本发明涉及口用组合物,如酸性饮料和口用保健品,还涉及钙在该组合物中的用途,用于减轻或预防酸性物质对牙齿造成的损伤。本发明特别是要减少因钙的加入而产生的口味问题。
酸性食品从牙齿中浸出钙要比代之以正常的补充矿质过程快,这尤其被认为是造成腐蚀牙齿的原因。将按照本发明制得的产品(如饮料)送入口腔饮用或达到保健目的时,能显著减少由化学过程所引起的钙及磷酸盐从牙齿溶解或去除。
钙是人体内最丰富的矿物质。大多数钙沉积在骨骼和牙齿中,不过它对其他身体功能来说也是必要的矿物质,如神经功能的调节、肌肉的收缩和血液的凝集。钙是来源于水果和硬水的饮料中常见成分,其中该硬水在用于饮料制备时没有经过预先软化处理。以这种方式得到的钙浓度值一般在0.005-0.02%w/w范围内。人们对钙离子使饮食的一般营养价值增加甚为关注,因而找到了一种实用方法,就是在饮料中加入0.02%w/w至2%w/w的高浓度钙离子。钙作为饮料添加剂的用途已如WO88/03762所述。
加入苹果酸将有助于保持加钙饮料中的钙溶解度,从而减少了由沉淀带来的损失。这是因为生成了一种可溶性配合物“柠檬酸苹果酸钙”。另一方面,Lussi等人(1995,Caries Res 29,349-354)已经将饮料的可滴定酸度与其侵蚀能力联系起来;饮料中酸浓度越大,对牙齿造成的损伤也越大。
PCT US91/07117公开了通过饮用一种酸性饮料(pH小于5.5)预防牙釉质侵蚀的方法,该酸性饮料含有0.02%至0.15%以柠檬酸苹果酸钙配合物形式存在的钙,柠檬酸盐与苹果酸盐的摩尔比为1∶0.5至1∶4.5。在柠檬酸苹果酸钙配合物中,总钙摩尔数∶总柠檬酸盐摩尔数∶总苹果酸盐摩尔数的这种摩尔比可以从约2∶1∶1至6∶3∶4。优选的饮料用配合物的这个摩尔比为4∶2∶3。
我们已经发现,饮料中含有高浓度的钙在口味上会产生问题。降低钙相对于酸化剂的用量,同时控制组合物的pH,可实现在酸性口用组合物中有效减少牙齿腐蚀,本发明是基于上述发现而完成的。
一方面,本发明提供了含有一种钙化合物和一种酸化剂的液体口用组合物,其特征在于钙含量范围为0.3至0.8摩尔每摩尔酸,选择组合物中钙和酸化剂的用量使组合物的pH为3.5至4.5。
另一方面,本发明提供了钙作为牙齿腐蚀抑制剂在酸性液体口用组合物中的用途,是通过向组合物中加入一种钙化合物,使钙含量范围为0.3至0.8摩尔每摩尔酸,所选择的组合物中钙和酸化剂用量要使组合物的pH为3.5至4.5。
再一方面,本发明提供了一种减少酸性口用组合物对牙齿腐蚀的方法,该方法包括向酸性液体口用组合物中加入一种钙化合物,使钙含量范围为0.3至0.8摩尔每摩尔酸,如果需要或必要的话,通过加入一种碱来调节pH,使组合物的pH为3.5至4.5。
本发明更进一步地提供了一种制备本发明组合物的方法,该方法包括向酸性液体口用组合物中加入一种钙化合物,使钙含量范围为0.3至0.8摩尔每摩尔酸,如果需要或必要的话,通过加入一种碱来调节pH,使组合物的pH为3.5至4.5。
本发明适用于口服的含水酸性物质,如酸性饮料、果汁、苹果汁、葡萄酒、醋和腌制品及各种酸性乳制品,以及其他口腔用的液体物质,如酸性漱口剂和药物。
实施本发明所生产的饮料没有口味上的缺点。尽管估计到饮料pH升高至pH4左右时酸化剂会鲜明降低口味,但令人惊讶的是,按照本发明加入钙,对此起到了缓和作用。
按照本发明,以碱性盐的形式使用低浓度的钙产生了另一个优点。酸的部分中和作用降低了制剂的缓冲能力,使唾液能更快地中和口腔中的残余物。
用于本发明的钙的绝对浓度并不严格,它将根据所含有的酸性物质的性质和浓度的不同而不同。酸溶液可含有有机和/或无机酸,并可添加维生素,如抗坏血酸。在饮用前被多至五倍量的水稀释的浓缩饮料中,钙浓度可以从0.001摩尔每升至0.05摩尔每升以上不等。即饮饮料中钙离子浓度可以从0.0002摩尔每升至0.01摩尔每升以上不等。
钙可以以任何合适的盐形式加入,如碳酸钙、氢氧化钙、柠檬酸钙、苹果酸钙、乳酸钙、氯化钙、甘油磷酸钙或甲酸钙,或任何其他的盐,以使任何由组合物产生的不良口味减少到最小程度。
通过将酸(如柠檬酸)与其相应的钙盐(如柠檬酸钙)或其他钙盐混合,可以实施本发明。将酸与诸如碳酸钙的碱性钙盐或氢氧化钙混合是有利的,这样使制剂中所用的酸浓度最小。也可将酸与无机钙盐混合,如氯化钙。
钙与酸的摩尔比可以是0.3-0.75,一般为0.3-0.65,优选为0.3-0.55。最优选的摩尔比至少为0.4,已发现该比值约为0.5时尤为有效。
通过加入相对酸来说是适当比例的钙化合物,可以将制剂的pH调节在所需的范围内。如果需要的话,通过使用一种碱、如氢氧化钠,或一种适当的盐、如柠檬酸钠、苹果酸钠或乳酸钠,可以进一步调节pH,这取决于所存在的酸。
组合物的pH优选不大于4,最优选为3.7至3.9。已发现pH约为3.8的组合物尤为有效。
果汁饮料浓缩物中柠檬酸或苹果酸的浓度一般在0.1%w/w至4%w/w的范围内。即饮饮料中酸浓度一般在0.01%w/w至1%w/w的范围内。也可以使用饮料常用的其他可饮用酸,如乳酸。可以使用可饮用酸的混合物。
在一种优选的实施方式中,酸组合物是从诸如茶藨子果汁的天然果汁制得的饮料浓缩物,如加味糖浆浓缩物。钙可以以适当的形式加入到浓缩物中,尤其是当饮料作为饮用前稀释的浓缩物出售给消费者时加入,或者在稀释用于制备“即饮”前被稀释的浓缩浆时也可加入钙。产品中优选含有低浓度的糖或碳水化合物,或者是含有高效甜味剂的低卡路里型产品。
口用组合物可以含有镁或其他离子作为补充矿质的添加剂。为了防止不溶性钙盐沉淀出来,或沉淀最少,也可以含有有效量的苹果酸或其可饮用的盐,以保持钙的溶解度。所加入的苹果酸可以仅提供饮料总酸度的10%,其余酸度由优选为天然存在的其他酸、如柠檬酸或由抗坏血酸提供。
本发明可以用在多种饮料中,如饮料浓缩物、蒸馏果汁饮料、或碳酸软饮料,特别是保健饮料,如茶藨子(blackcurrant)果汁饮料或加维生素饮料。将本发明用在天然含有柠檬酸的饮料或加柠檬酸的饮料中是有利的。饮料可以用糖或高效甜味剂、如糖精、天冬氨酰苯丙氨酰甲酯、或其他本领域已知的甜味剂进行甜味处理,或者不经甜味处理。饮料也可含有其他常用添加剂,如苯甲酸钠、山梨酸、偏亚硫酸氢钠、抗坏血酸、香料、色素和二氧化碳。
饮料的制备可以按照常规方法将各成分混合而成。可以先将固体成分溶于水,如果必要的话溶于热水,再加入其他成分。一般,饮料是在装瓶或罐头或其他包装之前进行巴氏灭菌,或者在灌装之后进行“带包装巴氏灭菌”。

在所进行的体外测平仪法测定中,在37℃温度下使牙釉质平截面在供试溶液中浸泡30分钟。用物理方法测量试验过程中釉质损伤的深度,借此评价侵蚀能力。含有14mM柠檬酸、pH3.2的对照制剂造成釉质损伤4微米,另一含有14mM柠檬酸、pH3.85的对照制剂除去1.8微米釉质,而含有14mM柠檬酸、7mM钙、且pH调为3.85的供试加钙制剂仅除去0.17微米釉质,这证明了本发明的实用性。实施例2

将氢氧化钙加到含一定比例的水的饮料浆料中,并使所制得的饮料中钙与柠檬酸的摩尔比为0.5∶1。所得饮料的pH为3.8。这批饮料进行急速巴氏灭菌,并灌装在250ml“利乐砖”(Tetra-Brik)容器中。
在本研究中,比较三种饮料对人牙釉质损伤:本例饮料、作为阳性对照的橙汁(pH3.8)和作为阴性对照的水。
十二名志愿者参加了本次研究,经拉丁十字形交叉设计分成三组。每个研究周期由三个星期组成,每星期由五个工作日组成。在每个研究周期中,选取一颗健康牙齿的釉质截面在一种装置中每工作日磨蚀七小时。在此期间内,在指导下逐渐吸啜250ml供试饮料四次,吸啜在十分钟内完成。允许受试者移去装置进食午餐,但在放装置期间不允许进食或饮用其他饮料。在处理周期的开始和每星期结束时用测平仪法测量釉质试样(该方法原理记载在Davis & Winter(1977)《英国牙科杂志143,116-119)。全部读取操作均重复进行。冲洗干净后,每个受试者用新鲜的釉质样品开始下一次处理周期。结果列在下表中,数值代表按给定处理方法在给定浸泡时间后釉质损伤的微米数,为十二名受试者的平均值。

本试验清楚地说明,向0.3%柠檬酸溶液中加入钙显著降低了其侵蚀能力。该作用在钙与酸的摩尔比约为0.5(大约7.5mM钙)时达到最大。将钙:酸摩尔比升高至远大于此值后,侵蚀能力没有达到合理的降低。实施例6

饮料浓缩物在93℃下急速巴氏灭菌42秒钟,灌装在600ml瓶中。钙与酸的摩尔比为0.4,最终pH为3.7。
经五倍量水稀释至饮用浓度后,发现组合物的pH为3.85,饮料的口味类型属于典型的茶藨子果味。
分别测定饮料浓缩物在室温和30℃下的贮存稳定性。9个月后没有观察到有不溶性钙沉淀产生。实施例7
体内研究
混合下列成分制得饮料:
茶藨子浓缩物                10.07升
天冬氨酰苯丙氨酰甲酯        0.21kg
乙酰舒泛K                   0.07kg
抗坏血酸                    0.27kg
乳酸80                      0.66升
山梨酸钾                    0.1kg
偏亚硫酸氢钠                0.02kg
香料                        0.56升
氢氧化钙                    0.52kg
水          加至            600.00升
将氢氧化钙与一定比例的水配制成悬液,最后加入,使所制得的饮料中钙与柠檬酸/乳酸的摩尔比为0.45∶1。所得饮料的pH为3.85。将这批饮料包装在250ml容器中,进行“带包装”巴氏灭菌。
在本研究中,比较四种饮料之间的人牙釉质损伤:本例饮料、没有加入钙的pH3.0的商业上可得到的茶藨子果汁饮料、作为阳性对照的橙汁(pH3.9)和作为阴性对照的水。
十二名志愿者参加了本次研究。每个研究周期由三个星期组成,每星期由五个工作日组成。在每个研究周期中,选取一颗健康牙齿的釉质截面在一种装置中每工作日磨蚀七小时。在此期间内,在指导下逐渐吸啜250ml供试饮料四次,吸啜在十分钟内完成。允许受试者移去装置进食午餐,但在放装置期间不允许进食或饮用其他饮料。在处理周期的开始和每星期结束时用测平仪法测量釉质样本(该方法原理记载在Davis & Winter(1977)《英国牙科杂志》143,116-119)。全部读取操作均重复进行。冲洗后,每个受试者用新鲜的釉质样本开始下一次处理周期。
结果列在下表中,数值代表按给定处理方法在给定浸泡时间后釉质损伤的微米数。

    水,加至     1000l
将氢氧化钙与一定比例的水配制成悬液,最后加入,然后将可乐糖浆用碳酸水稀释,装罐头并带包装巴氏灭菌,制得pH3.5的钙与酸的摩尔比为0.6的最终产品。实施例9

将乙醇、氯化鲸蜡基吡啶鎓、脂肪酸酰氨烷基甜菜碱CK-KB5(柯卡酰氨丙基甜菜碱的商标名称)和香料一起混合,形成一透明溶液。在另一个容器中将其他成分一起混合。然后向含水溶液中加入乙醇溶液,制得pH4.5的钙与酸的摩尔比为0.4的漱口剂。实施例10

发现所得组合物的pH一般为3.85,其钙与酸的摩尔比为0.5。
在所进行的体外测平仪法测定中,在37℃温度下使牙釉质平截面在供试溶液中浸泡30分钟。用物理方法测量试验过程中釉质损伤的深度,借此评价侵蚀能力。没有加入氢氧化钙的pH为2.5的对照制剂造成釉质损伤8.1微米,进一步的用氢氧化钠将饮料pH升至3.85的对照制剂除去1.65微米釉质,而上述组合物仅除去0.6微米釉质,这证明了它在减少牙侵蚀上具有实用性。

Claims (28)

1.含有一种钙化合物和一种酸化剂的液体口用组合物,其特征在于钙的含量范围为0.3至0.8摩尔每摩尔酸,选择该组合物中钙与酸化剂的比例使该组合物的pH为3.5至4.5。
2.权利要求1所要求保护的组合物,其中钙的含量范围为0.3-0.75摩尔每摩尔酸。
3.权利要求1所要求保护的组合物,其中钙的含量范围为0.3-0.65摩尔每摩尔酸。
4.权利要求1所要求保护的组合物,其中钙的含量范围为0.3-0.55摩尔每摩尔酸。
5.权利要求1至4中任一项权利要求所要求保护的组合物,其中钙的含量至少为0.4摩尔每摩尔酸。
6.权利要求1至5中任一项权利要求所要求保护的组合物,其中组合物的pH不大于4。
7.权利要求1至5中任一项权利要求所要求保护的组合物,其中组合物的pH为3.7至3.9。
8.权利要求1至7中任一项权利要求所要求保护的组合物,其中的酸是柠檬酸或苹果酸或乳酸或它们的混合物。
9.权利要求1至8中任一项权利要求所要求保护的组合物,其中的钙化合物是碳酸钙、氢氧化钙、柠檬酸钙、苹果酸钙、乳酸钙、氯化钙、甘油磷酸钙或甲酸钙。
10.权利要求1至9中任一项权利要求所要求保护的组合物,该组合物是一种饮料。
11.权利要求10所要求保护的组合物,其中饮料是蒸馏果汁饮料、或碳酸软饮料或优选的保健饮料。
12.权利要求11所要求保护的组合物,其中保健饮料是茶藨子果汁饮料或加维生素饮料。
13.权利要求1至9中任一项权利要求所要求保护的组合物,该组合物是用于制备饮料的饮料浓缩物。
14.权利要求13所要求保护的组合物,该组合物是果汁饮料或保健饮料浓缩物。

16.权利要求15所要求保护的组合物,该组合物是漱口剂。
17.钙在酸性液体口用组合物中作为牙齿腐蚀抑制剂的用途,是通过向该组合物中加入一种钙化合物,使钙含量范围为0.3至0.8摩尔每摩尔酸,所选择的该组合物中钙和酸化剂用量使该组合物的pH为3.5至4.5。
18.权利要求17所要求保护的用途,其中钙的含量范围为0.3-0.75摩尔每摩尔酸。
