Description
米国特許第7407811号明細書は、順次印加される複数の周波数のACブロックと、その後のDCブロックとの使用について教示している。たとえば、米国特許第7407811号明細書で記載される実施例5では、順次印加されるAC励起と、その後印加されるDC励起とを利用している。この励起信号は、およそ1.8間秒印加される10kHzのAC信号、およそ0.2秒間印加される20kHzのAC信号、およそ0.2秒間印加される2kHzのAC信号、およそ0.2秒間印加される1kHzのAC信号、および、およそ0.5秒間印加されるDC信号を含んでいた。トータルテストタイムは3秒であった。
「実施例5に対して上述した同一のテストストリップ1700および試薬を用いて、図24中に示された励起プロファイルを、トータルテストタイムを減少させるために利用した。図5に対して上述されたように、20kHzおよび10kHzにおける位相角は、ヘマトクリット推定値と最も密接に相関していることが決定された。したがって、トータルテストタイムを減少させるために、実施例6中のこれら2つの周波数にAC部分の励起を限定することが決められた。トータルテストタイムをさらに減少させるために、10kHzのAC励起がDC信号と同時に印加され(すなわち、DCオフセットを持ったAC信号)、またこの理論は、この組み合わされたモードが、DC電流、AC位相およびACアドミタンスについて同時の結果の収集を可能にし、最速の可能な結果を提供している。したがって、20kHzの信号は0.9秒間印加された。その後、10kHzおよびDC信号が、0.1秒の間隔後、1.0秒間同時に印加された。」(米国特許第7407811号明細書、23段、23〜40行)

以下の実施例2の記載から示される実施形態を含めて、ここに開示される実施形態のいくつかにおいては、たとえば、スロットダイコーティングのような、バイオセンサーの表面に試薬を均一に塗布する方法を使って、バイオセンサーの試薬の厚みを正確に調節することも利用されている。本文にて開示される試薬コーティングの厚みは、概しておよそ1.6〜5μmである。試薬コーティングの均一性、およびその結果生じる、試薬膜の体液試料との均一な溶解/水和が、AC測定値と良好に相関する再現性を可能にするため、正確に補償されたグルコースを提供することができる。試薬の厚みが均一でなければ、特に短時間では、測定値のばらつきが増えるため、より高速な方法およびパフォーマンスの向上を達成するにあたって支障がある。強固な性能を求めて、我々は非常に均一な膜を目指した努力を行ってきた。均一な膜のコーティングに関する方法についての記載、および開示については、上述した米国特許出願公開第2005/008537を参照されたい。
米国インディアナ州、インディアナポリスにあるロシュ・ダイアグノスティックス社のACCU−CHEK(登録商標)AVIVA(トレードマーク)バイオセンサーと類似した、電極およびテスト構成を使って、複数の全血試料のグルコース濃度をテストする共変量の調査が行われた。表1(上記)と同一または実質的に類似した配合の、ピロロキノリンキノン依存性グルコース脱水素酵素(PQQ−GDH)をベースとする試薬が、3種類の厚み、2μm、2.5μm、および4μmのうち、いずれか1つの厚みでバイオセンサーに適用された。共変量の調査は、実施例1と類似した全血試料で、ただし、図10に詳細に記載されるとおり、6種類のグルコース濃度、5種類のヘマトクリット値、および5種類の温度で行われた。

ここで述べたように、ここに開示される実施形態のいくつかにおいては、多周波励起波形技術を使うことによって、複数の周波数での、より短い時間にわたるACテストデータの収集を利用している。こうした多周波励起波形が、周波数が異なる個々の波形を複数、一緒に加えることによって形成されるため、体液試料が、順次ではなく実質的に同時に、複数の周波数によって励起される。
工程1:血液が、測定器内のバイオセンサーに塗布される。
工程2:滴下の検出および/または用量の充足性のため、試料のAC測定が行われる。
工程3:ヘマトクリットおよび温度に対する補正係数の算出を可能にするために、AC測定が、1つの期間にわたって行われる。多くの例では、複数のAC励起周波数が試料に順次印加される。
工程4:手を加えていない(未補正の)グルコース応答を測定するために、DC測定が行われる。
工程5:AC測定で得られた補正係数を使って、手を加えていないDC応答が、ヘマトクリットおよび温度の影響に対して補償される。
工程6:測定結果が、ユーザに対して表示される。
実施例3について実施された測定は、ACCU−CHEK(登録商標)AVIVA(トレードマーク)バイオセンサーの電極構造に類似した電極構造、および、(上記)表1に記載された配合と同一または類似した試薬で行われた。スパッタリング、レーザーアブレーション、試薬スロットダイコーティング、およびラミネート加工を含んだ工程を組み合わせて、ACCU−CHEK(登録商標)AVIVA(トレードマーク)バイオセンサーと概して同じ技術を使って、こうしたセンサーは製造された。

本実施例5は、図30に示されるとおり、2.45μmの厚みに略相当する、名目上30g/m2の塗装重量の塗布に基づいた試薬膜厚を有するテストセンサーを使って、(上記)実施例2と同様に実施された。ただし、実施例2とは違い、アジレント社のVXIコンポーネントをベースに構成され、かつ、要請された組み合わせおよび順序で、センサーにACおよびDC電位を印加して、その結果として生じるセンサーの電流応答を測定するようにプログラム可能な電気化学試験スタンドを使って、データ取得が実施された。このようにしたのは、実施例1および2に関して述べたように、こうした測定のために、DATSとともに使用される既存の測定器は、波形の予備安定化、各周波数ブロック後のトレーリングの通信、および、DC信号印加の最初の100ミリ秒における、あらかじめ設定された「スキップ」時間のために、その間電流応答を測定することができない強制的な時間ブロックが要求される、あらかじめ設定されたパラメータを備えるからである。しかし、この実施例5に関しては、DATSの既存の測定器によって課される、あらかじめ設定されたタイミング条件の制限なく、連続的な多周波数のACを印加することが望まれた。実施例3および4において使用された、アジレントをベースにした試験スタンドによって、このように、所望の測定シーケンスをプログラムする柔軟性がもたらされた。
Claims (6)
- 生体の試料における、対象の検体の濃度の決定をする方法であって、
a)4μm未満の厚さの試薬コーティングを有するバイオセンサーに試料を適用すること、
b)前記試料に、第1のAC成分を有する第1の信号を100ミリ秒間だけ印加すること、
c)前記第1の信号に対する第1の応答を測定すること、
d)前記試料に、第2のAC成分を有する第2の信号を100ミリ秒間だけ印加すること、
e)前記第2の信号に対する第2の応答を測定すること、
f)前記試料に、第3のAC成分を有する第3の信号を100ミリ秒間だけ印加すること、
g)前記第3の信号に対する第3の応答を測定すること、
h)前記試料に、第4のAC成分を有する第4の信号を100ミリ秒間だけ印加すること、
i)前記第4の信号に対する第4の応答を測定すること、
j)前記試料に、第5のDC信号を印加すること、
k)前記第5の信号に対する第5の応答を測定すること、および、
l)前記試料内の少なくとも1つの干渉物の影響に対して前記第5の応答を補正するために少なくとも前記第1、第2、第3、および第4の応答を使って、前記試料の検体の濃度を決定すること
を含んでおり、
前記決定が、0.5秒と2.0秒との間のトータルテストタイムおよび7.5%と11%との間のトータルシステムエラーを有している方法。 - 前記干渉物が、ヘマトクリットおよび温度のいずれか、または両方を含んでいる請求項1記載の方法。
- 前記第1の信号が10kHzのAC信号であり、前記第2の信号が20kHzのAC信号であり、前記第3の信号が1kHzのAC信号であり、前記第4の信号が2kHzのAC信号である請求項1記載の方法。
- 前記第5の信号が、200ミリ秒間だけ印加される請求項1記載の方法。
- 生体の試料における、対象の検体の濃度の決定をする方法であって、
a)試薬コーティングを有する電気化学的なバイオセンサーに、電気化学的分析における使用のために構成された電極システムを設けること、ここで、前記試薬コーティングは4μm未満の厚さを有している、
b)生体試料を前記バイオセンサーに供給すること、ここで、前記バイオセンサーは前記試料を前記試薬コーティングに接触させるように構成されている、
c)前記試料が前記試薬コーティングに接触する時を電気的に検出すること、
d)前記試料に、第1の信号を印加すること、
e)前記第1の信号に対する第1の応答を測定すること、
f)前記試料に、第2の信号を印加すること、
g)前記第2の信号に対する第2の応答を測定すること、および、
h)少なくとも前記第1および第2の応答を使って、前記試料の前記検体の濃度を決定すること
を含んでおり、
前記第1の信号が順次印加される第1AC成分を有する第1ブロックと、第2AC成分を有する第2ブロックと、第3AC成分を有する第3ブロックと、第4AC成分を有する第4ブロックとを含み、前記第1の応答が前記各AC成分のそれぞれに対する応答データを含み、前記応答データが補正係数の情報を含んでおり、
前記第2の信号がDC信号を含み、前記第2の応答が前記検体の濃度を示す電流応答を含んでおり、
前記第1の信号が、前記試料が前記試薬コーティングに接触するときに、800ミリ秒の第1の時間帯で印加され、前記第1〜第4ブロックがそれぞれ200ミリ秒間だけ印加され、
前記第2の信号が、前記第1の信号の時間帯の後、すぐに、100ミリ秒〜300ミリ秒の第2の時間帯で印加され、
前記決定が、1.1秒以下のトータルテストタイム、および、8.5%以下のTSEを有している方法。 - 前記第2の時間帯が100ミリ秒であり、前記トータルテストタイムが900ミリ秒であり、前記TSEが8.0%以下である請求項5記載の方法。
