminosäure-Sequenz aufweist,...
a) Nukleinsäure, die ein Protein kodiert, das eine Aminosäure-Sequenz enthält, die in SEQ ID NO: 2, 4, 6, 8, 10, 12, 14, 16, 18, 20 oder 22 gezeigt ist;
b) Nukleinsäure, die eine Nukleotid-Sequenz enthält, die in SEQ ID NO: 1, 3, 5, 7, 9, 11, 13, 15, 17, 19 oder 21 gezeigt ist;
c) Nukleinsäure, die sich unter den Bedingungen der Härte mit der Nukleinsäure (a) oder (b) hybridisiert, die oben angegeben sind;
d) Nukleinsäure, die ein Protein kodiert, das mindestens etwa eine 75%-ige Sequenz-Identität mit der oben angegebenen Aminosäüre-Sequenz (a) aufweist;
e) Nukleinsäure, die mindestens etwa eine 70%-ige Sequenz-Identität mit der oben angegebenen Nukleotid-Sequenz (b) aufweist;
f) Nukleinsäure, die ein Protein kodiert, das mindestens eine Aminosäure-Substitution, Deletion oder Insertion in der Aminosäure-Sequenz aufweist,...
Claims (24)
- Isoliertes Molekül der Nukleinsäure, das ein fluorenszierendes Protein oder Chromoprotein kodiert und aus der Gruppe gewählt ist, bestehend aus: a) Nukleinsäure, die ein Protein kodiert, das eine Aminosäure-Sequenz enthält, die in SEQ ID NO: 2, 4, 6, 8, 10, 12, 14, 16, 18, 20 oder 22 gezeigt ist; b) Nukleinsäure, die eine Nukleotid-Sequenz enthält, die in SEQ ID NO: 1, 3, 5, 7, 9, 11, 13, 15, 17, 19 oder 21 gezeigt ist; c) Nukleinsäure, die sich unter den Bedingungen der Härte mit der Nukleinsäure (a) oder (b) hybridisiert, die oben angegeben sind; d) Nukleinsäure, die ein Protein kodiert, das mindestens etwa eine 75%-ige Sequenz-Identität mit der oben angegebenen Aminosäüre-Sequenz (a) aufweist; e) Nukleinsäure, die mindestens etwa eine 70%-ige Sequenz-Identität mit der oben angegebenen Nukleotid-Sequenz (b) aufweist; f) Nukleinsäure, die ein Protein kodiert, das mindestens eine Aminosäure-Substitution, Deletion oder Insertion in der Aminosäure-Sequenz aufweist, wie es in SEQ ID NO: 2, 4, 6, 8, 10, 12, 14, 16, 18, 20 oder 22 gezeigt ist; g) Derivat oder der Mimetik der Nukleinsäure (a), (b), (c), (d), (e) oder (f), die oben angegeben sind; h) Mutante der Nukleinsäure (a), (b), (c), (d) oder (e), die oben angegeben sind; i) Nukleinsäure, die sich von der Nukleinsäure (b), (c), (d), (e), (f), (g) oder (h) , die oben angegeben sind, infolge der Degeneration der genetischen Kode unterscheidet; j) Fragment der Nukleinsäure (a) oder (b), die oben angegeben sind, das das Peptid kodiert, mit einer Größe mindestens aus 15 Aminosäure-Resten.
- Molekül der Nukleinsäure nach Anspruch 1, worin die angegebene Nukleinsäure aus dem Organismus aus der Klasse Hydrozoa isoliert ist.
- Molekül der Nukleinsäure nach Anspruch 1, worin die angegebene Nukleinsäure aus dem Organismus aus der Subklasse Anthomedusae isoliert ist.
- Molekül der Nukleinsäure nach Anspruch 1, worin die angegebene Nukleinsäure aus der Gattung Phialidium isoliert ist.
- Vektor, der ein Molekül der Nukleinsäure nach Anspruch 1 einschließt.
- Expressierende Kassette, enthaltend (a) ein Molekül der Nukleinsäure nach Anspruch 1; und (b) Regulatorelemente für die Expression des angegebenen Moleküls der Nukleinsäure in der erwünschten Wirtzelle.
- Zelle, die ein Molekül der Nukleinsäure nach Anspruch 1, einen Vektor nach Anspruch 5 oder eine expressierende Kassette nach Anspruch 6 enthält.
- Stabile Zellinie, die ein Molekül der Nukleinsäure nach Anspruch 1, einen Vektor nach Anspruch 5 oder eine expressierende Kassette nach Anspruch 6 enthält.
- Transgenpflanze, die ein Molekül der Nukleinsäure nach Anspruch 1, einen Vektor nach Anspruch 5 oder eine expressierende Kassette nach Anspruch 6 enthält.
- Transgentier, das ein Molekül der Nukleinsäure nach Anspruch 1, einen Vektor nach Anspruch 5 oder eine expressierende Kassette nach Anspruch 6 enthält.
- Verfahren zur Herstellung eines fluorenszierenden Proteins oder eines Chromoproteins, das (a) Versorgung des Moleküls der Nukleinsäure nach Anspruch 1, das mit den passenden, die Expression regelnden Elementen operativ verbunden ist, (b) Expression des fluorenszierenden Proteins oder Chromoproteins aus der angegebenen Molekül der Nukleinsäure und (c) isolierung des Proteins umfaßt, der im wesentlichen keine anderen Proteine enthält.
- Molekül der Nukleinsäure, enthaltend ein Molekülfragment der Nukleinsäure nach Anspruch 1, worin das angegebene Fragment das Peptid kodiert, mit einer Größe von mindestens 100 Aminosäuren.
- Molekül der Nukleinsäure, das eine Sequenz aufweist, die im wesentlichen der Nikleotid-Sequenz gleich oder mit dieser identisch ist, die mindestens aus 300 Molekülresten der Nukleinsäure nach Anspruch 1 besteht.
- Isoliertes fluorenszierender Protein oder Chromoprotein, das aus der Gruppe gewählt ist, bestehend aus: a) Protein, das eine Aminosäure-Sequenz umfaßt, die in SEQ ID NO: 2, 4, 6, 8, 10, 12, 14, 16, 18, 20 oder 22 gezeigt ist; b) Protein, das bei Molekül der Nukleonsäure kodiert wird, das eine Nukleotid-Sequenz enthält, die in SEQ ID NO: 1, 3, 5, 7, 9, 11, 13, 15, 17, 19 oder 21 gezeigt ist; c) Protein, der mindestens eine etwa 75% -ige Sequenzidentität zur Aminosäure-Sequenz (a) oder (b) aufweist, die oben angegeben sind; d) Mutante vom Protein (a), (b) oder (c), die oben angegeben sind; e) Protein, der mindestens eine Aminosäure-Substitution, Deletion oder Insertion in der Aminosäure-Sequenz aufweist, wie es in SEQ ID NO: 2, 4, 6, 8, 10, 12, 14, 16, 18, 20 oder 22 gezeigt ist; f) Derivat des Proteins (a), (b), (c), (d) oder (e), die oben angegeben sind; g) Fragment des Proteins (a), (b), (c), (d), (e) oder (f), die oben angegeben sind, das in sich mindestens 15 Aminosäure-Reste enthält; und h) Protein, das eine Sequenz aufweist, die im wesentlichen der Aminosäure-Sequenz gleich oder mit dieser identisch ist, die mindestens aus 100 Resten, (a) oder (b), besteht, die oben angegeben sind.
- Fusionsprotein, das den Protein nach Anspruch 14 enthält.
- Antikörper, der sich mit dem Protein nach Anspruch 14 spezifisch verbindet.
- Satz, der die Nukleinsäure nach Anspruch 1, den Vektor nach Anspruch 5, eine expressierende Kassette nach Anspruch 6, das Protein nach Anspruch 14, den Fusionsprotein nach Anspruch 15 oder Mittel zur Herstellung derselben enthält.
- Oligonukleotidsonde oder Primer, enthaltend eine Nukleotid-Sequenz, die zur Hybridisation mit der Nukleotid-Sequenz fähig ist, die aus der Gruppe gewählt ist, welche aus SEQ ID NO: 1, 3, 5, 7, 9, 11, 13, 15, 17, 19 oder 21 besteht.
- Verfahren zur Markierung eines biologischen Moleküls, das das Binden des angegebenen biologischen Moleküls mit dem Protein nach Anspruch 14 einschließt.
- Verfahren zur Markierung einer Zelle, das das Produzieren des Proteins nach Anspruch 14 in der Zelle einschließt.
- Verfahren zur Markierung einer Zellenorganelle, das das Produzieren des Proteins nach Anspruch 14 einschließt, der mit dem passenden intrazellulären Signal der Lokalisationen in der Zelle fusioniert ist.
- Verfahren zur Analyse von dem biologischen Molekül, der Zelle oder der Zellenorganelle, das die Detektion des Fluoreszenzsignals des Proteins nach Anspruch 14 oder 15 einschließt.
- Verfahren zur Analyse von dem biologischen Molekül, der Zelle oder der Zellenorganelle, das die Expression des Moleküls der Nukleinsäure nach Anspruch 1 in der Zelle einschließt.
- Verfahren zur Detektion vom biologischen Molekül, das die Detektion des Fluoreszenzsignals des Proteins nach Anspruch 14 oder 15 einschließt.
