Description
A für Alkyl mit 1 bis 12 C-Atomen, Phenylalkyl mit 7 bis 10 C-Atomen oder für eine Gruppe der Formel RCO- steht, wobei
- i) R Wasserstoff, Alkyl mit 1 bis 11 C-Atomen, Phenyl oder Phenylalkyl mit 7 bis 10 C-Atomen, oder
- ii) RCO-
- a) einen gegebenenfalls im Ring durch Fluor, Chlor, Brom, NO2, NH2, OH, Alkyl mit 1 bis 3 C-Atomen und/oder Alkoxy mit 1 bis 3 C-Atomen substituierten L- oder D-Phenylalaninrest,
- b) den Rest einer anderen als oben unter a) definierten natürli chen α-Aminosäure oder einer entsprechenden D-Aminosäure, oder
- c) einen Dipeptidrest, worin die einzelnen Aminosäurereste gleich oder verschieden sind und aus den oben unter a) und/oder b) definierten Resten ausgewählt werden, bedeuten, wobei die α-Aminogruppe der Aminosäurereste a) und b) und die N-termi nale Aminogruppe des Dipeptidrestes c) gegebenenfalls durch Alkyl mit 1 bis 12 C-Atomen mono- oder di- alkyliert sind,

E für Thr, Ser, Val, Phe, Ile oder Aminoisobuttersäurerest
F für COOR7
R10 für Wasserstoff oder den Rest eines physiologisch annehm baren, physiologisch hydrolysierbaren Esters
R11 für Wasserstoff, Alkyl mit 1 bis 3 C-Atomen, Phenyl oder Phenylalkyl mit 7 bis 10 C-Atomen, jedoch falls R12 für -CH(R13)-X1 steht, nur für Wasserstoff oder Methyl
R12 für Wasserstoff, Alkyl mit 1 bis 3 C-Atomen oder
R15 für Wasserstoff, Alkyl mit 1 bis 3 C-Atomen, Phenyl oder Phenylalkyl mit 7 bis 10 C-Atomen, und
R16 für Wasserstoff oder Hydroxy stehen,
wobei die Reste B, D und E in L-Form und die Reste in den Stellungen 2 und 7 in D- oder L-Form vorliegen können,
sowie die Salze und Komplexe dieser Polypeptidderivate.
- 1. A steht vorzugsweise für Phenylalkyl mit 7 bis 10 C-Atomen, beson
ders Phenethyl, oder für einen Rest RCO-. Ganz bevorzugt steht A
für RCO-.
- 1. 1.1. R bedeutet vorzugsweise Alkyl mit 1 bis 11 C-Atomen oder Phenyl alkyl mit 7 bis 10 C-Atomen, besonders Phenylalkyl mit 7 bis 10 C-Atomen, ganz besonders Phenethyl, oder RCO- hat die Bedeutung a), b) oder c).
- 2. 1.2. Falls RCO- die Bedeutung a), b) oder c) hat, sind die α-Amino gruppe der Aminosäurereste a) und b) und die N-terminale Amino gruppe des Dipeptidrestes c) vorzugsweise nicht alkyliert oder durch Alkyl mit 1 bis 12, besonders 1 bis 8 C-Atomen monoalky liert, ganz bevorzugt monomethyliert. Ganz besonders ist die N-terminale Aminogruppe nicht alkyliert.
- 3. 1.3. Falls RCO- die Bedeutung a) hat, steht es vorzugsweise für einen gegebenenfalls durch Alkyl mit 1 bis 12 C-Atomen alkylierten L- oder D-Phenylalanin- oder -Tyrosinrest. Vorzugsweise steht RCO- für einen L- oder D-Phenylalaninrest oder einen L-oder D-N-(C1-8 Alkyl)-Phenylalaninrest, besonders für einen D-Phenylalaninrest oder D-N-(C1-8 Alkyl)-Phenylalaninrest, ganz besonders für einen D-Phenylalanin- oder D-(N-Methyl)-Phenylalaninrest.
- 4. 1.4. Falls RCO- die Bedeutung b) oder c) hat, ist es vorzugsweise ein lipophiler Rest. Bevorzugte Bedeutungen für b) sind α-Aminosäure reste mit Seitenkette, z. B. Leucin- und Norleucinreste, wobei diese Reste die L- oder D-Konfiguration haben können. Bevorzugte Bedeutungen für c) sind Dipeptidreste, worin die einzelnen Aminosäurereste identisch oder verschieden sein können und eine Bedeutung wie oben für b) und c) als bevorzugt angegeben haben.
- 5. 1.5. RCO- hat ganz besonders eine Bedeutung wie oben unter a) defi niert, speziell die bevorzugten Bedeutungen von a).
- 2. B ist vorzugsweise B', wobei B' Phe oder Tyr ist.
- 3. C ist vorzugsweise C', wobei C' (D)Trp ist.
- 4. D ist vorzugsweise D', wobei D' Lys, MeLys, Lys(εMe), besonders Lys bedeutet.
- 5. E ist vorzugsweise E', wobei E' für den Rest einer natürlichen α-Aminosäure verschieden von Val steht. E' ist vorzugsweise Thr.
- 6. F ist vorzugsweise F', wobei F' einen Rest der Formel
besonders
bedeutet. Der Rest -CH(R13)-X1 liegt vorzugsweise in der L-Form vor. Vorzugsweise hat F' eine andere Bedeutung als ThrNH2 wenn E' Thr ist.- 1. 6.1. R11 ist vorzugsweise Wasserstoff.
- 2. 6.2. Als an das α-Kohlenstoffatom einer Aminosäure gebundene Substi tuent, beispielsweise wie H2N-CH(R13)-COOH, bedeutet R13 vorzugs weise -CH2OH, -(CH2)2-OH, -(CH2)3-OH, -CH(CH3)OH, Isobutyl oder Benzyl, besonders -CH2OH oder -CH(CH3)OH.
- 3. 6.3. X1 ist vorzugsweise eine Gruppe der Formel
insbesondere -CH2OR10, wobei R10 vorzugsweise Wasserstoff ist oder eine Bedeutung wie unten unter 7. definiert, hat. R10 bedeutet ganz bevorzugt Wasserstoff.
- 7. Als Rest eines physiologisch annehmbaren, physiologisch hydro lysierbaren Esters, steht R10 vorzugsweise für HCO, Alkylcarbonyl mit 2 bis 12 C-Atomen, Phenylalkylcarbonyl mit 8 bis 12 C-Atomen oder Benzoyl.
- 8. Die in Stellung 2 und 7 stehenden Reste haben vorzugsweise die L-Konfiguration.
- 9. Y1 und Y2 stehen vorzugsweise zusammen für eine Bindung.
Claims (7)
worin
A für Alkyl mit 1 bis 12 C-Atomen, Phenylalkyl mit 7 bis 10 C-Atomen oder für eine Gruppe der Formel RCO- steht, wobei
- a) R Wasserstoff, Alkyl mit 1 bis 11 C-Atomen, Phenyl oder Phenylalkyl mit 7 bis 10 C-Atomen, oder
- b) RCO-
- c) einen gegebenenfalls im Ring durch Fluor, Chlor, Brom, NO2 NH2, OH, Alkyl mit 1 bis 3 C-Atomen und/oder Alkoxy mit 1 bis 3 C-Atomen substituierten L- oder D-Phenylalaninrest,
- d) den Rest einer anderen als oben unter a) definierten natürli chen α-Aminosäure oder einer entsprechenden D-Aminosäure, oder
- e) einen Dipeptidrest, worin die einzelnen Aminosäurereste gleich oder verschieden sind und aus den oben unter a) und/oder b) definierten Resten ausgewählt werden, bedeutet, wobei die α-Aminogruppe der Aminosäurereste a) und b) und die N-termi nale Aminogruppe des Dipeptidrestes c) gegebenenfalls durch Alkyl mit 1 bis 12 C-Atomen mono- oder di- alkyliert sind,
Y1 und Y2 für Wasserstoff, oder
Y1 und Y2 zusammen für eine Bindung stehen,
B für Phe oder im Phenylrest durch Halogen, NO2, NH2, OH, Alkyl mit 1 bis 3 C-Atomen oder Alkoxy mit 1 bis 3 C-Atomen substituiertes Phe, oder NaphthylAla,
C für gegebenenfalls im Benzolring durch Halogen, NO2, NH2, OH, Alkyl mit 1 bis 3 C-Atomen oder Alkoxy mit 1 bis 3 C-Atomen substitu iertes L- oder D-Trp, wobei die α-Aminogruppe durch Methyl subs tituiert sein kann,
D für Lys, Thialys, γF-Lys, δF-Lys oder Orn, wobei die α-Aminogruppe durch Methyl substituiert sein kann, oder 4-AminocyclohexylAla oder 4-AminocyclohexylGly,
E für Thr, Ser, Val, Phe, Ile oder Aminoisobuttersäurerest,
F für COOR7, CH2OR10, CO-NR11R12 oder
R7 für Wasserstoff oder Alkyl mit 1 bis 3 C-Atomen),
R10 für Wasserstoff oder den Rest eines physiologisch annehm baren, physiologisch hydrolysierbaren Esters),
R11 für Wasserstoff, Alkyl mit 1 bis 3 C-Atomen, Phenyl oder Phenylalkyl mit 7 bis 10 C-Atomen, jedoch falls R12 für -CH(R13)-X1 steht, nur für Wasserstoff oder Methyl,
R12 für Wasserstoff, Alkyl mit 1 bis 3 C-Atomen oder
R13 für den am α-C-Atom befindlichen Rest einer natürlichen Aminosäure (inklusive Wasserstoff) oder für einen CH2OH, HO-CH2-CH2-, HO(CH2)3 oder -CH(CH3)OH-Rest,
R14 für Wasserstoff oder Alkyl mit 1 bis 3 C-Atomen)
R15 für Wasserstoff, Alkyl mit 1 bis 3 C-Atomen, Phenyl oder Phenylalkyl mit 7 bis 10 C-Atomen und
R16 für Wasserstoff oder Hydroxy stehen,
wobei die Reste B, D und E in L-Form und die Reste in den Stellungen 2 und 7 in D- oder L-Form vorliegen können,
in freier Form oder in einer pharmazeutisch akzeptablen Salz- bzw. Kom plexform, ist.
