Abstract
Mikrokügelchen
geeignet zur aktiven Embolisierung umfassend einen biokompatiblen,
quervernetzten und im Wesentlichen hydrophilen Polymer und eine oder
mehrere aktive Komponenten, wobei die Komponente ein Medikament,
einen Impfstoff oder eine beliebige Kombination davon umfasst.
Claims (44)
- Mikrokügelchen geeignet zur aktiven Embolisierung umfassend einen biokompatiblen, quervernetzten und im Wesentlichen hydrophilen Polymer und eine oder mehrere aktive Komponenten, wobei die Komponente ein Medikament, einen Impfstoff oder eine beliebige Kombination davon umfasst.
- Das Mikrokügelchen nach Anspruch 1, wobei das Mikrokügelchen ein oder mehr Elastomere umfasst.
- Das Mikrokügelchen von Anspruch 2, wobei die Elastomere ausgewählt sind aus der Gruppe bestehend aus akrylischen Polymeren, Akrylamidpolymeren, Vinylalkoholpolymeren, Akrylatpolymeren, Polysacharin, Silikonen und Mischungen davon.
- Das Mikrokügelchen von Anspruch 2, wobei der Durchmesser des Mikrokügelchens ungefähr 10 μm bis ungefähr 2000 μm beträgt.
- Das Mikrokügelchen von Anspruch 4, wobei der Durchmesser des Mikrokügelchens ungefähr 50 μm bis ungefähr 300 μm beträgt.
- Das Mikrokügelchen von Anspruch 1, wobei das Mikrokügelchen quellbar ist.
- Das Mikrokügelchen von Anspruch 6, wobei das Mikrokügelchen Polymere umfasst, die ausgewählt sind aus der Gruppe bestehend aus Natriumakrylatpolymer, Akrylamidpolymer, Akrylamidderivatpolymer oder -copolymer, Natriumakrylat- und Vinylalkohol-Copolymer, Vinylacetat und Akrylsäureester-Copolymer, Vinylacetat- und Methylmaleat-Copolymer, quervernetztes Isobutylen-Maleinanhydrid-Copolymer, Stärke-Akrylnitril Propf-Copolymer, quervernetztes Natriumpolyacrylat-Polymer und quervernetztes Polyethylenoxid.
- Das Mikrokügelchen nach Anspruch 6, wobei der Durchmesser des Mikrokügelchens vor der Quellung ungefähr 10 μm bis ungefähr 400 μm beträgt.
- Das Mikrokügelchen von Anspruch 6, wobei der Durchmesser des Mikrokügelchens vor der Quellung ungefähr 10 μm bis ungefähr 200 μm beträgt.
- Das Mikrokügelchen nach Anspruch 6, wobei der Durchmesser des Mikrokügelchens nach der Quellung ungefähr 10 μm bis ungefähr 2000 μm beträgt.
- Das Mikrokügelchen nach Anspruch 1, wobei das Medikament ausgewählt ist aus der Gruppe bestehend aus Antitumormedikamenten, Antiangiogenesemedikamenten, Antipilzmedikamenten, Antivirusmedikamenten, Medikamenten zur Bekämpfung von Entzündungen, antibakteriellen Medikamenten und Antihistaminmedikamenten.
- Das Mikrokügelchen nach Anspruch 1, wobei der Impfstoff ausgewählt ist aus der Gruppe bestehend aus Impfstoffen gegen Pneumokokkus, Poliomyelitis, Milzbrand, Tuberkulose (BCG), Hepatitis A, Cholera, Meningokokkus A, C, Y, W135, Pest, Tollwut (menschlich, diploid), Gelbfieber, Japanische Encephalitis, Typhus (Phenol und Hitze abgetötet), Hepatitis B, Diphterie, Tetanus, Keuchhusten, H. Influenzae TypB, Polio, Masern, Mumps, Röteln, Windpocken, Streptokokkus Pneumoniae Ty (lebendige Bakterienmutanten), Vi (Vi kapsuläres Polysacharid), DT (toxoid), Td (toxoid), aP (inaktives bakterielles Antigen/accelular (DtaP)), Hib (bakterielles Polysacharid-Protein Konjugat), Hepatitis B Virus (inaktives Serum-abgeleitetes, virales Antigen/rekombinantes Antigen), Influenza, Rotavirus, RSV (Repiratory Syncytial Virus), menschliches Astrovirus, Rotavirus, menschliches Influenza A und B Virus, Hepatitis A Virus, ein Impfstoff aus lebendigen abgeschwächten Parainfluenza Virus Typ 3, ein Enterovirusimpfstoff, Retrovirusimpfstoff und Pikornavirusimpfstoff.
- Das Mikrokügelchen von Anspruch 1, weiterhin enthaltend ein Kontrastmittel oder Kontrastmedium ausgewählt aus der Gruppe bestehend aus Fluoreszenz-Markerderivaten, chemischen Farbstoffen und magnetischen Resonanzkontrastmitteln.
- Das Mikrokügelchen nach Anspruch 1, wobei das Polymer etwa 0,5 bis etwa 20 Gew.% bezogen auf das Molekulargewicht an Quervernetzern enthält.
- Eine injizierbare Zusammensetzung enthaltend das Mikrokügelchen nach Anspruch 1 und einen biokompatiblen Träger.
- Die Zusammensetzung nach Anspruch 15, wobei die Zusammensetzung das Mikrokügelchen in einer Menge von ungefähr 10 bis ungefähr 90 Gew.% und den biokompatiblen Träger in einer Menge von ungefähr 10 bis ungefähr 90 Gew.% umfasst.
- Die Zusammensetzung nach Anspruch 16, wobei die Zusammensetzung Mikrokügelchen in einer Menge von ungefähr 10 bis 50 Gew.% und den biokompatiblen Träger in einer Menge von ungefähr 50 bis 90 Gew.% umfasst.
- Die Zusammensetzung nach Anspruch 15, wobei die Zusammensetzung eine Suspension der Mikrokügelchen in dem biokompatiblen Träger ist.
- Die Zusammensetzung von Anspruch 18, wobei das biokompatible Polymer in einer Emulsion vorliegt.
- Die Zusammensetzung nach Anspruch 18, wobei das biokompatible Polymer in einer organischen oder nichtwässrigen Lösung vorliegt.
- Die Zusammensetzung nach Anspruch 18, wobei das biokompatible Polymer in einer auf Wasser basierenden Lösung, einer hydroorganischen Lösung oder einer Mischung davon vorliegt.
- Die Zusammensetzung nach Anspruch 18, wobei der biokompatible Träger in einer Menge von ungefähr 0,01 M bis ungefähr 5 M Salze enthält, die zusammengesetzt sind aus Kationen, die ausgewählt sind aus der Gruppe bestehend aus Natrium, Kalium, Kalzium, Magnesium, Eisen, Zink und Ammonium.
- Die Zusammensetzung nach Anspruch 22, wobei das Salz in der Form eines Kontrastmittels vorliegt.
- Die Zusammensetzung nach Anspruch 18, wobei das Kontrastmittel monomerische (Akrylamido-3-Propionamido)-3-triiodo-2,4,6-Benzoesäure ist.
- Die Zusammensetzung nach Anspruch 15, wobei die Zusammensetzung durch eine Nadel vom Kaliber (gauge) 18 oder kleiner injizierbar ist.
- Ein Verfahren zur aktiven Embolisierung in einem Säugetier umfassend die Verabreichung eines Mikrokügelchens umfassend einen biokompatiblen, quervernetzten und substanziell hydrophilen Polymer, und eines oder mehrerer Medikamente, Impfstoffe oder eine Mischung davon an ein Säugetier, das einer Behandlung bedarf.
- Das Verfahren nach Anspruch 26, wobei das Mikrokügelchen ein oder mehrere Elastomere umfasst.
- Das Verfahren nach Anspruch 27, wobei die Elastomere ausgewählt sind aus der Gruppe bestehend aus Akrylpolymeren, Akrylamidpolymeren, Vinylalkoholpolymeren, Akrylatpolymeren, Polysachariden, Silikonen und Mischungen davon.
- Das Verfahren nach Anspruch 27, wobei der Durchmesser des Mikrokügelchens ungefähr 10 μm bis ungefähr 2000 μm beträgt.
- Das Verfahren nach Anspruch 29, wobei der Durchmesser des Mikrokügelchens ungefähr 50 μm bis ungefähr 300 μm beträgt.
- Das Verfahren nach Anspruch 26, wobei das Mikrokügelchen quellbar ist.
- Das Verfahren nach Anspruch 31, wobei das Mikrokügelchen ionische Polysacharide und ionische synthetische Polymere umfasst.
- Das Verfahren nach Anspruch 32, wobei die ionischen Polysacharide ausgewählt sind aus der Gruppe bestehend aus Carboxymethyldextran, Dextransulfat und Alginsäure.
- Das Verfahren nach Anspruch 32, wobei die ionischen synthetischen Polymere ausgewählt sind aus der Gruppe bestehend aus Natriumakrylatpolymer, Akrylamidpolymer, Akrylamidderivatpolymer oder -copolymer, Natriumakrylat und Vinylalkohol-Copolymer, Vinylazetat-Akrylsäureester-Copolymer, Vinylazetat-Methylmaleat-Copolymer quervernetztes Copolymer aus Isobutylen und Maleinsäureanhydrid, Stärke-Akrylnitril-Propfcopolymer, quervernetztes Natriumpolyakrylat und quervernetztes Polyethylenoxid.
- Das Verfahren nach Anspruch 31, wobei der Durchmesser des Mikrokügelchens vor der Quellung ungefähr 10 μm bis ungefähr 400 μm beträgt.
- Das Verfahren nach Anspruch 35, wobei der Durchmesser des Mikrokügelchens vor der Quellung ungefähr 10 μm bis ungefähr 200 μm beträgt.
- Das Verfahren nach Anspruch 31, wobei der Durchmesser des Mikrokügelchens nach der Quellung ungefähr 10 μm bis ungefähr 2000 μm beträgt.
- Das Verfahren nach Anspruch 26, wobei das therapeutisch aktive Medikament ausgewählt ist aus der Gruppe bestehend aus Antitumormedikamenten, Antianiogenesemedikamenten, Antipilzmedikamenten, Antivirusmedikamenten, Antientzündungsmedikamenten, antibakteriellen Medikamenten und Antihistaminmedikamenten, antiangiogenischen Faktoren, antineoplastischen Mitteln, Hormonen und Steroiden, Vitaminen, Peptiden und Peptidanaloga, Enzymen, antiallergenen Wirkstoffen, Kreislaufmedikamenten, Antituberkulosemitteln, Antivirusmitteln, antianginösen Mitteln, Mitteln gegen tierische Einzeller, Antirheumamitteln, Narkotika, Herzmitteln, Glykosemitteln, Beruhigungsmitteln, Lokalanästhetika, Generalanästhetika.
- Das Verfahren nach Anspruch 26, wobei der Impfstoff ausgewählt ist aus der Gruppe bestehend aus Impfstoffen gegen Pneumokokkus, Poliomyelitis, Milzbrand, Tuberkulose (BCG), Hepatitis A, Cholera, Meningokokkus A, C, Y, W135, Pest, Tollwut (menschlich diploid), Gelbfieber, Japanische Encephalitis, Typhus (Phenol und Hitze abgetötet), Hepatitis B, Diphterie, Tetanus, Keuchhusten, H. Influenzae Typ B, Polyo, Masern, Mumps, Röteln, Windpocken, Streptokokkus Pneumoniae Ty (lebendige Bakterienmutanten), Vi (Vi kapsuläres Polysacharid), DT (toxoid), Td (toxoid), aP (inaktives bakterielles Antigen/accelular (DtaP)), Hib (bakterielles Polysacharid-Protein-Konjugat), Hepatitis B Virus (inaktives, vom Serum abgeleitetes, virales Antigen rekombinantes Antigen), Influenza, Rotavirus, RSV (Repiratory Syncytial Virus), menschliches Astrovirus, Rotavirus, menschliches Influenza A und B Virus, Hepatitis A Virus, Impfstoff aus lebendigen, abgeschwächten Parainfluenza Virus Typ 3, Enterovirusimpfstoff, Retrovirusimpfstoff und Pikornavirusimpfstoff.
- Das Verfahren nach Anspruch 26, wobei das Mikrokügelchen weiterhin ein Kontrastmedium oder ein Diagnosemittel umfasst, ausgewählt aus der Gruppe bestehend aus fluoreszierenden Markerderivaten, chemischen Farbstoffen und magnetischen Resonanzkontrastmitteln.
- Das Verfahren nach Anspruch 26, wobei das Polymer etwa 0,5 bis etwa 20 Gew.% an Quervernetzern aufweist.
- Das Verfahren nach Anspruch 26, wobei die Verabreichung die Injektion an einer Stelle des Säugetiers, das einer Embolisierung bedarf, umfasst.
- Das Mikrokügelchen nach Anspruch 1, wobei das Antitumormedikament Taxol, Doxorubicin, Tamoxifen oder eine Kombination davon ist.
- Das Verfahren zur aktiven Embolisierung in einem Säugetier, umfassend die Verabreichung eines Mikrokügelchens an ein zu behandelndes Säugetier, wobei das Mikrokügelchen ein biokompatibles quervernetztes und im Wesentlichen hydrophiles Polymer und ein oder mehrerer Medikamente, Impfstoffe oder eine Kombination davon umfasst, und wobei besagtes Mikrokügelchen an seine Wirkstätte mit Hilfe von Zielantikörpern gebracht wird.
