Abstract
La presente invención se refiere a ácidos nucleicos, vectores, células hospedadoras, métodos y composiciones para conferir y/o aumentar respuestas inmunitarias mediante inmunoterapia celular, tal como al transferir de manera adoptiva células T CD8+ de memoria central o combinaciones de células T de memoria central con células T CD4+ que se modifican genéticamente para expresar un receptor quimérico bajo el control de un promotor inducible. En algunas alternativas, la célula hospedadora genéticamente modificada comprende un ácido nucleico que comprende un polinucleótido que codifica para un receptor de antígeno quimérico que comprende un dominio de unión a ligando, un polinucleótido que comprende una región separadora, un polinucleótido que comprende un dominio transmembrana, y un polinucleótido que comprende un dominio de señalización intracelular bajo el control de un promotor inducible por fármaco. El control de la expresión del receptor quimérico proporciona la capacidad de activar y desactivar la expresión dependiendo del estado del paciente.
