Description
Die Erfindung betrifft Zwischenprodukte, die zur Herstellung von einer neuen Klasse von ß-Lactam-Antibiotika verwendbar sind. Diese ß-Lactam-Antibiotika, die wertvolle Arzneistoffe mit Wirkung gegen gram-negative und gram-po-sitive Keime sind, sind im Schweizer Patent Nr. 651 020 beschrieben.
ß-Lactame mit einer Sulfonsäuregruppe in der freien Säureform oder in der Salzform, einschliesslich innerer Salze, in der 1-Stellung und einer Acylaminogruppe in der 3-Stellung sind Antibiotika mit Wirkung gegen gram-negative und gram-positive Keime.
Die bevorzugten oben erwähnten ß-Lactam-Antibiotika haben die allgemeine Formel I

Zusätzlich zu den vorstehend beschriebenen ß-Lactamen mit einer Sulfonsäuregruppe in der Säureform oder Salzform in der 1-Stellung und einer Acylaminogruppe in der 3-Stel-lung betrifft die Erfindung auch ß-Lactame mit einer Sulfonsäuregruppe in der Säureform oder Salzform (einschliesslich innerer Salze) in der 1-Stellung sowie einer Amino- oder Azi-dogruppe in der 3-Stellung. Die bevorzugten Verbindungen dieses Typs haben die allgemeine Formel Ia
In den allgemeinen Formeln I und Ia und in der Beschreibung haben die Substituenten folgende Bedeutung:
Ri ist ein Acylrest,
R2 ein Wasserstoffatom oder ein C^-Alkoxyrest, 5 R3 und R4, die gleich oder verschieden sind, bedeuten Wasserstoffatome, Alkyl- oder Cycloalkylreste, Phenyl- oder substituierte Phenylgruppen oder einer der Reste R3 und R4 bedeutet ein Wasserstoffatom und der andere Rest eine Alkoxycarbonyl-, Alken -1- yl-, Alkin -1- yl-, 2-Phenyläthenyl-10 oder 2-Phenyläthinylgruppe.
M® bedeutet ein Wasserstoffatom oder ein Kation.
Als Alkyl- und Alkoxyreste kommen unverzweigte und verzweigte Reste in Frage. Reste mit 1 bis 10 Kohlenstoffatomen sind bevorzugt. Als Cycloalkyl- und Cycloalkenylreste 15 kommen Reste mit 3 bis 7 Kohlenstoffatomen in Frage. Der Ausdruck «Alkenylrest» bedeutet unverzweigte oder verzweigte Reste. Reste mit 2 bis 10 Kohlenstoffatomen sind bevorzugt. Als Halogenatome kommen Fluor-, Chlor-, Chlor-, Brom- und Jodatome in Frage. Der Ausdruck substituierte 20 Phenylgruppe bedeutet eine durch bis zu drei Halogenatome oder Amino-, Hydroxyl-, Trifluormethyl-, niederer Alkyl-oder niederer Alkoxygruppen substituierte Phenylgruppe.
Der Ausdruck «geschützte Carboxylgruppe bedeutet eine Carboxylgruppe, die durch eine übliche Esterschutzgruppe 25 verestert worden ist. Die Gruppen sind an sich bekannt; vgl. z.B. US-PS 4 144 333. Die bevorzugten Carboxylschutzgrup-pen sind die Benzyl-, Benzhydryl- und tert.-Butylgruppe.
Der Ausdruck «Acylrest» umfasst sämtliche Reste die sich von einer organischen Säure, d.h. einer Carbonsäure, durch 30 Abspaltung der Hydroxylgruppe ableiten. Bestimmte Acyl-gruppen sind bevorzugt, doch soll dies hier keine Einschränkung der Erfindung bedeuten. Beispiele für Acylgruppen sind solche Reste, wie sie bisher zur Acylierung von ß-Lactam-Antibiotika, einschliesslich 6-Aminopenicillansäure und deren 35 Derivaten sowie 7-Aminocephalosporansäure und deren Derivaten, verwendet wurden; vgl. z.B. Cephalosporins and Penicillins, herausgegeben von H. Flynn Academic Press (1972), DE-OS 2 716 677, BE-PS 867 994, US-PSen 4 152 432, 3 971 778 und 4 172 199 sowie GB-PS 1 348 894. Auf den In-40 halt dieser Druckschriften, soweit er verschiedene Acylreste beschreibt, wird hier ausdrücklich Bezug genommen. Die nachstehend listenmässig angegebenen Acylreste dienen lediglich zur weiteren Erläuterung des Ausdrucks Acylrest. Bevorzugte, durch aromatische Reste substituierte Acylreste haben die Formel in der n den Wert 0,1,2 oder 3 hat, R9 die vorstehende Bedeutung hat und R10 einen unsubstituierten oder substituierten 5-, 6- oder 7-gliedrigen heterocyclischen Ring mit einem, 20 zwei, drei oder vier, vorzugsweise einem oder zwei Stickstoffatomen, Sauerstoffatomen oder Schwefelatomen ist. Spezielle Beispiele für heterocyclische Reste sind die Thienyl-, Furyl-, Pyrrolyl-, Pyridinyl-, Pyrazinyl-, Thiazolyl-, Morpholinyl-, Pyrimidinyl- und Tetrazolylgruppe. Spezielle Beispiele für 25 Substituenten sind Halogenatome, Hydroxy-, Nitro-, Amino-, Cyano oder Trifluormethylgruppen, CM-Alkyl- und Q 4-Alkoxyreste.
Bevorzugte, durch heteroaromatische Reste substituierte Acylgruppen sind die Gruppen der vorstehend angegebenen 3o allgemeinen Formeln, in denen R10 eine 2-Amino -4- thiazolyl-, 2-Amino -5- halogen -4- thiazolyl-, 4-Aminopyrimidin-2-yl-, 5-Amino-l,2,4-thiadiazolyl -5- yl-, 2-Thienyl- oder 2-Fur-anylgruppe ist;

Der Ausdruck «Kation» bedeutet hier jedes positiv geladene Atom oder jede positiv geladene Atomgruppe. Der Rest -SOe3M® am Lactam-Stickstoffatom umfasst sämtliche
Salze der Sulfonsäure. Bevorzugt sind pharmakologisch verträgliche Salze, doch sind auch andere Salze brauchbar zur Reinigung der Verbindungen der Erfindung oder als Zwischenprodukte zur Herstellung pharmakologisch verträglicher Salze. Der kationische Teil der Sulfonsäurcsalze kann sich von organischen oder anorganischen Basen ableiten. Beispiele für den kationischen Teil sind das Ammoniumion, substituierte Ammoniumionen, z.B. Alkylammoniumionen, beispielsweise das Tetra-n-butylammoniumion, nachstehend kurz als Tetrabutylammoniumion bezeichnet, Alkalimetallkationen wie Lithium-, Natrium- und Kaliumionen, Erdalkalimetallkationen, wie Calcium- und Magnesiumionen, das Pyridinium-, Dicyclohexylammonium-, Hydrabaminium-, Benzathinium- und N-Methyl-D-glucaminiumion.
M® kann auch ein Wasserstoffatom bedeuten.
Die oben erwähnten ß-Lactame können auf chemischem Wege hergestellt werden. Die nicht-alkoxylierten, 4-unsubsti-tuierten ß-Lactame der allgemeinen Formel I, d.h. die Verbindungen der allgemeinen Formel I, in der R2, R3 und R4 Wasserstoffatome bedeuten, können aus 6-Aminopenicillansäure oder einer 6-Acylaminopenicillansäure hergestellt werden. Die ß-Lactame der allgemeinen Formel I, in der R2 einen Alk-oxyrest bedeutet, können aus den entsprechenden nicht-alk-oxylierten ß-Lactamen hergestellt werden. Einige der Verbindungen der Erfindung können aus wasserhaltigen Lösungsmitteln zur Kristallisation gebracht oder umkristallisiert werden. In diesen Fällen können die Produkte als Hydrate anfallen. Die Erfindung umfasst die stöchiometrischen Hydrate sowie Verbindungen, die unterschiedliche Mengen an Wasser enthalten, die beispielsweise durch Gefriertrocknen erhalten werden.
Einige der ß-Lactame der allgemeinen Formel I können auch auf fermentativem Wege hergestellt werden. Bei der Züchtung eines Stammes von Chromobacterium violaceum SC 11 378 wird ein Salz der (R) -3- (Acetylamino) -3- meth-oxy -2- oxo -1- azetidinsulfonsäure gebildet. Die Züchtung verschiedener Essigsäurebakterien, wie Arten von Glucono-bacter SC 11 435, liefert ein Salz der (R) -3[[N-(D-y-Glut-amyl)-D-alanyl]-amino] -3- methoxy -2- oxo -1- azetidinsulfonsäure.
ß-Lactame mit einer Sulfonsäuregruppe in der 1-Stellung und einer Amino- oder Acylaminogruppe in der 3-Stellung enthalten mindestens ein chirales Zentrum, nämlich das Kohlenstoffatom in der 3-Stellung, an das die Amino- oder Acylaminogruppe gebunden ist. Die Erfindung betrifft die ß-Lactame, deren Stereochemie am chiralen Zentrum in der 3-Stellung des ß-Lactamringes die gleiche ist wie die Konfiguration des Kohlenstoffatoms in der 6-Stellung der natürlich vorkommenden Penicilline, wie Penicillin G, und die Konfiguration am Kohlenstoffatom in der 7-Stellung der natürlich vorkommenden Cephamycine, wie Cephamycin C.
Im Hinblick auf die bevorzugten ß-Lactame der allgemeinen Formel I und Ia wurden die Strukturformeln derart gezeichnet, dass die Stereochemie am chiralen Zentrum in der 3-Stellung ersichtlich ist. Nach den derzeitigen Regeln der Nomenklatur werden diejenigen Verbindungen der allgemeinen Formel I und Ia, in denen R2 ein Wasserstoffatom bedeutet, mit der S-Konfiguration und diejenigen Verbindungen der allgemeinen Formel I und Ia, in denen der Rest R2 einen Alkoxyrest bedeutet, mit der R-Konfiguration bezeichnet.
ß-Lactame mit einem Sulfonsäurerest in der 1-Stellung des ß-Lactamrings und einer Aminogruppe oder einer Acylaminogruppe in der 3-Stellung des ß-Lactamrings zeigen Aktivität gegen die verschiedensten gram-negativen und gram-positiven Keime. Die Sulfonsäuregruppe ist für die Aktivität der Verbindungen der Erfindung von entscheidender Bedeutung. ß-Lactame der vorstehend angegebenen allgemeinen Formeln, in denen der Rest R3 und/oder R4 ein Wasserstoffatom

oder einen Alkylrest, insbesondere eine Methylgruppe bedeutet, zeigen eine besonders günstige Aktivität.
Die Verbindungen der Formel (I) können zur Bekämpfung bakterieller Infektionen, einschliesslich Infektionen der 5 Gallenwege und der Atemwege eingesetzt werden, und zwar oral oder parenteral, beispielsweise intravenös oder intramuskulär oder in Form von Suppositorien.
Die ß-Lactame der Formel (I) werden in der Regel durch Einführung einer Sulfonsäuregruppe (einer Sulfogruppe 10 -SO3- an das Stickstoffatom in der 1-Stellung des ß-Lactam-rings hergestellt. Diese Sulfonierung lässt sich leicht durch Behandlung mit einem Schwefeltrioxidkomplex oder mit einem äquivalenten Sulfonierungsmittel, beispielsweise einem Chlorsulfonat, erreichen.
15 Die üblichsten Schwefeltrioxidkomplexe für Sulfonie-rungsreaktionen sind die Komplexe mit Pyridin, Lutidin, Pi-colin und Dimethylformamid. Anstelle eines vorgebildeten Komplexes kann der Komplex auch in situ gebildet werden, beispielsweise durch Verwendung eines Chlorsulfonyltri-20 methylsilylesters und Pyridin als Reagentien. Alternativ kann die Sulfonierung auch über ein Zwischenprodukt bewirkt werden, beispielsweise dadurch, dass man zunächst das Lac-tam-Stickstoffatom silyliert und anschliessend die erhaltene silylierte Verbindung einer Silyl-Austauschreaktion mit Tri-25 methylsilylchlorsulfonat oder einem ähnlichen Reagenz unterwirft. Beispiele für Silylierungsmittel sind Monosilyltriflu-oracetamid, Trimethylsilylchlorid/Triäthylamin und Bis-tri-methylsilyltrifluoracetamid.
Im allgemeinen wird die Sulfonierung in Gegenwart eines 30 organischen Lösungsmittels, wie Pyridin, oder eines Gemisches von organischen Lösungsmitteln, vorzugsweise eines Gemisches aus einem polaren Lösungsmittel, wie Dimethylformamid, und einem halogenierten Kohlenwasserstoff, wie Dichlormethan, durchgeführt.
35 Das zunächst bei der Sulfonierung gebildete Produkt ist ein Salz des sulfonierten ß-Lactams. Bei Verwendung von Py-ridin-Schwefeltrioxid fällt als Produkt zunächst als Pyridini-umsalz der ß-Lactam -1- sulfonsäure an. M® bedeutet also das Pyridiniumion. Das Produkt hat die Teilstruktur

Diese Salze oder Komplexe können nach üblichen Methoden in Salze anderer Sulfonsäuren überführt werden, beispielsweise durch Behandlung mit Ionenaustauscherharzen, Kristallisation oder Ionenpaarextraktion. Diese Methoden so eignen sich auch zur Reinigung der Produkte. Besonders geeignet ist die Umwandlung des Pyridiniumsalzes in das Kaliumsalz mittels Kaliumphosphat oder Kalium-äthylhexanoat, in das Tetrabutylammoniumsalz mittels Tetrabutylammoni-umhydrogensulfat oder in ein Zwitterion (Betain) (M® = 55 Wasserstoff) mit Ameisensäure.
Die Sulfonierung zur Einführung der Sulfonsäuregruppe an das Lactam-Stickstoffatom kann auf verschiedenen Herstellungsstufen erfolgen, einschliesslich der Einführung vor der Bildung des ß-Lactamringes. Die Verfahren werden nach-60 stehend erläutert. Die Sulfonierungsreaktion wird vorzugsweise in Gegenwart eines Lösungsmittels der vorstehend beschriebenen Art und gewöhnlich bei Raumtemperatur durchgeführt. Sofern in der eingesetzten Verbindung eine Aminogruppe vorliegt, wird diese vorzugsweise geschützt. Unter fis Verwendung eines beispielsweise durch eine Benzyloxycarbo-nylgruppe geschützten 3-Amino-ß-lactams der nachstehend angegebenen allgemeinen Formel II wird die Sulfonierungsreaktion nach folgendem Reaktionsschema durchgeführt.:

Zum Schutz der Aminogruppe können natürlich auch andere Schutzgruppen verwendet werden, beispielsweise eine tert.-Butyloxycarbonylgruppe, einfache Acylreste, wie die Acetyl-, Benzoyl- oder Phenylacetylgruppe. Ferner kann die Aminogruppe durch eine Triphenylmethylgruppe blockiert sein, schliesslich kann die Aminogruppe in Form einer Azi-dogruppe vorliegen, die auf einer späteren Stufe zur Aminogruppe reduziert wird. Nach Abspaltung der Schutzgruppe oder der Umwandlung der Azidogruppe in die Aminogruppe lässt sich das Molekül in üblicher Weise acylieren. Die Acylie-rung einer Verbindung der allgemeinen Formel III kann beispielsweise mittels einer Carbonsäure (Rj-OH) oder dem entsprechenden Carbonsäurehalogenid oder Carbonsäureanhydrid erfolgen. Wenn der Rest R2 einen Alkoxyrest darstellt,
wird die Acylierung vorzugsweise mit einem Säurechlorid oder Säurebromid durchgeführt. Die Acylierung mit einer Carbonsäure verläuft glatt in Gegenwart eines Carbodiimids, 15 wie Dicyclohexylcarbodiimid, und einer Verbindung, die in situ einen aktiven Ester bildet, wie N-Hydroxybenzotriazol. Wenn der Acylrest Rj eine reaktionsfähige funktionelle Gruppe enthält, beispielsweise eine Amino- oder Carboxylgruppe, kann es erforderlich sein, diese Gruppen zunächst zu 20 schützen, sodann die Acylierung durchzuführen und schliesslich die Schutzgruppen aus dem erhaltenen Produkt abzuspalten. Alternativ kann die Sulfonierung auch bereits mit dem 3-Acylamino-ß-lactam der allgemeinen Formel IV nach folgendem Reaktionsschema durchgeführt werden:

Die Abspaltung der Schutzgruppe aus der 1-Stellung der Verbindungen der allgemeinen Formel XVI kann durch Reduktion mit Natrium erfolgen, wenn der Rest Y einen Alkylrest darstellt. Es wird ein Zwischenprodukt der allgemeinen 5 Formel XVII

Sodann wird die Hydroxylgruppe durch Behandlung mit üblichen Reagenzien, wie Methansulfonylchlo-rid (Methansulfonyl wird nachstehend kurz als Ms bezeichnet) in eine Leaving-Gruppe V verwandelt. Andere Leaving-Gruppen sind die Benzolsulfonyl- oder Toluolsulfonylgruppe oder das Chlor-, Brom- oder Jodatom.
Die vollständig geschützte Verbindung der allgemeinen Formel XV
erhalten, in der mindestens einer der Reste R3 und R4 ein Wasserstoffatom bedeutet. Wenn der Rest Y eine Benzyl-15 gruppe darstellt, wird bei der katalytischen Hydrierung beispielsweise mit Palladium-auf-Kohlenstoff, zunächst die entsprechende N-Hydroxyverbindung erhalten, die nach Behandlung mit Titantrichlorid das Zwischenprodukt der allgemeinen Formel XVII liefert, in der mindestens einer der Reste 20 R3 und R4 ein Wasserstoffatom bedeutet.
Die Cyclisierungsreaktion des vorstehend beschriebenen Typs hat eine Inversion der stereochemischen Konfiguration der Reste R3 und R4 zur Folge.
Wie vorstehend bereits erwähnt, lässt sich das ß-Lactam 25 (Azetidinon) auch zu einer Verbindung der allgemeinen Formel XVIII

Die Cyclisierung der Verbindungen der allgemeinen Formel XXI wird mit einer Base, wie Kaliumcarbonat, durchgeführt. Die Umsetzung wird vorzugsweise in einem Gemisch von Wasser und einem organischen Lösungsmittel, z.B. einem halogenierten Kohlenwasserstoff, wie 1,2-Dichloräthan, unter Rückfluss durchgeführt. Es werden Verbindungen der allgemeinen Formel XXII
erneuten Abkühlen auf — 5 °C werden 59,5 g 1-Threonin zugegeben, und das Gemisch wird auf Raumtemperatur er-wärmt und 16 Stunden gerührt. Danach wird das Reaktions-(XXI) gemisch eingedampft und 2 Stunden bei einem Druck von
510—1 Torr evakuiert. Es hinterbleibt ein viskoses Öl. Das Produkt wird unmittelbar in die nächste Stufe eingesetzt.
B) Threoninamid
Das Rohprodukt von Stufe (a) wird in 2,5 Liter Methanol io gelöst und auf — 5 °C in einem Eis-Kochsalzbad abgekühlt. Die erhaltene Lösung wird mit Ammoniakgas gesättigt. Danach wird das Kühlbad entfernt, das Reaktionsgefäss verschlossen und 3 Tage stehengelassen. Hierauf wird der grösste Teil des nicht umgesetzten Ammoniaks an der Wasserstrahl-is pumpe entfernt. Der Rückstand wird mit 100 g Natriumbi-carbonat und 50 ml Wasser versetzt und das Gemisch eingedampft. Es hinterbleibt ein viskoses Öl.

überführt werden, in der mindestens einer der Reste R3 und R4 ein Wasserstoffatom ist. Dies kann durch Behandlung mit einer Säure, wie Ameisensäure, erreicht werden.
Wenn die Schutzgruppe A eine Boc-Gruppe ist, kann die Abspaltung dieser Schutzgruppe aus den Verbindungen der allgemeinen Formel XXII durch Behandlung mit einer Säure, wie Ameisensäure, erfolgen. Es wird das entsprechende Zwitterion der allgemeinen Formel XXIV erhalten.
Das Beispiel erläutert die Erfindung.
Beispiel
(3S-trans) -3- Amino -4- methyl -2- oxo -1- azetidinsülfonsäure A) Threoninmethylester-hydrochlorid
In einem Kolben werden 500 ml Methanol vorgelegt und auf - 5 °C in einem Eis-Kochsalzbad abgekühlt. Unter Stickstoff als Schutzgas werden 130 ml Thionylchlorid in derartiger Geschwindigkeit eingetragen, dass die Temperatur des Reaktionsgemisches zwischen 0 und 10 °C bleibt. Nach dem

20 Das Rohprodukt der Stufe (B), das bereits die erforderliche Menge Natriumbicarbonat enthält, wird mit Wasser auf 1 Liter verdünnt. Danach wird die Lösung unter starkem Rühren innerhalb 1 Stunde mit einer Lösung von 94 g (88 ml 90prozentig reines Material) Benzyloxycarbonylchlorid in 25 80 ml Tetrahydrofuran versetzt. Danach wird das Reaktionsgemisch weitere 16 Stunden gerührt und hierauf zunächst mit 500 ml und danach zweimal mit jeweils 250 ml Äthylacetat extrahiert. Die Äthylacetatextrakte werden vereinigt, über Magnesiumsulfat getrocknet und eingedampft. Der erhaltene 30 kristalline Rückstand wird in 250 ml heissem Äthylacetat gelöst, mit 300 ml Hexan versetzt und aufgekocht, bis eine klare Lösung erhalten wird. Nach dem Abkühlen werden die entstandenen Kristalle abfiltriert und getrocknet. Es werden 104 g der Titelverbindung erhalten.

D) Benzyloxycarbonylthreoninamid -0- mesylat
Unter Argon als Schutzgas werden 100 g Benzyloxycarbonylthreoninamid in 400 ml wasserfreiem Pyridin gelöst und in einem Eis-Kochsalzbad abgekühlt. Danach wird die erhal-40 tene Lösung unter Rühren innerhalb 15 Minuten mit 36,8 ml (54,5 g) Methansulfonylchlorid versetzt. Das Gemisch wird weitere 2 Stunden gerührt und danach nochmals mit 0,3 Äquivalenten Methansulfonylchlorid versetzt. Das Reaktionsgemisch wird 1 Stunde gerührt, sodann in ein Gemisch 45 aus 1,5 Liter Eis und 2 Liter Wasser gegossen. Die erhaltene Aufschlämmung wird noch weitere 30 Minuten gerührt und danach filtriert. Hierauf wird das Rohprodukt etwa 16 Stunden bei 60 °C in einem Vakuumtrockenschrank getrocknet. Es werden 109 g der Titelverbindung erhalten.

E) N-Sulfonylbenzyloxycarbonylthreoninamid -0- mesylat-Tetrabutylammoniumsalz
Eine Lösung von 17,8 ml 2-Picolin in 90 ml Methylenchlorid wird auf - 5 °C in einem Eis-Kochsalzbad abgekühlt 55 und mit 5,97 ml Chlorsulfonsäure in derartiger Geschwindigkeit versetzt, dass die Temperatur des Reaktionsgemisches unter 5 °C bleibt. Hierauf wird die erhaltene Lösung mittels einer Injektionsspritze mit einer Suspension von 7,56 g Benzyloxycarbonylthreoninamid -0- mesylat in 120 ml Methyl-60 enchlorid versetzt. Das erhaltene heterogene Gemisch wird etwa 16 Stunden unter Rückfluss erhitzt. Die entstandene klare Lösung wird in 500 ml einer 0,5 molaren Phosphatpufferlösung vom pH-Wert 4,5 gegossen und mit 120 ml Methylenchlorid versetzt. Die organische Phase wird abgetrennt und « einmal mit 100 ml der Phosphatpufferlösung gewaschen. Die wässrigen Phasen werden vereinigt und mit 10,2 g Tetra-n-butylammoniumhydrogensulfat behandelt und einmal mit 300 ml sowie zweimal mit jeweils 150 ml Methylenchlorid ex

trahiert. Die Methylenchloridextrakte werden vereinigt und über Natriumsulfat getrocknet. Danach wird die Lösung eingedampft. Es hinterbleiben 12,7 g eines Schaums.
F) (3S-trans) -3- Amino -4- methyl -2- oxo -1- azetidinsul-fonsäure
Ein Gemisch aus 5,52 g Kaliumcarbonat in 20 ml Wasser und 160 ml 1,2-Dichloräthan wird unter Rückfluss erhitzt und mit einer Lösung von 15,5 mMol N-Sulfonylbenzyloxy-carbonylthreoninamid -0- mesylat-Tetrabutylammoniumsalz in 20 ml 1,2-Dichloräthan versetzt. Weitere 20 ml werden zum Spülen verwendet. Danach wird das Gemisch 30 Minuten unter Rückfluss erhitzt, sodann in einen Scheidetrichter gegeben, mit 50 ml Wasser und 100 ml Methylenchlorid verdünnt und zur Phasentrennung stehengelassen. Die erhaltene organische Phase wird über Natriumsulfat getrocknet und eingedampft. Es wird rohes (3S-trans) -3- Benzyloxycarbor ylamino -4- methyl -2- oxo -1- azetidinsulfonsäure-Tetrabut-ylammoniumsalz erhalten. Das Rohprodukt wird in 250 ml Äthanol gelöst, mit 0,8 g 5prozentigem Palladium-auf-Koh-lenstoff versetzt und Wasserstoff durch die Lösung geleitet, s Nach 90 Minuten wird der Katalysator durch Kieselgur abfiltriert. Der Filterrückstand wird mit 50 ml Äthanol gewaschen. Das Filtrat und die Waschlösung werden vereinigt und mit 1,2 ml Ameisensäure versetzt. Es scheidet sich die Titelverbindung aus. Das Gemisch wird noch 1 Stunde gerührt io und danach filtriert. Das Produkt wird 1 Stunde bei 10"1 Torr getrocknet. Ausbeute 1,1g. Eine zweite Kristallausbeute wird nach dem Eindampfen des Filtrats und Zusatz weiterer Ameisensäure zum Rückstand erhalten. Es werden 1,3 g der Titelverbindung erhalten. Die Verbindung schmilzt oberhalb is 218 °C unter Zersetzung. [a]D - 41,1 (c = 1 inH20). NMR (D20) 1,58 (3H, d = 7), 4,80 (2H, M).

Claims (3)
1. Zwischenprodukte der allgemeinen Formel —
R.J-NH-
-NH-SO
in der R) ein Wasserstoffatom ist oder die Gruppe Rp-NH-eine geschützte Aminogruppe bedeutet, R3 und R4, die gleich oder verschieden sind, Wasserstoffatome, Alkyl- oder Cyclo-alkylreste, Phenyl- oder substituierte Phenylgruppen bedeuten oder einer der Reste R3 und R4 ein Wasserstoffatom und der andere Rest eine Alkoxycarbonyl-, Alken -1- yl-, Alkin -1-yl-, 2-Phenyläthenyl- oder 2-Phenyläthinylgruppe, M® ein Wasserstoffatom oder ein Kation und V eine abspaltbare Gruppe bedeuten.
2. Zwischenprodukte nach Anspruch 1, wobei die geschützte Aminogruppe eine Acyl-amino-gruppe ist.
3. Zwischenprodukte nach einem der Ansprüche 1 und 2, wobei die abspaltbare Gruppe V eine Methansulfonyl-, Ben-zolsulfonyl- oder Toluolsulfonylgruppe ist oder ein Chlor-, Brom- oder Jodatom bedeutet.
