Claims (20)
- REIVINDICAÇÕES1,- Célula de uma planta, cujo genoma nuclear é transformado com uma sequência de DNA estranha, de preferência uma sequência de DNA quimérica estranha, caracterizada pelo facto de incorporar :a) um DNA de esterilidade masculina que codifica um primeiro RNA, proteína ou polipéptido o qual, quando produzido ou sobreproduzido numa célula de estame da planta referida, perturba signi ficativamente o metabolismo, o funcionamento e/ou o desenvolvimento da referida célula de estame; eb) um primeiro promotor susceptivel de orientar a expressão do referido DNA de esterilidade masculina nas células dos estames da planta referida, de preferência em células de antera, pólen e/ou filamento, particularmente em células de tapetum e/ou epidérmicas de antera; estando o referido DNA de esterilidade masculina na mesma unidade transcricional, e sob o controlo, do referido primei ro promotor.
- 2.- Célula de acordo com a reivindicação 1, caracterizada pelo facto de a referida sequência de DNA estranha incluir também, de preferência no mesmo sítio genético que o referido DNA de esterilidade masculina:c) um DNA marcador que codifica um segundo RNA, proteína ou polipeptido o qual, quando presente pelo menos num tecido específico ou pelo menos em células específicas da referida planta, torna a referida planta facilmente separável de outras plantas que não contêm o segundo RNA, proteína ou polipeptido pelo menos nas referidas células ou tecido específico; ed) um segundo promotor susceptivel de orientar a expressão do referido DNA marcador pelo menos no referido tecido ou células específicas; estando o referido DNA marcador na mesma unidade transcricional, e sob o controlo, do referido promotor.
- 3.- Célula de acordo com a reivindicação 1 ou 2, caracterizada pelo facto de a referida sequência de DNA estranha incorporar adicionalmente:e) um primeiro DNA que codifica um péptido de transição susceptível de transportar a primeira proteína ou polipeptido para um cloroplasto ou mitocondria da referida célula de estame; estando o referido primeiro DNA na mesma unidade transcricional que o refe rido DNA de esterilidade masculina e referido o primeiro promotor e entre o referido DNA de esterilidade masculina e o referido-70/ /primeiro promotor;e/ouf) um segundo DNA que codifica um péptido de transição susceptível de transportar a referida segunda proteína ou polipéptido para um cloroplasto ou mitocondria de pelo menos o referido tecido específico ou células específicas; estando o segundo DNA na mesma unidade transcricional que o referido DNA marcador e o referido segundo promotor e entre o referido DNA marcador e o referido segundo promotor.
- 4. - Célula de acordo com uma qualquer das reivindicações 1 a 3, caracterizada pelo facto de o referido DNA de esterilidade masculina codificar: uma RNase, particularmente RNase TI ou Barnase; uma DNase, especialmente uma endonuclease,particularmente Ecorj uma protease, especialmente uma papaina, particularmente Ziinogeno de papaína ou proteína activa de papaína; uma glucanase; uma lipase, particularmente fosfolipase A^; uma peroxidase lipídica; um inibidor de parede da célula; uma toxina bacteriana;ou um ribozimo, particularmente o ribozimo contra mRNA codificado pelo gene TA2 9, o gene TA26 ou o gene TA13; ou é um DNA anti-sentido, particularmente o DNA anti-sentido do gene TA29, do gene TA26 ou do gene TA13.
- 5. - Célula de acordo com uma qualquer das reivindicações1 a 4, caracterizada pelo facto de o DNA de esterilidade masculina /-71codificar um enzima que catalisa a síntese de uma fito-hormona, particularmente um enzima codificado pelo gene 1, gene 2 ou gene 4 do DNA de Agrobacterium T.
- 6.- Célula de acordo com uma qualquer das reivindicações 2 a 5, caracterizada pelo facto de o referido DNA marcador ser: um gene de resistência herbicida, particularmente um gene sfr ou sfrv; um gene qne codifica um enzima alvo modificado para um herbi^ cida com uma afinidade menor para o herbicida, particularmente uma 5-enol-piruvilshiquimato-3-fosfato-sintase como um alvo para glifosato ou uma glutamina-sintetase modificada como um alvo para um inibidor de glutamina-sintetase tal como fosfinotricina; um gene que codifica uma proteína ou um polipéptido que confere uma cor pelo menos ao referido tecido específico ou ãs células específicas, particularmente o gene Al ou o gene GUS; um gene que codifi ca uma proteína ou um polipéptido que confere uma tolerância de tensão à planta referida, particularmente o gene que codifica Mn-superõxido-dismutase; ou um gene que codifica uma proteína ou polipéptido que confere uma resistência ãs doenças ou pestes, particularmente um gene que codifica uma endotoxina de Bacillus thuringiensis que confere resistência aos insectos ou um gene que codifica um péptido bacteriano que confere uma resistência bacteriana .
- 7.- Células de acordo com uma qualquer das reivindica-72ções 1 a 6, caracterizada pelo facto de o referido primeiro promotor ser PTA29, PTA26, PTA13 ou um promotor de um DNA que codifica um mRNA específico de tapetum hibridizãvel em TA29, TA26 ou TA13.
- 8. - Célula de acordo com uma qualquer das reivindicações2 a 7, caracterizada pelo facto de o referido segundo promotor ser um promotor fundamental, particularmente um promotor 35S, um promotor 35S'3, um promotor PNOS ou um promotor POCS; um promotor indutível nos tecidos feridos, particularmente um promotor TRI' ou TR2'; um promotor que orienta a expressão de gene selectivamente num tecido de plantas possuindo actividade fotossintética, particularmente um promotor SSLJ; ou um promotor que orienta a expressão de gene selectivamente nas células das folhas, pétalas ou sementes, particularmente células de revestimento de sementes.
- 9. - Vector adequado para transformar uma célula de uma planta,particularmente uma planta susceptivel de ser infectada com Agrobacterium, caracterizado pelo facto de ser constituído por a referida sequência de DNA estranha de acordo com uma qualquer das reivindicações 1 a 8, particularmente PTTM4, PTTM6, PTTM6A, PTTM8, PTVEP1, PTVEP2, PTVE62 ou PTVE63.
- 10.- Processo para a produção de uma planta estéril masculina e material de reprodução da planta referida, tendo a referida sequência de DNA estranho de acordo com uma qualquer das-73reivindicações 2 a 8 estavelmente integrada no genoma nuclear das suas células, pelo que o referido DNA de esterilidade masculina é susceptivel de ser expresso selectivamente nas células de estame da referida planta para produzir o referido primeiro RNA, proteína ou polipéptido nas células de estame e podendo assim o referido DNA marcador ser expresso em pelo menos o referido tecido específico ou células específicas da referida planta para a tornar separável das plantas não transformadas, caracterizado pelo facto de possuir os seguintes passos não biológicos: a) transformação de uma célula da referida planta por introdução da sequência de DNA estranho no genoma nuclear da referida célula; e depois b) regeneração da referida planta e dos materiais de reprodução a partir da célula referida.
- 11.- Cultura de células de planta, caracterizado pelo facto de conter a célula de planta de acordo com uma qualquer das reivindicações 1 a 8.
- 12,- Planta, particularmente milho, batata, tomate, colza, alfafa, girassol, algodão, aipo, cebola, trevo, soja, tabaco, vegetais do tipo brassicãceas ou beterraba sacarina, caracterizada por incluir a célula da planta de acordo com uma qualquer das reivindicações 1 a 8.
- 13.- Semente, caracterizada pelo facto de pertencer a uma ί -74planta de acordo com a reivindicação 12.
- 14.- Processo para produzir uma semente híbrida, caracterizado pelo facto de incluir os seguintes passos:a) polinização cruzada i) de uma planta estéril masculina que contém a sequência de DNA estranho de acordo com uma qualque das reivindicações 2 a 8,incluindo tanto o referido segundo promo tor como o referido DNA marcador, especialmente o DNA marcador que confere uma resistência ao herbicida, particularmente um gene sfr ou sfrv, estavelmente integrado no genoma nuclear das células da referida planta estéril masculina, com ii) uma planta fértil masculina homozigõtica sem o referido DNA marcador, especialmente sem o referido DNA marcador que confere a referida resistência herbicida; e depois b) separação da referida planta fértil masculina da referida planta estéril masculina tirando vantagem da ausência de expressão do referido DNA marcador pelo menos no referido tecido específico ou células específicas da referida planta fértil masculina.
- 15.- Processo para produzir uma semente híbrida de acordo com a reivindicação 14, caracterizado pelo facto de a referida planta estéril masculina conter pelo menos dois DNAs marcadores diferentes estavelmente integrados no genoma nuclear das suas células e por a referida planta fértil masculina conter um, mas não o outro, dos dois referidos DNAs marcadores; e por a referida planta fértil masculina ser separada da referida planta estéril masculina tirando vantagem da ausência de expressão do outro DNA marcador pelo menos no referido tecido específico ou células específicas da referida planta fértil masculina; conferindo de preferência o outro DNA marcador uma resistência a um herbicida.
- 16. - Semente híbrida, caracterizada pelo facto de ser obtida pelo processo de acordo com a reivindicação 14 ou 15.
- 17. - Planta híbrida, caracterizada pelo facto de ser obtida pelo desenvolvimento da semente híbrida de acordo com a reivin dicação 16.
- 18. - Primeiro promotor de acordo com a reivindicação 7, caracterizado pelo facto de ser um PTA29, PTA26, PTA13 ou um promotor de um DNA que codifica para um mRNA específico de tapetum hibridizável para TA29, TA26 ou TA13.
- 19. - Sequencia de DNA quimérica estranha de acordo com uma qualquer das reivindicações 1 a 8, caracterizada pelo facto de o referido DNA de esterilidade masculina nao estar naturalmente sob o controlo do primeiro promotor e/ou não estar naturalmente no mesmo sítio genético que o referido DNA marcador.
- 20. - Processo para produzir plantas e material de reprodução, tal como sementes, das referidas plantas incluindo um material genético estranho estavelmente integrado num seu genoma nuclear-76e susceptivel de ser expresso como um RNA, proteína ou polipeptido, incluindo os seguintes passos não biológicos: a) produção de células de plantas transformadas ou tecidos de plantas incluindo o referido material genético estranho de células de plantas de partida ou tecidos de plantas que não exprimem o referido RNA, proteina ou polipéptido, b) produção de plantas regeneradas e/ou material de reprodução das referidas plantas a partir de células de plantas transformadas ou tecidos de plantas que incluem o referido material genético estranho, e c) eventualmente, replicação biológica das referidas plantas regeneradas e/ou material de reprodução; em que o referido passo de produção de células de plantas transformadas ou tecidos de plantas incluindo o referido material genético estranho é caracterizado pelo facto: de se transformar o genoma nuclear das referidas células de planta de partida ou tecidos de plantas com uma sequência de DNA estranha de acordo com uma qualquer das reivindicações 1 a 8, bem como elementos reguladores que são susceptíveis de permitir a expressão da sequên cia de.DNA estranho nas referidas células de plantas ou tecidos de plantas, para proporcionar a integração estável da sequência de DNA estranho em células de planta transformadas ou tecidos de plantas, bem como em plantas e material de reprodução produzido através de gerações subsequentes,
