Claims (43)
- 以下によってコードされるヒトCD38に結合する抗体
(i)それぞれ、配列番号:1および配列番号:6に示されたようなそれらの可変領域におけるヌクレオチド配列を含むヒト軽鎖核酸およびヒト重鎖核酸;または
(iii)それぞれ、配列番号:21および配列番号:26に示されたようなそれらの可変領域におけるヌクレオチド配列を含むヒト軽鎖核酸およびヒト重鎖核酸。 - 配列番号:3に示されたような配列を有するVL CDR1、配列番号:4に示されたような配列を有するVL CDR2、および配列番号:5に示されたような配列を有するVL CDR3を含むヒト軽鎖可変領域と、配列番号:8に示されたような配列を有するVH CDR1、配列番号:9に示されたような配列を有するVH CDR2、および配列番号:10に示されたような配列を有するVH CDR3を含むヒト重鎖可変領域とを含む、ヒトCD38に結合する抗体。
- 配列番号:2に示されたようなアミノ酸配列を有するVL領域を含む、請求項2記載の抗体。
- 配列番号:7に示されたようなアミノ酸配列を有するVH領域を含む、請求項2または3記載の抗体。
- 配列番号:23に示されたような配列を有するVL CDR1、配列番号:24に示されたような配列を有するVL CDR2、および配列番号:25に示されたような配列を有するVL CDR3を含むヒト軽鎖可変領域と、配列番号:28に示されたような配列を有するVH CDR1、配列番号:29に示されたような配列を有するVH CDR2、および配列番号:30に示されたような配列を有するVH CDR3を含むヒト重鎖可変領域とを含む、ヒトCD38に結合する抗体。
- 配列番号:22に示されたようなアミノ酸配列を有するVL領域を含む、請求項5記載の抗体。
- 配列番号:27に示されたようなアミノ酸配列を有するVH領域を含む、請求項5または6記載の抗体。
- 以下の結合特徴を保有する、ヒトCD38(配列番号:31)に結合するヒトモノクローナル抗体:
(i)それがヒトCD38(配列番号:31)に結合するのと同じ程度に、位置274のセリン残基がフェニルアラニン残基で置換された、突然変異体ヒトCD38(配列番号:34)に結合し、
(ii)それがヒトCD38(配列番号:31)に結合するのと同じ程度に、位置272のグルタミン残基がアルギニン残基で置換された、突然変異体ヒトCD38(配列番号:33)に結合し、および
(iii)それがヒトCD38(配列番号:31)に結合するのと同じ程度に、位置237のスレオニン残基がアラニン残基で置換された、突然変異体ヒトCD38(配列番号:32)に結合する。 - CD38との結合について、請求項1記載の抗体と競合するヒトモノクローナル抗体。
- エピトープが請求項1記載の抗体によっても特異的に結合される、CD38エピトープに特異的に結合するヒトモノクローナル抗体。
- ヒトCD38(配列番号:31)の領域SKRNIQFSCKNIYRおよび領域EKVQTLEAWVIHGGに特異的に結合するヒトモノクローナル抗体。
- ヒトCD38(配列番号:1)に結合し、かつカニクイザルリンパ系組織と交差反応する、ヒトモノクローナル抗体。
- それが、IgG1抗体、好ましくはIgG1、κ抗体、またはIgM抗体、好ましくはIgM、κ抗体などの、全長IgG1、IgG2、IgG3、IgG4、IgD、IgA、IgE、またはIgM抗体であるという点で特徴付けられる、請求項1〜12のいずれか一項記載の抗体。
- 抗体断片または単鎖抗体である、請求項1〜13のいずれか一項記載の抗体。
- 実質的に単離された形態にある、請求項1〜14のいずれか一項記載の抗体。
- 請求項1〜15のいずれか一項記載の抗体を産生する真核宿主細胞または原核宿主細胞。
- 任意で、細胞毒性薬剤、放射性同位元素、または薬物に結合した単量体IgM抗体である、細胞毒性薬剤、放射性同位元素、または薬物に結合した請求項1〜15のいずれか一項記載の抗体を含む免疫コンジュゲート。
- 請求項1〜15のいずれか一項記載の抗体およびヒトエフェクター細胞に対する結合特異性を含む二重特異性分子または多重特異性分子であって、任意で、該結合特異性がCD3、CD4、CD138、IL-15R、膜結合TNF-αもしくは受容体結合TNF-α、ヒトFc受容体、または膜結合IL-15もしくは受容体結合IL-15に対するものである、前記二重特異性分子または多重特異性分子。
- 請求項1〜15のいずれか一項記載の抗体または請求項17記載の免疫コンジュゲートおよび薬学的に許容される担体を含む薬学的組成物。
- CD38を発現する細胞の成長および/または増殖を阻害するための医薬の製造における、請求項1〜15のいずれか一項記載の抗体、請求項17記載の免疫コンジュゲート、または請求項19記載の薬学的組成物の使用。
- 対象におけるCD38を発現する細胞が関与する疾患または障害を治療または予防するための医薬の製造における、請求項1〜15のいずれか一項記載の抗体、請求項17記載の免疫コンジュゲート、または請求項19記載の薬学的組成物の使用。
- 疾患または障害が関節リウマチである、請求項20または21記載の使用。
- 疾患または障害が多発性骨髄腫である、請求項20または21記載の使用。
- 前記医薬が、1つまたは複数のさらなる治療薬剤を含む、請求項20〜23のいずれか一項記載の使用であって、任意で、該1つまたは複数のさらなる治療薬剤が、化学療法薬剤、抗炎症薬剤、または免疫抑圧薬剤および/もしくは免疫調整薬剤より選択される、前記使用。
- 1つまたは複数のさらなる治療薬剤が、シスプラチン、ゲフィチニブ、セツキシマブ、リツキシマブ、ベバシズマブ、エルロチニブ、ボルテゾミブ、サリドマイド、パミドロネート、ゾレドロン酸、クロドロネート、リセンドロネート、イバンドロネート、エチドロネート、アレンドロネート、チルドロネート、三酸化ヒ素、レナリドマイド、フィルグラスチム、ペグフィルグラスチム、サルグラモスチム、スベロイラニリドヒドロキサム酸、およびSCIO-469からなる群より選択される、請求項24記載の方法。
- 試料における、CD38抗原の存在、またはCD38を発現する細胞を検出するためのインビトロ法であって、以下の工程を含む方法:
a)抗体とCD38の間の複合体の形成を可能にする条件下で、試料を請求項1〜15のいずれか一項記載の抗体と接触させる工程;および
b)複合体の形成を検出する工程。 - 対象における、CD38抗原、またはCD38を発現する細胞を検出するためのインビボ法であって、以下の工程を含む方法:
a)抗体とCD38の間の複合体の形成を可能にする条件下で、請求項1〜15のいずれか一項記載の抗体を投与する工程;および
b)形成された複合体を検出する工程。 - 請求項1〜15のいずれか一項記載の抗体に結合する抗イデオタイプ抗体。
- ヒトCD38(配列番号:31)に結合し、および
それがヒトCD38(配列番号:31)に結合するのと同じ程度には、位置274のセリン残基がフェニルアラニン残基で置換された、突然変異体ヒトCD38(配列番号:34)に結合しない、抗体。 - 配列番号:20に示されたような配列を有するV H CDR3を含む、ヒトCD38に結合する抗体である、請求項29記載の抗体。
- 配列番号:15に示されたような配列を有するV L CDR3、および配列番号:20に示されたような配列を有するV H CDR3を含む、請求項30記載の抗体。
- 配列番号:17に示されたようなアミノ酸配列を有するV H 領域を含む、ヒトCD38に結合する抗体である、請求項29記載抗体。
- 位置274のセリン残基がフェニルアラニン残基で置換された、突然変異体ヒトCD38(配列番号:34)への抗体の結合のEC 50 が、ヒトCD38(配列番号:31)への抗体の結合のEC 50 の10%未満、5%未満、または1%未満などの、50%未満である、請求項29記載の抗体。
- ヒトCD38(配列番号:31)に結合するのと同じ程度には、位置272のグルタミン残基がアルギニン残基で置換された、突然変異体ヒトCD38(配列番号:33)に結合しない、請求項29記載の抗体。
- 位置272のグルタミン残基がアルギニン残基で置換された、突然変異体ヒトCD38(配列番号:33)への抗体の結合のEC 50 が、ヒトCD38(配列番号:31)への抗体の結合のEC 50 の10%未満、5%未満、または1%未満などの、50%未満である、請求項34記載の抗体。
- それがヒトCD38(配列番号:31)に結合するのと同じ程度に、位置237のスレオニン残基がアラニン残基で置換された、突然変異体ヒトCD38(配列番号:32)に結合する、請求項29〜35のいずれか一項記載の抗体。
- 位置237のスレオニン残基がアラニン残基で置換された、突然変異体ヒトCD38(配列番号:32)への抗体の結合のEC 50 が、ヒトCD38(配列番号:31)への抗体の結合のEC 50 の75%〜125%に相当する、請求項36記載の抗体。
- 以下の結合特徴を保有する、ヒトCD38(配列番号:31)に結合する抗体:(i)それがヒトCD38(配列番号:31)に結合するのと同じ程度には、位置274のセリン残基がフェニルアラニン残基で置換された、突然変異体ヒトCD38(配列番号:34)に結合せず、(ii)それがヒトCD38(配列番号:31)に結合するのと同じ程度には、位置272のグルタミン残基がアルギニン残基で置換された、突然変異体ヒトCD38(配列番号:33)に結合せず、(iii)それがヒトCD38(配列番号:31)に結合するのと同じ程度に、位置237のスレオニン残基がアラニン残基で置換された、突然変異体ヒトCD38(配列番号:32)に結合する。
- 本明細書の実施例17に記載されたようなELISAの使用によってEC 50 を決定する、請求項33、35、または37のいずれか一項記載の抗体。
- それぞれ、配列番号:11および配列番号:16に示されたようなそれらの可変領域におけるヌクレオチド配列を含むヒト軽鎖核酸およびヒト重鎖核酸によってコードされるヒトCD38に結合する抗体と、CD38との結合について競合する、請求項29記載の抗体。
- 本明細書の実施例8または9に記載されたようなELISAの使用によって競合を決定し、吸収によって、または本明細書の実施例7に記載されたような交差遮断(cross-blocking)測定の使用によって評価されるような少なくとも90%のシグナルによって競合を定義し、蛍光によって評価されるような少なくとも90%のシグナルによって競合を定義する、請求項40記載の抗体。
- エピトープが、それぞれ、配列番号:11および配列番号:16に示されたようなそれらの可変領域におけるヌクレオチド配列を含むヒト軽鎖核酸およびヒト重鎖核酸によってコードされるヒトCD38に結合する抗体によっても特異的に結合される、CD38エピトープに特異的に結合する抗体。
- ヒトCD38(配列番号:31)の領域SKRNIQFSCKNIYRおよび領域EKVQTLEAWVIHGGに特異的に結合する、請求項29記載の抗体。
