Description
- Die vorliegende Erfindung betrifft allgemein das Einführen von neuropharmazeutischen Agentien in das Hirn durch Transcytose durch die Blut-Hirn-Barriere (Blut-Hirn-Schranke). Insbesondere betrifft die vorliegende Erfindung schimärische Peptide, die in der Lage sind, neuropharmazeutische Agentien in das Hirn durch die Blut-Hirn-Barriere mittels Rezeptor-vermittelter Transcytose zu befördern.
- Das Gehirn der Wirbeltiere hat ein einzigartiges Kapillarsystem, das sich von allen anderen Organen des Körpers unterscheidet. Dieses einzigartige Kapillarsystem hat morphologische Merkmale, die die Blut-Hirn-Barriere (BBB) bilden. Die Blut-Hirn-Barriere wirkt als eine systemumfassende Zellmembran, die den Hirnzwischenraum vom Blut trennt.
- Die außerordentlichen morphologischen Merkmale der Hirn-Kapillaren, die die Blut-Hirn-Barriere bilden, sind
- a) epithelartige hochbeständige Zellkontakte, die pedantisch genau alle Endothelien der Hirnkapillaren miteinander befestigen und
- b) spärliche Pinocytose- oder transendothele Kanäle, die reichlich in den Endothelien der peripheralen Organe vorhanden sind. Infolge der einzigartigen Merkmale der Blut-Hirn-Barriere werden hydrophile Medikamente und Peptide, die ohne weiteres Zugang zu anderen Geweben in dem Körper gewinnen, von dem Eintritt in das Hirn abgehalten oder das Ausmaß ihres Eintrittes ist sehr gering.
- Verschiedene Strategien sind für das Einbringen solcher Medikamente in das Hirn entwickelt worden, die ansonsten die Blut-Hirn-Barrieren nicht überwinden würden. Die am meisten benutzten Strategien umfassen Invasionsprozeduren, bei denen das Medikament direkt in das Hirn eingebracht wird. Die am meisten benutzte Prozedur ist die Implantierung eines Katheters in das Ventrikelsystem, um die Blut-Hirn-Barriere zu umgehen und das Medikament direkt in das Hirn zu befördern. Diese Prozeduren wurden bei der Behandlung von Hirnkrankheiten benutzt, die eine Neigung für die Hirnhaut, wie leukämische Attacken des Hirns, zeigen.
- Obgleich Invasionsprozeduren für die direkte Einführung von Medikamenten in die Hirnventrikel einigen Erfolg gebracht haben, waren sie letztlich doch nicht erfolgreich, da sie nur die Medikamente an der Oberfläche des Hirngewebes verteilen und nicht in die Strukturen tief innerhalb des Hirns eindringen. Auch sind die Invasionsprozeduren potentiell schädlich für den Patienten.
- Andere Versuche, die Blut-Hirn-Barriere zu umgehen, benutzen pharmakologisch aufgebaute Prozeduren mit Anpassung der Medikamente oder Umwandlung hydrophiler Medikamente in lipid-lösliche Medikamente. Die Mehrzahl der Anpassungsversuche setzen das Blockieren der Hydroxyl-, Carboxyl- und primären Aminogruppen des Medikamentes ein, um dieses mehr lipid-löslicher zu machen, so daß es leichter durch die Blut-Hirn-Barriere transportiert wird. Obgleich die pharmakologischen Versuche mit einigem Erfolg benutzt wurden, sind sie nicht völlig zufriedenstellend für die Beförderung von Peptiden durch die Blut-Hirn-Barriere hindurch, wie die Erfahrung des Erfinders mit dem Transport von Cyclosporin durch die Blut-Hirn-Barriere lehrt. Cyclosporin ist ein hoch angepaßtes (lipid-lösliches) Peptid, das die Blut-Hirn-Barriere relativ langsam durchquert.
- Ein weiterer Ansatz, um die Blut-Hirn-Barriere zu umgehen, benutzt die intraarterielle Infusion von hypertonischen Substanzen, die die Blut-Hirn-Barriere für den Durchgang öffnen, um hydrophilen Medikamenten den Durchgang zu ermöglichen. Jedoch sind hypertonische Substanzen potentiell toxisch und es besteht die Gefahr, daß sie die Blut-Hirn-Barriere beschädigen.
- Es besteht daher z.Zt. ein Bedarf, verbesserte Substanzen und Methoden für die Beförderung hydrophiler Medikamente und Peptide durch die Blut-Hirn-Barriere hindurch und in das Hirn hinein zu schaffen. Es ist wünschenswert, daß solche verbesserten Substanzen und Methoden für eine gleichmäßige Einführung des hydrophilen Peptides oder Medikaments im ganzen Hirn sorgen und ein geringstmögliches Risiko für den Patienten enthalten.
- Erfindungsgemäß werden neue Prozeduren und Substanzen vorgeschlagen, die eine gleichmäßige Verteilung von Neuro-Peptiden und anderen Medikamenten im ganzen Hirn ermöglichen, die die den früheren invasiven und pharmakologischen Medikamenteneinführungsprozeduren anhaftenden Probleme reduzieren.
- Die Erfindung basiert auf der überraschenden Erkenntnis, daß hydrophile Peptide physiologisch durch die Blut-Hirn-Barriere durch Kopplung oder Konjugation des Medikaments an ein transportables Peptid, das fähig ist, die Blut-Hirn-Barriere durch Rezeptor-vermittelte Transcytose zu durchqueren, transportiert werden können. Diese Entdeckung ist besonders überraschend im Hinblick auf die hergebrachte Vorstellung, daß die Blut-Hirn-Barriere eine passive Barriere sei, die für hydrophile Medikamente oder Peptide undurchdringbar ist.
- Die Erfindung betrifft neue schimärische Peptide, die angepaßt sind, ein neuropharmazeutisches Agens in das Hirn durch Transcytose durch die Blut-Hirn-Barriere hindurch zu befördern. Die schimärischen Peptide enthalten ein transportfähiges Peptid, das in der Lage ist, die Blut-Hirn-Barriere mit einer relativ hohen Rate mittels Rezeptor-vermittelter Transcytose zu durchqueren. Das transportfähige Peptid ist mit einem neuropharmazeutischen Agens konjugiert, um das schimärische Peptid zu bilden. Das neuropharmazeutische Agens ist im allgemeinen ein hydrophiles Peptid, das für sich genommen die Blut-Hirn-Barriere nicht im wesentlichen Umfange überwindet. Die Konjugation transportfähiger Peptide mit neuropharmazeutischen Agentien wurde überraschend zum Herstellen schimärischer Peptide aufgefunden, die befähigt sind, durch die Blut-Hirn-Barriere transportiert zu werden.
- Histone bilden eine Gruppe von natürlich vorkommenden Proteinen, die als gut geeignet für die Benutzung als transportfähiges Peptid gemäß der Erfindung gefunden wurden. Da Histone natürlich vorkommende Substanzen sind, erfordern sie keine organische Synthese und die Möglichkeit einer Immunantwort, wie sie bei synthetisch abgeleiteten Materialien auftritt, ist größtenteils reduziert.
- Es wird angenommen, daß die schimärischen Peptide durch die Blut-Hirn-Barriere mittels des physiologischen Prozesses der Transcytose über Rezeptoren in der Blut-Hirn-Barriere befördert werden. Das sichert die gleichmäßige Verteilung des schimärischen Peptids auf alle Teile des Hirns verteilt wird. Des weiteren reduziert die Einführung des schimärischen Peptids in das Hirn mittels eines physiologischen Weges die schädlichen Nebeneffekte und Risiken, die den traditionellen invasiven und pharmakologischen Versuchen innewohnen.
- Die vorliegende Erfindung umfaßt auch Methoden zum Verabreichen der schimärischen Peptide subkutan oder intranasal und Kompositionen, enthaltend das schimärische Peptid, das in solchen Methoden der Behandlung benutzt werden kann.
- Die vorangehenden erläuterten und zahlreiche weitere Eigenschaften und der Erfindung anhaftende Vorteile werden nachfolgend in der detaillierten Beschreibung und in Verbindung mit der Zeichnung dargelegt, die zum besseren Verständnis der Erfindung dient.
- Fig. 1 zeigt eine graphische Darstellung der Ergebnisse der im Beispiel 2 beschriebenen Versuche.
- Fig. 2 zeigt eine graphische Darstellung der Ergebnisse der Versuche gemäß Beispiel 10, die die Aufnahme der Histone durch die Hirnkapillaren zeigt.
- Fig. 3 (-a,b-) zeigen graphische Darstellungen der Ergebnisse von Histon-Aufnahmeversuchen gemäß Beispiel 10, die die Temperatur- und Zeitabhängigkeit des Transportmechanismus aufzeigen.
- Fig. 4 zeigt eine graphische Darstellung der Linearität von Histon in bezug auf die Menge von kapillarem Protein.
- Fig. 5 zeigt eine graphische Darstellung, bei der die Bindung (% gebunden/mgp) von (¹²&sup5;I)-Histon gegen die Konzentration des nichtgebundenen Histons in dem Inkubationskessel aufgezeichnet ist und wobei gebunden (B)/frei (F) gegen (¹²&sup5;I)-Histon, gebunden an die Rinderhirnkapillaren (nmol/mgp), aufgetragen ist.
- Fig. 6 zeigt eine graphische Darstellung der Radioaktivität A(t) von Serum (³H)-Albumin und (¹²&sup5;I)-Histon (DPM/ml/Prozent injiziert), aufgetragen gegen die Zeit nach einer einzigen intravenösen Injektion beider Isotope.
- Die erfindungsgemäßen schimärischen Peptide sind zur Beförderung einer großen Vielzahl von neuropharmazeutischen Agentien in das Hirn geeignet. Die Erfindung ist insbesondere für die Einführung neuropharmazeutischer Agentien in Gestalt hydrophiler Peptide gut geeignet. Diese hydrophilen Peptide werden im allgemeinen nicht in nennenswerterm Maße durch die Blut-Hirn-Barriere transportiert.
- Beispielhafte hydrophile Peptide als neuropharmazeutische Agentien sind: Thyrotropin freigebendes Hormon (TRH) - zur Behandlung von Rückenmarkserkrankungen und Lou Gehrig's Krankheit; Vasopressin - zur Behandlung von Amnesie; Alpha - Interferon - zur Behandlung von Multipler Sklerose; Somatostatin - zur Behandlung von Alzheimer's Krankheit; Endorphin - zur Behandlung von Schmerzen; L-methionyl (sulfon)-L-glutamyl-L-histidyl-L-phenylalanyl-D-lysyl-L- phenylalanin (ein Analogon des adrenocorticotrophen Hormons (ACTH)-4-9) - zur Behandlung der Epilepsie; und Muramyl - Dipeptid - zur Behandlung von Schlaflosigkeit. Alle diese neuropharmazeutischen Peptide sind handelsüblich erhältlich oder sie können aus natürlichen Resourcen mittels bekannter Techniken isoliert werden.
- Die folgende Beschreibung beschränkt sich auf die schimärischen Peptide, bei denen die neuropharmazeutischen Agentien hydrophile Peptide (Neuropeptide) sind, wobei jedoch die Erfindung auf alle neuropharmazeutischen Agentien Anwendung finden kann, die selbst nur in geringer oder nicht feststellbarer Rate durch die Blut-Hirn-Barriere transportiert werden. Die Erfindung findet auch Anwendung dort, wo die Rate, mit der das neuropharmazeutische Agens durch die Blut-Hirn-Barriere transportiert wird, erhöht werden soll.
- Das schimärische Peptid umfaßt das hydrophile Peptid- Medikament, konjugiert mit einem transportablen Peptid, das in der Lage ist, die Blut-Hirn-Barriere mittels Transcytose mit einer wesentlich höheren Rate als die hydrophilen Neuropeptide zu durchqueren. Geeignete transportfähige Peptide umfassen: Histone, Insulin, Transferrin, insulin-like growth factor I (IGF-I), insulin-like growth factor II (IGF-II), basisches Albumin und Prolactin.
- Transferrin ist ein 80 K Glykoprotein, es ist das prinzipale Eisen-Transportprotein im Kreislauf. Transferrin ist auch ein Protein, das in der Hirn-Rückenmarks-Flüssigkeit (CSF) angereichert ist. Transferrin ist umfangreich erhältlich und kann gekauft werden oder aus Blut oder der Hirn-Rückenmarksflüssigkeit mittels bekannter Methoden isoliert werden.
- Insulin, IGF-I und IGF-II sind ebenfalls allgemein erhältlich. Insulin ist in großem Umfange handelsüblich erhältlich und kann ebenfalls aus natürlichen Quellen mittels bekannter Techniken gewonnen werden. IGF-I und IGF-II sind von kommerziellen Verkaufsstellen, wie Amgen oder Peninsula Labs erhältlich oder sie können aus natürlichen Quellen, entsprechend der Methode von Rosenfeld et al (J. Clin Endocrinol. Metab. 55, 434, 1982) isoliert werden.
- Basisches Albumin oder kationisiertes Albumin hat einen isoelektrischen Punkt (pI) von 8.5 im Vergleich zu einem pI von 3.9 für natürliches Albumin. Kationisiertes Albumin tritt im Gegensatz zu natürlichem Albumin schnell in das Hirn durch die Blut-Hirn-Barriere ein. Kationisiertes Albumin (pI = 8.5) wird bevorzugt durch kovalente Kopplung von Hexamethylen-diamin (HMD) an Rinderserum Albumin (pI = 3.5) nach Bergmann, et al, "Cationized Serum Albumin Enhances Response of Cultured Fetal Rat Long Bones to Parathyroid Hormone", Endocrinologie, 116:1727-1733 (1985), hergestellt. Nachfolgend eine beispielhafte Synthese: 10 ml einer 10%-igen Lösung von Albumin in Wasser wird langsam 60 ml von 2.0 M HMD zugegeben und der pH der Lösung auf 6 bis 7 mit 1N HCl eingestellt. Nach 30 Minuten wird 1 g von N-ethyl-N'-3-(dimethylaminopropyl)-Carbodiimid- Hydrochlorid (EDAC) zugegeben, um die Carboxylgruppen des Albumin zu aktivieren, wobei weitere 1 g EDAC 1 Stunde später zugegeben werden. Der pH wird stetig auf 6 bis 7 mit 0.2 N HCl eingestellt. Die Lösung wird über Nacht unter konstantem Rühren stehengelassen. Am nächsten Tag wird die Lösung einer ausgedehnten Dialyse mit destilliertem Wasser unterzogen. Diese Lösung wird dann mittels chromatischer Fokussierung unter Verwendung des polybuffer exchanger 94 resin und des polybuffer 96 elution buffer von Pharmacia gereinigt.
- Histone sind besonders gut geeignet für den Einsatz als transportfähiges Peptid, da es sich um natürlich vorkommende Proteine handelt, die keine organische Synthese erfordern, wie die vorgenannte Prozedur zum Herstellen von basischem Albumin. Des weiteren macht die Abwesenheit einer Antikörper-Antwort zu den natürlich vorkommenden Histonen sie für viele Gelegenheiten geeignet, bei denen Immunantworten auf synthetische Materialien, wie kationisiertes Albumin, deren Einsatz potentiell begrenzt. Histone sind eine Gruppe Lysin-reicher, hoch kationischer Proteine, die in fünf Klassen (H1, H2, H3, H4 und H5) eingeteilt sind. Es gibt zahlreiche Subtypen in jeder der fünf Klassen. Diese Proteine kommen in allen Zellkernen vor und sind fest an die Phosphatgruppen des Chromatins gebunden. Die Histonmoleküle spielen eine wichtige Rolle in der Chromatin-Struktur. Die Histone werden routinemäßig aus der säurelöslichen Fraktion der Zellkerne aus Kalbsthymus, Hühnererythrocyten oder anderem Ausgangsmaterial isoliert. Die verschiedenen Subtypen werden mittels bekannter Techniken, wie isoelektrischer Fokussierung oder Ionenaustausch- Chromatographie separariert.
- Eine wichtige Eigenschaft der Histonmoleküle, die wichtig für die Verbindungschemie ist, ist - mit Ausnahme von H3 - das Fehlen einer Cystein-sulfhydryl-Gruppe. Histone sind dafür bekannt, daß sie eine große Anzahl von chemischen Modifikationen in der normalen Zelle eingehen, welche die N-methylierung, O-Methylierung, Acetylierung, Phosphorylierung, Adenosin-Diphosphat (ADP) Ribosylierung, Ubiquitinierung und enzymatische Hydrolyse von spezifischen Peptidbindungen einschließt.
- Histone sind in großer Vielzahl aus kommerziellen Quellen erhältlich oder sie können nach bekannten Prozeduren, wie sie nachfolgend aufgeführt werden, isoliert werden.

- Histone, die aus einer der konventionellen Quellen isoliert sind, können benutzt werden und die einzelnen Klassen oder Subtypen sind dabei nicht besonders kritisch. Es werden bevorzugt Histone, isoliert aus menschlichen Quellen für die Herstellung schimärischer Peptide, für die Anwendung zur Behandlung von Menschen eingesetzt.
- Ein anderes transportfähiges Peptid ist Prolactin. Prolactin ist ein Hormon, das von dem Vorderlappen der Hypophyse abgesondert wird. Prolactin ist umfangreich kommerziell erhältlich und es kann aus der Hirnanhangdrüse mittels bekannter Methoden isoliert werden.
- Die schimärischen Peptide werden durch Konjugation eines transportablen Peptids mit dem neuropharmazeutischen Peptid geschaffen. Die Konjugation kann unter Benutzung bifunktionaler Reagentien, die fähig sind, mit jedem der Peptide zu reagieren und eine Brücke zwischen den beiden Peptiden zu bilden, ausgeführt werden. Die bevorzugte Methode der Konjugation umfaßt die Peptid-Thiolierung, bei der die beiden Peptide mit einem Reagens, wie N-Succinimidyl 3-(2-pyridyl-dithio) Propionat (SPDP), behandelt werden, um eine Disulfid-Brücke zwischen den beiden Peptiden zum Ausbilden des schimärischen Peptides zu bilden. Andere bekannte Konjugationsagentien können benutzt werden, solange sie eine Verbindung der beiden Peptide, d.h. des hydrophilen Peptidmedikaments und des transportablen Peptids, ohne diese zu denaturieren, ermöglichen. Bevorzugt kann die Verbindung leicht auseinanderbrechen, sobald das schimärische Peptid in das Hirn eingedrungen ist. Geeignete Beispiele von Konjugationsreagentien schließen Glutaraladehyd und Cystamin und EDAC ein.
- Die Konjugation von Peptiden unter Einsatz von Glutaraldehyd ist beschrieben in Poznansky et al, Insulin: Carrier potential for enzyme and drug therapy, Science 233:1304-1306, 1984. Die Konjugation von Peptiden unter Einsatz von Cystamin und EDAC ist beschrieben in Ito et al, Transmembrane delivery of polypeptide hormones bypassing the intrinsic cell surface receptors: eine Kopplung von Insulin mit a2-Macroglobulin, (a2M), erkennend beide Insulin und a2-M Rezeptoren und ihre biologische Aktivität in Beziehung auf endocytische Durchgänge, siehe Mol Cell Endocrinol 36:165, 1984.
- Beispiele bevorzugter schimärischer Peptide umfassen diejenigen der folgenden allgemeinen Strukturformel
- A-NH (CH&sub2;)&sub2;S-S-(CH&sub2;)&sub2; HN-B
- worin A Somatostatin, Thyrotropin auslösendes Hormon (TRH), Vasopressin, Alpha-Interferon, Endorphin, Muramyl-Dipeptid oder ACTH 4-9 Analoge; und B Histon, Insulin, IGF-I, IFG-II, Transferrin, kationisiertes (basisches) Albumin oder Prolactin ist.
- Andere Beispiele bevorzugter schimärischer Peptide schließen die vorgenannten ein, bei denen die Disulfid-Konjugationsbrücke zwischen A und B durch Brücken der folgenden Struktur ersetzt ist:
- A-NH(CH&sub2;)&sub2;S-S-B (spaltbare Bindung)
- die gebildet werden, wenn Cystamin und EDAC als Brückenreagentien eingesetzt werden;
- A-NH=CH(CH&sub2;)&sub3;CH=NH-B (nicht spaltbare Bindung)
- die gebildet werden, wenn Glutaraldehyd als Brückenreagens eingesetzt wird.
- Die schimärischen Peptide können in den Körper mittels jeder konventionellen Methode einschließlich parenteraler Injektion oder intranasaler Inhalation eingebracht werden. Bevorzugt werden die schimärischen Peptide mit einem verträglichen pharmazeutischen Träger kombiniert und parental injiziert oder, falls gewünscht, mit einem geeigneten Träger kombiniert und intranasal verabreicht gemäß bekannter konventioneller Prozeduren, die für die intranasale Verabreichung von Insulin benutzt werden. Geeignete Trägerlösungen schließen die üblicherweise in injizierbaren oder nasalinhalierten Hormonpräparationen verwendeten Lösungen ein, wie sterile Salzlösung mit einem pH von etwa 5, die ein übliches Bakteriostatika enthält. Die Konzentration eines schimärischen Peptids in dem Träger hängt von dem spezifischen transportfähigen Peptid und dem spezifizischen neuropharmazeutischen Peptid ab. Bevorzugt sind die Gehalte des schimärischen Peptids in dem Träger zwischen 0.001 Gewichts-Prozent bis zu 0,01 Gewichts-Prozent. Als allgemeiner Anhalt sollen Höhe der Gehalte und Prozente der in der Injektions- oder intranasalen Lösung gegenwärtigen schimärischen Peptide den akzeptierten und etablierten Mengen für die einzelnen neuropharmazeutischen Peptide ebenso wie der transportfähigen Peptide entsprechen.
- Es folgen Ausführungsbeispiele:
- Somatostatin ist ein Peptid, das bei Alzheimer's Krankheit nicht ausreichend im Hirn vorhanden ist und das nicht durch die Blut-Hirn-Barriere transportiert wird. Umgekehrt ist Insulin ein Peptid, das durch die Blut-Hirn-Barriere transportiert wird. Die Transportfahigkeit von Insulin durch die Blut-Hirn-Barriere ist in meinem Artikel "Receptor-Mediated Peptide Transport Through The Blood-Brain Barrier" (Endocrine Reviews, Vol. 7, No. 3, August 1986) beschrieben, dessen Inhalt hier mit einbezogen wird.
- Somatostatin und Insulin werden durch Peptid-Thiolierung konjugiert, wobei eine reversible Peptid-Peptid- Konjugationsmethode benutzt wird, die bei Carlsson, et al. in "Protein thiolation and Reversible Protein-Protein Conjugation (Biochem. J. (1978) 173, 723-737) beschrieben ist. Ein heterobifunktionales Reagens N-succinimidyl 3-(2-pyridyldithio)Propionat (SPDP) wurde zum Koppeln eines Lysin- oder freien N-Terminus am Insulin an einen freien Lysin- oder Amino-Terminus am Somatostatin benutzt. Etwa 0.3 mg Insulin und 26 uCi von ¹²&sup5;I-Insulin in 2 ml einer phosphatgepufferten Salzlösung wurden präpariert. Zur Hälfte dieser Menge wurden 4 Lambdas von 20 mM frischer SPDP zugefügt und dieses wurde bei Raumtemperaturen während 45 Minuten einer Inkubation unterworfen.
- Getrennt hiervon wurde 180 uCi von tritiiertem Somatostatin in 180 uL von 0.01 N HCl gelöst und 180 uL von 0.2 M phosphatgepufferter Salzlösung zugefügt. Zur Hälfte dieser Lösung wurden 4 uL von 20 mM SPDP zugefügt und dann während 45 Minuten einer Inkubation unterworfen, gefolgt von einer Ansäuerung mit 20 uL von 0.75 M Natriumacetat (pH = 4.5), gefolgt von einer Reduktion mit 20 uL von 0.25 M Dithiothreit. Dies Produkt wurde bei Raumtemperatur während 30 Minuten einer Inkubation unterworfen und danach einer kurzen Dialyse zum Entfernen der nichtreagierten kleinen Moleküle. Das konjugierte Insulin und konjugierte Somatostatin wurden dann über Nacht bei Raumtemperaturen einer Inkubation unterworfen, anschließend einer Dialyse unterzogen und die Tritium- und ¹²&sup5;I - Radioaktivität gezählt. Hierbei ergab sich insgesamt 53 uCi von ³H-Somatostatin gekoppelt an 5.3 uCi von ¹²&sup5;I-Insulin in 2 ml einer phosphatgepufferten Salzlösung.
- Die Struktur der Somatostatin-Insulin-Schimäre ist nachfolgend dargestellt.
- Somatostatin hat die fölgende Aminosäuresequenz:
- Ala-Gly-Cys-Lys-Asn-Phe-Phe-Trp-Lys-Thr-Phe-Thr-Ser-Sys.
- Insulin ist ein zweikettiges Proteinhormon, dessen Struktur gut bekannt ist.
- Somatostatin wurde von den Peninsula Laboratories erhalten und mittels reduktiver Methylierung unter Einsatz von ³H-Natriumborhydrid tritiiert. Insulin der Sigma Chemical Company wurde mittels oxidativer Iodierung unter Einsatz von Chloramin T und ¹²&sup5;I-Jod iodiert. Die beiden Komponenten wurden unter Einsatz von SPDP, wie in Beispiel 1 beschrieben, miteinander gekoppelt. Die Rinderhirn-Mikrogefäße wurden wie bei Pardridge, et al., "Rapid Sequestration And Degradation Of Somatostatin Analogues By Isolated Brain Microvessels", (Journal of Neurochemistry, Vol. 44, No. 4, 1985, pp. 1178-1184) beschrieben, isoliert.
- ³H-Somatostatin wurde einer Menge Mikrogefäße für eine 60-minütige Inkubation bei Raumtemperatur zugegeben. Einer weiteren Menge Mikrogefäße wurde die ³H-Somatostatin-¹²&sup5;I-Insulin-Schimäre zur Inkubation zugegeben. Wie in der Fig. 1 dargestellt, ist die Aufnahme der Schimäre mehr als doppelt so groß wie die des freien Somatostatins. Die Aufnahme des freien Somatostatins spiegelt die nichtspezifische Bindung, wie sie in dem oben angeführten Artikel erwähnt ist (Journal of Neurochemistry, Vol. 44, No. 4, 1985) wieder.
- Dies Beispiel zeigt die Rezeptor-vermittelte Transcytose oder Endocytose der Somatostatin-Insulin-Schimäre via Insulin- Rezeptor. Frühere Studien haben gezeigt, daß die Rezeptor-vermittelte Endocytose von Peptiden in den isolierten Hirn-Mikrogefäßen ein verläßlicher Index der in vivo Blut-Hirn-Barriere Rezeptor-Transportaktivität von Peptiden ist (siehe mein vorgenannter Artikel in Endocrine Reviews, Vol. 7, No. 3, August 1986).
- Ein schimärisches Peptid wird gemäß der im Beispiel 1 beschriebenen Prozedur hergestellt mit der Ausnahme, daß Transferrin für Insulin eingesetzt ist. Das erhaltene schimärische Peptid wird mit einer sterilen Salzlösung kombiniert, so daß eine Lösung, enthaltend 0.01 Gewichts-Prozent schimärisches Peptid erhalten wird, die dem Patienten parenteral oder intranasal verabreicht wird.
- Ein schimärisches Peptid wird gemäß der in Beispiel 1 beschriebenen Prozedur hergestellt mit der Maßgabe, daß an Stelle Somatostatin Vasopressin steht. Das erhaltene schimärische Peptid wird mit einer sterilen Salzlösung kombiniert, so daß eine Lösung, enthaltend 0.01 Gewichts-Prozent schimärisches Peptid erhalten wird, die dem Patienten parenteral verabreicht wird.
- Ein schimärisches Peptid wird gemäß der in Beispiel 1 beschriebenen Prozedur hergestellt mit der Maßgabe, daß Transferrin an Alpha-Interferon gekoppelt ist. Das erhaltene schimärische Peptid wird mit einer sterilen Salzlösung kombiniert, so daß eine Lösung, enthalten 0.01 Gewichts-Prozent schimärisches Peptid erhalten wird, die dem Patient oder Subjekt parenteral oder intranasal verabreicht wird.
- Ein schimärisches Peptid wird gemäß der in Beispiel beschriebenen Prozedur hergestellt mit der Maßgabe, daß IGF-II an Beta-Endorphin gekoppelt ist. Das erhaltene schimärische Peptid wird mit einer sterilen Salzlösung kombiniert, so daß eine Lösung, enthaltend 0.01 Gewichts-Prozent schimärisches Peptid erhalten wird, die dem Patienten oder Subjekt parenteral oder intranasal verabreicht wird.
- Ein schimärisches Peptid wird gemäß der gleichen Prozedur wie in Beispiel 1 hergestellt mit der Maßgabe, daß Insulin an ACTH 4-9 Analogon gekoppelt ist. Das erhaltene schimärische Peptid wird mit einer sterilen Salzlösung kombiniert, so daß eine Lösung, enthaltend 0.01 Gewichts-Prozent schimärisches Peptid, erhalten wird, die dem Patienten oder Subjekt parenteral oder intranasal verabreicht wird.
- Ein schimärisches Peptid wird gemäß der gleichen Prozedur wie in Beispiel 1 hergestellt mit der Maßgabe, daß kationisiertes Albumin an Hexosaminidase A gekoppelt ist. Das erhaltene schimärische Peptid wird mit einer sterilen Salzlösung kombiniert, so daß eine Lösung, enthaltend 0.01 Gewichts-Prozent schimärisches Peptid erhalten wird, die dem Patienten oder Subjekt parenteral oder intranasal verabreicht wird.
- Ein schimärisches Peptid wird nach der gleichen Prozedur wie in Beispiel 1 hergestellt mit der Maßgabe, daß handelsüblich erhältliches Rinder-Histon Type V für Insulin substituiert ist. Das erhaltene schimärische Peptid wird mit einer sterilen Salzlösung kombiniert, um eine Lösung, enthaltend 0.01 Gewichts-Prozent schimärisches Peptid zu erhalten, die dem Patienten parenteral oder intranasal verabreicht wird.
- Um die Eignung von Histon als einem polykationischen transportablem Peptid zu zeigen, wurden die folgenden Aufnahmetests durchgeführt.
- Ein Radio-Rezeptor-Test wurde mit Rinderhirnkapillaren gemäß der Prozedur wie in Beispiel 2 durchgeführt, außer daß handelsüblich erhältliches Rinder-Histon Type V an Stelle der Somatostatin-Insulin-Schimäre eingesetzt und mit ¹²&sup5;I-Jod und Chloramin-T radiomarkiert wurde. Das Histon wurde mit den Hirnkapillaren während 60 Minuten einer Inkubation unterworfen und die Prozentaufnahme per mg Hirnkapillaren wurde bestimmt. Das Ergebnis dieser Tests ist in Fig. 2 dargestellt und es zeigt, daß die Aufnahme des Histon hoch ist, nahezu 110 % gebunden per mg Protein (Grundbindung ist 3-5 % per mg Protein). Des weiteren wurde gefunden, und Fig. 2 zeigt, daß die Aufnahme des Histons durch Erhöhung der Konzentrationen von nichtmarkiertem Histon sättigbar ist und daß die 50 % Hemmschwelle bei einer Konzentration von 300 ug/ml oder etwa 14 uM erreicht ist. Diese Sättigungsantwort ist typisch für einen Rezeptor-vermittelten oder absorptiv-vermittelten Endocytose-Mechanismus.
- Die Menge der Aufnahme an Histon V wurde in Intervallen von 2, 10, 30 und 60 Minuten gemessen. Es wurde gefunden, daß die Aufnahme des Histons mit der Zeit, wie in Fig. 3a und b dargestellt, ansteigt, wobei die Inkubationen des Histons V bei Temperaturen von 37ºC bzw. 4ºC durchgeführt wurden. Die Aufnahme von Histon wurde bei 4ºC gehemmt, wie in der Fig. 3b ersichtlich. Des weiteren wurde der Histon-Aufnahmewiderstand bei Säurespülung gemessen, indem die Rinder-Hirnkapillaren mit einer milden Säurespülung nach Beendigung der Histonaufnahme behandelt wurden. Die Säurespülung wurde durch Spülen der Hirnkapillaren in einer Lösung von kaltem 0.028 molarem Natriumacetat, 0.02 molarem Natriumbarbital und 0.12 molarem Natriumchlorid mit einem pH von etwa 3.0 ausgeführt. Die Ergebnisse dieser Versuche zeigen, daß die Aufnahme von Histon durch isolierte Hirnkapillaren partiell resistent gegenüber milder Säurespülung ist, siehe die graphischen Ergebnisse in Fig. 3a und b, die die säurebeständige Histonaufnahme zeigen. Die Ergebnisse weisen nach, daß etwa ein Drittel des aufgenommenen Histons tatsächlich in die Hirnkapillaren durch Endocytose gelangt ist. Die vorgenannten Ergebnisse zeigen, daß beide, Bindung und Endocytose, bei Inkubation bei 4ºC verlangsamt sind, was typisch für einen Rezeptor-vermittelten oder adsorptiv-vermittelten Aufnahmemechanismus für den Transport quer durch die Blut-Hirn-Barriere ist.
- Eine nanomolare Konzentration von ¹²&sup5;I-Histon V wurde bei Raumtemperatur während 10 Minuten in der Gegenwart von Hirnkapillaren inkubiert. Die Menge des von den Hirnkapillaren aufgenommenen markierten Histons wurde dann bestimmt. Die Ergebnisse der Versuche zeigen, daß die Aufnahme des ¹²&sup5;I-Histons V linear in bezug zu der Menge von kapillarem Protein in der Inkubationsflasche ist. Die Ergenisse dieser Tests sind graphisch in der Fig. 4 dargestellt.
- Dieses Beispiel demonstriert die Herstellung eines Histon/Meerrettich-Peroxidase-Konjugates, das getestet und bei dem gezeigt wird, daß es von dem Hirn-Mikrogewebe abgesondert wird. Das Histon/Meerrettich-Peroxidase -Konjugat wurde folgendermaßen hergestellt:
- Sulfhydryl-Gruppen wurden in das Histon mittels einer Modifikation des Prozesses, wie er bei Jue et al. (1978) Biochemistry 17-5399-5405 beschrieben ist, eingeführt. Sechzehn Milligramm Histon (Sigma-Type II-AS) wurden in 3.0 ml von 50 mM Triethanolamin Puffer, pH 8.25, in einem abgeschlossenen Reaktionsgefäß aufgelöst. Die Protein-Lösung wurde erschöpfend entgast und mit Stickstoff gereinigt. Hierzu wurden 0,074 ml von einer 0.5 M Lösung von 2-Iminothiolan (Pierce) mittels Spritze zugegeben. Die Mischung konnte während 45 Minuten bei 25 ºC reagieren. Um alle Sulfhydryl-Gruppen zu reduzieren, wurde das Produkt dann mit 0.5 ml von 0.1 M Dithiotreit (Calbiochem) während 30 Minuten bei 37ºC unter mildem Schütteln (75 Upmin) einer Inkubation unterworfen. Das modifizierte Protein wurde dann von der Reaktionsmischung mittels Chromatographie über Sephadex G-25 (1.5 x 30 cm) unter Verwendung von entgastem von Stickstoff gereinigtem Triethanolaminpuffer gereinigt. Die Proteinelution wurd mittels UV-Absortion bei 280 nm (5-5'Dithio-bis(-2-nitrobenzoe-Säure), Ellman (1959) Arch. Biochem. Biophys. 82:70-77) gemessen. Charakteristisch wurden bei diesen Konditionen 1.1 Mol von Sulfhydryl/Mol Protein eingeführt.
- Meerrettich-Peroxidase (HRP) wurde mit Maleimid-Gruppen unter Einsatz von N-γ-Maleimidobutyryloxysuccinimid (GMBS) nach einer Methode, beschrieben bei Hashida et al. (1984) J. Appl. Biochem., 6:56-63, gewonnen. 20 mg von Peroxidase (0.5 mmol, Boeringer Mannheim EIA Grad) wurden 1.5 ml von 0.1 M Natriumphosphatpuffer, pH 7.0 zugegeben. Sechzehn mg (57 mmol) GmbS wurden in 0.2 ml von DMF aufgelöst und der Peroxidaselösung zugegeben. Die Mischung wurde während 30 Minuten bei 30ºC geschüttelt. Das hergeleitete HRP wurde mittels Chromatographie der Reaktionsmischung über Sephadex G-25 (1.5 x 30 cm) unter Einsatz von 0.1 M Phosphat, pH 6 als Eluat isoliert. Die HRP-Elution wurde bei ihrer Absorption von 403 nM überprüft.
- Der pH der thiolierten Histonlösung wurde auf 6 mit 1 N HCl eingestellt. Dieses wurde unmittelbar dem vom Maleimid abgeleiteten HRP zugefügt. Die Lösung wurde erschöpfend entgast, vom Stickstoff gereinigt und versiegelt. Die Komponenten konnten während 48 Stunden bei 25ºC reagieren. Die Trennung des Konjugats von den nichtreagierten Proteinen wurde an einer Sephadex G 100 SF Säule (2.5 x 54 cm) unter Einsatz von 0.1 M Phosphat, pH 6 als Eluat durchgeführt. Die von der Säule isolierten Fraktionen wurden auf ihre Absorption bei 230 nm und 403 nm geprüft. Das zuerst absorbierende Eluat wurde gesammelt, gegen Wasser bei 4ºC über Nacht dialysiert und lyophilisiert. Die folgende Rekonstitution der Peroxidase in 2 mL von PBS, 80 % (16 mg) wurde mit dem Konjugat-Peak isoliert. SDS-Page Analyse des Konjugats zeigte mehrere Bänder von MW 54 kD und darüber auf und konnte kein freies HRP oder Histon anzeigen. Des weiteren konnte das ganze Protein, das mit der Methode von Lowry als 1/3 der Masse bestimmt wurde, für das HRP angesehen werden unter Annahme einer 6/1 Histon/HRP- Konjugationsrate. Das Konjugat wurde zu 1 mg/ml (Total-Protein nach Lowry-Methode) eingestellt und bei 4ºC für eine in vivo Analyse aufbewahrt.
- Das Histon/HRP-Konjugat wurde wie nachfolgend beschrieben getestet:
- BALB/C Mäuse (6 Wochen, weiblich) wurden intravenös mit 0.2 ml Histon/HRP Konjugat in einer phosphatgepufferten Salzlösung (PBS) injiziert. Die Tiere wurden nach 15 Minuten mit einer tödlichen Injektion von Chlorhydrat getötet, mit 5.0 ml von PBS, enthaltend 4 % Paraformaldehyd, übergossen und dann wurde das Hirn entfernt und für weitere 15 Minuten in Fixativ gelegt. Gefrorene Abschnitte (30 Micron dick) wurden präpariert und mindestens 20 Minuten in PBS getaucht. Die Abschnitte wurden von PBS befreit und in eine Inkubationslösung (20 mm Natriumacetat, pH 3.3, 2.5 % Ethanol, 4 mM Natriumnitroprussiat, 250 mM 3,3',5,5' - Tetramethylbenzidin (TMB)) 20 Minuten lang gelegt. Die Reaktion wurde durch Zugabe von 1 ml von 0.3 % H&sub2;O&sub2; auf je 100 ml Inkubationslösung initiiert. Die Reaktion wurde für 10 Minuten bei Raumtemperatur fortgesetzt und dann wurden die Abschnitte in 20 mM Natriumacetat pH 3.3 transferiert. Die Abschnitte wurden sechsmal gewaschen, jeweils fünf Minuten lang. Gewebeabschnitte wurden auf mit Gelatine überzogene Träger montiert und während sieben Stunden bei Raumtemperatur getrocknet. Die Träger wurden bei 60ºC für eine Stunde erhitzt und während 30 Sekunden mit 0.5 % Toluidinblau pH 4.5 gebeizt. Die Träger wurden in einer graduierten Alkoholserie dehydriert, mit Xylol gewaschen und mit Permount montiert.
- Die Kapillaren des Hirns wurden in Gegenwart des TMB-Reaktionsproduktes in Verbindung mit der lumenalen Oberfläche der Kapillaren sichtbar gemacht. Das Produkt wurde bei Tieren, injiziert mit dem Histon/HRP-Konjugat, beobachtet, jedoch wurde es nicht bei Tieren beobachtet, die weder mit HRP allein injiziert waren noch bei mit PBS injizierten Tieren.
- Beide weißen und grauen Massen wurden mit TMB markiert. Es gab keine offenkundige Erscheinung spezifizischer Lokalisation nur an hoch mit Gefäßen angereicherten Bereichen des Hirns.
- Dieses Beispiel setzt die Experimente, die zeigen, daß Histon in der Lage ist, die Blut-Hirn-Barriere in vivo zu penetrieren, fort. Kalbsthymus-Histon wurde mit ¹²&sup5;I-Jod iodiert und es wurde gefunden, daß es schnell durch isolierte Rinderhirnkapillare aufgenommen wird, die als ein in vitro Modellsystem der Blut-Hirn-Barriere über einen zeit- und temperaturabhängigen Mechanismus benutzt wurden. Die Bindung war sättigbar und ein Scatchard-Diagramm der Bindungsdaten war linear mit einer Ausbeute von KD = 15.2 +/- 2.8 uM und einer maximalen Bindung (Bmax) = 7.7 +/- 1.0 nmol/mg Protein (Fig. 5).
- Andere Polykationen, wie Protamin oder Polylysin behindern merkbar die Aufnahme von (¹²&sup5;I)-Histon, aber kationisiertes Albumin zeigt minimale Unterbindung und kationisiertes Immunglobulin verursacht keine Behinderung der Aufnahme von (¹²&sup5;I)-Histon bei Rinderhirnkapillaren. Das bei in vivo Hirn erreichte Volumen der Verteilung von (¹²&sup5;I)-Histon betrug 159 +/- 70 uL/g während 10 Minuten Halsschlagader-Perfusion, verglichen dem 10 Minuten Volumen der Verteilung für (³H)-Albumin von 17 +/- 7 uL/g. Der größte Anteil dieser Aufnahme entspricht einer Sequestration durch das Gewebe, aber etwa 8 % des gesamten durch das Hirn aufgenommenen Histons wurde in nicht metabolisiertem Zustand in die Hirnzwischenräume transportiert aufgefunden (basierend auf Trichloressigsäure (TCA) Fällbarkeit).
- Die Experimente wurden wie nachfolgend durchgeführt:
- (¹²&sup5;I)-Jod war erhältlich von DuPont-New England Nuclear Corporation (Boston, MA). (³H)-NaBH&sub4; wurde von Amersham Corporation (Chicago, Il) gekauft. Rinderalbumin (Pentex fraction V) war erhältlich von Miles Laboratories (Elkhart, IN). Männliche Sprague-Dawley Ratten (200-300 g) waren erhältlich von Batin und Kingman (Fremont, CA). Kalbsthymus-Histon VS (Lysin-reich) und alle anderen Reagenzien waren erhältlich von Sigma Chemical Company (St. Louis, MO).
- Histon wurde für eine spezifizische Aktivität von 10-20 uCi/ug unter Einsatz von (¹²&sup5;I)-Jod und Chloramin T iodiert. 50 ug von Histon (2.3 nmol) wurden mit 2.5 mCi von (¹²&sup5;I)-Jod (1.2 nmol) und 2.1 nmol Chloramin T reagiert, gefolgt von einer 60 Sekunden währenden Inkubation bei Raumtemperatur. Die Mischung wurde mit 0.01 N HCl angesäuert und auf eine 0.7 x 28 cm Säule von Sephadex G-25 (mittel) und 0.01 N HCl gegeben. Das iodierte Histon spült in das freie Volumen und war zu 98 % ausfällbar mit Trichchloressigsäure (TCA). Das (¹²&sup5;I)-Histon wurde bei 4ºC in 0.01 N HCl gelagert, aber es unterlag einer relativ schnellen Deiodierung während des Verlaufs einer Woche. Daher wurden die in vivo Halsschlagader-Perfusionsexperimente mit 24 Stunden Iodierung durchgeführt und die isolierten Kapillarexperimente wurden mit drei bis vier Tagen Iodierung durchgeführt.
- Rinder-Albumin (Pentex franction V) wurde auf eine spezifische Aktivität von 0.4 uCi/ug mit (³H)-NaBH&sub4;, wie vorangehend beschrieben (Pardridge et al., 1985a) tritiiert. Die TCA- Ausfällbarkeit dieser Präparation war größer als 99 %.
- Rinderhirn-Mikrogefäße wurden mit einer mechanischen Homogenisierungstechnik, wie sie vorangehend beschrieben wurde (Pardridge et al.: Rapid sequestration and degradation of somatostatin analogues by isolatet brain microvessels. J. Neurochem. 44: 1178-1184, 1985) aus frischer Rinder-Großhirnrinde, erhalten aus dem lokalen Schlachterhaus, isoliert. Das endgültige Mikrogefäß-Pellet wurde cryo-konserviert in 0.28 M Saccharose, 0.02 M Tris (pH 7.4) and 2 mM Dithiothreit in flüssigem Stickstoff (- 70ºC). Am Tage des Experimentes wurden die Mikrogefäße aufgetaut, zentrifugiert und in Ringer-HEPES-Puffer (RHB) (10 mM HEPES, pH 7.4, 141 mN NaCl, 4 mM KCl, 2.8 mM CaCl&sub2; und 0.1 gm/dl Rinderserum Albumin) wieder aufgelöst. Die Aufnahme von (¹²&sup5;I)-Histon durch Rinderhirn-Mikrogefäße wurde, wie vorangehend beschrieben (Pardridge et al.: Cationization of immunoglobulin G (IgG) as a new strategy for enhanced IgG delivery through the bloodbrain barrier. Clin. Res. 37: 140A) durchgeführt. Kurz, etwa 100 ug von kapillarem Protein wurde mit 0.2 uCi/ml von (¹²&sup5;I)-Histon in einem Volumen von 0.45 ml RHB bei 37ºC oder 4ºC während Zeitperioden von 5 Sekunden bis 60 Minuten inkubiert. Konkurrierende Bindungsstudien wurden durch Hinzufügen verschiedener Konzentrationen von entweder unmarkiertem Histon, kationisiertem Immunglobulin, natürlichem oder kationisiertem Rinderserum Albumin, Protamin oder Polylysin (59,000 Molekulargewicht) durchgeührt. Am Ende der Inkubationsperiode wurde die Mischung bei 10,000 g für 45 Sekunden zentrifugiert und das Kapillar-Pellet in 0.5 ml von 1 N NaOH aufgelöst, gefolgt vom Zählen der (¹²&sup5;I) und der Proteinbestimmung durch die Methode von Lowry et al. (Protein measurement with the Folin phenol reagent. J. Biol. Chem. 193: 262-275, 1951).
- Die Internalisation des markierten Histons durch die isolierten Rinderhirnkapillaren wurde mittels einer milden Säure-Waschprüfung, wie vorangehend beschrieben (Pardridge et al., Rapid sequestration and degradation of somatostatin analogues by isolated brain microvessels. J. Neurochem. 44: 1178-1184, 1985), abgeschätzt. Die Säure-Waschlösung bestand aus 0.12 M NaCl, 0.02 M Natriumacetat (pH = 3) und 0.028 M von Natriumbarbital.
- Die Quantifizierung des in vivo Transportes von (¹²&sup5;I)-Histon durch die Blut-Hirn-Barriere in vivo wurde mit einer inneren Halsschlagaderperfusionstechnik gekoppelt mit einer kapillaren Erschöpfungsmethode bestimmt. Ratten wurden anästhesiert mit Ketamin/Xylazin (Ketamin 200 mg/kb, i.p./Xylazin 2 mg/kg i.p.) und folgend dem Verlauf der rechten Haupthalsschlagader wurden die Hinterhaupt-, obere Schilddrüsen- und Gaumenarterien durch Elektrokoagulation geschlossen. Die rechte äußere Halsschlagader wurde mit einem Polyethylen-Katheter (PE-10) katheterisiert. Die Haupthalsschlagader wurde, kurz bevor die Perfusion gestartet wurde, zugezogen und geschlossen gehalten. Das perfusat bestand aus Krebs-Henseleit Puffer, pH 7.4 (118 mM NaCl, 4.7 mM KCl, 2.5 mM CaCl&sub2;, 1.2 mM MgSO&sub4;, 1.2 mM KH&sub2;PO&sub4;, 25 mM NaHCO&sub3;, 10 mM D-Glucose und 3 gm/dl Rinderserum Albumin), enthaltend 2.5 uCi/ml von (¹²&sup5;I)-Histon und 25 uCi/ml von (³H)-Albumin. Das Perfusat wurde bei 37 º C gehalten und kontinuierlich während der Perfusion oxygeniert, die mit einer Flow-Rate (Harvard peristaltic pump Model 1210) von 1-1.2 ml/min für 1 bis 10 Minuten durchgeführt wurde. Für Perfusionszeiten länger als 2.5 Minuten wurde das Rattenblutvolumen konstant durch Abziehen von Blut aus der Oberschenkelarterie (durch einen PE-50 Katheter, gefüllt mit Heparin) mit derselben Flow-Rate (Harvard syringe pump Model 940) konstant gehalten. Nach der Perfusion wurden die Tiere geköpft und die ipsilateralen Hirnhemisphären wurden entfernt. Das choroide Nervengeflecht wurde entfernt und das Hirn gewogen und dann in 3.5 ml eines physiologischen Puffers, pH 7.4 (10 mM HEPES , 141 mM NaCl, 4 mM KCl, 2.8 mM CaCl&sub2;, 1 MM MgSO&sub4;, 1 mM NaH&sub2;PO&sub4;, 10 mM D-Glucose) homogenisiert. Vier ml von 26 %-iger Dextranlösung (79,000 Molekulargewicht) wurde einer abschließenden Dextrankonzentration von 13 % zugegeben und das Material wurde re-homogenisiert (3 Schläge). Alle Homogenisierungsprozeduren wurden bei 4ºC durchgeführt.
- Nach Entfernung eines Aliquots des Homogenates für die Radioisotopenzählung wurde das Verbleibende bei 5,400 g während 15 Minuten bei 4ºC in einem schwingenden Bucket-Rotor (Beckmann JA-7.5 Rotor, Beckman J2-21 Zentrifuge) zentrifugiert. Der Supernatant und das Pellet wurden sorgfältig separiert. Mikroskopische Prüfung des Pellets zeigte, daß es aus Hirngewebe, roten Zellen und Hirnnucleus bestand, wohingegen der Supernatant im wesentlichen bar von Gewebe war. Aliquote der Supernatant-Fraktion und des Perfusats (dem eine Menge von Dextran gleich der in dem Supernatant gegenwärtigen Menge zugegeben wurde) wurden für eine 25 %-ige TCA Ausfällbarkeit genommen. Beispiele von Homogenat, Pellet, Supernatant und Perfusat wurden in 2 ml Soluene-350 (Packard Instrument Co., Downers Grove, IL) gelöst und präpariert für (¹²&sup5;I), (³H) zweifache Isotopen-Flüssig-Szintillation-Spektrometrie, wie vorangehend beschrieben (Pardridge, Carrier-mediated transport of thyroid hormones through the blood-brain barrier. Primary role of albumin-bound hormone. Endocrinology 105: 605-612, 1979).
- Volumen der Verteilung für beide der (¹²&sup5;I)- und (³H)-markierten Proteine wurden für das Homogenat, Pellet und das postvasculare Supernatant kalkuliert:
- VD= DPM-f/g Gewebe/DPM-ml Perfusat
- wobei DMP-f das DPM in der entsprechenden Fraktion (d.h. Homogenat, Pellet oder postvasculares Supernatant) ist.
- Die Supernatantvolumen der Verteilung werden wie nachfolgend korrigiert:
- Supernatant VD = ((¹²&sup5;I) VD x % TCA Ausfällbarkeit)-((3H) VD).
- Das korrigierte Supernatant VD ist ein quantitatives Maß der Proteinverteilung in die Hirnzwischenräume durch Transcytose durch die Blut-Hirn-Barriere. Die Subtraktion des VD für (³H)-Albumin wurde für die Korrektur für radioaktives Protein, enthalten in dem Lumen des Hirngewebes, das aus den Gefäßen infolge der Homogenisierung und Zerstörung dieser Gefäße ausläuft, für notwendig erachtet.
- Ein 0.5 ml Aliquot eines physiologischen Puffers, enthaltend 5 uCi von (¹²&sup5;I)-Histon und 50 uCi von (³H)-natürlichem Albumin wurde schnell in eine Oberschenkelvene durch eine 27-Gauge-Nadel injiziert. Bei 0,25, 5, 30, 60, 120 und 180 Minuten nach der Injektion wird das Tier schnell laparotomisiert und ein 0.5 ml Aliquot von arteriellem Blut aus der absteigenden Aorta entnommen, danach das Tier geköpft und das Hirn und neun andere Organe (Herz, Leber, Milz, Hoden, Dünndarm, Skelettmuskel, Fett, Niere und Lunge) entnommen.
- Die Gewebe werden in Soluene-350 gelöst und mittels zweifach Isotopen-Flüssig-Szintillations-Zählung analysiert. Die Blut (³H) und (¹²&sup5;I) Radioaktivitäten werden in bezug auf DPM/ml als Prozent der injizierten Dose, das ist A(t), normalisiert und diese Daten genügen der folgenden biexponentialen Funktion:
- benutzend eine von Derivativ freie, nichtlineare Regressions-Analyse (PAR von BMDP, Biomedical Computer P Series Programs, entwickelt bei UCLA Health Sciences Computing Facility). Da die Standardabweichung etwa proportional zu den Mitteln ist, wurden die Daten gewichtet, benutzend Gewicht = 1/(Clearance). Das Integral der arteriellen Radioaktivitätskurve wurde aus diesen Daten wie folgt bestimmt:
- wobei t = die Zeit nach der Injektion darstellt. Das Volumen der Verteilung von Histon oder Albumin im Hirn und den neun anderen Organen wurde aus dem Verhältnis von DPM/gm Gewebe, geteilt durch das integrierte DPM/ml Blut bestimmt. Die TCA-Ausfällbarkeit des Serums (³H)-Albumin war größer als 98 % zu allen Zeitpunkten. Jedoch gab es einen progressiven Abfall in der TCA-Ausfällbarkeit des (¹²&sup5;I)-Histons. Deshalb wurden nur arteriell TCA ausfällbare (¹²&sup5;I) Zählpunkte in der Berechnung der Räumung (Clearance) von Histon aus oder den Volumen der Verteilung im Blut oder den Organen benutzt.
- (¹²&sup5;I)-Histon wurde schnell durch isolierte Rinderhirn-Kapillaren bei 37ºC aufgenommen und etwa 25 % dieser Aufnahme war resistent gegenüber einer milden Säure-Spülung und soll das internalisierte Histon (siehe Fig. 3) repräsentieren.
- Beide, die Bindung und Internalisierung, werden durch Inkubation bei 4 ºC (siehe Fig. 3) verlangsamt. Die Aufnahme des (¹²&sup5;I)-Histons durch isolierte Rinderhirnkapillaren war linear in bezug auf die Menge der Kapillaren, die dem Inkubationsgefäß im Bereich von 47-210 ug von kapillarem Protein (Daten nicht dargestellt) zugegeben wurden. Die Bindung des (¹²&sup5;I)-Histons an die isolierten Hirn-Mikrogefäße war sättigbar mit einem ED&sub5;&sub0; von etwa 300 uG/ML (14 uM), wie durch die Daten in Fig. 2 gezeigt. Diese Sattigungsdaten wurden mittels Scatchard Analyse analysiert, um das Diagramm in Fig. 5 zu erhalten. Die Dissoziationskonstante (KD) = 15.2 +/- 2.8 uM und die maximale Bindung oder Bmax = 7.7 +/- 1.0 nmol/mg Protein. Die Bindung von (¹²&sup5;I)-Histon an isolierte Rinderhirn-Mikrogefäße wurde durch andere polykationische Proteine, wie Protamin oder Polylysin inhibiert, aber sie wurde nur minimal inhibiert durch kationisiertes Albumin und wurde nicht inhibiert durch kationisiertes Immunglobulin G oder natürliches Albumin (siehe Tabelle 1). - Das Homogenat VD für (¹²&sup5;I)-Histon erhöht sich mit der Zeit und erreicht 159 +/- 70 uLg&supmin;¹ nach 10 Minuten (siehe Tabelle 2). Das meiste dieses (¹²&sup5;I)-Histons wird durch das Hirn aufgenommen, jedoch wird es in den Gefäßabteilen abgesondert, da der postvasculare Supernatant VD von (¹²&sup5;I)-Histon nach 10 Minuten 12 +/- uLg&supmin;¹ war, was 8 % der gesamten Aufnahme darstellt. Das (¹²&sup5;I)-Histon in dem Supernatant, das TCA ausfällbar war, war 90 +/- 3 % bei 10 minütiger Perfusion. Das Homogenat VD für (¹²&sup5;I)-Histon war nahezu zehnfach größer als das Homogenat VD für (³H)-Albumin (siehe Tabelle 2). Gemäß Definition ist der Supernatant VD für (³H)-Albumin = 0. Das Pellet VD für (³H)-Albumin war 0.76 +/- 0.20 uLg&supmin;¹ bei 10 minütiger Perfusion, das ist mehr als 100-fach weniger als das Pellet VD für (¹²&sup5;I)-Histon (siehe Tabelle 2). Tabelle 2. - In der Fig. 6 ist der Zerfall in Plasma (¹²&sup5;I)-Histon, das in TCA ausfällbar ist und der Zerfall in Plasma (³H)-Albumin nach einer einzigen intravenösen Injektion dargestellt. Die (³H)-Albumin Daten konnten nicht mit einer biexponentiellen Funktion in Verbindung gebracht werden, jedoch genügten sie einer monoexponentialen Funktion. Die (¹²&sup5;I)-Histon Blutdaten konnten weder einer monoexponentialen noch einer triexponentialen Funktion angepaßt werden, sondern genügten einer biexponentialen Funktion und die Abschnitte und Anstiege der beiden exponentialen Zerfälle sind in der Fig. 6 dargestellt.
- Nach der schnellen Freigabe vom Blut wurde (¹²&sup5;I)-Histon monoexponential mit einer Halbzeit von 2.0 +/- 0.5 Stunden freigesetzt und dieser Wert entsprach etwa 40 % der Halbzeit für die (³H)-Albumin Freisetzung, 4.8 +/- 1.8 Stunden (siehe Fig. 6). Das 60-Minuten Hirn VD von (¹²&sup5;I)-Histon und die Raten der 60 Minuten (¹²&sup5;I)-Histon VD/(³H)-Albumin VD für Hirn und die weiteren neun Organe sind in der Tabelle 3 dargestellt. Die 60 Minuten VD Daten werden gezeigt, da mit Ausnahme von Hoden und Dünndarm das Hirn und die anderen Organe die maximale Körperverteilung nach 60 Minuten erreichten. Die 180 Minuten VD war 41 % und 53 % höher als die 60 Minuten VD für Hoden bzw. Dünndarm. Die 180 Minuten VD war 40 % kleiner als die 60 Minuten VD für Hirn, Lunge und Milz. Die VD war im wesentlichen unverändert bei 60 oder 180 Minuten für Leber, Herz, Nieren, Muskel oder Fett. Tabelle 3. - Dies Beispiel zeigt, daß (¹²&sup5;I)-Histon beide, die lumenale und die antilumenale, Seiten der Hirnkapillaren bindet. Die in vivo Perfusionsstudien von Tabelle 2, die die sehr hohe microvasculare Pellet VD für (¹²&sup5;I)-Histon in bezug auf (³H)-Albumin zeigen, demonstrieren, daß (¹²&sup5;I)-Histon von der lumenalen Membran der Hirnkapillaren gebunden ist. Umgekehrt muß auch eine Bindung an die antilumenale Membran vorhanden sein, um die schnelle Bindung innerhalb 5 Sekunden nach Inkubation mit den isolierten Rinderhirn-Mikrogefäßen zu erklären (siehe Fig.3).
- Die Bindung des (¹²&sup5;I) Histons an die Hirngefäße ist temperaturabhängig (Fig. 3) und sättigbar (Fig. 2). Die Sättigung ED&sub5;&sub0; von 14 uM Histon zeigt, daß die Kapazität des Histonaufnahmesystems sehr hoch ist. Zum Beispiel war die Sättigung ED&sub5;&sub0; für kationisiertes Albumin 0.05 mg/ml (0.07 uM) (Kumagai et al.: Absorptive-mediated endocytosis of cationized albumin and a β-endorphin-cationized albumin chimeric peptide by isolated brain capillaries. Model System of blood-brain barrier transport. J. Biol.- Chem. 262:15214-15219, 1987) und für kationisiertes Immunglobulin 1 mg/ml (6 uM) (Triguero et al.: Cationization of immunoglobulin G (IgG) as a new strategy for enhanced IgG delivery through the blood-brain barrier. Clin. Res. 37: 140A). Das Bmax für Histon von 7.7 +/- 1.0 nmol/mg Protein ist annähernd das Fünffache des Bmax von kationisiertem Immunglobulin G und ist annähernd 90 mal größer als das Bmax für die Bindung von kationisiertem Albumin. Die voneinander verschiedenen Bmax-Werte für die verschiedenen polykationischen Proteine lassen vermuten, daß diese Moleküle sich an verschiedene Gruppen negativer Ladungen an der Endothelmembrane der Hirnkapillaren binden. Zum Beispiel inhibitiert kationisiertes Albumin nur wenig die Histonaufnahme, während kationisiertes Immunglobulin keine Wirkung hat, ebenso wie wie Polylysin (siehe Tabelle 1).
- Nachfolgend seiner Bindung an die Oberfläche der Kapillaren, wie durch die Resistenz gegen die milde Säurewaschung (Fig. 3) aufgezeigt, macht Histon eine absorptivvermittelte Endocytose in das endothele Cytoplasma der Hirnkapillaren durch. Mehr noch, die Daten der Tabelle 2 zeigen, daß annährend 8 % des gesamten Histon, gebunden und durch Endocytose durch die Hirnkapillaren aufgenommen, eine Exocytose in die Hirnzwischenräume in vivo durchmacht, wodurch ein kompletter Durchgang von Transcytose durch das Kapillarendothel vollendet wird.
Claims (11)
1. Schimärisches Peptid zum Befördern eines
neuropharmazeutischen Agens in das Hirn durch Transcytose
durch die Blut-Hirn-Barriere, enthaltend ein transportables
zum Durchqueren der Blut-Hirn-Barriere durch Transcytose
befähigtes Peptid, das mit dem neuropharmazeutischen Agens
konjugiert ist, wobei als transportables Peptid ein Histon
vorgesehen ist.
2. Schimärisches Peptid nach Anspruch 1,
bei dem das Histon aus einer Humanquelle isoliert ist.
3. Schimärisches Peptid nach Anspruch 1 oder 2,
bei dem das Histon ausgewählt ist aus den Histonen der
Klassentypen I - V.
4. Schimärisches Peptid nach einem der vorangehenden
Ansprüche, bei dem das neuropharmazeutische Agens ein
hydrophiles Peptid ist.
5. Schimärisches Peptid nach Anspruch 4,
bei dem das neuropharmazeutische Agens ausgewählt ist aus
der Gruppe, bestehend aus Somatostatin, Thyrotropin
freisetzendes Hormon (TRH), Vasopressin, Alpha-Interferon,
Endorphin, Muramyl-Dipeptid und
L-methionyl(sulfon)L-glutamyl-L-histidyl-L-phenylalanyl-D-
lysyl-L-phenylalanin.
6. Schimärisches Peptid nach einem der vorangehenden
Ansprüche, bei dem das transportable Peptid und das
neuropharmazeutische Agens über ein Kopplungsagens
konjugiert sind.
7. Schimärisches Peptid nach Anspruch 6,
bei dem das Kopplungsagens fähig ist, das transportable
Peptid mit dem neuropharmazeutischen Agens über
Peptid-Thiolierung oder Lysin-Kopplung über Glutaraldehyd
zu konjugieren.
8. Schimärisches Peptid nach Anspruch 1 einer Formel
A-NH (CH&sub2;)&sub2;S-S-(CH&sub2;)&sub2; NH-B
worin A ein neuropharmazeutisches Agens und B ein Histon
ist.
9. Schimärisches Peptid nach Anspruch 8,
bei dem A ausgewählt ist aus der Gruppe, bestehend aus
Somatostatin, Thyrotropin freisetzendes Hormon (TRH),
Vasopressin, Alpha-Interferon,
Endorphin, Nuramyl-Dipeptid und
L-methionyl(sulfon)L-glutamyl-L-histidyl-L-phenylalanyl-D-
lysyl-L-phenylalanin.
10. Komposition, enthaltend ein schimärisches Peptid gemäß
einem der vorangehenden Ansprüche und einen pharmazeutisch
akzeptablen Träger für das schimärische Peptid.
11. Komposition nach Anspruch 10,
bei der der pharmazeutisch akzeptable Träger eine sterile
Salzlösung ist.
