Abstract
Description
このようなメラニン色素の異常沈着の防止、改善を目的として、美白剤を皮膚外用剤に配合することが行われている。現在、美白剤として皮膚外用剤に配合されている成分としては、ビタミンC誘導体、コウジ酸、アルブチン(4-ヒドロキシフェニル-β-D-グルコピラノシド)やルシノール(4-n-ブチルレソルシノール)、エラグ酸などがあり、これらはメラニン生成抑制作用を有することが知られている。
しかしながら、その効果や安全性などの点において、未だ十分に満足し得るものは得られておらず、新しい美白剤の開発が望まれていた。
しかしながら、これら文献にはメラニン生成抑制作用や美白効果については全く記載されていない。
すなわち、本発明にかかる美白剤は、下記一般式(1)で示されるヘテロ環化合物又はその薬理学的に許容される塩を有効成分とする。
また、本発明は、前記美白剤において、R2及びR2’はメチルである美白剤を提供する。
また、本発明は、前記美白剤において、pが1又は2である美白剤を提供する。
また、本発明は、前記美白剤において、R1が-NH-基に対してオルト位及び/又はパラ位に置換している美白剤を提供する。
また、本発明は、前記美白剤の有効成分がメラニン生成を抑制する美白剤を提供する。
また、本発明は、前記何れかに記載のヘテロ環化合物又はその薬理学的に許容される塩を配合した皮膚外用剤、ならびに化粧料を提供する。
本発明において、「C1-3アルキル」とは、炭素数1~3の直鎖状、分枝状又は環状の飽和炭化水素基である。例えば、メチル基、エチル基、n-プロピル基、イソプロピル基、シクロプロピル基等が挙げられる。好ましいアルキルとしては、メチル、エチルが挙げられ、特に好ましくはメチルである。
YはS又はOである。従って、一般式(1)中、Yを含む5員ヘテロ環は、チアゾリン(Y=S)、又はオキサゾリン(Y=O)である。
pは0~3の整数であり、pが2~3の場合、R1は同一又は異なっていてもよい。
また、本発明の美白剤の有効成分として好適な化合物の例として、R2及びR2’はメチルであるものが挙げられる。

合成する場合には、官能基が分子内に存在し、この官能基が反応の妨害となる、あるいはその恐れのある場合には、適切な保護基を用いて効率的に反応を進行させることが好ましい。保護基の利用は、例えば、Theodora W. Greene、Peter G. M. WutsによるProtective Groups in Organic Synthesis等に従って実施できる。
また、特開平6-25197号に記載される方法のように、2-イミノ(又は2-アミノ)チアゾリン化合物(又はオキサゾリン化合物)と、(R1)p-C6H4-X(Xはハロゲン;R1、pはそれぞれ前記定義の通り)で示される化合物との反応を採用することもできる。本反応は、通常、適当な溶媒中で、トリエチルアミンなどの塩基存在下、必要に応じて加熱して行うことができる。
一般式(1)の化合物を美白剤として皮膚外用剤に配合する場合、外用剤全量中における配合量は、通常0.0002質量%以上、好ましくは0.002質量%以上である。配合量が少なすぎると効果が十分に発揮されない。上限は特に規定されないが、通常30質量%以下、好ましくは20質量%以下、さらに好ましくは5質量%以下である。過剰に配合しても増量に見合った顕著な効果が得られないことがあり、また、製剤設計や使用性などにおいて影響を及ぼすことがある。
また、本発明の皮膚外用剤には、一般式(1)の化合物に加えて、本発明の効果を損なわない範囲内で、通常化粧品や医薬品等の皮膚外用剤に用いられる他の成分、例えば油分、湿潤剤、紫外線防止剤、酸化防止剤、金属イオン封鎖剤、界面活性剤、防腐剤、保湿剤、香料、水、アルコール、増粘剤、粉末、色材、生薬、その他各種薬効成分等を必要に応じて適宜配合することができる。
さらに、ビタミンC、アスコルビン酸リン酸マグネシウム、アスコルビン酸グルコシド、アルブチン、コウジ酸、ルシノール、エラグ酸、トラネキサム酸、リノール酸等の他の美白剤も適宜配合することができる。
以下、具体例を挙げてさらに本発明を説明するが、本発明はこれらに限定されるものではない。
(1)細胞播種・試験物質の添加
マウスB16メラノーマ細胞を6ウェルプレートに100,000細胞/ウェルで播種した。翌日、試験物質溶液(溶媒:DMSO)を添加した。
(2)細胞増殖試験
試験物質溶液添加から3日後に培地を吸引除去した後、10%のアラマブルー溶液を含むEMEM培地を1ml添加して、37℃で反応させた。30分後に100μlを96ウェルプレートに移し、励起波長544nm、測定波長590nmで蛍光を測定した。その値を細胞数の相対値として、試験物質無添加群(溶媒のみ添加)に対する試験物質添加群の細胞数比率(%細胞数)を算出した。%細胞数が高いほど細胞毒性が低いことを意味する。%細胞数が80%以上の場合を毒性なし、80%未満は毒性ありと判断した。
細胞増殖試験後の細胞をPBSにて3回洗浄した後、1M NaOH 200μLを添加して細胞を溶解し、475nmの吸光度を測定した。この値をメラニン量の相対値として、試験物質無添加群(溶媒のみ添加)に対する試験物質添加群のメラニン量比率(%)を算出した。メラニン量比率が低いほどメラニン生成抑制効果が高いことを意味する。毒性なしと判断した試験物質終濃度において、メラニン量比率(%)が80%以下となった最小の試験物質終濃度をメラニン生成抑制最小濃度(ppm)とし、下記の基準でメラニン生成抑制効果を評価した。
◎:メラニン生成抑制最小濃度が1ppm以下である。
○:メラニン生成抑制最小濃度が1ppmより大きく10ppm以下である。
×:10ppm以下でメラニン生成抑制効果がない(10ppm以下ではメラニン量比率が80%以下にならない)。
表1の化合物には何れもメラニン生成抑制効果が認められ、ほとんどの化合物が1ppm以下という極めて低濃度で効果を示した。
2-アミノ-2-メチル-1-プロパノール(61.6g,0.69moL)をクロロホルム500gに溶解し、イソチオシアン酸フェニル(81.6g,0.6moL)のクロロホルム300g溶液を攪拌しながら1時間かけてゆっくりと滴下した。滴下後、室温で12時間攪拌した後、析出した結晶を加熱還流にて溶解し再結晶した。得られた結晶をろ取し、20mLのジエチルエーテルで3回洗浄し、室温で減圧乾燥してN-(1-ヒドロキシ-2-メチルプロパン-2-イル)-N’-フェニルチオ尿素80.1g(収率60%)を得た。
N-(1-ヒドロキシ-2-メチルプロパン-2-イル)-N’-フェニルチオ尿素(80.1g,0.36mol)を2400mLの35%HClに溶解し、90℃で1.5時間加熱攪拌した。冷却後、NaOHで中和してジエチルエーテルにて抽出し、飽和食塩水にて洗浄した後に無水硫酸ナトリウムを加えた。その有機相を減圧留去し、ヘキサンで3回洗浄して得られた結晶を室温で減圧乾燥した。その後、メタノールから再結晶を2回行うことにより表題化合物22.4g(収率18%)を得た。

イオン交換水にプロピレングリコールと苛性カリを加え溶解し、加熱して70℃に保つ(水相)。他の成分を混合し加熱融解して70℃に保つ(油相)。水相に油相を徐々に加え、全部加え終わってからしばらくその温度に保ち反応を起こさせる。その後、ホモミキサーで均一に乳化し、よくかきまぜながら30℃まで冷却する。

- 請求項1又は2記載の美白剤において、R2及びR2’はメチルであることを特徴とする美白剤。
- 請求項1~3の何れかに記載の美白剤において、pが1又は2であることを特徴とする美白剤。
- 請求項4記載の美白剤において、R1が-NH-基に対してオルト位及び/又はパラ位に置換していることを特徴とする美白剤。
- 請求項1~5の何れかに記載の美白剤において、有効成分がメラニン生成を抑制することを特徴とする美白剤。
- 請求項1~6の何れかに記載のヘテロ環化合物又はその薬理学的に許容される塩を配合したことを特徴とする皮膚外用剤。
- 請求項1~6の何れかに記載のヘテロ環化合物又はその薬理学的に許容される塩を配合したことを特徴とする化粧料。
